Die vorliegende Erfindung betrifft ein Verfahren zur Her-30 Stellung von neuen in 2-Position substituierten 1,4-Dihydropyridin-Derivaten, die als Arzneimittel, insbesondere als kreislaufbeeinflussende Mittel verwendet werden können.
Es ist bereits bekannt geworden, dass man 2,6-Dimethyl-4-phenyl-1,4-dihydropyridin-3,5-dicarbonsäurediäthylester 35 erhält, wenn man Benzylidenacetessigsäureäthylester mit ß-Amino-crotonsäure-äthylester oder Acetessigsäureäthyl-ester und Ammoniak umsetzt (Knoevenagel, Ber. dtsch. ehem. Ges. 31,743 [1898]). Weiterhin ist bekannt, dass bestimmte 1,4-Dihydropyridine interessante pharmakolo-40 gische Eigenschaften aufweisen (F. Bossert, W. Vater, Naturwissenschaften 58,578 [1971]).
Die erfmdungsgemäss herstellbaren Verbindungen weisen die folgende Formel auf

Die erhaltenen Verbindungen können in ihre pharmazeutisch unbedenklichen Säureadditionssalze überführt werden.
Es wurde gefunden, dass man die neuen in 2-Position substituierten Verbindungen der Formel I erfindungsgemäss erhält, wenn man Aldehyde der Formel

Bei der Kombination der Formeln (II) und (IX) und bei der Kombination der Formeln (V) und (VIII) entstehen 20 jeweils nur symmetrische Dihydropyridinester.
Eine spezielle Ausführungsform des erfindungsgemässen Verfahrens für die Herstellung symmetrischer Endverbindungen, d.h. solcher Endverbindungen, in denen R4 die Bedeutung von A-R1 hat und Y und Z identisch ist, besteht 25 darin, ohne Isolierung der während der Reaktion entstehenden Enamine direkt zwei Teile Ketoverbindung der Formel (II) bzw. (V) mit einem Teil eines Amins oder Amin-salzes der Formel (III) und mit einem Teil Aldehyd der Formel (VII) umzusetzen.
30 Die Herstellung der Salze kann in an sich bekannter Weise durch Umsetzung der erfindungsgemäss erhaltenen Verbindungen mit geeigneten Säuren erfolgen.
Die erfindungsgemäss hergestellten, in 2-Position substituierten 1,4-Dihydro-pyridinderivate zeigen deutliche pharma-35 kologische Wirkungen, insbesondere zeichnen sie sich durch eine starke gefässbeeinflussende Wirkung aus. Aufgrund ihrer neuartigen und vielseitigen Wirkungsweise stellen die genannten Verbindungen eine Bereicherung der Pharmazie dar.
40 Verwendet man z.B. 4-Äthylthiobenzaldehyd, ß-Amino-y-methylthiocrotonsäurepropylester und Propionylessigsäure-butylester als Ausgangskomponenten, so kann der Reaktionsablauf durch das folgende Formelschema wiedergegeben werden:

Die Ketoverbindungen der Formeln (II) und (V) sowie die entsprechenden Enamine der Formeln (VIII) und (IX) sind teilweise bereits bekannt oder können nach allgemein bekannten Verfahren hergestellt werden (vgl. B. Johnson und H. Chesnoff, J.A.C.S. 36,1774 [1914]; US-Patentschrift 2.351.366; A.C. Cope, J.A.C.S. 67,1017 [1945]).

