Description
BESCHREIBUNG Die Erfindung betrifft ein Mundpflegemittel mit einem Gehalt eines oral verträglichen Trägers und einer Zahnstein verhindernden Komponente.
Zahnstein ist eine harte mineralische Ablagerung auf den Zähnen, dessen schnelle Ablagerung durch regelmässiges Zähneputzen zwar verhindert wird, jedoch nicht zur Entfernung aller Zahnsteinablagerungen am Zahn ausreicht. Zahnstein bildet sich durch Ablagerung von Calciumphosphatkri-stallen auf den Häutchen und der extracellularen Matrix von Zahnbelag und entwickelt sich zu hinreichend eng miteinander verbundenen Aggregaten, die sich nicht mehr deformieren lassen. Es bestehen verschiedene Auffassungen über das Entstehen des kristallinen als Hydroxylapatit (HAP) anzusprechenden Zahnbelages aus Calcium und Orthophosphat. Jedoch wird allgemein angenommen, dass bei höheren Sättigungen d.h. oberhalb der kritischen Sättigungsgrenze der Vorläufer des kristallinen HAP ein amorphes oder mikrokristallines Calciumphosphat ist. Dieses mit dem Hydroxylapatit verwandte amorphe Calciumphosphat unterscheidet sich dennoch in seiner Atomstruktur, in der Morphologie der Teilchen und stöchiometrisch voneinander. Die Röntgen-Brechungsmuster von amorphem Calciumphosphat zeigen breite Peaks, die für amorphe Produkte typisch sind, und nicht die breiten Bereiche geordneter Atome, wie sie für alle kristallinen Materialien einschliesslich HAP typisch sind. Daraus ergibt sich, dass Verbindungen, die wirksam das Kristallwachstum von HAP stören als Zahnstein verhindernde Komponenten wirksam sein können.
Untersuchungen haben gezeigt, dass ein erkennbarer Zusammenhang zwischen der Fähigkeit einer Verbindung zur Verhinderung eines HAP-Kristallwachstums in vitro und der Fähigkeit zur Verhinderung einer Kalkausbildung in vivo bestehen, vorausgesetzt, dass eine derartige Verbindung im Zahnbelag, im Speichel und in dessen Bestandteilen stabil ist.
Aus dem Stand der Technik ist zu entnehmen, dass wasserlösliche Hexametaphosphate, Tripolyphosphate und Py-rophosphate wirksam sind, Calcium- und Magnesiumionen abzufangen und Inhibitoren, Sequestriermittel und/oder Ge-latiermittel für diese sind und in vitro eine HAP-Bildung verhindern. So beschreibt US-PS 4 515 772 Mundpflegemittel mit Zahnstein verhindernden Eigenschaften, die einen Fluo-ridionen-Lieferanten und lösliches Dialkalipyrophosphat allein oder zusammen mit Tetraalkalipyrophosphat enthalten. Wenngleich in dieser Druckschrift kein experimenteller Nachweis für eine wirksame Zahnsteinverhinderung in vivo offenbart ist, wird unter Hinweis auf frühere Literaturstellen angegeben, dass Pyrophosphate durch Pyrophosphatase in vivo nämlich durch/lieses im Speichel enthaltende Enzym inhibiert werden würde; darüber hinaus zeigen die zahlreichen Literaturstellen in dieser Druckschrift, dass diese Poly-phosphate bislang in Mundpflegemitteln in vielfaltiger Hinsicht mit vielen Funktionen eingesetzt worden ist.
Wie dieser Stand der Technik und die folgenden Nachweise zeigen, erleiden diese linearen, molekular dehydrati-sierten Polyphosphate wie Hexametaphosphate, Tripolyphosphate und Pyrophosphate in der Mundhöhle und/oder im Zusammenhang mit Speichel alle eine deutliche Hydrolyse durch die im Speichel vorhandenen Enzyme wie Phosphatase; diese hydrolysieren zu Orthophosphaten, die als Inhibitoren für eine HAP-Bildung unwirksam sind.
Die Erfindung hat sich die Aufgabe gestellt, ein verbessertes Mundpflegemittel zu schaffen, das die obigen Nachteile nicht besitzt und die Umwandlung des amorphen Calciumphosphat zu einer HAP-Kristallstruktur, die mit der Zahnsteinbildung einhergeht, zu verhindern.
Zur Lösung dieser Aufgabe wird daher ein Mundpflegemittel der eingangs geschilderten Art vorgeschlagen, welches gemäss Kennzeichen des Anspruches 1 zusammengesetzt ist, wobei besondere Ausführungsformen in den abhängigen Ansprüchen aufgeführt sind.

