Description
L'invention a pour objet un procédé de préparation du diméthyl-2,3-pentène-4-al tel que défini à la revendication 1.
Une utilisation particulière du diméthyl-2,3-pentène-4-al obtenu par ce procédé est celle pour la synthèse du méthyl-2-sec.-butyI-2-propanediol-1,3 que l'on peut transformer en mébutamate (dicarba-mate de méthyl-2-sec.-butyl-2-propanediol-l,3) par réaction avec le phosgène puis traitement par l'ammoniac. Le mébutamate est utile comme dépresseur du système nerveux central ou comme antihyper-tenseur. Plus particulièrement, l'invention concerne un nouveau procédé selon lequel on fait réagir l'alcool crotylique avec le chlorure d'allyle pour obtenir l'éther d'allyle et de crotyle que l'on transpose ensuite en diméthyl-2,3-pentène-4-al. On transforme l'aldéhyde en méthyl-2-sec.-butyl-2-propanediol-l,3 puis en mébutamate selon des procédés connus.
Le mébutamate que l'on connaît depuis plus de 20 ans a été décrit pour la première fois dans le brevet US N° 2878280.
On prépare le mébutamate à partir du méthyl-2-sec.-butyl-2-propanediol-3 (mébutamatediol). Bien que l'on dispose de nombreux procédés pour préparer le propanediol-l,3-2,2-disubstitué désiré tels que ceux décrits dans les brevets US Nos 3359324 et 2937119, les brevets GB N<* 803059,991978 et 1015911, le brevet BE N° 615484, le brevet FR N° 1377140 et de nombreuses autres publications telles que celle de H. Yale («J. Am. Chem. Soc.», 72, 3710-6,1950), les procédés actuels présentent des défauts tels qu'un rendement global faible, un coût de production élevé ou l'impossibilité de se procurer l'intermédiaire nécessaire. On recherche donc une nouvelle voie d'obtention du méthyl-2-sec.-butyl-2-propanediol-l,3.
Par réaction de l'alcool crotylique et du chlorure d'allyle, on produit un mélange des isomères de l'éther d'allyle et de crotyle et le chauffage de l'éther crotylique pour obtenir le diméthyl-2,3-pentène-4-al. Pour transformer ensuite l'aldéhyde en méthyl-2 sec.-butyl-2-propanediol-1,3, on le fait réagir avec du formaldéhyde pour produire le diméthyl-2,3-hydroxyméthyl-2-pentène-4-ol que l'on hydrogène en présence d'un catalyseur pour produire le méthyl-2-sec.-butyl-2-propanediol-1,3.
Selon une variante, si on le désire, on hydrogène partiellement le diméthyl-2,3-pentène-4-al pour produire le diméthyl-2,3-pentanal que l'on fait ensuite réagir avec le formaldéhyde en présence d'un catalyseur basique pour produire le méthyl-2-sec.-butyl-2-propane-diol-1,3.
On peut également, selon une autre variante, hydrogéner totalement le diméthyl-2,3-pentène-4-al dans des conditions d'hydrogénation telles que la liaison oléfinique soit saturée et que la fonction aldéhyde soit réduite en fonction alcool pour produire le diméthyl-2,3-pentanol que l'on déshydrogène ensuite pour produire le diméthyl-2,3-pentanal que l'on fait réagir avec le formaldéhyde pour produire le mébutamatediol.
On peut utiliser les réactions par lesquelles on produit le mébutamatediol par la série de schémas réactionnels suivants. On combine tout d'abord l'alcool crotylique et le chlorure d'allyle en mélange homogène et on les condense pour produire l'oxyde d'allyle et de crotyle :

On effectue la réaction ci-dessus en présence d'un agent de condensation tel que les métaux du groupe i de la classification périodique, l'hydrure de sodium, l'hydroxyde de potassium, le carbonate de potassium et similaires en des quantités de l'ordre de 1 mol environ par équivalent molaire, dans le cas de l'hydrure de sodium et du sodium métallique, à un grand excès dans le cas du carbonate de potassium. Les quantités d'alcool crotylique utilisées par rapport au chlorure d'allyle peuvent varier d'environ 1,0 mol à environ 1,3 mol de chlorure d'allyle par mole d'alcool crotylique.

On effectue la réaction à une température de l'ordre d'environ 50° c et environ 80° c. La gamme des températures n'a pas de limite particulière et ne dépend que de la vitesse de réaction désirée.
Le solvant utilisé est le diméthylformamide, le diméthylacét-amide, le toluène, le benzène ou un autre composé semblable constituant un solvant des composants réactionnels ne gênant pas la réaction.
On transforme ensuite l'éther d'allyle et de crotyle en diméthyl-2,3-pentène-4-al selon une transposition catalysée par un métal de transition correspondant au schéma réactionnel suivant:

De façon surprenante, on peut effectuer la réaction de transposition avec des températures de la vapeur relativement basses de 125 à • 136°C et à la pression ordinaire. Les catalyseurs convenant à cette réaction sont les métaux de transition tels que le rubidium, le molybdène, le ruthénium et le palladium, de préférence le ruthénium bivalent, comme décrit par Reuter et Solomon, («J. Org. Chem.», vol. 42, N° 21,1977, pp. 3360-3364).
La transformation du diméthyl-2,3-pentène-4-al en mébutamatediol est décrite dans le brevet US N° 3359324.
La séquence de ces diverses opérations est illustrée par les exemples suivants.
Exemple 1:
Oxyde d'allyle et de crotyle
On introduit 72 g d'hydrure de sodium à 50% en dispersion dans l'huile dans un ballon à trois cols à fond rond de 11 contenant 100 ml de diméthylformamide. On ajoute goutte à goutte en agitant

un mélange homogène de 108 g d'alcool crotylique et de 143 g de chlorure d'allyle. On maintient la température réactionnelle entre 50 et 60° C et on effectue l'addition en 1 h. On agite le mélange réac-tionnel pendant 1 h puis on décompose avec 300 ml d'eau. On ajuste le pH du mélange entre 4 et 5 avec de l'acide acétique glacial et on s sépare la couche aqueuse et la couche organique. On extrait la couche aqueuse avec 75 ml de pentane et on combine l'extrait à la phase organique. On lave deux fois la phase organique avec 100 ml d'eau puis on sèche sur sulfate de sodium. On filtre le mélange puis on le distille pour obtenir 122 g d'oxyde d'allyle et de crotyle. P. E. io 125-127° C.
Exemple 2:
Dimêthyl-2,3-pentène-4-al
On introduit 122 g d'oxyde d'allyle et de crotyle dans un ballon à 15 trois cols à fond rond de 300 ml. On ajoute 10 mg de dichlorure de tris(triphénylphosphine)ruthénium et on porte le mélange à reflux pendant une nuit avec une température de la vapeur de 125 à 136°C. On peut utiliser le produit obtenu qui est le diméthyl-2,3-pentène-4-al, ayant un point d'ébullition de 133-134°C, tel quel ou après distil- 2o lation.
Exemple 3:
Méthyl-2-sec.-butyl-2-propanediol-l,3
Dans un ballon à fond rond de 250 ml, on ajoute 50 g de dimé- 25
thyl-2,3-pentène-4-al à 100 ml de formaldéhyde à 37%. On introduit goutte à goutte dans le mélange 70 g d'une solution à 50% en poids d'hydroxyde de sodium en agitant et en maintenant la température entre 35 et 40° C avec un bain glacé. Lorsque l'addition est achevée, on agite le mélange pendant 'A h à la température ordinaire et on sépare les deux couches. On extrait la couche aqueuse avec 75 ml d'acétate d'éthyle et on combine l'extrait à la couche organique. On lave deux fois la couche organique avec 100 ml d'eau. On place la couche organique dans un appareil d'hydrogénation de Parr et on dilue à 150 ml avec du méthanol. On introduit 1 g de nickel de Raney humide comme catalyseur et on recuit le mélange sous une pression de 3,4 bar jusqu'à ce qu'il n'y ait plus d'absorption d'hydrogène. On sépare le catalyseur par filtration, on concentre le filtrat et on le distille. On obtient 45,4 g de méthyl-2-sec.-butyl-2-propane-diol-1,3. P. E. 139-140° C/10 mmHg; indice de réfraction ri2ß =1,4610.
On produit l'agent antihypertensif qu'est le mébutamate selon des procédés bien connus qui consistent, par exemple, à faire barboter du phosgène gazeux à travers un mélange liquide contenant du mébutamatediol jusqu'à ce qu'au moins 1 mol de phosgène ait été consommée par équivalent d'hydroxyle et que le dichloroformiate de méthyl-2-sec.-butyl-2-propanediol-l,3 soit produit. On fait réagir ce composé avec l'ammoniac pour produire le mébutamate.
Selon une variante, on peut faire réagir l'ammoniac et le phosgène gazeux avec le diol dans un solvant anhydre pour produire le mébutamate.

Claims (5)
1. Procédé de préparation du diméthyl-2,3-pentène-4-al, caractérisé en ce qu'il comprend le chauffage de l'éther d'allyle et de crotyle à une température de 125 à 136°C à la pression ordinaire en présence d'un catalyseur à base d'un métal de transition.
2. Procédé selon la revendication 1, caractérisé en ce que le catalyseur est le dichlorure de tris(triphénylphosphine)ruthénium, de formule C54H4.5P3C12Ru.

REVENDICATIONS
3. Utilisation du diméthyl-2,3-pentène-4-al obtenu par le procédé selon l'une des revendications 1 ou 2 pour la préparation du méthyl-2-sec.-butyl-2-propanediol-l,3, caractérisée en ce qu'on fait réagir du diméthyl-2,3-pentène-4-al avec du formaldéhyde et qu'on hydrogène le composé résultant en présence d'un catalyseur.
4. Utilisation du diméthyl-2,3-pentène-4-al obtenu par le procédé selon l'une des revendications 1 ou 2 pour la préparation du méthyl-2-sec.-butyl-2-propanediol-l,3, caractérisée en ce qu'on hydrogène partiellement le diméthyl-2,3-pentène-4-al, ce qui conduit au diméthyl-2,3-pentanal, que l'on fait ensuite réagir avec le formaldéhyde en présence d'un catalyseur basique.
5. Utilisation du diméthyl-2,3-pentène-4-al obtenu par le procédé selon l'une des revendications 1 ou 2 pour la préparation du méthyl-2-sec.-butyl-2-propanediol-l,3, caractérisée en ce qu'on hydrogène le diméthyl-2,3-pentène-4-al dans des conditions d'hydrogénation assurant la saturation de la liaison oléfinique et la réduction de la fonction aldéhyde en fonction alcool, qu'on déshydrogène le diméthyl-2,3-pentanol obtenu pour produire le diméthyl-2,3-pent-anal et qu'on fait réagir ce dernier dérivé avec du formaldéhyde.
