Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung einer Verbindung der Formel II

worin Rx den Rest einer Carbonsäure RjCOOH darstellt und R' und B die angegebene Bedeutung haben, und Ac eine Acylgruppe ist, mit einem Salz eines Metalls der Gruppe IB oder IIb des Periodischen Systems umsetzt, wobei im Falle einer Verbindung der Formel Ia als Ausgangsverbindung die Umsetzung in Gegenwart eines hydroxylgruppenhaltigen organischen Lösungsmittels oder Wasser und im Falle einer Verbindung der Formel Ib die Umsetzung in Gegenwart von Wasser durchgeführt wird.
2. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass das Salz eines Metalls der Gruppe Ib oder IIb des Periodischen Systems AgC104, AgI04, AgPFe, CF3|COOAg, CF3S03Ag, AgBF4, CH3S03Ag, AgNOa, CH3COOAg, Hg(C104)2, Hg(I04)2, Hg(C103)2, Hg(PFc)2, (CF3COO)2Hg, (CF3S03)2Hg, Hg(BF4)2, (CH3S03)2Hg, Hg(N03)2, (CH3COO)2Hg, Cu(C104)o, Zn(C104)2, Zn(BF4)2, Cd(NOs)2 oder ZnSiFc ist.
3. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass die Reaktion in einem Lösungsmittel, in welchem die hergestellte Metallthio-Verbindung weniger löslich ist, vorzugsweise Methanol, Äthanol, Propanol oder Butanol,
oder mehr löslich ist, vorzugsweise in Methylenchlorid oder Chloroform, durchgeführt wird.
4. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass die Acylgruppe eine von organischen Säuren mit 1-12 C-Atomen abgeleitete Acylgruppe ist, vorzugsweise von Carboxylsäuren gewählt aus Alkansäure, Alkensäure, Alkyn-säure, Cycloalkan- oder Aroylsäure, die gegebenenfalls durch eine inerte Gruppe gewählt aus Halogen, Alkyl, Phenyl, Halo-genphenyl, Alkoxy, Hydroxy, Aryloxy, Acyloxy und Oxo sub-

Das 4-ständige Acyl des Acylthio kann abgeleitet sein von organischen Säuren mit 1-12 C-Atomen, beispielsweise einer Carbonsäure, wie Alkansäure, Alkensäure, Alkynsäure, Cyclo-alkansäure, Aralkansäure, Aralkensäure, aromatische Carb-oxylsäure, oder andere gegebenenfalls durch beispielsweise eine inerte Gruppe gewählt aus Halogen, Alkyl, Phenyl, Halo-genphenyl, Alkoxy, Aryloxy, Acyloxy und Oxo substituierte Carbonsäuren, ferner Sulfonsäure, Sulfinsäure, Phosphon-säure oder andere organische Säuren oder kann das Acyl von anorganischen Säuren abgeleitet werden, z.B. von Kohlensäure, Schwefelsäure, Phosphorsäure, Halogensäure, Halogenwasserstoffsäure oder anderen anorganischen Säuren.
Der Amino-Substituent im 3-ständigen R kann ein Amino-substituent sein, der bekannte Seitenketten in natürlichen oder synthetischen Penicillinen oder Cephalosporinen, oder ihre Äquivalente, bildet. Diese Gruppe kann in weitem Ausmass variieren, da sie im allgemeinen eine weniger direkte Beziehung zu der erfindungsgemässen Hydrolyse der 4-Acylthio-gruppe hat. Diese Gruppe kann ein Acyl, Hydrocarbyl, Hy-drocarbyliden, Silyl, Sulfenyl oder ähnliche übliche bei Cephalosporinen oder Penicillinen verwendbaren Substituenten mit bis zu 20 C-Atomen sein.