Die erfindungsgemäss erhältlichen Salze der 1,4-Dihydro-pyridinderivate können nach allgemein bekannten Methoden, z.B. durch Auflösen der Base in Äther und Ver-20 setzen der Lösung mit geeigneten Säuren hergestellt werden.
Als anorganische und organische Säuren, die mit den Dihydropyridinen der Formel (I) bevorzugt physiologisch verträgliche Säureadditionssalze bilden, seien beispielhaft genannt: Halogenwasserstoffsäuren wie Chlor- und Brom-25 wasserstoffsäure, insbesondere Chlorwasserstoffsäure, Phosphorsäuren, Schwefelsäure, Salpetersäure, mono- und bifunktionelle Carbonsäuren und Hydroxycarbonsäuren wie Essigsäure, Maleinsäure, Bernsteinsäure, Fumarsäure, Weinsäure, Zitronensäure, Salicylsäure, Sorbinsäure, Milchsäure, 30 1,5-Naphthalindicarbonsäureund Naphthalindisulfonsäure.
Als Verdünnungsmittel kommen beim erfindungsge-mässen Verfahren bevorzugt Wasser und alle inerten organischen Lösungmittel in Frage. Hierzu gehören vorzugsweise Alkohole, z.B. niedere Alkylalkohole mit vorzugsweise 1 bis 4 35 Kohlenstoffatomen, wie Äthanyl, Methanol, Isopropanol, Äther, z.B. niedere Dialkyläther (vorzugsweise 3 bis 5 Kohlenstoffatome), wie Diäthyläther oder Ringäther wie Tetra-hydrofuran, Dioxan, niedere aliphatische Carbonsäuren (vorzugsweise 2 bis 5 Kohlenstoffatome), wie Essigsäure, 40 Propionsäure, niedere Dialkylformamide (vorzugsweise 1 oder 2 Kohlenstoffatome je Alkylgruppe), wie Dimethylfor-mamid, niedere Alkylnitrile (vorzugsweise 2 bis 4 Kohlenstoffatome), wie Acetonitril, Dimethylsulfoxid, flüssige heteroaromatische Basen wie Pyridin, sowie Gemische dieser 45 Lösungsmittel einschliesslich Wasser untereinander.
Die Reaktionstemperaturen können in einem grösseren Bereich variiert werden. Im allgemeinen arbeitet man zwischen etwa 20 und etwas 150°C, vorzugsweise zwischen 50 und 100°C, insbesondere bei der Siedetemperatur des verso wendeten Lösungsmittels.
Die Umsetzung kann bei Normaldruck, aber auch bei erhöhtem Druck durchgeführt werden. Im allgemeinen arbeitet man bei Normaldruck.
Bei der Durchführung des erfindungsgemässen Verfahrens 55 werden die an der Reaktion beteiligten Ausgangsstoffe vorzugsweise jeweils etwa in molaren Mengen eingesetzt. Die Molverhältnisse können jedoch über einen weiten Bereich variiert werden, ohne dass das Ergebnis nachteilig beeinflusst wird.

Die therapeutisch wirksamen Verbindungen sollen in den oben aufgeführten pharmazeutischen Zubereitungen vorzugsweise in einer Konzentration von etwa 0,1 bis 99,5, vorzugsweise von etwa 0,5 bis 95 Gewichtsprozent der Gesamtmischung vorhanden sein.
Die oben aufgeführten pharmazeutischen Zubereitungen können ausser Verbindungen der Formel (I) und/oder deren Salzen auch andere pharmazeutische Wirkstoffe enthalten.
Die Herstellung der oben aufgeführten pharmazeutischen Zubereitungen kann in üblicher Weise nach bekannten Methoden, z.B. durch Mischen des oder der Wirkstoffe mit dem oder den Trägerstoffen, erfolgen.
Die Wirkstoffe oder die pharmazeutischen Zubereitungen können vorzugsweise oral, parenteral und/oder rectal, vorzugsweise oral und parenteral, insbesondere perlingual und intravenös appliziert werden.
Im allgemeinen hat es sich als vorteilhaft erwiesen, den oder die Wirkstoffe bei parenteraler (intravnöser) Applikation in Mengen von etwa 0,01 bis etwa 50, vorzugsweise von 0,1 bis 5 mg/kg Körpergewicht je 24 Stunden und bei oraler Applikation in Mengen von etwa 0,1 bis etwa 100, vorzugsweise 1 bis 30 mg/kg Körpergewicht je 24 Stunden, gegebenenfalls in Form mehrerer Einzelgaben zur Erzielung der gewünschten Ergebnisse zu verabreichen. Eine Einzelgabe enthält den oder die Wirkstoffe, vorzugsweise in Mengen von etwa 0,005 bis 50, insbesondere 0,5 bis 20 mg/kg Körpergewicht. Es kann jedoch erforderlich sein, von den genannten Dosierungen abzuweichen, und zwar in Abhängigkeit von der Art und dem Körpergewicht des zu behandelnden Objektes, der Art und der Schwere der Erkrankung, der Art der Zubereitung und der Applikation des Arzneimittels sowie dem Zeitraum bzw. Intervall, innerhalb welchem die Verabreichung erfolgt. So kann es in einigen Fällen ausreichend sein, mit weniger als der oben genannten Menge Wirkstoff auszukommen, während in anderen Fällen die oben angeführte Wirkstoffmenge überschritten werden muss. Die Festlegung der jeweils erforderlichen optimalen Dosierung und Applikationsart der Wirkstoffe kann durch jeden Fachmann aufgrund seines Fachwissens leicht erfolgen.

