Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Guanidinderivaten. Die erfindungsgemäss herstellbaren Verbindungen hemmen beispielsweise die Magen-saftsekretion und senken den Blutzuckerspiegel.
30 In der GB-PS 1 409 768 sind verschiedene hetero-cyclische Derivate von Guanidin beschrieben, bei denen der heterocyclische Rest ein 5- oder 6-gliedriger gesättigter 1,3-diazacarbocyclischer 2-Ylidenrest ist. Diese Derivate sind am Amino-Stickstoffatom des Guanidinrestes unsub-35 stituiert.
Weitere ähnliche Verbindungen sind in den DE-OS 2 321 330 und 2 502 397 beschrieben.

Der Ausdruck «nieder» bedeutet, dass die entsprechende Gruppe, vorzugsweise 1 bis 4 Kohlenstoffatome enthält. Als 15 Halogenatome kommen gewöhnlich Fluor-, Chlor-, Brom-und Jodatome in Frage. Wenn einer der Reste R2 oder R3 einen Alkyl- oder Arylrest bedeutet, ist der andere Rest vorzugsweise ein Wasserstoffatom. Wenn n den Wert 2 oder 3 hat, ist der Rest R2 und R3 vorzugsweise ein Wasserstoff-20 atom.
Die Verbindungen der allgemeinen Formel I können aufgrund der Gegenwart eines tertiären Stickstoffatoms Salze mit Säuren und quartäre Ammoniumsalze bilden. Beispiele für die zur Salzbildung verwendbaren Säuren sind anorgani-25 sehe Säuren, wie Halogenwasserstoffsäuren, beispielsweise Salzsäure und Bromwasserstoffsäure, Schwefelsäure, Salpetersäure, und Phosphorsäure, und organische Säuren, wie Essig-, Propion-, Glykol-, Pamoin-, Brenztrauben-, Malon-, Bernstein-, Malein-, Fumar-, Äpfel-, Wein-, Citronen-, Ben-30 zoe-, Zimt-, Mandel-, Methansulfon-, Äthansulfon-, Benzol-sulfon-, p-Toluolsulfon-, Cyclohexansulfamin-, Salicyl- und p-Aminosalicylsäure. Aus den Salzen können die freien Basen durch Behandlung mit einer Base gewonnen werden.
Beispiele für die zur Quaternisierung verwendbaren Ver-35 bindungen sind Alkylierungsmittel, wie Alkyl-, Alkenyl- und Aralkylhalogenide-, -sulfate und -sulfonate, wie Methyl-jodid, Äthylbromid, Propylbromid, Allylchlorid, Benzyl-chlorid, Dimethylsulfat, Diäthylsulfat, p-Toluolsulfonsäure-methylester und Fluorsulfonsäuremethylester. Die Quater-40 nisierung kann in Gegenwart oder Abwesenheit eines Lösungsmittels bei Raumtemperatur oder unter Kühlung, bei Atmosphärendruck oder Überdruck durchgeführt werden. Beispiele für verwendbare inerte organische Lösungsmittel sind Äther, wie Diäthyläther und Tetrahydrofuran, Kohlen-45 Wasserstoffe, wie Benzol und Heptan, Ketone, wie Aceton und Butanon, niedere aliphatische Alkohole, wie Äthanol, Propanol und Butanol, sowie Säureamide, wie Formamid und Dimethylformamid. Das Anion des quartären Ammoniumsalzes lässt sich im allgemeinen durch Ionen-50 austausch leicht gegen ein anderes Anion austauschen.
Aufgrund ihrer sehr hohen blutzuckersenkenden Wirkung sind erfindungsgemäss herstellbare Verbindungen der allgemeinen Formel II

Ausgangsstoffe der Formel XVII erhält man z.B. aus einer 2-Iminoverbindung der nachstehend angegebenen allgemeinen Formel, in der n, Rls R2 und R3 die vorstehend angegebene Bedeutung haben. Diese 2-Iminoverbindungen sind grösstenteils bekannt oder können nach dem in der CAPS 950 464 beschriebenen Verfahren, beispielsweise gemäss Beispiel 14 dieser PS, hergestellt werden. Diese 2-Iminover-bindungen werden vorzugsweise mit einem Isothiocyanat der allgemeinen Formel XV, in der R die vorstehend angegebene Bedeutung hat und vorzugsweise keine 9-Fluorenyl-gruppe darstellt, in einem inerten organischen Lösungsmittel, wie Benzol, Methylenchlorid oder Chloroform, bei Temperaturen von 0 bis Raumtemperatur während etwa 2 bis 24 Stunden in etwa äquimolaren Mengen umgesetzt. Sodann wird im allgemeinen die Thiogruppe in den erhaltenen Thio-harnstoffen der allgemeinen Formel XVI, von denen mehrere in der US-PS 3 717 648 beschrieben sind, durch Umsetzung der Verbindungen der allgemeinen Formel XVI mit einem Alkylierungsmittel der allgemeinen Formel R'X, in der R' eine Äthyl- oder vorzugsweise Methylgruppe darstellt

Die Umsetzung wird vorzugsweise in einem Lösungsmittel durchgeführt. Beispiele s für verwendbare Lösungsmittel sind Äther, vorzugsweise Diäthyläther, Tetrahydrofuran oder Dioxan, niedere Ketone, wie Aceton oder 2-Butanon, Halogenkohlenwasserstoffe und niedere aliphatische Alkohole, vorzugsweise Methylen-dichlorid und Methanol.