Repräsentative Acylgruppen umfassen folgende Gruppen:
1) Alxanoyl mit 1-5 C-Atomen;
2) Halogenalkanoyl mit 2-5 C-Atomen;
3) Azidoacetyl;
4) Cyanoacetyl;
5) Acylgruppen der Formel

worin Q und Q' jedes Wasserstoff oder Methyl und Ar Phenyl, Dihydrophenyl oder eine monocyclische heterocyclische aromatische Gruppe mit 1-4 Heteroatomen, gewählt aus Stickstoff, Sauerstoff und/oder Schwefel ist und kann gegebenenfalls durch eine inerte Gruppe substituiert sein, z.B. ein Alkyl oder Alkoxy mit 1-3 C-Atomen, Chlor, Brom, Jod,
Fluor, Trifluormethyl, Hydroxy, Cyano, Aminomethyl, Amino oder Nitro;
6) Acylgruppen der Formel
Ar-G-CQQ'-CO-
worin G Sauerstoff oder Schwefel ist und Ar, Q und Q' die obige Bedeutung haben;

worin L eine leicht abspaltbare, gegebenenfalls substituierte Kohlenwasserstoffgruppe mit 1-8 C-Atomen, wie 2,2,2-Tri-chloräthyl, Isobornyl, tertiäres Butyl, 1-Methycyclohexyl, 2-Alkoxy-tertiäres-Butyl, Benzyl, p-Nitrobenzyl oder p-Meth-25 oxybenzyl ist.
Der Substituent kann ferner eine Diacylgruppe sein, die sich ableitet von einer polybasischen Carbonsäure mit 4-12 C-Atomen, von Alkyliden mit 1-6 C-Atomen, oder von Aryl-methyliden mit 7-9 C-Atomen. In obiger Aufzählung von 30 Acylgruppen umfassen Beispiele von Ar Furyl, Thienyl, Pyr-ryl, Oxazolyl, Isoxazolyl, Oxadiazolyl, Oxatriazolyl, Thiazolyl, Isothiazolyl, Thiadiazolyl, Thiatriazolyl, Pyrazolyl, Imidazolyl, Triazolyl, Tetrazolyl, Phenyl, Pyridyl, Pyrimidyl, Pyrazinyl, Pyridazinyl, Triazinyl und Dihydrophenyl, wobei jede dieser 35 Gruppen gegebenenfalls substituiert ist durch Halogen, Alkyl mit 1-3 C-Atomen, Hydroxy, Aminomethyl oder Alkoxy mit 1-3 C-Atomen.
Andere Aminosubstituenten umfassen ein Kohlenwasserstoff mit bis zu 20 C-Atomen, z.B. Methyl, Propyl, tertiäres 40 Butyl, Benzyliden, o-Oxybenzylideii, Trityl, ferner Silyl mit 1-10 C-Atomen, wie Trialkylsilyl, Sulfenyl, wie o-Nitrophe-nylsulfenyl, welche Gruppen womöglich ungesättigt oder durch eine inerte Gruppe, z.B. Halogen, Alkyl, Acyloxy, Alkoxy oder Acylamino mit 1-8 C-Atomen, Oxo, Nitro und 45 ähnliche Substituenten substituiert sein können. Die in Amino oder Amido umwandelbare Gruppe, z.B. Azido, Isocyanato, Isocyano, wird gleichfalls von obiger Definition des substituierten Amino umfasst. Die Silyl- und Sulfenylgruppen sind übliche Schutzgruppen für Amino. Es können auch zwei so Aminosubstituenten des Amino eine Ringstruktur bilden.
Von der Definition der genannten 4-Acylthio-3-amino--2-oxo-a-(substituierten oder unsubstituierten)-azetidin-l-essig-säure der Formel (I) wird eine 3-substituierte-4-Thia-2,6--d:aza-7-oxo-bicyclo[3,2,0]-hept-2-en-6-ylessigsäure umfasst, 55 welche eine Verbindung mit kombinierten 3-Amino- und 4-Thio-Gruppen durch ein Acyl ist.
Die Carboxy-Schutzgruppe B kann enthalten 1-20 C-Ato-me und kann eine Sauerstoff-Funktion sein, wie beispielsweise Alkoxy mit 1-8 C-Atomen, z.B. Methoxy, Äthoxy oder 6o tertiäres Butoxy, ein Aralkoxy mit 7-20 C-Atomen, z.B. Ben-zyloxy, Methoxybenzyloxy, Nitrobenzyloxy, Diphenyl-methoxy oder Trityloxy, mono- oder bicyclisches Aryloxy, z.B. Phenoxy oder Naphthyloxy, oder Organometalloxy, z.B. Trimethylstannioxy oder Trimethylsilyloxy, ein organisches 65 oder anorganisches Acyloxy mit bis zu 8 C-Atomen oder Me-talloxy von Gruppen I, II oder III des Periodischen Systems, z.B. Natriumoxy, Kaliumoxy oder Magnesiumoxy; oder B kann eine Schwefel-Funktion sein wie jene die ein Thiolester,

Thiocarboxy oder ähnliche Gruppen bildet; oder eine Stickstoff-Funktion, wie jene die Amide, Hydrazide, Azide oder ähnliche Gruppen bildet oder B kann schliesslich auch eine andere Carboxy-Schutzgruppe sein.
