Description

Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von fetthaltigen kosmetischen Zubereitungen und auf die erhaltene Zubereitung.
Es ist bekannt, dass die UV-Strahlen des Sonnenlichts nachhaltige Wirkungen auf die menschliche Haut haben. So verursachen Strahlen des Wellenlängenbereichs 280 bis 320 nm Sonnenbrand bzw. Erytheme. Aber auch die oft als erwünscht angesehenen «bräunenden» Strahlen des Wellenlängenbereichs 320 bis 400 nm (UV-A) können Zustand und Aussehen der Haut nachhaltig negativ beeinflussen. Sie können krankhafte Veränderungen der Haut, wie z. B. Lichtdermatosen, hervorrufen. Auch sind sie, vor allem bei langfristiger Einwirkung, verantwortlich für die im Vergleich zu von Kleidung geschützter Haut sehr viel raschere Alterung der dem Licht ausgesetzten Haut, insbesondere der Gesichtshaut. Für den Schutz vor Strahlen im Bereich 280 bis 320 nm (UV-B) sind bereits wirksame Mittel bekannt (z.B. aus der DE-PS 2 051 824).
Dianisoylmethan, das in jüngster Zeit als UV-Filter eingesetzt wurde, absorbiert UV-Strahlung bis etwa 380 nm. Allerdings zeigt Dianisoylmethan im Bereich unterhalb 330 nm nur eine geringe Absorption und ist ausserdem in gebräuchlichen kosmetischen Ölen und Fetten nur schwer löslich. Auch die als «Breitbandfilter» eingesetzten Methoxy- und Hydroxy-substi-tuierten Benzophenone sind sehr schwer fettlöslich und zeigen ausserdem auch nur bis etwa 340 nm ein befriedigendes Absorptionsverhalten.
Die Aufgabe bestand also darin, ein Verfahren zur Herstellung von Mitteln zu finden, die die menschliche Haut vor schädlicher UV-Strahlung schützen. Solche Mittel sollten in einem geeigneten Medium einen Wirkstoff enthalten, der die Strahlen des Wellenlängenbereichs 285 bis 380 nm möglichst vollständig absorbiert, dabei ungiftig und hautverträglich ist, eine gute Löslichkeit in kosmetischen Ölen und vor allem auch eine hohe Lichtstabilität besitzt.
Es wurde nun gefunden, dass alkylsubstituierte Diben-zoylmethane sich hervorragend als Wirkstoffe zur Absorption von UV-A- und UV-B-Strahlung eignen, da sie Strahlung mit einer Wellenlänge von 285 bis 380 nm in geringer Konzentration praktisch vollständig absorbieren, eine sehr gute Lichtstabilität besitzen und vor allem sehr gut löslich sind in den in der Kosmetik gebräuchlichen Lösungsmitteln, vor allem auch in fetthaltigen Zubereitungen.
Gegenstand der Erfindung ist somit ein Verfahren, das im Patentanspruch 1 definiert ist.
Die Verbindungen der Formel I können auch in der zur Formel I tautomeren Form der Formel Ia vorliegen.
Es besteht dabei ein Gleichgewicht zwischen beiden For-10 men, das in vielen Fällen weitgehend auf Seite der Verbindungen der Formel Ia liegt. Werden im vorligenden Fall die Verbindungen der Formel I genannt, so sollen damit gleichzeitig auch die Verbindungen der Formel Ia umfasst sein.
Gegenstand der Erfindung sind auch die nach dem be-15 schriebenen Verfahren hergestellten fetthaltigen kosmetischen Zubereitungen.
Die Verbindungen der Formel I können nur in einem oder aber auch beiden Phenylkernen substituiert sein. Bevorzugt sind jedoch solche, die nur in einem Kern substituiert sind. 20 Ist n oder n' gleich 1, so ist der Substituent R oder R1 bevorzugt in 2- oder 4-Stellung. Bei mehrfacher Substitution (n oder n' gleich 2 oder 3) ist die 2,4-, 2,6- oder 2,4,6-Substi-tution bevorzugt. Substitution in 3-Stellung oder auch z. B. 2,5- oder 3,5-Disubstitution ist jedoch ebenso möglich, und 25 diese Verbindungen sind für die erfindungsgemässen Zwecke ebenfalls sehr gut geeignet.
Wenn beide Phenylkerne substituiert sind, so kann die Substitution in beiden Kernen gleich sein, dies muss jedoch nicht der Fall sein. Es können ohne Schwierigkeiten Diben-30 zoylmethane hergestellt werden, die in beiden Phenylkernen unterschiedliche Substituenten tragen.
Als Alkylreste sind vor allem zu nennen:
Methyl, Äthyl, Propyl, Isopropyl, n-Butyl, iso-Butyl, sec-Butyl, t-Butyl, n-Pentyl, iso-Pentyl, neo-Pentyl, Hexyl, Hep-35 tyl und Octyl. Bei den höheren Alkylsubstituenten sind jeweils mehrere Isomere möglich.
Aus der Gruppe der Verbindungen der Formel I seien im einzelnen genannt:

Die Verbindungen der Formel I sind gut bis ausgezeichnet löslich in kosmetischen Ölen und Fetten. 4-Methyldibenzoylmethan z. B. ist bereits dreimal so gut löslich in Paraffinöl wie das unsubstituierte Dibenzoylmethan, während z. B. 4-Isopro-50 pyldibenzoylmethan damit praktisch unbegrenzt mischbar ist.
Überraschend ist auch die gute Alkalistabilität der Verbindungen der Formel I im Gegensatz zu dem unsubstituierten Dibenzoylmethan. Auch alkalische kosmetische Formulierun-55 gen sind daher sehr gut lagerfähig.
Bemerkenswert ist auch die sehr gute Lichtstabilität der beschriebenen Verbindungen. So wurde im Xenotestversuch nach 24stündiger Bestrahlung nur eine sehr geringe Zunahme der Durchlässigkeit beobachtet.
Die gute Fett- und Öllöslichkeit der beschriebenen Verbindungen ist sehr vorteilhaft, wenn Zubereitungen hergestellt werden sollen, die höhere Konzentrationen der Verbindungen enthalten. Obwohl die Absorptionsfähigkeit der Verbindungen 65 sehr gut ist, können höhere Konzentrationen in Mitteln angebracht sein, die für sehr empfindliche Haut bestimmt sind oder die in Gegenden mit sehr intensiver Sonnenbestrahlung angewendet werden sollen.

Die Verbindungen der Formel I sind zum Teil bekannt, können aber alle nach an sich bekannten Methoden hergestellt werden.
So kann z. B. an Benzylidenacetophenone (durch Kondensation von substituierten Benzaldehyden mit gegebenenfalls ebenfalls substituierten Acetophenonen erhältlich) die in einem oder beiden Phenylkernen gleich oder ungleich durch Al-kylgruppen substituiert sind, Brom addiert werden. Umsetzung des entstehenden Dibromids mit Basen, wie z. B. Natriumme-thylat oder Äthylat, liefert dann die entsprechenden substituierten Dibenzoylmethane.
Ausgehend von substituierten Acetophenonen können unsymmetrisch oder symmetrisch substituierte Dibenzoylmethane hergestellt werden durch Umsetzung mit, gegebenenfalls substituierten, Benzoesäureestern unter Einwirkung von starken Basen, wie z. B. Natriumamid.
Symmetrisch substituierte Dibenzoylmethane können auch durch Umsetzung von Arylcarbonsäurevinylestern mit Aluminiumchlorid unter Friedel-Crafts-Bedingungen hergestellt werden [siehe J. Org. Che. 36, 1447 (1971)].
Man kann aus den Verbindungen der Formel I durch Zusatz von Salben- oder Cremegrundlagen fettende oder nicht-fettende Lichtschutzsalben und Hautcremes oder durch Mischung mit Lösungsmitteln, gegebenenfalls unter Zusatz von Emulgatoren, flüssige Lichtschutz- und Hautpflegepräparate herstellen. Geeignete Lösungsmittel sind beispielsweise: Kohlenwasserstoffe, wie festes oder flüssiges Paraffin, Kristallöl, Ceresin, Ozokerit, Montanwachs und pflanzliche oder tierische Öle, Fette und Wachse, wie Oliven-, Erd- oder Carnauba-wachs, Wollfett, Walrat; ausserdem Fettsäuren und Fettsäureester wie Stearinsäure, Palmitinsäure, Ölsäure, Glycerinmo-no- oder -distearat, Glycerinmonooleat, Isopropylmyristat, Isopropylstearat, Butylstearat, Alkohole, wie Äthyl-, Isopro-pyl-, Cetyl-, Stearyl-, Palmityl-, Hexyldodecylalkohol. Auch mehrwertige Alkohole wie Glykol, Glycerin, Sorbit, die gleichzeitig als Feuchthaltemittel dienen, können hierbei verwendet werden. Geeignete Zusatzstoffe sind z. B. Emulgatoren der Systeme Öl in Wasser und Wasser in Öl, wobei die handelsüblichen ionogenen oder nichtionogenen, kationen- oder anionen-aktiven bzw. ampholytischen Emulgatoren in Frage kommen. Auch Verdickungsmittel, wie Methyl-, Äthyl- oder Carboxy-methylcellulose, Polyacrylsäure, Traganth, Agar-Agar, Gelatine, können zugesetzt werden. Ferner können nach Bedarf oder nach Belieben noch weitere Zusatzstoffe wie Parfums, Konservierungsmittel und/oder physiologisch unbedenkliche Farbstoffe hinzugefügt werden.
Die erfindungsgemäss hergestellten Zubereitungen können zusätzlich einen oder mehrere andere UV-Absorber enthalten, z. B. p-Methylbenzyliden-D,L-campher bzw. dessen Sulfonsäu-re-Na-Salz oder 2-Phenylbenzimidazol-5-sulfonsäure-Natrium-salz, 3,4-Dimethylphenylglyoxylsäure-Natriumsalz, 4-Phenyl-benzophenon, 4-Phenylbenzophenon-2'-carbonsäure-isooctyl-ester, p-Methoxyzimtsäureester, 2-Phenyl-5-methylbenzoxa-zol, p-Dimethylaminobenzoesäureester.
Die Verbindungen der Formel I sind in den erfindungsgemäss hergestellten Zubereitungen zweckmässig in wirksamer Konzentration enthalten. Der Gehalt ist an sich nicht kritisch und weitgehend vom Verwendungszweck abhängig. Im allgemeinen enthalten die Zubereitungen 0,5-15 Gew.% an Verbindungen der Formel I. Bevorzugt sind solche Zubereitungen, die 1,5-8,0 Gew.% an Verbindungen der Formel I enthalten. Enthalten die Zubereitungen zusätzlich noch andere UV-Absorber, so bewegt sich der Gesamtgehalt an UV-absorbieren-den Verbindungen meist zwischen 0,5 und 15 Gew.%, vorzugsweise zwischen 0,5 und 10 Gew.%.