io Besonders geeignet ist Methyljodid als Alkylierungsmittel in Methanol. Im allgemeinen wird das Alkylierungsmittel in mindestens äquimolarer Menge verwendet. Der Mengenanteil hängt normalerweise von der Reaktionsfähigkeit des Thioharnstoffs der allgemeinen Formel XVI oder seiner Lös-15 lichkeit im verwendeten Lösungsmittel ab. Die Alkylierungs-reaktion kann bei Raumtemperatur oder bei höheren Temperaturen in einem geschlossenen Reaktionsgefäss durchgeführt werden. Die Alkylthioverbindungen der allgemeinen Formel XVII werden sodann in Form ihrer Salze mit einer 20 Säure (HX) vorzugsweise bei Temperaturen von Raumtemperatur bis zur Rückflusstemperatur oder in einem geschlossenen Reaktionsgefäss bei höheren Temperaturen mit einem Amin der allgemeinen Formel HNR4RS umgesetzt, in der R4 und R5 die vorstehend angegebene Bedeutung haben. 25 Die Umsetzung wird in einem niederen aliphatischen Alkohol, wie beispielsweise Isopropanol oder tert.-Butanol, und bei der Rückflusstemperatur des Reaktionsgemisches durchgeführt. Es werden die Guanidinderivate der allgemeinen Formel I in Form ihrer Salze erhalten, die mit Alkali 30 in die entsprechenden freien Basen überführt werden. Dieses Verfahren kann durch folgendes Reaktionsschema erläutert werden:

Die Isothiocyanate der allgemeinen Formel XV, von denen zahlreiche Verbindungen bekannt sind, können in an sich bekannter Weise hergestellt werden.
Bei der Umsetzung der Verbindungen der allgemeinen Formel XVII mit dem Amin der allgemeinen Formel HNR4R5 wird das Amin vorzugsweise im Überschuss über die stöchiometrische Menge verwendet, beispielsweise in einr nem Molverhältnis von 1: 1,05 bis 1 :2,0. Bei Verwendung nur eines geringen Überschusses des Amins kann es zweckmässig sein, eine stöchiometrisch äquivalente Menge eines tertiären Alkylamins, wie Triäthylamin, zuzusetzen, um die Reaktionsgeschwindigkeit zu erhöhen. Während der Umsetzung können sich Nebenprodukte, beispielsweise der allgemeinen Formel

bilden. Diese Verbindungen können durch fraktionierende Umkristallisation von den Verbindungen der allgemeinen Formel I abgetrennt werden.
Die Verbindungen der allgemeinen Formel I und ihre Salze mit Säuren und quartären Ammoniumsalze sind beispielsweise wertvolle Arzneistoffe, die die Magensaftsekretion hemmen und den Blutzuckerspiegel senken, wie nachfolgend erläutert:
Die Hemmung der Magensaftsekretion wurde an Ratten im Magenfisteltest nachgewiesen. Für die Versuche wurden weibliche Ratten des Sprague-Dawley-Stammes verwendet, denen die zu untersuchende Verbindung intraduodenal in einer Dosis von 2,5 bis 40 mg/kg Körpergewicht injiziert wurde. 24 Stunden vor dem Versuch werden die Ratten nicht mehr gefüttert, es wird ihnen jedoch Wasser angeboten. Die Ratten werden einzeln gehalten. Am Versuchstag werden die Ratten gewogen und so ausgewählt, dass für jede Versuchsgruppe das Körpergewicht im Bereich von + 20 g liegt.