Diese Gruppen können womöglich durch ein Heteroatom in ihrer Struktur unterbrochen sein oder sie können ungesättigt oder gegebenenfalls durch die Stickstoff-, Sauerstoff-, Schwefel-, Kohlenstoff- oder Phosphor-Funktionen oder Halogen substituiert sein.
Unter den Carboxy-Schutzgruppen können genannt werden jene, die Halogenalkylester mit 1-5 C-Atomen, Acyl-alkylester mit 2-10 C-Atomen, Alkoxy alkyl oder Aminoalkyl-ester mit 2-8 C-Atomeni, den Phenylester, Aralkylester mit 7-20 C-Atomen, Ester mit einem Oxim mit 2-10 C-Atomen, N-Alkoxyamid mit 1-5 C-Atomen, Imid mit Saccharin, Imid mit Phthalimid, N,N'-Diisobutylhydrazid, Metallsalze, Alkyl-aminsalze mit 1-6 C-Atomen oder gleichwertige Gruppen zu jenen Gruppen, bilden, wobei in obigen Gruppen die dort angeführten Zahlen der Kohlenstoffatome für die Gruppe B gelten.
Antibakteriell bevorzugte Carboxy-Schutzgruppen B umfassen Acyloxymethylester, Phenacylester, den Benzaldoxim-ester, N,N-Dimethylaminoäthylester, Methansulfonyläthyl-ester, Alkalimetallsalze, Erdalkalimetallsalze, acylierte Erdalkalimetallsalze und andere gleichwertige Gruppen im Bezug auf diese Gruppen. Bevorzugte Carboxy-Schutzgruppen B umfassen Benzhydryloxy, p-Nitrobenzyloxy, p-Methoxy-benzyloxy, 2,2,2-Trichloräthoxy, und Alkalimetalloxy.
Das vorliegende erfindungsgemässe Verfahren zur Herstellung oben definierter 4-Metallthio-Verbindungen der Formel II ist dadurch gekennzeichnet, dass man eine 4-Acyl-thio-Verbindung der Formel I mit einem Salz eines Metalls der Gruppe Ib oder IIb des Periodischen Systems behandelt.
Das genannte Salz eines Metalls der Gruppe Ib oder IIb des Periodischen Systems kann ein Salz eines Metalls der genannten Gruppe Ib wie Kupfer, Silber, oder IIb wie Zink, Cadmium, Quecksilber, oder deren Gemische sein, die befähigt sind, die Acylthiogruppe abzuspalten und die gewünschte Metall-substituierte Thiogruppe zu liefern. Das Salz kann ein Salz des Metalls mit einer organischen Säure sein, wie einer aliphatischen Säure, z.B. Ameisensäure, Essigsäure, Propionsäure, Bernsteinsäure, Oleinsäure, Laurinsäure, Stearinsäure, oder einer aromatischen Carbonsäure, z.B. Benzoesäure, Salicylsäure, Naphthensäure, einer Sulfonsäure, z.B. Phenolsulfonsäure, Toluol-p-sulfonsäure, Trifluormethan-sulfonsäure, Methansulfonsäure oder anderer organischer Säuren, z.B. Zitronensäure, Milchsäure, Oxalsäure, Trifluor-essigsäure; oder einer anorganischen Säure, wie Mineralsäure, z.B. Chlorsäure, Perchlorsäure, Bromwasserstoffsäure, Jodwasserstoff säure, Perjodsäure, schweflige Säure, Perschwefelsäure, Selensäure, Salpetersäure, salpetrige Säure, Arsensäure, Phosphorsäure, Pyrophosphorsäure, Carbonsäure usw. oder andere anorganische Säuren, z.B. Borsäure, Sulf-aminsäure, Carbaminsäure oder Doppelsalze oder Komplexsalze, z.B. Nitroprussidsalze, Silicofluorid, Fluoroborat, ein Komplex von Mineralsäuresalzen mit Alkalimetallsalzen oder Ammoniumsalzen, oder ihre Gemische.
