Description
Die vorliegende Erfindung betrifft neue Retinoide, deren Herstellung und Verwendung und pharmazeutische Präparate auf der Basis dieser Retinoide. Die neuen Retinoide sind Verbindungen der allgemeinen Formel worin R einen esterartig gebundenen Rest eines Zuckers 25 oder einen amidartig gebundenen Rest eines Amino-Zuckers oder eines Derivates eines solchen Zuckers, nämlich eines O-oder N-Acylats, Glycosids, Aminoalkylglykosids, Deoxy-zuckers, Acetals oder einer Uronsäure, darstellt und n 1 oder 2 ist.
30 Die in den Verbindungen der Formel I in Form des Restes R enthaltenen Zucker sind insbesondere Monosaccharide und Aminozucker wie Glucose, Fructose, Mannose, Galaktose, Ribose, Xylose, Glucosamin, Fructosamin, Manno-samin und Galaktosamin; und Disaccharide wie Saccharose, 35 Lactose, Maltose und Trehalose. Vorzugsweise liegen die Zucker in der natürlichen (D)-Konfiguration vor. Beispiele von Acylaten sind Zucker oder Aminozucker, in denen eine oder mehrere Hydroxygruppen oder die Aminogruppe einen Acylrest, z.B. einen Q 6-Alkanoylrest wie den Acetylrest tra-40 gen, wie 2,3,4-tri-O-Acetylglucose. Beispiele von Glycosiden sind Alkylglycoside, wie Methylglucosid, von Aminoalkyl-glykoside [Aminoäthyl]-glucosid, [Aminoäthyl]-2-deoxy-2-aminoglucosid und [Aminoäthyl]- N-acetyl- tri-O-acetyl-2-deoxy- 2-aminoglucosid.
45 Die Doppelbindungen in der Polyenkette, d.h. dem 9-(4-Methoxy-2,3,6-trimethylphenyl)- 3,7-dimethyl- 2,4,6,8-nona-tetraenylrest in den Verbindungen der Formel I können cis-oder trans-Konfiguration haben. Bevorzugt sind Verbindungen der Formel I, in denen die Polyenkette all-trans-Konfi-50 guration hat.
Die Verbindungen der Formel I können erfindungsge-mäss dadurch hergestellt werden, dass man einen Zucker oder einen Aminozucker oder ein Derivat eines solchen Zuk-kers, der mindestens eine freie Hydroxy- oder Aminogruppe 55 enthält, mit einem reaktiven Derivat der 9-(4-Methoxy-2,3,6-trimethylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetraensäu-re umsetzt und gewünschtenfalls im Reaktionsprodukt enthaltene reaktionsfähige Gruppen funktionell abwandelt.
Beispiele reaktiver Derivate der 9-(4-Methoxy-2,3,6-tri-6o methylphenyl)- 3,7-dimethyl- 2,4,6,8-nonatetraensäure sind die Halogenide, insbesondere das Chlorid, reaktive Amide wie das Imidazolid, und gemischte Anhydride. Die erfin-dungsgemässe Acylierung kann in einem inerten organischen Lösungsmittel, z.B. einem Kohlenwasserstoff, wie Benzol ss oder Toluol oder einem Äther wie Dioxan oder Tetrahydro-furan in Gegenwart einer Base, z.B. einem Amin, wie Pyridin (das gleichzeitig als Lösungsmittel dienen kann) durchgeführt werden. Die Reaktionstemperatur ist nicht kritisch.

Zweckmässig arbeitet man bei Temperaturen zwischen 0 °C und 50 C, insbesondere bei 0 °C bis 30 °C.
Im Reaktionsprodukt enthaltene reaktionsfähige Gruppen können gewünschtenfalls funktionell abgewandelt werden. Beispielsweise können in Reaktionsprodukten der Formel I, in denen R amidartig gebunden ist, im Rest R enthaltene O-Acylgruppen durch Behandlung mit Basen, wie Al-kalimetallalkoholaten, verseift werden.
Die Verbindungen der Formel I sind als Wirkstoffe für Pharmazeutika, z.B. zur Behandlung von Neoplasmen, insbesondere zur topischen Verabreichung von Wert. Sie können weiterhin zur Behandlung von Akne und Psoriasis, und zur Behandlung von entzündlichen und allergischen Dermatosen eingesetzt werden. Die Verbindungen zeichnen sich besonders durch eine gute Verträglichkeit, z.B. das Fehlen von Hautirritationen bei der topischen Verabreichung, aus.
Die tumorhemmende Wirkung der Verbindungen wurde an Mäusen geprüft, bei denen durch Behandlung mit Dime-thylbenzanthracen und Krotonöl Papillome der Haut erzeugt wurden. Durch Verabreichung von Verbindungen der Formel I wurde eine Rückbildung der Papillome beobachtet. Weiterhin wurde die Toxizität der Verbindungen anhand ihrer Vitamin A-Wirksamkeit ermittelt. Die Versuchsmethodik für diese Untersuchungen ist in Europ. J. Cancer Vol. 10, 731-737 (1974) beschrieben. Die Versuchsresultate sind in Tabelle I zusammengefasst.