Die Verbindungen, die als Fluoridionen-Lieferant dienen können, sind im Stand der Technik ausführlich als Antika-riesmittel beschrieben.
An sich sind synthetische anionische Carboxygruppen aufweisende Polymere, auch polymere Polycarboxylate genannt, und deren Komplexe mit verschiedenen kationischen Germiciden, Zink und Magnesium als Zahnstein verhindernden Mittel z.B. aus US-PS 3 429 963,4 152 420, 3 956 480, 4 138 477 und 4 138 914 bekannt. Keine dieser Literaturstellen und auch nicht der Stand der Technik offenbart die Verwendung derartiger Polycarboxylate als solche zur Inhibierung einer speichelbedingten Hydrolyse der linearen Pyro-phosphate, und schon gar nicht in Kombination mit einer Verbindung, die Fluoridionen liefert. Es ist davon auszugehen, dass die synthetischen anionischen linearen polymeren Polycarboxylate gemäss Stand der Technik in den Zusammensetzungen gemäss Erfindung wirksam sind.
Die linearen molekular dehydratisierten Polyphosphate, die hier als Zahnstein verhindernde Komponente wirksam sind, sind allgemein bekannt und werden im allgemeinen in Form ihrer vollständig oder teilweise neutralisierten wasserlöslichen Alkalisalze z.B. als Kalium- oder vorzugsweise Natriumsalz oder als Ammoniumsalze alleine oder in Mischungen eingesetzt. Typische Beispiele dieser Verbindungen sind Natriumhexametaphosphat, Natriumtripolyphosphat, Di-natriumdihydrogen-, Trinatriummonohydrogen- und Tetra-natriumpyrophosphate und dergleichen. Die linearen Polyphosphate entsprechen der Formel (NaPC>3)n, in der n einen Wert von etwa 2 bis 125 hat. Sie werden allgemein bei den vorliegenden Mundpflegemitteln in Anteilen von etwa 0,1 bis 7 Gew.% und vorzugsweise in Mengen von 0,1 bis 6 und insbesondere 2 bis 6 Gew.% eingesetzt. Wenn n einen Wert von mindestens 3 besitzt, sind die Polyphosphate von glasartigem Charakter.
Die synthetischen anionischen linearen polymeren Polycarboxylate, die im vorliegenden Fall einsatzfähig sind, sind bekannt und werden vorzugsweise eingesetzt, als teilweise oder insbesondere als voll neutralisierte wasserlösliche Alkalisalze oder Ammoniumsalze. Bevorzugt sind Copolymere von Maleinsäureanhydrid oder -säure mit anderen polymeri-sierbaren ethylenisch ungesättigten Monomeren und vorzugsweise Methylphenylether (Methoxyethylen) mit einem Molekulargewicht von etwa 30 000 bis etwa 1 000 000 in einem Verhältnis von 1:4 bis 4:1. Diese Copolymere sind beispielsweise mit einem Molekulargewicht von 500 000 als «Gantrez AN 139» oder mit einem Molekulargewicht von 250 000 als «Gantrez AN 119» und auch insbesondere mit einem Molekulargewicht von 70 000 als Produkt S-97 in pharmazeutischer Qualität von der GAF Corporation ent-hältlich. Diese Produkte werden als synthetische Verbindungen bezeichnet, um bekannte Verdickungsmittel oder Gelie-rungsmittel auszuschliessen, die Carboxymethylzellulose und andere Zellulosederivate oder natürliche Gumme enthalten.
Andere einsetzbare polymere Polycarboxylate sind in der US-PS 3 956 180 offenbart, wie Copolymere von Maleinsäureanhydrid mit Ethylacrylat, Hydroxyethylmethacrylat, N-Vinyl-2-pyrrolidon oder Ethylen (Monsanto EMA Nr. 1103, Molgewicht 10 000 und EMA Nr. 61) und Copolymere von Acrylsäure mit Methyl- oder Hydroxyethylmethacrylat, Methyl* oder Ethylacrylat, Isobutylvinylether oder N-Vinyl-2-pyrrolidon im Verhältnis von 1:1.
Weitere geeignete polymere Polycarboxylate gemäss US-PS 4 138 477 und 4 183 914 sind Copolymere von Maleinsäureanhydrid mit Styrol, Isobutylen oder Ethylvinylether, Polyacrylsäure, Polyitaconsäure und Polymaleinsäuren und Sulfoacryl-Oligomere mit einem Molekulargewicht bis herab zu 1000 (die als «Uniroyal ND-2» erhältlich sind).