Unter diesen Salzen werden am meisten bevorzugt im Reaktionsmedium sehr lösliche Salze, stark dissoziierende Salze von starken Säuren, und Salze von Silber oder Quecksilber mit einer hohen Fähigkeit sich gebildete Thiogruppe zu binden. Spezifische Beispiele dieser bevorzugten Salze umfassen AgClOi, AgI04, AgPFc, CF3COOAg, CF3S03Ag, AgBF4, CH,S03Ag, AgN03, CH3COOAg, Hg(C104)2, Hg(I04)2, Hg(C10.3)2, Hg(PF0)2, CF3COO)2Hg, (CF3S03)2Hg, Hg(BF4)2, (CH3S03)2Hg, Hg(N03,)2, (CH,COO)2Hg, Cu(C104)2, Zn(C104)2, Zn(BF4)2, Cd(NOg)2 und ZnSiF0.
Die genannte Reaktion erfolgt auf befriedigende Weise mit Vorteil in einem Lösungsmittel, bei Zimmertemperatur oder bei gekühlter oder erhöhter Temperatur. Das genannte Lösungsmittel kann mit Vorteil sein ein polares organisches Lösungsmittel, z.B. Alkohol, Keton, Äther, Amid, Sulfoxid, Nitrii, Ester oder Wasser oder deren Gemische, welche andere organische Lösungsmittel enthalten können. Die Reaktion kann unter Verwendung eines Katalysators oder Radikalinitiators und falls erforderlich unter Bestrahlung zwecks leichter Umsetzung durchgeführt werden. Es wird ein Lösungsmittelsystem, in welchem die erzeugten Metallthio-Ver-bindungen weniger löslich sind bevorzugt, und zwar zwecks Vereinfachung der Abtrennung des Produktes. Das Lösungsmittelsystem, in welchem die Metallthio-Verbindungen löslich sind, z.B. Methylenchlorid, Chloroform, wird für die chromatographische Trennung oder darauffolgende Behandlung der nächsten Stufe bevorzugt.
Als Lösungsmittel für das Reaktionsmedium kann verwendet werden ein Kohlenwasserstoff, wie Pentan, Hexan, Benzol, Toluol, ein Halogenkohlenwasserstoff, z.B. Methylenchlorid, Chloroform, Kohlenstofftetrachlorid, Dichlorbenzol, ein Ester, z.B. Äthylacetat, Butylacetat, Methylbenzoat, Bu-tylacetat, Methylbenzoat, ein Keton, wie Aceton, Cyclohexa-non, Benzophenon, ein Äther wie Diäthyläther, Äthylengly-kol-dimethyläther, Tetrahydrofuran, Tetrahydropyran, Di-oxan, Morpholin, Anisol, ein Alkohol, z.B. Methanol, Äthanol, Äthylenglykol, Benzylalkohol, eine Carbonsäure, z.B. Essigsäure, Propionsäure, eine Base, z.B. Butylamin, Tri-äthylamin, Pyridin, Picolin, ein Amid, z.B. Dimethylform-amid, Dimethylacetamid, Hexamethylphosphortriamid, ein Nitrii, wie Acetonitril, Benzonitril, ein Nitrokohlenwasser-stoff, Sulfoxid, wie Dimethylsulfoxid, Wasser, flüssiges Ammoniak-Lösungsmittel oder ihre Gemische.
In einem Reaktionsmedium, in welchem das Produkt weniger löslich ist, trennt sich das Reaktionsprodukt im Medium ab, und kann durch Filtrieren gesammelt, dann gewaschen und getrocknet werden und liefert die gewünschte Verbindung in hoher Ausbeute. In einem Reaktionsmedium, in welchem das Reaktionsprodukt löslich ist, kann das Reaktionsgemisch, zwecks Entfernung der Lösungsmittel, abgedampft werden u. mit einem Lösungsmittel behandelt werden, in welchem das Produkt weniger löslich ist oder das Produkt kann mit Hilfe üblicher Methoden, wie Extraktion, Waschen, Trocknen oder Abdampfen des Lösungsmittels, isoliert werden. Für die Reinigung wird gewöhnlich Umkristallisieren, Umfällen und weitere bekannte Methoden, verwendet.
