Description
Aufgabe der Erfindung war es daher, ein Verfahren zur Herstellung einer Klasse von Verbindungen mit entsprechender Aktivität aufzufinden, die besser verteilt werden, jedoch nach oraler Verabreichung nicht toxischer sind. Die Aufgabe wird durch die Erfindung gelöst.
Gegenstand der Erfindung ist somit das im Patentanspruch 1 definierte Verfahren.
In der Formel II kann der Alkylenrest X geradkettig oder verzweigt sein, bevorzugt ist ein geradkettiger Rest, insbesondere ein Rest der Formel -(CH2)„-, in der n eine ganze Zahl von 1 bis 6 ist;

Bevorzugte Reste der Formel a sind Reste der Formel b

in der R3 ein Wasserstoff-, Fluor-, Chlor- oder Bromatom, ein Methyl- oder Methoxyrest ist.
Für R3 in Formel b besonders geeignet ist das Wasserstoff*, Chlor- oder Bromatom, bevorzugt ist das Wasserstoffatom.
Die Verbindungen der Formel II können Additionssalze bilden, z.B. mit pharmakologisch verträglichen anorganischen oder organischen Säuren, wie mit Chlorwasserstoffsäure, o-Phosphorsäure, Essigsäure, Bernsteinsäure, Milchsäure, Zitronensäure, Fumarsäure und Weinsäure.
Für Z in den Ausgangsverbindungen geeignet ist das Chlor-, Brom- oder Jodatom, die Methylsulfonyl- oder To-luolsulfonylgruppe.
Geeignete basische Salze der Verbindungen der Formeln IV oder V sind Alkalimetallsalze, wie das Natriumsalz.
Die erfindungsgemässen Reaktionen werden im allgemeinen in einem relativ polaren organischen Lösungsmittel durchgeführt, wie Dimethylformamid, Dimethylsulfoxid oder Acetonitril, bei nicht extremen Temperaturen, wie —20 bis 180°C, insbesondere 10 bis 100°C, z.B. 25 bis 60°C.
Die Verbindungen der Formel II oder deren Salze eignen sich als Wirkstoffe und ergeben, in Kombination mit üblichen pharmakologisch verträglichen Trägerstoffen und/ oder Verdünnungsmitteln, Arzneipräparate mit antimikro-bieller Aktivität.
Derartige Arzneipräparate können oral verabreicht oder injiziert werden, die orale Verabreichung ist bevorzugt.
Diese Arzneipräparate können in herkömmlicher Weise konfektioniert werden, z.B. gemäss der US-PS 3 723 429.
Besonders geeignet sind Tabletten, Kapseln oder andere Einzeldosisformen, die 5 bis 500 mg Wirkstoff enthalten und ein- bis mehrmals täglich verabreicht werden, um eine tägliche Dosis von 20 bis 1000 mg, insbesondere 50 bis 500 mg, für einen 70 kg schweren Patienten zu erreichen.
Solche Arzneipräparate sind für die Behandlung von Malaria und/oder bakteriellen Infektionen geeignet und sind besonders wertvoll, da sie eine 3ehandlung von Ma-
aria-Infektionen möglich machen, die gegenüber anderen Antimalaria-Mitteln resistent sind. Die guten oralen Absorptionseigenschaften sind ein weiterer Vorteil, da sie ihre Verwendung erleichtern.
5 Die beschriebenen Arzneipräparate kann man aber auch für die Behandlung von bakteriellen Infektionen verwenden, da sie antibakterielle Aktivität besitzen, z.B. gegen gram-positive Bakterien, wie Staphylococcus aureus, und gegen gram-negative Bakterien, wie Escherichia coli und io Proteus mirabilis.
Gegebenenfalls können solche Arzneipräparate noch ein weiteres antibakterielles Mittel enthalten, z.B. ein Sulfonamid, wie Sulfamethoxazol.
Die Ausgangsverbindung der Formel V, worin R2 Was-15 serstoff bedeutet, kann gemäss dem Verfahren von R. Hull, «J. Chem. Soc.» 1965, Seite 2033, hergestellt werden.
Die Beispiele erläutern die Erfindung.