Die Operation wird unter leichter Äthernarkose durchgeführt. Sobald die Ratte narkotisiert ist, werden die Zähne mittels einer kleinen Kneifzange entfernt. Es wird eine etwa 1,5 cm lange mediane Laparatomie ausgeführt und der Magen und das Duodenum dargestellt. Wenn zu diesem Zeitpunkt der Magen mit Nahrung oder Faeces gefüllt ist, wird die Ratte verworfen. Mit einer 4-0 Naht wird eine Tabaksbeutelnaht im Fundusbereich angelegt, wobei darauf geachtet wird, keine Blutgefässe in diesem Gebiet anzustechen. In das Zentrum der Tabaksbeutelnaht wird ein kleiner Schnitt in die Magenwand angelegt und eine Kanüle aus einem kleinen Vinylschlauch mit einem Flansch an einem Ende in den Magen eingeführt. Sodann wird die Tabaksbeutelnaht fest um den Flansch zugezogen.
Unmittelbar danach wird die zu untersuchende Verbindung in einer Menge von 0,5 ml/100 g Ratte intraduodenal zugeführt. Für jede Testdosis werden im allgemeinen drei Ratten verwendet. Kontrollratten erhalten die im Test benützte Trägersubstanz, im allgemeinen 0,5%ige wässrige Methylcelluloselösung.
Nach der Zufuhr der Testflüssigkeit wird die Bauchwand und Haut gleichzeitig mit drei bis vier 18 mm Wundklipps verschlossen. Ein Sammelschlauch wird an die Kanüle angeschlossen. Jede Ratte wird dann in einen Käfig gesetzt, in den ein Längsschlitz geschnitten wurde, um die Kanüle frei hängen lassen zu können und der Ratte freie Bewegung zu ermöglichen. Nachdem sich die Ratte 30 Minuten lang stabilisieren und erholen konnte, wird der Sammelschlauch von der Kanüle verworfen und durch einen sauberen Schlauch 65 zur Aufnahme des Magensaftes ersetzt. Jede Stunde wird die sezernierte Menge gesammelt. Am Ende dieser Untersuchung wird die Kanüle entfernt und die Ratte getötet.
Die Probe des Mageninhalts wird in ein Zentrifugenröhr-chen entleert und zentrifugiert, um das Sediment niederzuschlagen. Das Volumen wird abgelesen, und 1 ml des Überstandes wird in ein Becherglas gegeben, das 10 ml destilliertes Wasser enthält, und mit 0,01 n Natronlauge bis zu einem pH-Wert von 7 titriert. Es werden das Volumen, die titrierbare Säure und die Gesamtazidität bestimmt. Das Volumen berechnet sich aus dem Gesamtvolumen an Magensaft, abzüglich dem Sediment. Die titrierbare Säure in Milliäquiva-lent/Liter ist diejenige Menge an 0,01 n Natronlauge, die zur Titration der Säure bis zum pH-Wert 7 erforderlich ist. Die Gesamtazidität ist das Produkt aus titrierbarer Säure mal Volumen. Die Ergebnisse werden in % Hemmung gegenüber dem Wert von Kontrolltieren angegeben. Ein Mindestwert von 5% Hemmung ist ein Anzeichen für eine signifikante Hemmung der Magensaftsekretion.
Die Verbindungen der allgemeinen Formel I und ihre Salze senken im allgemeinen auch den Blutzuckerspiegel, d.h. sie haben hypoglykämische Eigenschaften. Dies wird vorzugsweise anhand des Glucosetoleranztests an Ratten nachgewiesen. Besonders ausgeprägte hypoglykämische Eigenschaften haben gewöhnlich die Verbindungen der allgemeinen Formel II.