Die so erfindungsgemäss erzeugte 4-Metallthio-Verbindung der Formel II kann mit einem ein Metall entfernenden Reagens behandelt werden und liefert die oben angeführte 4-Mercapto-Verbindung der Formel III.
Das genannte Metall abspaltende Reagens kann eine Thio!-verbindung sein, z.B. Schwefelwasserstoff oder eine Verbindung, die befähigt ist, eine unlösliche Verbindung mit genanntem Metall zu bilden, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder ihre Salze, oder Verbindungen, die befähigt sind, das Metall durch Komplexbildung zu entfernen. Diese Reaktion kann in einem oben angeführten Lösungsmittel, bei niedriger Temperatur, gekühlt oder bei erhöhter Temperatur bewerkstelligt werden. Das Produkt kann hernach mittels üblicher Methoden aus dem Reaktionsgemisch durch Entfernen von unreagiertem Material, von Nebenprodukten, Lösungsmitteln oder anderen Verunreinigungen, isoliert und durch übliche Methoden, z.B. Umkristallisieren, Chromatographie, Umfällung, gereinigt werden.
Die erfindungsgemäss erzeugte 4-Metallthio-Verbindung der Formel II ist eine neue Verbindung, die als wichtige Zwischensubstanz für die Synthese z.B. von Cephalosporin-Ver-

bindungen durch Cyclisieren, und als medizinischer Wirkstoff, nützlich ist.
Das oben beschriebene Verfahren wird im nachfolgenden durch einige bevorzugte Ausführungsbeispiele näher dargelegt.
Beispiel 1
Zu einer Lösung von 734 mg von Methyl-4-cyclopropyl--methoxycarbonylthio-3-phthalimido-2-oxo-a-isopropyliden-azetidin-l-acetat in 3 ml Methanol wurde eine Lösung von 464 mg Silberperchlorat in 3 ml Methanol zugesetzt und das Gemisch wurde bei Zimmertemperatur in einer Argonatmosphäre gerührt. Nach Ablauf von 8 h wurde die sich abgeschiedene Substanz filtriert und mit Äther gewaschen. Die Kristalle wurden in Methylenchlorid gelöst, durch eine Silica-gelschicht filtriert, abgedampft und aus einem Gemisch von Äther und Pentan umkristallisiert. Man erhielt 546 mg an Kristallen, die ein Silbersalz am Mercapto des Methyl-4-mer-capto-3-phthalimido-2-oxo-a-isopropylidenazetidin-1 -acetats mit F = 196-198°C sind. Ausbeute 73%.
Beispiel 2
Zu einer Lösung von 464 mg von Methyl-4-benzoylthio--3-phthaIimido-2-oxo-a-isopropenylazétidin-l-acetat in 8 ml Dioxan wurde eine Lösung von 456 mg Silberperchlorat in 2 ml Methanol zugefügt und das Gemisch wurde bei Zimmertemperatur unter Stickstoff gerührt. Nach Ablauf von 10 h wurde das Reaktionsgemisch nach Zugabe von Methanol eingeengt und das sich abgeschiedene Silbersalz am Mercapto von Methyl-4-mercapto-3-phthalimido-2-oxo-a-isopropenyl-azetidin-l-acetat wurde filtriert. Man erhielt 401 mg an Substanz mit F. von 165-169°C. Die Ausbeute betrug 86%.
Beispiel 3
Zu einer Lösung von 429 mg von Methyl-4-phenylacetyl-thio-3-phthalimido-2-oxo-a-(l-methoxyäthyliden)-azetidin-l--acetat in 3ml Aceton wurde eine Lösung von 334 mg Silberperchlorat in 2 ml Methanol zugesetzt und das Gemisch wurde bei Zimmertemperatur unter Stickstoff gerührt. Nach Ablauf von 20 h wurde das Reaktionsgemisch mit Methanol verdünnt und eingeengt. Der Rückstand wurde in Methylenchlorid gelöst, mit einem Gemisch von Äther und Pentan verdünnt, wobei sich Kristalle von Silbersalz am Mercapto von Methyl-4--mercapto-3-phthalimido-2-oxo-<x-(l-methoxyäthyliden)-aze-tidin-l-acetat in einer Menge von 189 mg abschieden und dann durch Filtrieren gesammelt. Die Ausbeute betrug 76%.