Beispiel 1
2,4-Diamino-6-methyl-5-(4-chlor-2-cyclohexylphenoxypro-pyloxy)-pyrimidin
Eine Lösung von 0,8 g Natrium und 30 ml Äthanol wird mit 2,13 g 2,4-Diamino-6-methyl-5-hydroxypyrimidin-dihy-25 drochlorid versetzt, 1 Stunde lang bei Zimmertemperatur gerührt und dann portionsweise mit 3,32 g 4-Chlor-2-cyclo-hexylphenoxypropylbromid versetzt. Das Gemisch wird über Nacht gerührt, dann erwärmt und bei 50°C 24 Stunden lang gehalten, abgekühlt und filtriert. Das Filtrat wird ein-30 gedampft, der Rückstand wird mit Aceton behandelt und filtriert. Das Rohprodukt wird in Äthanol gelöst und mit einigen Tropfen konzentrierter Salzsäure versetzt, wobei das Produkt als weisser Feststoff auskristallisiert. Das Produkt wird aus Methanol/Äthanol umkristallisiert, Fp. 246 bis 35 247°C. Ausbeute: 1,65 g (40%). Die Analyse zeigt, dass das Produkt als Monohydrochlorid, möglicherweise als Mo-nohydrat, vorliegt.

Claims (2)
- 6373852PATENTANSPRUCH Verfahren zur Herstellung einer Verbindung der Formel IIAr - Y - X - 0(II)worin R Wasserstoff, Methyl oder Äthyl; X Alkylen mit 1-10 C-Atomen, Y Sauerstoff, Schwefel oder eine Bindung und Ar Aryl bedeuten und von deren pharmakologisch verträglichen Salzen, dadurch gekennzeichnet, dass man eine Verbindung der Formel IVfür Y geeignet ist das Sauerstoffatom;Ar bedeutet zweckmässigerweise einen Naphthyl-, An-thranyl-, Phenanthryl- oder Phenylrest, wobei letzterer mit 1 bis 5 Substituenten in Form von Fluor-, Chlor- oder s Bromatomen, niederen Alkoxy-, Acyloxy-, Alkyl- oder Alkenylresten, Cycloalkyl- oder Cycloalkenylresten mit 5 bis 6 Kohlenstoffatomen oder niederen Alkylthioresten substituiert sein kann; und der Ausdruck «niederer» bedeutet einen Rest mit 1 bis io 6 Kohlenstoffatomen.Besonders geeignete, erfindungsgemäss herstellbare Verbindungen sind Verbindungen der Formel III und deren pharmakologisch verträgliche Salze15Ar—R1mit einer Verbindung der Formel V(IV)Ar - 0 - (CH2)m - 020^ NH2 (III)(V)umsetzt, wobei in den Formeln IV und V Ar und R wie oben definiert sind, entweder R1 Hydroxy und R2 die Gruppe Z-X- oder R1 die Gruppe -Y-X-Z und R2 Wasserstoff bedeutet, worin X und Y die oben genannten Bedeutungen haben, Z ein durch einen nukleophilen Rest leicht verdrängbarer Rest ist, und die Ausgangsstoffe, die eine Hy-droxygruppe aufweisen, ebenfalls als basische Salze eingesetzt werden können, und die erhaltenen Verbindungen gegebenenfalls in ihre pharmakologisch verträglichen Salze überführt.In der US-PS 3 723 429 sind unter anderem Verbindungen der Formel I beschrieben in der R und Ar wie in Formel II definiert sind und m 1, 25 2, 3 oder 4 bedeutet.Für Ar bevorzugt sind der Naphthylrest oder ein Phenylrest, der gegebenenfalls mit einem oder mehreren Chloroder Bromatomen, niederen Alkyl-, Alkoxy- oder Alkenylresten, Cycloalkyl- oder Cycloalkenylresten mit 5 bis 6 Koh-30 lenstoffatomen substituiert ist.Besonders geeignet sind Verbindungen der Formeln II und III, in denen R das Wasserstoffatom oder der Methylrest ist.Bevorzugt sind Verbindungen der Formeln II und III, 35 in denen Ar ein Phenylrest, ein mono-, di- oder trisubsti-tuierter Phenylrest ist, insbesondere ein Phenylrest, der mit einem Cyclohexyl- oder Cyclopentylrest und gegebenenfalls mit weiteren 1 oder 2 Gruppen substituiert ist.Besonders bevorzugt sind Verbindungen der Formel II 40 und III, in denen Ar ein Rest der Formel a ist45B
- -0.1.o/XCDCE, CH, 5 350in der A ein divalenter aliphatischer Rest und B ein gegebenenfalls substituierter Kohlenwasserstoffrest ist. Diese Verbindungen haben antimikrobielle Aktivität, sie werden jedoch im Körper von Säugetieren nach oraler Verabreichung nicht besonders gut verteilt.