Der Glucosetoleranztest an Ratten ist im allgemeinen ein extrem empfindlicher Standardtest zur Diagnose von Diabetes und hypoglykämischen Krankheitsbildern und sei nachfolgend erläutert:
Für den Versuch werden männliche Ratten des Sprague-Dawley-Stammes mit einem Körpergewicht von 184 bis 250 g verwendet. Die Ratten werden 24 Stunden vor dem Versuch nicht mehr gefüttert, jedoch wird ihnen Wasser angeboten. Nach oraler Gabe von 1 g Glucose/kg Körpergewicht in 1 ml Wasser wird ihnen nach 0 Minuten, 30 Minuten, 60 Minuten, 90 Minuten, 120 Minuten, 150 Minuten und 180 Minuten 0,1 ml Blut aus der Schwanzvene ohne Anästhesie entnommen. Die Blutproben werden sofort mit wässrigen Lösungen von Bariumhydroxid und Zinksulfat entproteinisiert, und die Glucosespiegel werden nach dem Glucoseoxidase-Test von L.P. Cawley u. Mitarb., «Ultra Micro Chemical Analysis of Blood Glucose with Glucose Oxidase» Amer. J. Clin. Path., Bd. 32 (1959), S. 195, bestimmt. Für jeden Versuch werden 2 bis 5 Ratten sowie eine Kontrollgruppe verwendet. Die zu untersuchenden Verbindungen werden in einer Dosis von 1 bis 200 mg/kg entweder subcutan oder intraperitoneal in 0,5 oder 1 ml einer 0,5 bis lprozentigen Methylcelluloselösung gegeben. Den Kontrollgruppen wird die gleiche Volumenmenge Träger subcutan gegeben. Die Blutglucosewerte werden zu jedem Zeitpunkt ausgedruckt in mg % (mg Glucose/100 ml Blut). Die mittleren Glucosewerte der Kontrollgruppen werden statistisch nach dem t-Test von Student mit den Mittelwerten der Ver25

suchsgruppe zu jedem entsprechenden Zeitpunkt verglichen. Wenn die zu untersuchende Verbindung den Blutglucose-wert signifikant zu jedem Zeitpunkt bei einer Vertrauensgrenze von 95% vermindert, hat die Verbindung hypoglykämische Aktivität.
Ausführungsbeispiele Beispiel 1
A) N-( 1 -Methyl-2-pyrrolidinyliden)-N'-phenylthioharnstoff Eine Suspension von 6,73 g (0,05 Mol) 2-Imino-l-me-
thyl-pyrrolidin-hydrochlorid in Benzol wird unter Rühren mit 5 ml 50prozentiger Natronlauge versetzt. Nach 2minutigem Rühren wird die Benzollösung auf wasserfreies Kaliumcarbonat dekantiert und die wässrige Phase zweimal mit frischem Benzol extrahiert. Die Benzolextrakte werden vereinigt und nach dem Trocknen rasch durch ein Bett aus Diatomeenerde unter möglichst geringem Luftzutritt filtriert. Das Filtrat wird mit 6,76 g (0,05 Mol) Phenylisothio-cyanat rasch versetzt. Nach 3stündigem Rühren wird der erhaltene Feststoff abfiltriert. Aus der Mutterlauge wird eine weitere Menge der Titelverbindung erhalten. Nach Ümkri-stallisation aus Äthylacetat schmilzt die Verbindung bei 142 bis 143,5 °C.
B) N-(l-Methyl-2-pyrrolidinyliden)-N'-phenylcarbamimi-dothiomethylester-hydrojodid
Eine Lösung von 34,86 g (0,15 Mol) der in (A) erhaltenen Verbindung in 500 ml Aceton wird mit einer Lösung von 21,3 g (0,15 Mol) Methyljodid in Aceton versetzt. Die Lösung wird 30 Minuten unter Rückfluss erhitzt und eine weitere Stunde bei Raumtemperatur stehengelassen. Beim Abkühlen in einem Eisbad erfolgt Kristallisation. Die Kristalle werden abfiltriert und aus einem Gemisch von Methanol und Isopropanol umkristallisiert. Es wird die Titelverbin-dung vom F. 145 bis 147 °C erhalten.
C) Die Umsetzung der in (A) erhaltenen Verbindung in einem niederen aliphatischen Alkohol, Aceton oder einem niederen Halogenkohlenwasserstoff mit p-Toluolsulfon-säuremethylester, Dimethylsulfat, Fluorsulfonsäuremethyl-ester, Trimethyloxoniumfluoborat oder Methansulfonsäure-methylester liefert die Methylthioverbindung (B) als entsprechendes Salz.
D) N-( 1 -Methyl-2-pyrrolidinyliden)-N'-phenyl-1 -pyrrolidin-carboximidamid-hydrojodid (I)
Eine Lösung von 15,02 g (0,04 Mol) der in (B) erhaltenen Verbindung und 4,31 g (0,06 Mol) 99prozentigem Pyrrolidin in 100 ml Isopropanol wird 24 Stunden unter Rückfluss erhitzt. Beim Abkühlen im Eisbad erfolgt Kristallisation. Die Kristalle werden abfiltriert und die Mutterlauge aufbewahrt; vgl. Beispiel 26. Die Kristalle werden aus Isopropanol sowie einem Gemisch von Methanol und Diäthyläther umkristallisiert. Es wird die Titelverbindung vom F. 206 bis 208 °C erhalten.
