Description
Die vorliegende Erfindung betrifft ein Verfahren zur 15 Herstellung neuer Derivate des Piperazins und dessen Homologen der Formel I:

Norbornyl dadurch gekennzeichnet, dass man ein entsprechendes Chlor-acylderivat einer Verbindung der Formel R, -NH2 mit einem entsprechenden Piperazin oder dessen Homologen in Wasser oder einem organischen Lösungsmittel kondensiert.
2. Verfahren nach Anspruch 1 zur Herstellung von N-(Piperazinyl-[l]-methylencarbonyl)-cyc!ohexylamin, dadurch gekennzeichnet, dass man N(2-ChIoracetyl)-cyclohexyl-amin und Piperazin-dichlorhydrat in Wasser bei einer Temperatur von 80-120°C während 7 bis l/i Stunden kondensiert.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass die Kondensation von N(2-Chloracetyl)-cyclohexylamin mit Piperazin im Verhältnis von 1:1 bis 1:3 Mol erfolgt.

Diese Verbindungen besitzen eine antiulzeröse und antisekretorische Wirkung, die vollständig frei von anticholin-ergischen Wirkungen ist.
Man stellt diese Verbindungen her, indem man zunächst ein entsprechendes Chloracylderivat (beispielsweise mit Chloracetylchlorid, Chlorpropionylchlorid oder Chlorbuty-rylchlorid) eines Amins der Formel RrNH2 mit dem entsprechenden Piperazin oder dessen Homologen kondensiert. Beispielsweise kann man die Reaktion wie folgt durchführen:

Man kann so vorgehen, dass man z.B. N(2-ChIoracetyI)--cyclohexylamin mit 2 Mol Piperazin und 2 Mol HCl in wässeriger Lösung umsetzt.
Die Reaktion findet auch dann statt, wenn 1 Mol Piper-azindihydrochlorid eingesetzt wird.
Man kann auch einen Überschuss von Piperazin in einem Verhältnis von 1:3 bis 1:4 Mol in Wasser anwenden, jedoch verursacht das Verfahren in diesem Fall höhere Kosten. Ferner kann man die Kondensation in organischen Lösungsmitteln, insbesondere in Chloroform oder Benzol durchführen.
Die neuen Verbindungen der Formel I wirken auch in Form ihrer Salze mit organischen oder anorganischen Säuren, wie z.B. als Hydrochloride, Sulfate, Phosphate, Malea-te, Succinate und Tartrate.
Die Erfindung wird anhand der folgenden Ausführungsbeispiele erläutert.
Beispiel 1
178,28 g (2 Mol) wasserfreies Piperazin werden in 1000 ml destilliertem Wasser gelöst. Dann werden 166,6 ml HCl 37 % zugetropft, wodurch 2 Mol Piperazin Mono-Hy-drochlorid erhalten werden. Zur erhaltenen Lösung werden 175,66 g N-(2-Chloracetyl)-cyclohexylamin gegeben. Die Temperatur steigt auf 100°C, und die Reaktion ist in 2 Stunden durch Rückfluss beendet.
Nach dem Abkühlen wird das Reaktionsgemisch mit einer 35 %igen NaOH-Lösung alkalisch gemacht und mit Chloroform extrahiert.
Die Chloroform-Lösung wird mit Wasser gewaschen, mit wasserfreiem Na2S04 getrocknet, dann wird die Lösung filtriert und eingedampft.
Der Rückstand wird mit Petroläther gewaschen, filtriert und getrocknet.
Das erhaltene N-(Piperazinyl-[l]-methyIen-carbonyl)--cyclohexylamin (C/63) wird in Vakuum auf 190°C (0,5 mm Druck) abdestilliert.
Schmelzpunkt = 1110/112°C (Ausbeute = 65% der Theorie).

