PATENTANSPRÜCHE
1. Verfahren zur Herstellung von N-[(N'-methyl)-p-aminoben-zoyl]-L-glutaminsäure, dadurch gekennzeichnet, dass man a) in p-(N-methyl)aminobenzoesäure eine Formylgruppe einführt,
b) die entstandene p-(N-methyl-N-formyl)aminobenzoesäure mit einem Chlorierungsmittel in das Säurechlorid überführt,
c) das entstandene p-(N-methyl-N-formyl) aminobenzoesäurechlorid mit L-Glutaminsäure in Gegenwart einer Base als Säureakzeptor umsetzt, und d) die entstandene p-(N-methyl-N-formyl) aminobenzoylglutaminsäure durch alkalische Hydrolyse in 2,5 bis 3,5 normaler Natronlauge bei 30 bis 70°C in die N-[(N'-methyl)-p-aminobenzoyl]-L-glutaminsäure überführt.
2. Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass man die Formylgruppe durch Kochen der p-(N-methyl) aminobenzoesäure in HCOOH als Reaktionsmedium am Rückfluss einführt.
3. Verfahren gemäss Patentansprüchen 1 und 2, dadurch gekennzeichnet, dass man die p-(N-methyl-N-formyl)aminobenzoesäure mit Sulfonylchlorid bei 40 bis 80°C in das Säurechlorid überführt.
4. Verfahren gemäss Patentansprüchen 1 bis 3, dadurch gekennzeichnet, dass man das p-(N-methyl-N-formyl) aminobenzoesäurechlorid mit L-Glutaminsäure in Gegenwart von Carbonaten, Bicarbonaten oder Hydroxiden von Na, K, Mg, Ca, Ba oder in Gegenwart von tertiären Aminen oder Calcium- oder Magnesiumoxid umsetzt.
Erfindungsgemäss wird das durch ein Verfahren gemäss Patentanspruch 1 erreicht.
Die Formylgruppe eignet sich hervorragend zum Schutz der Methylaminofunktion und nicht nur die Einführung der Formyl-5 grappe, sondern auch deren Abspaltung ist leicht und verläuft ohne Racemisierung des Glutaminsäureteils.
Das Verfahren basiert auf der folgenden Reaktionsreihe:

Es ist bekannt, p-(N-methyl)aminobenzoylglutaminsäure aus p-Jodobenzoylglutaminsäure und Methylamin in Gegenwart von Natronlauge bei pH 8 bis 9 und einem Kupferkatalysator herzustellen (Cosulich, Smith, „J. Am. Chem. Soc." 70, S. 1922,1948). Dieses Verfahren zeichnet sich durch die sehr teuren Jodderivate als Ausgangsprodukte aus und die Reproduzierbarkeit der Versuche ist nicht gewährleistet.
Gemäss Fu, Reiner, Loo, „J. Org. Chem." 30, S. 1277 1965 kann p-(N-methyl)aminobenzoylglutaminsäure hergestellt werden aus p-Methylaminobenzoesäure, die in das Säurechlorid überführt wird, das seinerseits mit Diäthyl-L-glutamat zum entsprechenden Amino-benzoylglutaminsäureester und nach geeigneter Hydrolyse zur Säure reagiert.
Die Methylaminogruppe der p-Methylaminobenzoesäure wird durch Reaktion mit Carbobenzyloxidchlorid geschützt. Das bedeutet, dass ein für technischen Massstab zu teures Schutzmittel angewendet werden muss, zudem konnte das Verfahren bezüglich des ersten Schrittes nicht reproduziert werden. Es wurde nur eine durch p-(N-methyl)aminobenzoesäure stark verunreinigte Benzyloxy-carbonylmethylaminobenzoesäure erhalten.
Es ist auch bekannt (D.V. Santi, „J. Heterocyc. Chem." 4, S. 475, 1967), ausgehend von Äthyl-p-aminobenzoat über Äthyl-p-(N-tosyl)aminobenzoat und Äthyl-p-(N-methyl-N-tosyl)aminobenzoat die p-(N-methyl-N-tosyl)aminobenzoesäure herzustellen, diese zum p-(N-methyl-N-tosyl)aminobenzoylchlorid zu chlorieren, durch Reaktion mit Diäthyl-L-glutamat-hydrochlorid den Diäthyl-p-(N-me-thyl-N-tosyl)aminobenzoylglutaminsäureester herzustellen, um nach einem weiteren Reaktionsschritt zum Diäthyl-p-(N-methyl) aminobenzoyl-(L)-glutamat zu gelangen. Diese Methode ist bezüglich der Zahl der Syntheseschritte sehr umständlich und eignet sich deshalb wenig für eine industrielle Ausnützung.