Präparat A
Nach dem in der CA-PS 950 464 beschriebenen Verfahren werden folgende Salze der 2-Iminoverbindungen der allgemeinen Formel XIV hergestellt:
a. 2-Imino-l-methyl-5-phenylpyrrolidin-fluoborat, F. 128-131 °C;
b. 2-Imino-l-methylpyrrolidin-fluoborat, F. 109-111 °C;
c. 2-Imino-l-äthylpyrrolidin-hydrochlorid, F. 181-185 °C;
d. 2-Imino-l-n-butylpyrrolidin-cyclohexansulfamat, F. 110-114,5 °C;
e. 2-Imino-l-benzylpyrrolidin-fluoborat, F. 112-114 °C;
f. 2-Imino-l,5-dimethylpyrrolidin-fluoborat, F. 100-102°C;
g. 2-Imino-l -methylpiperidin-hydrochlorid;
h. Hxahydro-2-imino-1 -methyl-1 H-azepin-cyclohexansul-famat, F. 143-145 =C;
i. 1 -Hydroxyäthyl-2-iminopyrrolidin-cyclohexansulfamat,

Diese Salze werden durch Behandlung mit 50prozentiger Natronlauge gemäss Beispiel 1-A in die freien Basen überführt.
Präparat B
Nach dem von J. C. Jochims und A. Seeliger, Angew. Chemie, Internat. Ed., Bd. 6 (1967), S. 174, beschriebenen Verfahren werden folgende Isothiocyanate hergestellt:
a. 1- und 2-(2,3,3a,4,5,6,7,7a-Octahydro-4,7-methanoin-danyl)-isothiocyanat;
b. 1,1 -Dimethylphenäthylisothiocyanat;
c. 1-a-Phenäthylisothiocyanat;
d. d-a-Phenäthylisothiocyanat;
e. 1-Benzylcyclopentylisothiocyanat;
f. 7-(Bicyclo[4.2.0]octa-l,3,5-trienyl)-isothiocyanat;
g. 1-Adamantylmethylisothiocyanat;
h. 2-Adamantylisothiocyanat;
i. trans-2-Phenylcyclopentylisothiocyanat; j. cis-2-Phenylcyclopentylisothiocyanat;
k. trans-2-Cyclohexylcyclopentylisothiocyanat; 1. cis-2-Cyclohexylcyclopentylisothiocyanat und m. cis-2-Phenylcycloheptylisothiocyanat.
Präparat C
Gemäss Beispiel 1-A, jedoch unter Verwendung einer äquivalenten Menge der in Präparat A erhaltenen 2-Imino-verbindung, werden bei der Umsetzung mit einer äquivalenten Menge des entsprechenden Isothiocyanats folgende Thioharnstoffe der allgemeinen Formel XVI erhalten:

Beispiel 1-D wird wiederholt, jedoch werden die entsprechenden Methylthio-hydrojodide von Beispiel 1-B und Präparat D sowie die entsprechenden Amine der allgemeinen Formel HNR4R5 in einem Molverhältnis von 1 :1,5 bis 1: 3 in unter Rückfluss kochendem Isopropanol oder vorzugsweise tert.-Butanol zu den entsprechenden Verbindungen der allgemeinen Formel I in Form der Hydrojodide umgesetzt. Die Hydrojodide werden als solche isoliert oder mit wässrigem Alkali in die freien Basen überführt oder durch Umsetzung der freien Base mit der angegebenen Säure in ein anderes Salz überführt. Die Guanidine der allgemeinen Formel IV können als Nebenprodukte gebildet und aus dem Reaktionsgemisch beispielsweise durch fraktionierende Kristallisation als freie Basen oder als Salze einer Säure isoliert werden. Beispiel 3
Dieses Beispiel erläutert Verfahren zur Herstellung von quartären Ammoniumsalzen der Verbindungen der allgemeinen Formel I.
