Description
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen N-acylsubstituierten p-Aminobenzamiden der in Patentanspruch 1 angegebenen Formel I bzw. der in Patentanspruch 3 angegebenen Formel Ia.
In den Formeln I bzw. Ia bedeuten:
X =C(Rj)oder =N—
Rj Wasserstoff, Alkyl, Halogen, Trifluormethyl, Alkoxy, Phenoxy oder Acetoxy,
Y -O-,-NH-oder-S-,
Z Cj bis Cs-Alkylen oder C2 bis C5-Alkenylen, die gegebenenfalls durch Alkyl, Alkenyl, Phenyl, Cycloalkyl, Acetyl, Amino oder Halogenphenoxy substituiert sein können,
R2 gegebenenfalls halogen-, phenyl- oder halogenphenylsubstituiertes Cj bis Q-Alkyl oder C2 bis Q-Alkenyl und n, m, p den Wert 0 oder 1.
Als Halogenatome kommen Fluor-, Chlor-, Brom- und Jodatome, vorzugsweise Chlor- und Fluoratome, in Frage. Ri kann ein-oder mehrfach vorhanden sein; wenn es nicht Wasserstoff bedeutet und nur einfach vorhanden ist, steht es vorzugsweise meist in p- oder so o-Stellung, insbesondere in p-Stellung; wenn Rt eine Trifluormethyl-gruppe darstellt, steht diese bevorzugt in m-Stellung.
Beispiele für geeignete Alkylreste, die sich als Rx eignen, sind die Methyl-, Ethyl, Propyl-, Isopropyl-, n-Butyl-, tert.-Butyl- oder n-Pentylgruppe, ferner verzweigte Pentylgruppen. Bevorzugt werden 55 Alkylreste mit 1 bis 3 C-Atomen. Die Methylgruppe ist besonders bevorzugt.
Beispiele für geeignete geradkettige oder verzweigte Alkenylen-gruppen sind die Ethenylen-, Propenylen-, Butenylen- oder Pentenylengruppen mit jeweils einer Doppelbindung. 60 Als quaternäre Verbindungen (Ia) werden insbesondere solche bevorzugt, die Halogenalkyl, z.B. Methojodid und Ethojodid oder Butylbromid enthalten.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel I ist gekennzeichnet durch die im Kennzeichen «5 von Patentanspruch 1 angegebenen Merkmale.

Das als Reaktionskomponente verwendete 4-Amino-5-chlor-2-methoxy-N-[2-(diethylamino)ethyl]benzamid ist eine unter der Freibezeichnung Metoclopramid bekannte Verbindung.
Die als Reaktionskomponente dienende Carbonsäure der Formel (II)

kann als solche oder als reaktionsfähiges Derivat verwendet werden.
Geeignete Säurederivate sind z.B. Säurechloride, Säureanhydride oder Ester. Die direkte Umsetzung der Carbonsäure der Formel II mit Metoclopramid erfolgt bevorzugt in aromatischen Lösungsmitteln oder Halogenkohlenwasserstoffen unter Erwärmung auf Rückflusstemperatur. In gleicher Weise kann die Umsetzung auch in anderen geeigneten indifferenten Lösungsmitteln oder auch lösungsmittelfrei unter Einsatz einer wasserabspaltenden Komponente, wie z.B. Phosphoroxychlorid, Phosphortrichlorid, Phosphorpenta-chlorid, Dicyclohexylcarbodiimid erfolgen.
Das Lösungsmittel wird in allen Fällen vorzugsweise in wasserfreier Form verwendet. Das Molverhältnis zwischen der Carbonsäure der Formel II und Metoclopramid im Reaktionssystem beträgt vorzugsweise 1:1 bis 1:2.
Das Metoclopramid ist eine in der Literatur bekannte Verbindung mit insbesondere antiemetischer Wirksamkeit. Darüber hinaus werden in den deutschen Offenlegungsschriften Nrn. 2330425 und 2331262 sowie in den deutschen Auslegeschriften Nrn. 1518271 und 1518310 noch neuroleptische, psycholeptische, antiarrhytmische, analgetische, spasmolytische, choleretische, sedative, antipruriginöse und antihistaminische Wirkungen beschrieben. Die akute Toxizität von Metoclopramid ist aus der deutschen Offenlegungsschrift Nr. 2330425 bekannt und mit folgenden Werten angegeben: LD50 oral Ratte 235 mg/kg LD50 i.p. Ratte 180 mg/kg LD50 i.p. Maus 242 mg/kg
Die erfindungsgemäss erhaltenen Verbindungen bieten eine wesentlich geringere Toxizität. Die Toxizität der erfindungsgemäss erhaltenen Verbindungen nach Beispiel 9,43 und 48, jeweils in Form ihrer Hydrochloride sowie von Metoclopramid Dihydrochlorid, wurde sowohl oral (p.o.) als auch intraperitoneal (i.p.) an der Maus überprüft und brachte folgende Ergebnisse:

Wie die Ergebnisse zeigen, besitzen die erfindungsgemäss erhaltenen Verbindungen sowohl oral als auch intraperitoneal eine wesentlich geringere akute Toxizität bei gleichzeitig veränderten Toxizitäts-symptomen.
Die erfindungsgemäss erhaltenen Verbindungen besitzen interessante pharmakologische Eigenschaften. Die Pharmakodynamik der Verbindungen nach Beispiel 9,43 und 48 (Hydrochloride), sowie der Referenzverbindung Metoclopramid Dihydrochlorid, wurde im pharmakologischen Screening an 54 Gruppenaktivitäten getestet. Ermittelte Aktivitäten wurden durch Sekundärtests abgesichert. Die Ergebnisse dieses Vergleichstests sind in Tabelle 1 zusammengefasst. Zusätzlich ist die ED100 bekannter gruppenspezifischer Vergleichsverbindungen angegeben.

Wie die Testergebnisse zeigen, besitzen die erfindungsgemäss erhaltenen Verbindungen im Vergleich zu Metoclopramid besondere pharmakologische Eigenschaften. So zeigen die Verbindungen nach Beispiel 40 und 45 eine sehr starke ausgeprägte antiinflammatorische Aktivität im Adjuvans Arthritismodell und im akuten Carrageneen-Versuch. Diese Hauptwirksamkeit wird noch ergänzt durch schwache antimikröbielle, sowie insbesondere durch antiallergische Eigenschaften.
Die erfindungsgemäss erhaltenen Verbindungen bieten daher wertvolle Therapeutika zur Behandlung entzündlicher und allergischer Erkrankungen, insbesondere rheumatischer und arthritischer Formen. Darüber hinaus können sie auch für Erkrankungen des Magen-Darm-Traktes verwendet werden.
Die Anwendung der entsprechenden Arzneimittel erfolgt vorzugsweise oral, z.B. in Form von Tabletten, Kapseln oder Liquida, gegebenenfalls zusammen mit üblichen pharmazeutischen Trägerstoffen und Hilfsmittel, wie z.B. Laktose, Stärke, Talk oder Ma-gnesiumstearat.
Die Verabreichung der erfindungsgemäss erhaltenen Verbindungen erfolgt, je nach dem Fall, z.B. in oralen oder rektalen Tagesdosen von 50 bis 500 mg, vorzugsweise 100 bis 200 mg, oder in parenteralen Tagesdosen von 30 bis 150 mg, vorzugsweise 50 bis 100 mg, in den üblichen pharmazeutischen Formen.
Die erfindungsgemässe Herstellung der neuen Verbindungen wird anhand folgender Beispiele beschrieben:
Beispiel 1:
6,82 g (0,075 mol) Nicotinsäure und 14,9 g (0,05 mol) Metoclopramid Base vom Fp. 148°C werden in 100 ml Chloroform gelöst. Bei Raumtemperatur werden 5,6 g Phosphoroxychlorid zugetropft und der Ansatz noch 3 h unter Rückfluss erwärmt. Nach dem Ausschütteln mit 80 ml 10%iger Natronlauge sowie anschliessend mit Wasser wird die Chloroformlösung i.v. eingedampft und der Destillationsrückstand aus Benzin (Kp. 100-140° C) umkristallisiert.
Es werden 10,4 g (51,4% d.Th.) 4-Nicotinoylamino-5-chlor-2-me-thoxy-N-[2-(diethylamino)ethyl]benzamid vom Fp. 99-100°C erhalten.
Beispiel 2:
Hydrochlorid der Verbindung gemäss Beispiel 1 : Fp. 213-214°C aus Essigester-Isopropanolmischung.
IR(KBr): 3360,3300,1690,1644cm-'.
Beispiel 3:
14,9 g (0,05 mol) Metoclopramid Base werden in 100 ml Xylol suspendiert und nach Zugabe von 46,5 g (0,15 mol) 2-Phenylbutter-säureanhydrid (Kp. 150°C/1,15 Torr) 3 h auf 120°C erhitzt. Der Ansatz wird mit verdünnter Natronlauge sowie anschliessend mit Wasser extrahiert und eingedampft. Der Destillationsrückstand wird aus Diisopropyläther umkristallisiert und ergibt 11,7 g (56,2% d.Th.) 4-(2-Phenylbutyroylamino)-5-chlor-2-methoxy-N-[2-(diethylamino)-ethyl]benzamid vom Fp. 118-119° C.