Die Erfindungsaufgabe besteht in einem Verfahren zur Herstellung von N-[(N'-methyl)-p-aminobenzoyl]-L-glutaminsäure, welches in bezug auf Materialkosten, Einfachheit der Ausführung und Übertragbarkeit in den technischen Massstab grosse Vorteile bietet.
Die Einführung der Formylgruppe in einen ersten Verfahrensschritt (I) kann durch Kochen der p-(N-methyl)aminobenzoesäure in 20 HCOOH als Reaktionsmedium am Rückfluss erfolgen. Die p-(N-me-thyl-N-formyl)aminobenzoesäure wird in quantitativer Ausbeute gewonnen und kann durch Waschen mit Wasser in guter Reinheit isoliert werden. Die p-(N-methyl-N-formyl)aminobenzoesäure kann mit dem Sulfonylchlorid bei 40 bis 80°C in das Säurechlorid überführt 25 werden (II). Mit Vorteil arbeitet man mit dem Sulfonylchlorid als Reaktionsmedium in einer Verdünnung von 10 zu 1, bei Temperaturen von 60°C, wobei aber weder Temperatur noch Verdünnung kritisch sind. Anhand der Abgasentwicklung kann der Verlauf der Reaktion verfolgt werden. Es ist möglich, die p-(N-methyl) 30 aminobenzoesäure mit Ameisensäure umzusetzen und nach erfolgter destillativer Entfernung der überschüssigen Ameisensäure das Reaktionsprodukt p-(N-methyl-N-formyl)aminobenzoesäure in demselben Reaktor mit dem Sulfonylchlorid zu behandeln.
Das entstandene p-(N-methyl-N-formyl) 35 aminobenzoesäurechlorid wird dann durch Verdampfen des überschüssigen Thionylchlorids als kristalline Substanz erhalten, die sich in organischen Lösungsmitteln wie Benzol, Toluol, Methylenchlorid usw. gut löst.
Das in einem der genannten Lösungsmittel gelöste Säurechlorid 40 wird in einem weiteren Schritt (III) einer wässrigen Suspension von L-Glutaminsäure zudosiert.
Dabei wird eine Base, vorzugsweise eine nicht allzu starke Base, als Säureakzeptor eingesetzt. Als Base können Carbonate, Bicarbonate oder Hydroxide von Na, K, Mg, Ca, Ba, tertiäre Amine oder 45 Calcium- oder Magnesiumoxid angewendet werden.
Die entstandene p-(N-methyl-N-formyl)aminobenzoyl-L-glutaminsäure fällt zum grossen Teil nach Ansäuern auf pH 1 als Öl aus, welches durch Dekantierung isoliert werden kann. Zur Gewinnung des restlichen Produktes können dann verschiedene, an sich be-50 kannte Methoden, wie beispielsweise Flüssig/Flüssigextraktion der wässrigen Mutterlauge verwendet werden. Bei dem letzten Schritt (IV) wird die Formylgruppe durch alkalische Hydrolyse abgespalten. Dies darf nicht unter scharfen Bedingungen geschehen, wegen der Gefahr der Racemisierung des Glutaminsäureteils. Als optimaler 55 Kompromiss für die Konzentration der Base und die Temperatur wurde gefunden, dass die Hydrolyse der Formylgruppe in 2,5 bis 3,5 N, am besten in 3N NaOH, bei 30 bis 70°C, vorzugsweise bei 40 bis 50° C, quantitativ und ohne Racemisierung durchgeführt werden kann. Die N-[(N'-methyl-p-aminobenzoyl]-L-glutaminsäure kann 60 durch Neutralisieren und Eindampfen der Lösung mit anschliessender Methanolextraktion des Rückstandes isoliert werden, allerdings fällt dabei ein durch NaCl und Na-Formiat verunreinigtes Produkt an. Durch Behandlung der Methanollösung mit BaCl2 kann N-[(N'-methyl)-p-aminobenzoyl]-L-glutaminsäure als Bariumsalz in reiner 65 Form isoliert werden. Die aus der Hydrolyse erhaltene wässrige Lösung, die praktisch nur N-[(N'-methyl)p-aminobenzoyl]-L-glutamin-säurenatriumsalz, Natriumformiat und NaOH enthält, kann als solche auch direkt in der Methotrexatsynthese eingesetzt werden. N-[(N'-methyl)-p-aminobenzoyl]-L-glutaminsäure ist ein Bruchstück des wichtigen Krebsmittels Methotrexat und wird heute als Edukt im Methotrexatherstellungsverfahren eingesetzt (D.R. Seeger, „J. Am. Chem. Soc." 71, S. 1753,1949).