A) N-(2,6-Dichlorphenyl)-N'-(l-methyl-2-pyrrolidinyliden)-1-pyrrolidincarboximidamidinium-methofluorsulfonat Ein Gemisch von 6,83 g (0,02 Mol) N-(2,6-Dichlor-phenyl)-N'-( 1 -methyl-2-pyrrolidinyliden)-1 -pyrrolidincarb-oximidamid in wasserfreiem Methylenchlorid wird unter Stickstoff als Schutzgas und unter Rühren mit 2,70 g (0,023 Mol) Fluorsulfonsäuremethylester versetzt. Das Gemisch wird etwa 16 Stunden gerührt und sodann unter vermindertem Druck zur Trockene eingedampft. Der ölige Rückstand wird mit Diäthyläther digeriert. Die entstandenen Kristalle werden zunächst aus Aceton und sodann aus Äthylacetat umkristallisiert. Es wird die Titelverbindung vom F. 148-150 °C erhalten.
so B) N-(l-Methyl-2-pyrrolidinyliden)-N'-phenyl-l-pyrrolidin-carboximidamidinium-methojodid
Eine Lösung von 0,05 Mol N-(l-Methyl-2-pyrrolidinyli-den-N'-phenyl-l-pyrrolidincarboximidamid (Beispiel 1-D) in 30 ml Aceton wird mit 0,05 Mol Methyljodid versetzt. Nach 55 48stündiger Umsetzung werden die entstandenen Kristalle abfiltriert und aus Aceton umkristallisiert. Es wird die Titelverbindung vom F. (156) 162-164 °c erhalten.
c) Die Umsetzung von 0,05 Mol der Verbindung von Beispiel 1-D in wasserfreiem Diäthyläther mit 0,05 Mol 6o Fluorsulfonsäuremethylester liefert eine ölige Fällung, die kristallisiert. Nach Umkristallisation aus tert.-Butanol wird das Produkt von Beispiel 18-B in Form des Methofluorsul-fonats vom F. 135,5-137 °c erhalten.

2,46 g (0,0078 Mol) N-(l-Methyl-2-pyrrolidinyliden)-N'-(4-nitrophenyl)-l-pyrrolidincarboximidamid in 50 ml wasserfreiem Äthanol werden in Gegenwart von Raney-Nickel bei etwa 3 at Wasserstoffdruck hydriert. Die Wasserstoffaufnahme ist innerhalb etwa 30 Minuten beendet. Sodann wird der Katalysator abfiltriert und mit Äthanol ausgewaschen. Das Filtrat und die Waschlösung werden vereinigt und unter vermindertem Druck eingedampft. Der Rückstand wird in Äthanol gelöst und mit 1 Äquivalent 48prozentiger Bromwasserstoffsäure neutralisiert. Die entstandenen Kristalle werden abfiltriert und aus einem Gemisch von Äthanol und Diäthyläther umkristallisiert. Es wird die Titelverbindung vom F. 241-241,5 °C erhalten.
Beispiel 6
N-(4-Acetylaminophenyl)-N'-(l-methyl-2-pyrrolidinyliden)-1-pyrrolidincarboximidamid-hydrobromid
3,4 g (0,0119 Mol) N-(4-Aminophenyl-N'-(l-methyl-2-pyrrolidinyliden)-l-pyrrolidincarboximidamid (das Produkt von Beispiel 5) werden mit 25 ml Essigsäureanhydrid etwa 1 Stunde auf einem Dampfbad erhitzt. Danach wird das Reaktionsgemisch mit Äthanol verdünnt und eine weitere Stunde erhitzt. Hierauf werden die flüchtigen Bestandteile unter vermindertem Druck abdestilliert, und der Rückstand wird mit verdünnter Natronlauge behandelt und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt wird über Kaliumcarbonat getrocknet, filtriert und eingedampft. Es hinterbleibt ein Öl, das in einem Gemisch von Diäthyläther und Äthanol mit Bromwasserstoff in das entsprechende Hydrobromid überfuhrt wird. Nach Umkristallisation aus einem Gemisch von Äthanol und Diäthyläther schmilzt die Titelverbindung bei 246 bis 247 °C.
Beispiel 7
N-( 1 -Methyl-2-pyrrolidinyliden)-N'-(4-methylsulfonylphe-nyl)-4-morpholincarboximidamid
Eine Lösung von 1,98 g (0,0006 Mol) N-(4-Methylthio-phenyl)-N'-(l-methyl-2-pyrrolidinyliden)-4-morpholincarb-oximidamid in wasserfreiem Methylenchlorid wird bei 0 °C unter Rühren mit 3,6 g (0,018 Mol) m-Chlorperbenzoesäure versetzt. Die Temperatur soll nicht über 5 °C ansteigen. Danach wird das Gemisch 30 Minuten bei dieser Temperatur gerührt und sodann dreimal mit jeweils 40 ml gesättigter Na-triumbicarbonatlösung und dreimal mit jeweils 35 ml gesättigter Kochsalzlösung gewaschen. Die organische Phase wird über Kaliumcarbonat getrocknet und durch eine Filtrierhilfe filtriert. Das Filtrat wird unter vermindertem Druck nahezu zur Trockene eingedampft. Der Rückstand wird mit Methanol verdünnt. Unlösliche Stoffe werden abfiltriert und verworfen. Das Filtrat wird auf einem Dampfbad nahezu

zur Trockene eingedampft. Der Rückstand wird mit Diäthyläther verdünnt. Die entstandenen Kristalle (F.