Die Verbindungen derFormel I können als Heilmittel, z.B. in Form pharmazeutischer Präparate, Anwendung finden. Die zur systemischen Anwendung dienenden Präparate können z.B. dadurch hergestellt werden, dass man eine Verbindung der Formel I als wirksamen Bestandteil nicht-toxischen, inerten, an sich in solchen Präparaten üblichen festen oder flüssigen Trägern zufügt. Die Mittel können enterai oder parenteral verabreicht werden. Für die enterale Applikation eignen sich z.B. Mittel in Form von Tabletten, Kapseln, Dragées, Sirupen, Suspensionen, Lösungen und Suppo-sitorien. Für die parenterale Applikation sind Mittel in Form von Infusions- oder Injektions-Lösungen geeignet.
Die Dosierungen, in denen die Verfahrensprodukte verabreicht werden, können je nach Anwendungsart und Anwendungsweg sowie nach den Bedürfnissen der Patienten variieren.
Die Verfahrensprodukte können in Mengen von ca. 5 bis ca. 200 mg täglich in einer oder mehreren Dosierungen verabreicht werden. Eine bevorzugte Darreichungsform sind Kapseln mit einem Gehalt von ca. 10 mg bis ca. 100 mg Wirkstoff.
Die Präparate können inerte oder auch pharmakodyna-misch aktive Zusätze enthalten. Tabletten oder Granula z.B. können eine Reihe von Bindemitteln, Füllstoffen, Trägersubstanzen oder Verdünnungsmitteln enthalten. Flüssige Präparate können beispielsweise in Form einer sterilen, mit
Wasser mischbaren Lösung vorliegen. Kapseln können neben dem Wirkstoff zusätzlich ein Füllmaterial oder Verdik-kungsmittel enthalten. Des weiteren können geschmacksverbessernde Zusätze, sowie die üblicherweise als Konservie-5 rungs-, Stabilisierungs-, Feuchthalte- und Emulgiermittel verwendeten Stoffe, ferner auch Salze zur Veränderung des osmotischen Druckes, Puffer und andere Zusätze vorhanden sein.
Die vorstehend erwähnten Trägersubstanzen und Verdünnungsmittel können aus organischen oder anorganischen Stoffen, z.B. aus Wasser, Gelatine, Milchzucker, Stärke, Ma-gnesiumstearat, Talkum, Gummi arabicum, Polyalkylengly-kolen und dgl. bestehen. Voraussetzung ist, dass alle bei der Herstellung der Präparate verwendeten Hilfsstoffe untoxisch 15sind.
Zur topischen Anwendung werden die Verfahrensprodukte zweckmässig in Form von Salben, Tinkturen, Crèmen, Lösungen, Lotionen, Sprays, Suspensionen und dgl. verwendet. Bevorzugt sind Salben und Crèmen sowie Lösungen. 2( Diese zur topischen Anwendung bestimmten Präparate können dadurch hergestellt werden, dass man die Verfahrensprodukte als wirksamen Bestandteil nichttoxischen, inerten, für topische Behandlung geeigneten, an sich in solchen Präparaten üblichen festen oder flüssigen Trägern zumischt. 25 Für die topische Anwendung sind zweckmässig ca. 0,01 bis ca. 0,3%ige, vorzugsweise 0,02 bis 0,l%ige Lösungen, sowie ca. 0,05 bis ca. 5%ige, vorzugsweise ca. 0,1 bis ca. 2%ige, Salben oder Crèmen geeignet.
Den Präparaten kann gegebenenfalls ein Antioxydations-3omittel, z.B. Tocopherol, N-Methyl-y-tocopheramin sowie butyliertes Hydroxyanisol oder butyliertes Hydroxytoluol beigemischt werden.
Die folgenden Beispiele sollen die Erfindung weiter erläutern.