Man erhitzt die Lösung von 7,5 g 2-Nitrobenzaldehyd, 17,6 g y-Methylmercaptoacetessigsäureäthylester und 4 g Methylamin-Chlorhydrat in 30 ccm Pyridin 7 Std. auf90-100°C, gibt in Eiswasser, dekantiert die wässrige Phase ab und nimmt das Öl in Äther auf. Anschliessend wird die ätherische Lösung pyridinfrei gewaschen, über Natriumsulfat getrocknet und eingedampft.
Öl, Ausbeute: 90%.

Man erhitzt 5,3 g Benzaldehyd, 17,6 g y-Methylmercapto-acetessigsäureäthylester und 4 g Methylamin-Chlorhydrat in 70 ccm Pyridin 6 Stunden auf 90-100°C, gibt in Eiswasser und nimmt in Äther auf. Die ätherische Lösung wird pyridin-20 frei gewaschen, über Natriumsulfat getrocknet und eingedampft.

ester in 250 ccm Äthanol 3 x 24 Stunden am Rückfluss zum Sieden, lässt unter Rühren erkalten und erhält nach dem Absaugen hellgelbe Kristalle vom Fp. 135-136°C, Ausbeute: 55%.
Beispiel 43
2-Phthalimidomethyl-6-methyl-4-(2'-methylmercapto-phenyl)-1,4-dihydropyridin-3,5-dicarbonsäurediäthylester

7,5 g 4-Nitrobenzaldehyd, 14 g y-Phthalimidoacetessigsäure-äthylester und 6,5 g ß-Aminocrotonsäureäthylester werden 50 Stunden in 80 ccm Äthanol zum Sieden erhitzt.
Man lässt anschliessend unter Rühren erkalten, saugt ab. s Gelbe Kristalle vom Fp. 152-155°c,
Ausbeute: 45%.

Die Lösung von 10 g 2-Methylmercapto-benzaldehyd (77%), 14 g Phthalimidoacetessigsäureäthylester und 6,5 g ß-Amino-crotonsäureäthylester in 100 ccm Äthanol wird 2 x 24 Stunden am Rückfluss erhitzt, und nach anschliessendem Kühlen der Niederschlag abgesaugt.
Hellgelbe Kristalle, Ausbeute: 35%.

3,5 g 2-Chlorbenzaldehyd, 16 g 5-Phthalimidopropionyîes-sigsäureäthylester (Fp. 90°) und 6 ccm Ammoniak werden in 40 ccm Äthanol 70 Stunden zum Sieden erhitzt. Nach Abkühlen und Absaugen erhält man gelbe Kristalle vom Fp. 170°C,

Man erhitzt die Lösung von 5,2 ccm Pyridin-4-aldehyd, 14,0 g Phthalimidoacetessigsäureäthylester und 6,5 g ß-Ami-nocrotonsäureäthylester in 80 ccm Äthanol 24 Stunden am Rückfluss und erhält nach Kühlen und Absaugen weisse Kristalle vom Fp. 205-207°C (Äthanol),
Ausbeute: 65%.

Man erhitzt 13 g 2-Chlorbenzaldehyd, 28 g Phthalimidoacet-essigsäureäthylester und 13 g ß-Aminocrotonsäureäthylester in 250 ccm Äthanol ca. 70 Stunden am Rückfluss zum Sieden, lässt unter Rühren erkalten und erhält nach dem Absaugen hellgelbe Kristalle vom Fp. 174-176°C; Ausbeute: 90%