Beispiel 2
86,2 g (1 Mol) wasserfreies Piperazin und 156,2 g (1 Mol) Piperazin Dihydrochlorid werden in 1.000 ml destilliertem Wasser gelöst. Zur erhaltenen Lösung werden 175,66 g 30 (1 Mol) N-(2-Chloracetyl)-cyclohexylamin gegeben. Das Verfahren wird wie im Beispiel 1 beendet.
Beispiel 3
258,6 g (3 Mol) Piperazin werden in 1000 ml destillier-35 tem Wasser gelöst. Zur erhaltenen Lösung werden 176,66 g (1 Mol) N-(2-ChloracetyI)-cyclohexylamin gegeben. Das Verfahren wird wie im Beispiel 1 beendet.
Beispiel 4
172,28 g (2 Mol) wasserfreies Piperazin werden in 1100 ml destilliertem Wasser gelöst und 166,7 ml HCl 37 % unter Rühren zugetropft, wodurch 2 Mol Piperazin Mono-Hy-drochlorid erhalten werden.
Zur erhaltenen Lösung werden 190,7 g (1 Mol) N-(2--ChloracetyI)-4-methyl-cyclohexylamin gegeben. Die Temperatur steigt auf 100°C, und die Reaktion wird in 2 Stunden unter Rückfluss beendet.
Nach dem Abkühlen wird das Reaktionsgemisch mit einer 35%igen NaOH-Lösung alkalisch gemacht und mit Chloroform extrahiert. Die Chloroformlösung wird mit Wasser gewaschen, mit wasserfreiem Na2SO., getrocknet und nach Filtration eingedampft.
Der Rückstand wird mit Petroläther gewaschen, filtriert und getrocknet.
N-(Piperazinyl-[ 1 ]-methylen-carbonyl)-4-methyl-cyclo-hexylamin (C/82) wird mit guter Ausbeute (etwa 70% der Theorie) erhalten.
Schmelzpunkt = 123°/124°C.

Beispiel 5
86,1 g (1 Mol) wasserfreies Piperazin und 156,2 g (1 Mol) 65 Piperazin Dihydrochlorid werden in 1.100 ml destilliertem Wasser gelöst. Zur erhaltenen Lösung werden 190,7 g (1 Mol) N-(2-ChIoracetyl)-4-methyl-cycIohexylamin gegeben. Das Verfahren wird wie in Beispiel 4 beendet.

Pharmakologische Tätigkeit von (C/63) und (C/82)
1) Akute Giftigkeit (C/63)
Mäuse DL/50 271 (311,6 - 235,6) mg/kg i.v.
Mäuse DL/50 1974 (2058,8 - 1892,6) mg/kg os. Ratten DL/50 3900 (4251 - 3577,9) mg/kg os.
2) Subchronische Giftigkeit (CI63)
80 Wistar-Ratten waren in 4 Gruppen von jeweils 20 (10 männlichen und 10 weiblichen) Tieren unterteilt. Die Verbindung C/63 wurde durch perorale Darreichung 3 Gruppen von Tieren für die Dauer von 30 Tagen in Dosen von 100-200 und 400 mg/kg eingegeben. Bei den Ratten ruft die subchronische Verabreichung von C/63 in Dosen von 400 mg/kg bei peroraler Darreichung für die Dauer von 30 Tagen keine von Arzneigiftigkeit verursachten Erscheinungen bei allen in Betracht gezogenen Parametern wie Blutcrasis, biochemische Blutanalyse, Gewicht und histologische Prüfung der Organe hervor.

Spasmolytische Tätigkeit (C/82)
C/82 hat keine anticholinergische Wirkung.
Die vorliegende Erfindung liefert auch pharmazeutische Zusammensetzungen, die erfindungsgemäss die neuen Verbindungen in Mischung mit einem pharmazeutischen festen oder flüssigen Verdünnungsmittel oder Träger und nach Wahl in Mischung mit aktiven Bestandteilen oder Bindemitteln enthalten.
Als Beispiele pharmazeutischer Formen kann man 50 oder 100 mg C/63 oder C/82 enthaltende Kapseln oder 50-100 mg C/63 oder C/82 enthaltende Phiolen nennen. Die tägliche Dosis besteht in 1 bis 2 Phiolen oder 2 bis 4 Kapseln.

- 624944PATENTANSPRÜCHE 1. Verfahren zur Herstellung neuer Derivate des Piper-azins und dessen Homologen der Formel:

- 4. Verfahren nach Anspruch 1 zur Herstellung von N-(Piperazinyl-[l ]-methylencarbonyl)-4-methyl-cycIohexyl-amin, dadurch gekennzeichnet, dass man N(2-ChIoracetyl)--4-methyl-cyclohexylamin und Piperazindichlorhydrat in5 Wasser bei einer Temperatur von 80-120°C während 7 bis 3^ Stunden kondensiert.
- 5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass die Kondensation von N(2-Chloracetyl)-4-methyl-cyclo-hexylamin mit Piperazin im Verhältnis von 1:1 bis 1:3 Mol io erfolgt.worin bedeuten n gleich 2 oder 3, m gleich 1, 2 oder 3,X Wasserstoff oder Methyl,R Wasserstoff, C,-Cc-Niederalkyl oder C1-C3-Alkohol, R, eine der folgenden Gruppen