Beispiel 1
Eine Lösung von 15,6 g Methyl-a-D-glucopyranosid in 150 ml Pyridin wurde bei 0 mit einer Lösung von 9-(4-Me-thoxy- 2,3,6-trimethylphenyl)- 3,7-dimethyl- 2,4,6,8-nonate-4otraenoylchlorid (hergestellt aus 13 g der entsprechenden Säure) in 200 ml Toluol 2 Stunden unter Argon gerührt. Das Gemisch wurde weitere 2 Stunden bei 0: und weitere 2 Stunden bei Raumtemperatur gerührt, und dann zur Trockne eingedampft. Chromatographie an Silicagel mit Hexan-45Äthylacetat als Elutionsmittel lieferte vier Produkte. Als am wenigsten polare Fraktion wurden 2,1 g [Methyl]- 2,6-bis-0-[9-(4-methoxy- 2,3,6-trimethylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetraenoyl]- a-D-glucopyranosid, Schmelzpunkt

Als dritte Substanz wurden 2 g [Methyl]- 2-0-[9-(4-meth-oxy-2,3,6-trimethylphenyl)- 3,7-dimethyl- 2,4,6,8-nonate-sotraenoyl]- a-D-glucopyranosid, Schmelzpunkt 197 (aus 25
Äthylacetat), [a] ^ = +176,6 (c = 1 in Pyridin) erhalten.
Als letztes Produkt wurde 1 g [Methyl]-6-0- [9-(4-meth-65:oxy-2,3,6-trimethylphenyl)-3,7-dimethyl-2,4,6,8-nonatetra-

enoyl]- a-D-glucopyranosid als Schaum erhalten, [a] ^ = + 28,9 (c = 1 in Chloroform).

Beispiel 2
In Analogie zu Beispiel 1 wurden durch Acylierung von D-Trehalosedihydrat mit einem Äquivalent Säurechlorid die
6.6-bis-0-[9-(4-Methoxy- 2,3,6-trimethylphenyl)- 3,7-dime-thyl-2,4,6,8-nonatetraoyl]- D-trehalose, Schmelzpunkt
163-164° (aus Methanol), [a] ^ = +58,2° (c = 1 in
Chloroform) in einer Ausbeute von 8% erhalten.
Beispiel 3
Zu einer Suspension von 16,3 g 9-(4-Methoxy- 2,3,6- tri-methylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetraensäure in 200 ml Dioxan wurden bei 0° unter Rühren 6,9 ml Triäthyl-amin und 4,8 ml Chlorameisensäureäthylester gegeben. Das Gemisch wurde 2 Stunden bei Raumtemperatur gerührt und dann mit einer Lösung von 10,8 g D-Glucosamin-hydro-chlorid in 15 ml Wasser und 6,9 ml Triäthylamin versetzt. Nach 5 Stunden Rühren wurde der Niederschlag gesammelt, mit Äther gewaschen und an Silicagel chromatographiert. Elution mit Methanol-Dioxan lieferte 12,9 g 2-Deoxy-2-[9-(4-methoxy- 2,3,6-dimethylphenyl)- 3,7-dimethyl- 2,4,6,8-no-natetraenamido]- D-glucose, Schmelzpunkt 195° (aus Äthanol, Zersetzung), [a] ^ = +43,1° (c = 1 Pyridin) erhalten.
Beispiel 4
Eine Lösung von 19,5 g [2-Aminoäthyl]- 2-acetamido-3,4,6-tri-O-acetyl- 2-deoxy-ß- D-glucopyranosid in 400 ml Pyridin wurde wie in Beispiel 1 beschrieben acyliert. Der nach Eindampfen des Reaktionsgemisches erhaltene Trok-kenrückstand wurde in 3 1 Chloroform gelöst, die Lösung mit Wasser gewaschen, über Natriumsulfat getrocknet und zu einem Sirup eingedampft. Chromatographie an einer Sili-cagelsäule mit Chloroform und Äthanol als Elutionsmittel lieferten 19,8 g [2-[9-(4-Methoxy- 2,3,6-trimethylphenyl)-
3.7-dimethyl-2,4,6,8-nonatetraenamido]äthyl]- 2-acetamido-3,4,6-tri- O-acetyl- 2-deoxy- ß-D-glucopyranosid, Schmelzpunkt 228° (aus 2-Propanol), [a] ^ = —49,9° (c = X in Chloroform).
Beispiel 5
Eine Suspension von 12 g [2-[9-(4-Methoxy- 2,3,6-trime-thylphenyl)- 3,7-dimethyl- 2,4,6,8-nonatetraenamido]äthyl]-2-acetamido- 3,4,6-tri-O-acetyI- 2-deoxy- ß-D-glucopyrano-sid in 500 ml Methanol wurden mit 40 ml 0,5N methanolischem Natriummethoxid behandelt. Nach 20 Minuten Rühren bei Raumtemperatur wurden die Kristalle abfiltriert, mit Methanol gewaschen und aus Methanol umkristallisiert. Man erhielt 7,4 g [2-[9-(4-Methoxy- 2,3,6-trimethylphenyl)-3,7-dimethyl- 2,4,6,8-nonatetraenamidojäthyl]- 2-acetamido-