Im allgemeinen sind polymerisierte olefinisch oder ethylenisch ungesättigte Carbonsäuren geeignet, die eine olefmische aktivierte Kohlenstoff-Kohlenstoff Doppelbindung und mindestens eine Carboxylgruppe enthalten, nämlich eine Säure, die eine olefinische Doppelbindung enthält, die bei der Polymerisation zur Verfügung steht, und zwar wegen ihrer Anwesenheit in dem monomeren Molekül entweder in der — ß-Stellung gegenüber dem Carboxylrest oder als Teil einer endständigen Ethylengruppe. Beispiele derartiger Säuren sind Acryl-, Methacryl-, Ethacryl-, a-Chloracryl-, Croton-, ß-Acryloxypropion-, Sorbin-, a-Chlorsorbin-, Cinna-mon-, ß-Styrilacryl-, Mucon-, Itacon-, Citracon-, Mesacon-, Glutacon-, Aconit-, a-Phenylacryl-, 2-Benzylacryl-, 2-Cy-clohexylacryl-, Angelic-, Umbellic-, Fumar-, Maleinsäure und deren Anhydride. Andere olefinische Monomere, die mit diesen Carbonsäure-Monomeren copolymierbar sind, sind unter anderem Vinylacetat, Vinylchlorid, Dimethylma-leat und dergleichen. Die Copolymere enthalten hinreichend Carbonsäuresalzgruppen, um eine Wasserlöslichkeit zu ermöglichen.
Die synthetischen anionischen polymeren Polycarboxylate sind im wesentlichen Kohlenwasserstoffe, die vorzugsweise Halogen oder einen sauerstoffhaltigen Substituenten enthalten und Bindungen aufweisen, wie beispielsweise Ester, Ether und OH-Reste und die bei den erfindungsgemässen Zusammensetzungen in Mengen von etwa 0,05 bis 3, und vorzugsweise von 0,05 bis 2 und insbesondere von 0,1 bis 2 Gew.%, verwendet werden. Anteile in den höheren Bereichen werden in der Regel bei Zahnpflegemitteln eingesetzt, die ein dentales Reibmittel oder Putzmittel enthalten und zum Zähneputzen verwendet werden, wie beispielsweise Zahnpasten oder Zahncreme, Zahngele, Zahnpulver und Tabletten. Grössere Mengen ausserhalb dieser Bereiche können zum Verdicken oder zum Gelieren verwendet werden.
Als Lieferant der Fluoridionen oder der Fluorverbindungen, die gemäss Erfindung als wesentliche Komponente der Inhibitor-Kombination dienen, werden die bei der Kariesbekämpfung bekannten Verbindungen verwendet, die auch als solche bei den erfindungsgemässen Mundpflegemitteln wirken. Diese Verbindungen können gering oder vollständig wasserlöslich sein und besitzen die Fähigkeit, Fluoridionen in Wasser ohne unerwünschte Reaktionen mit anderen Verbindungen der Zahnpflegemittel freizusetzen. Hierzu gehören anorganische Fluoridsalze wie lösliche Alkali- und Erdalkalisalze wie beispielsweise Natriumfluorid, Kaliumfluo-rid, Ammoniumfluorid, Calciumfluorid, Kupferfluorid wie Kupfer(I)-fluorid, Zinkfluorid, Bariumfluorid, Natriumfluorsilikat, Ammoniumfluorsilikat, Natriumfluorzirkonat, Natriummonofluorphosphat, Aluminiummono- und di-fluorphosphat und fluorierte Natriumcalciumphosphate. Alkali- und Zinnfluoride, wie Natriumfluorid und Zinn(II)-fluorid, sowie Natriummonofluorphosphat und deren Mischungen werden bevorzugt.
Die Menge der Fluor liefernden Verbindung hängt in gewisser Weise von der Art der Verbindung, ihrer Löslichkeit und der Art des Mundpflegemittels ab; sie muss in einer nicht-toxischen Menge vorliegen, allgemein etwa in einer Menge von 0,05 bis etwa 3,0 Gew.% bezogen auf die Zusammensetzung. Bei einer Mundpflegezubereitung wie beispielsweise einem Zahngel einer Zahnpasta oder einer Zahncreme, einem Zahnpulver oder einer Zahntablette ist die Menge der entsprechenden Verbindung so bemessen, dass sie bis zu etwa 2000 ppm Fluorionen bezogen auf das Gewicht der Zubereitung zur Verfügung stellt. Jede geeignete Mindestmenge an einer derartigen Verbindung kann verwendet werden, jedoch wird vorzugsweise soviel der Fluorverbindung verwendet, dass etwa 300 bis 2000 ppm und insbesondere 800 bis 1500 ppm Fluoridionen zur Verfügung gestellt werden.

