Description
DESCRIZIONE La presente invenzione si riferisce a composizioni farmaceutiche e dermo-cosmetiche aventi come principio attivo acetylcy-steine opportunamente veicolate sotto forma di pomate, creme, unguenti, olii, latti, lozioni, spray, ecc., ad uso dermatologico nel trattamento e prevenzione degli eritemi solari e quali agenti acceleranti della pigmentazione della pelle (abbronzanti).
La N-Acetyl-L-Cysteine (NAC) è il derivato N-acetilato dell'aminoacido naturale L-Cysteine.
La N-Acetyl-L-Cysteine (USP), C5H9NO3S, M.W. = 163.20, C.A.S. 616-91-1, corrisponde alla formula

La sua preparazione è stata descritta per la prima volta da Smith e Gorin nel 1961 (J. Org. Chem. 26, 820, 1961) e successivamente rivendicata, insieme all'uso nelle malattie respiratorie, da Martin e Waller nel 1965 (U.S. Pat. 3 184 505 da Mead Johnson, 1965).
La NAC difatti, in ragione del suo gruppo sulfidrilico libero, è in grado di interagire con le macromolecole mucopolisac-caridi costituenti le secrezioni mucose, riducendone i legami di-solfurici con conseguente riduzione del peso molecolare delle proteine e quindi diminuzione della viscosità delle secrezioni stesse.
Ciò ha portato all'impiego terapeutico della NAC in tutte quelle condizioni patologiche, prevalentemente a livello dell'albero respiratorio, in cui vi fosse eccessiva presenza di secrezioni dense e vischiose (J. Ramirez, Archiv. Int. Med. 119, 147, 1967; S.R. Hirsh and R.C. Kory, J. Allergy 39, 265, 1967; R. Denton et al., Am. Rev. Resp. Dis., 95, 643, 1967; M. Bracey et al., Arch. Dis. Children 44, 404, 1969; C. Grassi et al. Curr.
Ther. Res. 15, 165, 1973) In anni più recenti è stato postulato un ruolo protettivo da parte degli agenti sulfidrilici verso quegli agenti tossici (metaboliti tossici, radicali ossidanti ecc.) il cui meccanismo di depurazione nell'organismo avviene attraverso vie «glutatione dipendenti» (H. Sprince, Agents and Actions 5, 164, 1975; H. Sprince, Agents and Actions, 9, 407, 1979; G.M. Green, Science 162, 810, 1968; C. Leuchtenberger, Br. J. Exp. Path., 58, 625, 1977).
Il glutatione è difatti un tripeptide sulfidrilato dell'organismo, di cui composti sulfidrilici, ed in particolare la cisterna, sono precursori. In questo rispetto la NAC, in quanto derivato della cisterna, più stabile della cisterna stessa (A.L. Schaffner et al., Biochem. Pharmacol. 15,1523, 1966; L. Bonanomi and A. Gazzaniga. Eur. J. Resp. Dis. 61 (suppl. Ili), 45, 1980), si sta dimostrando più efficace nel ripristinare le concentrazioni utili di glutatione endrogeno ai fini antidotici e protettivi (Proc. of Symposium on N. Acetylcysteine, a significant chemoprotective adjunct, Seminars in Oncology, 10 (suppl. 1) 1983.
In questo contesto si sono rivelati estremamente interessanti osservazioni che mostrano come la NAC, applicata topicamente, possa prevenire lesioni irritative della pelle causate da radiazioni X (J.A. Kim et al., Seminars in Oncology, 10 (suppl. 1) 86, 1983).
Non si conosce fino ad ora l'uso dermatologico della acetyl-cysteina. La richiedente dopo approfondite ricerche ha ora trovato che l'acetylcysteina possiede caratteristiche dermatologiche e che, opportunamente veicolata, può essere impiegata per usi dermatologici e come abbronzante, causa la sua attività anti-eritemica e la sua azione accelerante nel processo di pigmentazione della pelle.
La richiedente inoltre, ha riscontrato in una casistica sufficientemente probante che l'acetilcisteina sotto forma di preparazione topica, quali pomate, unguenti, creme, latti, lozioni, spray, etc., al 5%-15%, protegge un soggetto esposto alle radiazioni solari da eritemi e permette nello stesso tempo un più rapido aumento della pigmentazione (abbronzatura) della pelle.
Più precisamente la presente invenzione si riferisce a composizioni farmaceutiche di formula

caratterizzate dal fatto che detto principio attivo viene veicolato mediante supporti opportuni per ottenere preparati per uso esterno dermatologico.
Trattandosi di un derivato di un aminoacido, cioè di una sostanza fisiologica, già impiegata con successo per altri scopi terapeutici e priva di effetti secondari, essa risulta essere completamente atossica ed ipoallergica rispetto ad altri principi attivi usati nei prodotti per le cure solari.
L'acetylcisteina presenta inoltre una marcata azione nell'accelerazione dei processi di pigmentazione, ciò che le conferisce un ulteriore vantaggio sui prodotti noti, usati per l'abbronzatura. Si aggiunga che la stessa può essere abbinata nelle composizioni ad altri principi attivi per dare una composizione ad effetto multiplo.
Dalle medesime ricerche effettuate dalla richiedente è apparso che questo effetto accelerante della pigmentazione della pelle può essere ottenuto anche con altre sostanze aventi pure il gruppo tiolico libero.

caratterizzato dal fatto che detto principio attivo viene veicolato mediante supporti opportuni per ottenere preparati per uso esterno dermatologico.
2. Composizioni farmaceutiche topiche dermatologiche e dermo-cosmetiche secondo la rivendicazione 1, caratterizzata dal fatto che l'acetylcysteina di formula I è contenuta sotto forma di preparazione topica quali pomate, unguenti, latti, lozioni, olii, spray, ecc. al 5%-15%.

3. Utilizzazione delle composizioni farmaceutiche di cui alle rivendicazioni precedenti in preparati ad uso dermatologico per la prevenzione degli eritemi da sole e per accelerare il processo di pigmentazione della pelle.