Beispielsweise Herstellungsmethoden A-I für die Wirkstoffe.
A. In eine Lösung von 55,4 g p-Toluylidenacetophenon in 250 ml Chloroform tropft man bei 5 bis 10° C 40 g Brom ein und saugt die gegen Ende der Reaktion ausfallenden Kristalle ab, wäscht mit Methanol und trocknet. Die so erhaltenen 87,5 g l-Benzoyl-2-p-tolyl-l,2-dibrom-äthan, Fp. 168° C gibt man portionsweise in 180 ml methanolische Natriummethylat-Iösung, die 2 Mol Methylat pro Mol Bromverbindung enthält. Nach zweistündigem Kochen versetzt man mit HCl, kocht 5 Minuten lang, gibt Wasser zu und kühlt, wobei 35 g 4-Methyl-dibenzoylmethan auskristallisieren, Fp. 84° C.
B. Zu einer Suspension von 29,7 g Natriumamid in
400 ml Toluol tropft man zunächst 61,5 g Isopropylacetophe-non und dann 51,7 g Benzolsäuremethylester und lässt über Nacht bei Zimmertemperatur rühren. Nach Zugabe von Wasser und Ansäuern trennt man die organische Phase ab, entfernt das Lösungsmittel, destilliert, kristallisiert das übergehende Öl aus Methanol und erhält so 54 g 4-Isopropyldibenzoylmethan, Fp. 46° C.
C. Eine Suspension von 39 g Natriumamid in 540 ml To-
5 luol versetzt man mit 88 g 4-t-Butylacetophenon und anschliessend mit 87 g Benzoesäuremethylester, lässt über Nacht rühren, säuert dann an, trennt die organische Phase ab, wäscht sie, entfernt die Lösungsmittel, destilliert und kristallisiert das übergehende Öl. Man erhält so 80 g 4-t-ButyIdibenzoylme-
lo than, Fp. 97° C.
D. Analog Methode C erhält man, ausgehend von 2-Me-thylacetophenon und Benzoesäuremethylester, 2-Methyldi-benzoylmethan, Kp 0,o4 157° C.
E. Analog Methode C erhält man, ausgehend von 4-Me-
15 thylacetophenon und Benzoesäuremethylester, 4-Methyldi-
benzoylmethan, Fp. 84° C.
F. Analog Methode C erhält man, ausgehend von 4-Iso-propylacetophenon und 4-Isopropylbenzoesäuremethylester, 4,4'-Diisopropyldibenzoylmethan, Fp. 48° C.
20 G. Analog Methode C erhält man ausgehend von 4-n-Octylacetophenon und Benzoesäuremethylester, 4-n-Oc-tyldibenzoylmethan, Kp 0;15 232-240° C.
H. Analog Methode C erhält man, ausgehend von 2,4-Dimethylacetophenon und Benzoesäuremethylester, 2,4-Di-
25 methyldibenzoylmethan, Fp. 62° C.
I. Analog Methode C erhält man, ausgehend von 2,5-Dimethylacetophenon und Benzoesäuremethylester, 2,5-Dimethyldibenzoylmethan, Kp 0i0i 182° C.

    PATENTANSPRÜCHE 1. Verfahren zur Herstellung von fetthaltigen kosmetischen Zubereitungen, dadurch gekennzeichnet, dass man mindestens eine Verbindung der Formel

  2. 2. Nach dem Verfahren nach Anspruch 1 hergestellte Zubereitung.