Bei Verwendung von Alkalifluoriden und Zinn(II)-fluo-rid ist diese Komponente gewöhnlich in Mengen von bis zu 2 Gew.%, bezogen auf das Gesamtgewicht des Mundpflegemittels, vorhanden und liegt vorzugsweise im Bereich von etwa 0,05 bis 1 Gew.%. Bei Natriummonofluorphosphat liegt diese Verbindung in einer Menge von 0,1 bis 3 und vorzugsweise in einer Menge von etwa 0,76 Gew.% vor.
Bei anderen Mundpflegemitteln wie Mundwässern, Lutschtabletten und Kaugummi ist die fluorliefernde Verbindung gewöhnlich in solchen Mengen vorhanden, dass bis zu etwa 500 ppm und vorzugsweise etwa 25 bis 300 ppm, bezogen auf das Gewicht des Fluoridions, freigesetzt werden. Im allgemeinen sind 0,005 bis 1,0 Gew.% der Verbindung vorhanden.
Bei den vorzugsweise im wesentlichen flüssigen Mundpflegemitteln wie Mundwässern dient als Träger meist eine Wasser/Alkohol-Mischung, die vorzugsweise ein Feuchthaltemittel enthält. Im allgemeinen liegt das Gewichtsverhältnis von Wasser zu Alkohol in einem Bereich von etwa 1:1 bis 20:1, vorzugsweise bei 3:1 bis 10:1 und insbesondere bei 4:1 bis 6:1. Die Gesamtmenge des Wasser/Alkohol-Gemisches liegt gewöhnlich in einem Bereich von 70 bis etwa 99,9 Gew.% der Zusammensetzung.
Der pH-Wert der Zahnpflegemittel gemäss Erfindung Hegt im allgemeinen im Bereich von etwa 4,5 bis 9 und gewöhnlich bei 5,5 bis 8. Vorzugsweise liegt der pH-Wert in einem Bereich von etwa 6 bis 8,0. Es ist anzumerken, dass die erfindungsgemässen Mundpflegemittel in pH-Wert-Bereichen unter 0,5 oral eingesetzt werden können, ohne dass sie im wesentKchen decalzifierend wirken oder auf andere Weise den Zahnschmelz angreifen. Der pH-Wert kann mit einer Säure wie beispielsweise Zitronensäure oder Benzoesäure oder mit einer Base wie beispielsweise mit Natriumhydroxid eingestellt oder abgepuffert werden, wie beispielsweise mit Natriumeitrat, -benzoat, -carbonat oder -bicarbonat, Dina-triumhydrogenphosphat oder Natriumdihydrogenphosphat.
Das Mundpflegemittel kann im wesentlichen fest oder pastenförmig sein, wie bei einem Zahnpulver, in einer Zahntablette oder bei einer Zahnpasta oder einem Zahngel. Die Trägerstoffe dieser festen oder pastösen Zahnpflegemittel enthalten gewöhnlich ein oral- oder dentalverträgliches Po-liermittel, das beim Zähnebürsten wirkt. Beispiele für derartige Poliermittel sind wasserunlösliches Natriummetaphosphat, Kaliummetaphosphat, Tricalciumphosphat, Calcium-phosphat-Dihydrat, wasserfreies Dicalciumphosphat, Cal-ciumpyrophosphat, Magnesiumorthophosphat, Trimagne-siumphosphat, Calciumcarbonat, Aluminiumsilikat, Zirkon-silikat, Kieselsäure, Bentonit und deren Mischungen. Andere geeignete Poliermittel oder Putzkörper sind teilchenförmi-ge wärmehärtbare Harze gemäss US-PS 4 070 510 wie Mel-amin-, Phenol- und Harnstoff-Formaldehyd-Harze und vernetzte Polyepoxide und Polyester. Bevorzugte Poliermittel sind kristalline Kieselsäure mit einer Teilchengrösse bis zu 5 (im, einer mittleren Teilchengrösse bis zu etwa 1,1 um und einer Oberfläche bis zu etwa 50 000 cm2/g, Kieselgele oder kolloidale Kieselsäure und komplexe amorphe Alkalialumo-silikate.