Beispiel:
MeAmbs = p-(N-methyl)aminobenzoesäure
MeAmbsGls = N-[(N'-methyl)-p-aminobenzoyl]-L-glutamin-säure
10,0 g (0,066 mol) MeAmbs wurden in 100 ml Ameisensäure wasserfrei während 3 h am Rückfluss gekocht, dann zur Trockene eingedampft. Das Reaktionsprodukt, Formyl-MeAmbs, kann in reiner Form (weisse Kristalle Fp. 218°C) durch Suspendieren in Wasser oder besser durch Umfallen mittels NaOH/HCl in 97% Ausbeute isoliert werden, oder es kann als Rohware in der nächsten Stufe eingesetzt werden.
Das Rohprodukt der Formylierungsstufe wurde in 140 ml Thio-nylchlorid suspendiert und auf 60° C erhitzt, bis alles in Lösung ging und keine Abgase mehr entwickelt wurden. Nach destillativer Entfernung des Thionylchlorids unter Vakuum wurde der Rückstand in 60 ml Benzol gelöst und als gelbe Lösung für die weitere Stufe verwendet. Ein Beweis für den quantitativen Verlauf dieser Reaktion war die Abwesenheit von in Benzol unlöslichen Kristallen von Formyl-MeAmbs.
Eine Suspension von 10,6 g MgO und 14,2 g L-Glutaminsäure (0,096 mol) in 100 ml H20 wurde nun 1 h bei 70°C gerührt, dann auf 30° C abgekühlt und unter Rühren im Verlauf von etwa 15 min mit der benzolischen Lösung von Formyl-MeAmbs-chlorid aus Schritt II tropfenweise versetzt und über Nacht bei RT nachgerührt. Nach Trennung der Schichten wurde die wässrige Phase mittels konz. HCl auf pH 1 gebracht und über Nacht stehen gelassen. Die dabei gebildete Ölschicht, welche im wesentlichen aus Formyl-MeAmbsGls bestand, wurde von der wässrigen Mutterlauge getrennt. Sie machte 11,5 g aus (56% d.Th., bezogen auf eingesetztes MeAmbs).
In der Mutterlauge befanden sich nach HPLC (High Pressure Liquid Chromatography) nur Spuren von MeAmbs und Formyl-MeAmbs (< 1% Ausbeute); die Hauptkomponenten waren Formyl-MeAmbsGls und die sich durch Säurehydrolyse gebildete 5 MeAmbsGls im Verhältnis 3 zu 1. Bei dieser Reaktionsstufe betrug also die Ausbeute an Formyl-MeAmbsGls etwa 89% und an MeAmbsGls etwa 11 %, bezogen auf eingesetzte MeAmbs.
Das isolierte Öl von Stufe III wurde nun in 90 ml NaOH 3N gut gerührt und die entstandene Suspension bei 40 bis 52° C 1 h gerührt, io Die HPLC-Analyse zeigte, dass sich nach dieser Zeit MeAmbsGls als Na-Salz quantitativ gebildet hatte und dass also keine Abspaltung des Glutaminsäureteils geschehen war.
Die Lösung könnte nun direkt in der Methotrexatsynthese eingesetzt werden.
15 Zur Bestimmung der optischen Reinheit der MeAmbsGls wurde diese folgendermassen isoliert: Die Hydrolyselösung wurde auf pH 6 mittels konz. HCl gebracht und unter Vakuum zur Trockene eingedampft. Der Rückstand wurde in 300 ml Methanol aufgeschlämmt und 3 h gerührt, dann abfiltriert. Das klare Filtrat wurde unter Va-20 kuum konzentriert, bis es milchig wurde, dann mit einer 0,5M BaCl2-Lösung in H20/Me0H 1 zu 1 versetzt, bis kein Produkt mehr ausfiel. Nach Filtration und Trocknung erhielt man 15g (48%) MeAmbsGls-Ba-Salz, welches noch Spuren Ba-Formiat und BaCl2 enthielt. 10 g davon wurden aus H20/Me0 umgefällt und ergaben 25 7 g reines (>98%ig) MeAmbsGls-Ba-Salzdihydrat. 12 g des rohen Ba-Salzes von MeAmbsGls wurden in 120 ml 20% HCl in absolutem Äthanol 3 d gerührt, nachher mit 200 ml H20 und dann mit NH40H konz. bis pH 7,5 versetzt. Nach Extraktion mit Äther und Trocknung und anschliessender Verdampfung des Äthers wurde ein Rück-30 stand erhalten, welcher nach Umkristallisation aus Alkohol/H20 6,3 g MeAmbsGls-diäthylester als weisse Kristalle lieferte. Fp. 90°C;