Beispiels:
14,4 g (0,075 mol) 2-Phenyl-2-ethylbuttersäure und 14,9 g (0,05 mol) Metoclopramid Base werden in 100 ml Toluol suspendiert. Unter leichtem Erwärmen auf 30-40° C werden 5,6 g Phosphoroxychlorid zugetropft und nach vollständiger Zugabe noch 3 h unter Rückfluss erhitzt. Nach dem Ausschütteln mit 10%iger Natronlauge sowie Wasser wird die organische Phase eingedampft und der Rückstand aus Benzin (Kp. 100-140°C) umkristallisiert.
Ausbeute 12,2 g (51,5% d.Th.) 4-(2-Phenyl-2-ethylbutyroyl-amino)-5-chlor-2-methoxy-N-[2-(diethylamino)ethyl]benzamid vom Fp. 113°C.
IR (KBr): 3390, 1708, 1665 cm~i Beispiel 6:
59,8 g (0,2 mol) Metoclopramid Base und 30,0 g (0,3 mol) Tri-ethylamin werden in 400 ml Chloroform gelöst und tropfenweise mit 72,0 g (0,3 mol) 2-(4-Chlorphenoxy)-2-methylpropionsäurechlorid versetzt. Der Ansatz wird noch 2 h unter Rückfluss erwärmt und anschliessend mit 100 ml 20%iger Natronlauge sowie 200 ml Wasser gewaschen. Nach dem Eindampfen wird der Destillationsrückstand aus Methanol kristallisiert, wobei 73,2 g (73,7% d.Th.) 4-[2-(4-Chlor- aus Essigester umkristallisiert, wobei 5,1 g (78,2% d. Th.) 4-[2-(4-
phenoxy)isobutyroylamino]-5-chlor-2-methoxy-N-[2-(diethylamino)-
ethyl]benzamid vom Fp. 120-121°C erhalten werden.

4,74 g (0,01 mol) des unter Beispiel 5 beschriebenen Amids werden unter leichtem Erwärmen in 30 ml Acetonitril gelöst und mit 1,42 g (0,01 mol) Methyljodid versetzt. Der Ansatz wird noch 2 h auf 50° C erwärmt. Das erhaltene Kristallisat wird abgesaugt und aus we-m nig absolutem Äthanol umkristallisiert. Es werden 4,7 g (76,3% d.Th.) 4-(2-Phenyl-2-ethylbutyroylamino)-5-chlor-2-methoxy-N-[2-diethylamino)ethyl]benzamidmethojodid vom Fp. 150°C erhalten.
Beispiel 11:
65 4,9 g (0,01 mol) des unter Beispiel 6 beschriebenen Amids und 1,56 g (0,01 mol) Ethyljodid werden in 30 ml Aceton 3 h unter Rückfluss erhitzt. Nach dem Eindampfen wird der Destillationsrückstand

Entsprechend dem Verfahren nach Beispiel 1 sowie nach
Beispiel 10 im Falle der quartären Amide wurden weitere Verbindungen hergestellt, die zusammen mit den Verbindungen der vorgehenden Beispiele 1-11 in der nachstehenden Tabelle tabellarisch erfasst sind.

PATENTANSPRÜCHE 1. Verfahren zur Herstellung von neuen N-acylsubstituierten p-Aminobenzamiden der Formel (I)

worin X=C (Rj)— oder =N—, R! Wasserstoff, Alkyl, Halogen, Trifluormethyl, Alkoxy, Phenoxy oder Acetoxy, Y — O—, — NH — oder — S—, Z Cj bis C5-Alkylen oder C2 bis C5-Alkenylen, die gegebenenfalls durch Alkyl, Alkenyl, Phenyl, Cycloalkyl, Acetyl, Amino oder Halogenphenoxy substituiert sein können, n, m, p den Wert 0 oder 1 bedeuten und im Falle p = 1 der Salze mit pharmazeutisch verträglichen Säureresten, dadurch gekennzeichnet, dass man eine entsprechende Carbonsäure der Formel (II)

15 oder ein reaktionsfähiges Derivat einer solchen Carbonsäure mit 4-Amino-5-chlor-2-methoxy-N-[2-(diethylamino)ethyl]benzamid zur Reaktion bringt, und die erhaltene Verbindung der Formel (I), in der p gleich 0 ist, gegebenenfalls mit einer den pharmazeutisch verträglichen Säurerest liefernden Säure in das entsprechende Salz umwan-20 delt.
2. Nach dem Verfahren gemäss Anspruch 1 hergestellte Verbindung der Formel (I).
25 3. Verfahren zur Herstellung von neuen N-acylsubstituierten p-Aminobenzamiden der Formel (Ia)
(Ia)
worin X=C (Rj)— oder =N—, R! Wasserstoif, Alkyl, Halogen,
Trifluormethyl, Alkoxy, Phenoxy oder Acetoxy, Y —O—, — NH—
oder — S—, Z Q bis C5-Alkylen oder C2 bis C5-Alkenylen, die gege- 40
benenfalls durch Alkyl, Alkenyl, Phenyl, Cycloalkyl, Acetyl, Amino oder ein reaktionsfähiges Derivat einer solchen Carbonsäure mit 4-
oder Halogenphenoxy substituiert sein können, R2 gegebenenfalls Amino-5-chlor-2-methoxy-N-[2-(diethylamino)ethyl]benzamid zur halogen-, phenyl- oder halogenphenylsubstituiertes Q bis C4-Alkyl Reaktion bringt und die erhaltene Verbindung zur Einführung des oder C2 bis C4-Alkenyl und n, m den Wert 0 oder 1 bedeuten, da- Restes R2 mit einem entsprechenden Quaternisierungsmittel umsetzt,
durch gekennzeichnet, dass man eine entsprechende Carbonsäure der 45 4. Nach dem Verfahren gemäss Anspruch 3 hergestellte Verbin-