Beispiel 4
Im oben beschriebenen Verfahren von Beispiel 3 wurde das genannte Silberperchlorat durch 265 mg Silbernitrat oder 260 mg Silberacetat ersetzt und man erhielt ein Silbersalz am Mercapto von Methyl-4-mercapto-3-phthalimido-2-oxo-a-(l--methoxyäthyliden)-azetidin-l-acetat.
Beispiel 5
Zu einer Lösung von 4-Acetylthio-3-acylamino-2-oxo-a--substituiertes-azetidin-l-acetat in einem Lösungsmittel wurde Pulver oder eine Lösung des Silbersalzes zugefügt und das Gemisch wurde bei Zimmertemperatur unier Stickstoff oder Argon als inertem Gas gerührt. Nach Ablauf der Reaktion und nach Abtrennung der gewünschten Verbindung wurde das Produkt filtriert, gewaschen und getrocknet. Falls die gewünschte Verbindung sich nach Einengen nicht abtrennt, wird weniger Lösungsmittel zugesetzt, um die gewünschte Verbindung abzutrennen.
Das erhaltene Produkt ist ein Silbersalz am Mercapto von 4-Mercapto-3-acylamino-2-oxo-a-substituiertes-azetidin-1 --acetat.
Die Reaktionsbedingungen und die physikalischen Konstanten des Produktes sind in folgenden Tabellen I und II angeführt. In der Tabelle steht MeOH für Methanol, Di für Dioxan, EtOH für Äthanol, DMSO für Dimethylsulfoxid und Ph für Phenyl.
Beispiel 6
Zu einer Lösung von 3-substituiertem-4-Thia-2,6-diaza-7--oxo-a-substituiertem-bicyclo-[3,2,0]hept-2-en-6-ylacetat in einem Lösungsmittel wird ein Silbersalz zugefügt und das Gemisch wird bei Zimmertemperatur unter Stickstoff oder Argon als inertes Gas gerührt. Nach Ablauf der Reaktion und nach Ausscheiden der gewünschten Verbindung wird das Produkt filtriert; falls das Produkt sich auch nach Einengen nicht abscheidet, wird weniger Lösungsmittel zugesetzt, um die gewünschte Verbindung abzutrennen.
Das Produkt ist ein Silbersalz am Mercapto von 4-Mer-capto-3-acylamino-2-oxo-a-substituiertem-azetidin-l-acetat.
Die Reaktionsbedingungen sind in folgender Tabelle III angeführt. Die Abkürzungen und die physikalischen Konstanten sind identisch mit Angaben von Beispiel 5 oder Tabelle II.
Beispiel 7
Ein Silbersalz am 4-Mercapto von 4-Mercapto-3-acyl-amino-2-oxo-a-substituiertem-azetidin-l-acetat wird in einem Lösungsmittel gelöst und zum Gemisch wird Schwefelwasserstoffgas bei Zimmertemperatur, bis zur vollständigen Ausfällung, eingeleitet. Dann wird durch das Reaktionsgemisch Stickstoff perlen gelassen, um den überschüssigen Schwefelwasserstoff zu verdrängen und das unlösliche Silbersulfid wird filtriert. Durch Konzentrieren des Filtrats ergibt sich ein 4-Mercapto-3-acylamino-2-oxo-a-substituiertes-azetidin-l--acetat.
Die Reaktionsbedingungen und die physikalischen Konstanten des Produktes sind in Tabelle IV und V angeführt. Die Abkürzungen sind dieselben wie im Beispiel 5 angeführt.