2-deoxy-ß- D-glucopyranosid, Schmelzpunkt 223°, [a] ^ = — 25,0°(c = 1 in Dimethylsulfoxid).
Beispiel 6
Eine Lösung von 1,6 g l-[9-(4-Methoxy- 2,3,6-trimethylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetraenoyl]imidazol und 5,62 g ß-D-Glucose in 180 ml Pyridin wurde nach Zusatz einer Spatelspitze Imidazolnatrium 5 Stunden bei 30° gerührt. Danach wurde das Reaktionsgemisch unter vermindertem Druck eingedampft, der Rückstand in 500 ml n-Butanol gelöst und die Lösung zweimal mit je 150 ml Phosphatpuffer (pH 7) und viermal mit je 200 ml Eiswasser extrahiert. Die organische Lösung wurde dann unter vermindertem Druck eingedampft und der Rückstand an 90 g Silicagel chromatographiert. Elution mit Isopropanol-Methylenchlorid (1:1)
lieferte D-Glucopyranosyl- 9-(4-methoxy- 2,3,6-trimethyl-phenyl)- 3,7-dimethyl- 2,4,6,8-nonatetraensäureester als a,ß-Anomerengemisch.
Das als Ausgangsmaterial verwendete Imidazolid wurde s wie folgt hergestellt:
25,1 g 9-(4-Methoxy-2,3,6-trimethylphenyl)- 3,7-dime-thyl- 2,4,6,8-nonatetraensäure und 37,3 g N,N-Carbonyldi-imidazol wurden in 890 ml Tetrahydrofuran gelöst und 18 Stunden bei Raumtemperatur gerührt. Das Reaktionsge-io misch wurde dann unter Rühren auf 9 1 Wasser gegossen, 15 Minuten gerührt und abfiltriert. Der Rückstand wurde in 11 Methylenchlorid gelöst, über Natriumsulfat getrocknet und eingedampft. Umkristallisation aus Methylenchlorid-Pentan lieferte das l-[9-(4-Methoxy- 2,3,6-trimethylphenyl)- 3,7-di-15 methyl- 2,4,6,8-nonatetraenoyljimidazol in Form orangegelber Kristalle vom Schmelzpunkt 184-185°.
Beispiel 7
7,0 g l-[9-(4-Methoxy-2,3,6-trimethylphenyl)- 3,7-dime-2othyl- 2,4,6,8-nonatetraenoyljimidazol und 6,34 g D-(+)-Maltose wurden in 500 ml Pyridin gelöst und nach Zusatz einer Spatelspitze Imidazolnatrium 18 Stunden bei Raumtemperatur gerührt. Das Reaktionsgemisch wurde unter vermindertem Druck eingedampft, der Rückstand in 21 n-Butanol 25 gelöst und viermal mit je 250 ml Phosphatpuffer (pH 7) extrahiert. Die organische Phase wurde eingedampft und der Rückstand an 180 g Silicagel chromatographiert. Elution mit Isopropanol-Methylenchlorid lieferte [4-O-a-D-Gluco-pyranosyl- D-glucopyranosyl]- 9-(4-methoxy- 2,3,6-trime-30 thylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetraensäureester als a, ß-Anomerengemisch.
Beispiel 8
In Analogie zu Beispiel 7 wurde aus l-[9-(4-Methoxy-35 2,3,6-trimethylphenyl)- 3,7-dimethyl- 2,4,6,8-nonatetraeno-yljimidazol und D-Ribose der D-Ribofuranosyl- 9-(4-meth-oxy- 2,3,6-trimethylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetra-ensäureester als Gemisch der a- und ß-Anomeren erhalten.