160-166 °C) werden aus Diäthyläther umkristallisiert. Es wird die reine Titelverbindung vom F. (165) 167-170 °C erhalten.
Beispiel 8
N-(l-Methyl-2-pyrrolidinyliden)-N'-(4-methylsulfinylphe-nyl)-4-morpholincarboximidamid
Eine Lösung von 1,98 g (0,006 Mol) N-(4-Methylthio-phenyl)-N'-(l-methyl-2-pyrrolidinyliden)-4-morpholincarb-oximidamid in Methylenchlorid wird bei 5 °C mit 1,22 g (0,006 Mol) m-Chlorperbenzoesäure versetzt. Die Temperatur soll nicht über 5 °C ansteigen. Danach wird das Reaktionsgemisch auf Raumtemperatur erwärmt und weitere 2 Stunden gerührt. Hierauf wird die organische Phase nacheinander dreimal mit jeweils 30 ml gesättigter Natriumbicar-bonatlösung und zweimal mit jeweils 30 ml gesättigter Kochsalzlösung gewaschen und über wasserfreiem Kaliumcarbonat getrocknet. Nach dem Filtrieren wird das Lösungsmittel unter vermindertem Druck abdestilliert. Der Rückstand wird mit einem Gemisch von Diäthyläther und Hexan behandelt. Das kristalline Rohprodukt wird aus einem Gemisch von Diäthyläther und Hexan umkristallisiert. Es wird die reine Titelverbindung vom F. (115) 117-120 °C erhalten.
Beispiel 9
N-(4-Hydroxyphenyl)-N'-(l-methyl-2-pyrrolidinyliden)-l-pyrrolidincarboximidamid-hydrojodid
Eine Lösung von 26,6 g (0,072 Mol) N-(l-Methyl-2-pyr-rolidinyliden)-4'-(4-benzyloxyphenyl)-2-pyrrolidincarb-oximidamid in 50 ml Essigsäure wird in Gegenwart von lOprozentigem Palladium-auf-Kohlenstoff etwa 2 Stunden bei 3 at Wasserstoffdruck und bei Raumtemperatur hydriert. Sobald die Wasserstoffaufnahme beendet ist, wird der Katalysator abfiltriert und das Filtrat unter vermindertem Druck eingedampft. Es hinterbleibt ein viskoses Öl, das in Aceton gelöst und mit 1 Äquivalent konzentrierter Jodwasserstoffsäure versetzt wird. Die erhaltene rohe kristalline Titelverbindung wird aus einem Gemisch von Methanol, Aceton und Diäthyläther umkristallisiert. Die reine Titelverbindung schmilzt bei 174-178 °C.
Beispiel 10
N-(4-Acetoxyphenyl)-N'-(l-methyl-2-pyrrolidinyliden)-l-pyrrolidincarboximidamid-hydrojodid
Eine Lösung von 5,15 g (0,0124 Mol) N-(4-Hydroxyphe-nyl)-N'-( 1 -methyl-2-pyrrolidinyliden)-1 -pyrrolidincarb-oximidamid-hydrojodid und 41,2 g (0,20 Mol) N,N'-Di-cyclohexylcarbodiimid in 400 ml Aceton wird mit 12 g (0,20 Mol) Eisessig langsam versetzt. Das Gemisch wird 72 Stunden bei Raumtemperatur unter Stickstoff als Schutzgas gerührt. Sodann wird der auskristallisierte N,N'-Dicyclo-hexylharnstoff abfiltriert und das Filtrat unter vermindertem Druck eingedampft. Das zurückbleibende bernsteinfarbene Öl wird mehrmals mit Diäthyläther digeriert. Es wird ein gelber Feststoff erhalten, der aus einem Gemisch von Aceton und Diäthyläther umkristallisiert wird. Es werden weisse Kristalle vom F. (188) 195-197 °C erhalten. Nach weiterer Umkristallisation aus einem Gemisch von Methanol, Aceton und Diäthyläther wird die reine Titelverbindung in weissen Kristallen vom F. 196-199 °C erhalten.