Beispiel 8
Zu einer Lösung von 57 mg eines Silbersalzes am 4-Mercapto von 2,2,2-Trichloräthyl-4-mercapto-3-phenoxyacet-amido-2-oxo-a-(l,l-dimethoxyäthyI)-azetidin-l-acetat in 5 ml Chloroform werden 0,5 ml 10%ige Chlorwasserstoffsäure zugegeben und das Gemisch wird bei Zimmertemperatur während 10 min gerührt. Nach Entfernen von unlöslichem Material durch Filtrieren wird das Reaktionsgemisch eingeengt und man erhält 40 mg von 2,2,2-Trichloräthyl-4-mercapto-3--phenoxyacetamido-2-oxo-a-( 1,1 -dimethoxyäthyl)-azetidin-1--acetat. Die Ausbeute betrug 94,2%.
Beispiel 9
Gemäss ähnlichen Methoden wie in Beispielen 5-8 beschrieben, werden die folgenden Verbindungen aus den entsprechenden 4-Acetylthioazetidinessigsäure-Derivaten oder Thiadiazabicycloheptanessigsäure-Derivaten präpariert:
1) p-Methoxybenzyl-4-mercapto-3-(2,2,2-trichloräthoxy-carbonyI)-amino-2-oxo-a-( 1 -hydroxyäthyliden)-azetidin-1--acetat;
2) 4-Mercapto-3-benzyloxycarbonylamino-2-oxo-a-iso-propylidenazetidin-l-essigsäure;
3) p-Bromphenacyl-4-mercapto-3-(o-nitrophenylsulfenyl>--amino-2-oxo-a-isopropenylazetidin-l-acetat;
4) Acetoxymethyl-4-mercapto-3-(N-tertiäres-butoxycarbo-nyl-a-phenylglycyl)-amino-2-oxo-a-(l-methoxyäthyliden)-aze-tidin-1-acetat;
5) p-Nitrobenzyl-4-mercapto-3- [N-tertiäres-butoxycarbo-nyl-a-(l,4-cyclohexadien-l-yl)-glycyl]-amino-2-oxo-a-(l-hy-droxy äthyliden)-azetidin-1 -acetat;

Claims (4)
- 6213362PATENTANSPRÜCHE 1. Verfahren zur Herstellung einer Verbindung der Formel IIr ^S-Me f0S-N\-R'ICOBIIworin R ein gegebenenfalls substituiertes Amino, B eine Hy-droxygruppe oder eine Carboxyschutzgruppe, Me ist ein Metall der Gruppe Ib oder IIb des Periodischen Systems, R' ist ein ein- bzw. zweiwertiger Rest ausgewählt aus Äthyl, Vinyl oder Äthyliden, der gegebenenfalls durch Alkyl oder Alkoxy mit 1-3 C-Atomen, durch Halogen, Hydroxy, Carboxy, Sul-fon oder Carbonsäureacyloxy mit bis zu 8 C-Atomen, oder durch eine Aminogruppe, die Wasserstoff, Alkyl mit 1-8 C-Atomen oder Alkoxy mit 1-8 C-Atomen trägt, substituiert ist, dadurch gekennzeichnet, dass man eine Verbindung der Formel
- R.RfS-AcNoderO_N\c—R' ICOBIa'C —R'ICOBIb stituiert sind, oder abgeleitet von Sulfonsäure, Sulfinsäure, Phosphonsäure oder von anorganischen Säuren, insbesondere von Kohlensäure, Schwefelsäure, Phosphorsäure, einer Halogensäure oder Hydrohalogensäure.5 5. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass die Acylgruppe Acetyl, Cyclopropylmethoxycarbo-nyl, Benzoyl oder Phenylacetyl ist.

Verwendung der laut Verfahren gemäss Anspruch 1 erhaltenen 4-Metallthio-Verbindung der Formel II zur Herstel-lo lung einer 4-Mercapto-Verbindung der Formel III15200rKSH'C=R'ICOB(Hl)worin R, R' und B die obige Bedeutung haben, dadurch gekennzeichnet, dass man eine 4-Metallthio-Verbindung der Formel II mit einem Metallabspaltungsmittel behandelt.
- 7. Verwendung gemäss Anspruch 6, dadurch gekenn-25 zeichnet, dass das Metallabspaltungsreagens eine Thiolverbin-dung ist, insbesondere Schwefelwasserstoff oder eine zur Bildung einer unlöslichen Verbindung mit genanntem Metall befähigte Verbindung, insbesondere Chlorwasserstoffsäure, Bromwasserstoffsäure oder ihre Salze.30