In Analogie zu Beispiel 7 wurde aus l-[9-(4-Methoxy-2,3,6-trimethylphenyl)- 3,7-dimethyl- 2,4,6,8-nonatetraeno-yljimidazol und L-Rhamnose die l-0-[9-(4-Methoxy-2,3,6-trimethylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetraenoyl]- L-

rhamnose, [a] ^ = +6,9 (c = 1 in Chloroform) erhalten.
Beispiel 10
so In Analogie zu Beispiel 7 wurde aus 2-Acetamido- 2-de-oxy- D-glucose und l-[9-(4-Methoxy- 2,3,6-trimethylphenyl)- 3,7-dimethyl- 2,4,6,8-nonatetraenoyljimidazol die 2-Acetamido- 2-deoxy- l,6-bis-0-[9-(4-methoxy- 2,3,6-tri-methylphenyl)-3,7-dimethyl- 2,4,6,8-nonatetraenoyl]- D-glu-55 cose, Schmelzpunkt 128-130 C (aus Äthylacetat-Hexan) erhalten.
Beispiel 11
In Analogie zu Beispiel 7 wurde aus 4,6-O-Äthyliden- D-60 glucose und l-[9-(4-Methoxy- 2,3,6-trimethylphenyl)- 3,7-di-methyl-2,4,6,8-nonatetraenoyl]imidazol die 4,6-O-Äthyliden-l-O- [9-(4-methoxy- 2,3,6-trimethylphenyl)- 3,7-dimethyl-2,4,6,8-nonatetraenoyl]- D-glucose, Schmelzpunkt 146-147 C (aus Tetrahydrofuran-Hexan) erhalten.

- 2. Verbindungen gemäss Anspruch 1, in denen der Zuk-ker ein Mono- oder Disaccharid ist.2PATENTANSPRÜCHE 1. Verbindungen der allgemeinen Formel worin R einen esterartig gebundenen Rest eines Zuckers oder einen amidartig gebundenen Rest eines Amino-Zuckers oder eines Derivates eines solchen Zuckers, nämlich eines O-oder N-Acylats, Glycosids, Aminoalkylglykosids, Deoxy-zuckers, Acetals oder einer Uronsäure darstellt und n 1 oder 2 ist.
- 3. Verbindungen gemäss Anspruch 1, in denen sich der Zuckerrest von Glucose, Maltose, Trehalose oder Ribose oder einem Derivat davon, nämlich einem O- oder N-Acylat, Glucosid, Deoxyzucker, Acetal oder einer Uronsäure, ableitet.
- 4. Verbindungen gemäss einem der Ansprüche 1-3, in denen die Polyenkette all-trans Konfiguration hat.
- 5. [4-O-a-D-Glucopyranosyl-D- glucopyranosyl]- 9-(4-methoxy-2,3,6-trimethylphenyl)-3,7-dimethyl-2,4,6,8- nona-tetraensäureester, als Verbindung gemäss Anspruch 1.
- 6. - 7. Verbindungen gemäss einem der Ansprüche 1-6 als Arzneimittel.
- 8. Verbindungen gemäss einem der Ansprüche 1-6 als Mittel für die Behandlung von Neoplasien, Akne oder Psoriasis.
- 9. Verfahren zur Herstellung von Verbindungen der Formel I von Anspruch 1, dadurch gekennzeichnet, dass man einen Zucker oder einen Aminozucker oder ein wie im Anspruch 1 definiertes Derivat davon mit einem reaktiven Derivat der 9-(4-Methoxy-2,3,6-trimethylphenyl) 3,7-dimethyl-2,4,6,8-nonatetraensäure umsetzt.
- 10. Pharmazeutische Präparate, enthaltend eine Verbindung gemäss einem der Ansprüche 1-6 und ein Trägermaterial.
- 11. Verwendung einer Verbindung gemäss einem der An-5 Sprüche 1-6 zur Herstellung eines Mittels zur Behandlung von Neoplasien, Akne oder Psoriasis.