Nach dem erfindungsgemässen Verfahren lassen sich z.B. die folgenden Verbindungen herstellen (Me = Methyl; Et = Äthyl; Ph = Phenyl):

- 632 9942PATENTANSPRÜCHE 1. Verfahren zur Herstellung von heterocyclischen Gua-nidinderivaten der allgemeinen Formel I.(OUÌ NR(IJund der pharmazeutisch annehmbaren Säureadditionssalze davon, in der n den Wert 1,2 oder 3 hat, Rt ein Wasserstoffatom, einen C^g-Alkyl-, C^-Cycloalkyl-, C3_5-Alken-2-yl-, niederen Hydroxyalkyl-, Aralkyl-oder Arylrest, R2 ein Wasserstoffatom, einen C^-Alkylrest oder einen Arylrest, R3 ein Wasserstoffatom, einen C]_8-Alkylrest oder einen Arylrest, R4 ein Wasserstoffatom, eine Methyl- oder Äthylgruppe und R5 einen C^-Alkylrest oder einen C3_7-Cyclo-alkylrest bedeutet, oder R4 und R5 zusammen mit dem Stickstoffatom, an das sie gebunden sind, einen 3- bis 7-gliedrigen gesättigten heterocyclischen Ring bilden, wobei ein 6-gliedriger Ring (a) noch ein Sauerstoff- oder Schwefel-Ringatom oder ein gegebenenfalls durch einen niederen Alkylrest, eine Phenyl- oder Benzylgruppe substituiertes Stickstoff-Ringatom enthält oder (b) der Ring durch einen niederen Alkylrest an einem Kohlenstoffatom substituiert ist, das nicht unmittelbar benachbart zum Stickstoffatom steht, das an die Carboximidamid-Funktion gebunden ist, und R einen C4-C10-Alkylrest, einen C^g-Cycloalkylrest, einen C7_10-Bicycloalkylrest, einen C 7_10-Bicycloalkenylrest, einen C9_10-Tricycloalkylrest, eine 1-Adamantylmethyl-gruppe, eine C9_I0-T ricycloalkenylgruppe, einen Phenyl-C^-alkyl- oder Naphthyl-C^-alkylrest, eine a,a-Tetramethylen-phenäthylgruppe, einen Diphenyl-C^-alkylrest, eine a-oder ß-Naphthylgruppe, einen kondensierten Diarylcycloalkenyl-rest, einen kondensierten Arylcycloalkylrest, einen Phenyl-C5_7-cycloalkylrest, einen C^-Cycloalkyl-C^-cycloalkyl-rest, eine Phenyl- oder Methylendioxyphenylgruppe, eine durch Halogenatome, niedere Alkyl- oder Alkoxyreste mono-, di- oder trisubstituierte Phenylgruppe, eine durch eine Amino-, Dimethylamino-, Methyläthylamino-, Di-äthylamino-, nieder-Alkanoylamino-, Thio-nieder-alkyl-, Sulfinyl-nieder-alkyl-, Sulfonyl-nieder-alkyl-, Trifiuorme-thyl-, Hydroxyl-, Benzyloxy-, nieder-Alkanoyloxy-, nieder-Alkanoyl- oder Nitrogruppe substituierte Phenylgruppe,eine 3-Pyridyl- oder eine durch Halogenatome, niedere Al-kyl- oder Alkoxyreste mono- oder disubstituierte 3-Pyridyl-gruppe oder einen 5gliedrigen heterocyclischen Rest bedeutet, dadurch gekennzeichnet, dass man eine Verbindimg der allgemeinen Formel:N—C=NRHZHNR4R5in der R4 und R5 obige Bedeutungen besitzen, in einem niederen aliphatischen Alkohol bei der Rückflusstemperatur umsetzt und das erhaltene Salz der Verbindung der allgemeinen Formel I in die freie Base überführt und gegebenenfalls die erhaltenen Verbindungen in die pharmazeutisch annehmbaren Säureadditionssalze davon überführt.
- 2. Verwendung der heterocyclischen Guanidinderivate der allgemeinen Formel I15(XVII)in der R' eine Äthyl- oder Methylgruppe und HZ eine Säure darstellt und Rls R2, R3 und n obige Bedeutungen besitzen, mit einem Amin der allgemeinen Formel(I)20 worin die Substituenten in Anspruch 1 definiert sind, zur Herstellung von quaternären Salzen davon, dadurch gekennzeichnet, dass man diese Verbindungen mit einem Alkylie-rungsmittel quaternisiert.25