Wenn optisch klare Gele verwendet werden, werden als Poliermittel kolloidale Kieselsäure.(SYLOID-Handelspro-dukte wie Syloid 72 und Syloid 74 oder SANTOCEL-Produkte wie Santocel 100) und Alkalialumosilikatkomplexe bevorzugt, da deren Brechungsindices nah an dem Brechungsindex des Systems aus Gehermittel und flüssiger Phase einschliesslich Wasser und/oder Feuchthaltemittel liegen.
Zahlreiche der sogenannten wasserunlöslichen Poliermittel sind anionisch und enthalten kleine Mengen an löslichen Bestandteilen. So kann beispielsweise unlösliches Natriummetaphosphat auf behebige Weise hergestellt werden, wie es in «Thorpe's Dictionary of Applied Chemistry», Bd. 9,4. Aufl., S. 510—511 beschrieben ist. Beispiele für weitere geeignete unlösliche Natriummetaphosphate sind als Madrell'-sches Salz und als Kurrol'sches Salz bekannt. Diese Meta-phosphate zeigen nur eine geringe Löslichkeit in Wasser und werden demzufolge im allgemeinen als unlösliche Metaphos-phate bezeichnet. Diese enthalten eine kleine Menge an löslichen Phosphaten als Verunreinigungen von gewöhnlich einigen Prozent bis zu etwa 4 Gew.%. Die Menge an löslichem Phosphat, die vermutlich lösliches Natriumtrimetaphosphat beim unlöslichen Methaphosphat ist, kann gegebenenfalls durch Waschen mit Wasser verringert oder ausgeschaltet werden. Das unlösliche Alkalimetaphosphat wird gewöhnlich in pulveriger Form in einer solchen Teilchengrösse verwendet, dass nicht mehr als etwa 1 Gew.% des Materials grösser als etwa 37 (im ist.
Das Poliermaterial ist im allgemeinen in den festen oder pastösen Zusammensetzungen in Konzentrationen von etwa 10 bis 99 Gew.% vorhanden. Bei Zahnpasten oder Zahngelen liegt die Menge an Reibmitteln vorzugsweise in einem Bereich von 10 bis 75 Gew.% und bei Zahnpulver oder Zahntabletten in einem Bereich von etwa 70 bis 99 Gew.%.
Bei Zahnpasten kann der flüssige Träger Wasser und ein Feuchthaltemittel gewöhnlich in Mengen von etwa 10 bis 90 Gew.%, bezogen auf die Zusammensetzung, enthalten. Gly-cerin, Propylenglykol, Sorbit, Polypropylenglykol und/oder Polyethylenglykol z.B. mit einem Molekulargewicht von 400 bis 600 sind Beispiele für geeignete Feuchthaltemittel bzw. Träger; ferner sind flüssige Mischungen von Wasser, Glyce-rin und Sorbit von Vorteil. Bei klaren Zahngelen, bei denen der Refraktionsindex wesentlich ist, werden etwa 3 bis 30 Gew.% Wasser, 0 bis etwa 80 Gew.% Glycerin und etwa 20 bis 80 Gew.% Sorbit vorzugsweise eingesetzt.
Zahnpasten bzw. Zahncremes und Zahngele enthalten gewöhnlich ein natürliches oder synthetisches Verdickungs-oder Geliermittel in Mengen von etwa 0,1 bis 10 und vorzugsweise 0,5 bis 5 Gew.%. Ein geeignetes Verdickungsmittel ist synthetischer Hectorit, ein synthetischer, kolloidaler Magnesiumalkalisilikat-Ton, wie beispielsweise Laponite-Typen (wie sie von der Laporte Industries Limited als Type CP, SP 2002, D vertrieben werden), wobei Laponite D etwa 58,00 Gew.% Si02,25,40 Gew.% MgO, 3,05 Gew.% Na20, 0,98 Gew.% Li20 und etwas Wasser und Spurenmetalle enthält. Die absolute spezifische Dichte liegt bei 2,53; das Material hat eine Schüttdichte von 1,0 g/ml bei 8% Feuchtigkeit.
Andere geeignete Verdickungsmittel sind Irish Moos, Gum-Tragacanth, Stärke, Polyvinylpyrrolidon, Hydroxy-ethylpropylzellulose, Hydroxybutylmethylzellulose, Hy-droxypropylmethylzellulose, Hydroxyethylzellulose (Natro-sol), Natriumcarboxymethylzellulose und colloidale Kieselsäure (wie fein vermahlenes Syloid z.B. Type 244).
Die Zahnpasten, Zahncremes oder Zahngele können in üblichen zusammendrückbaren Tuben meist aus Aluminium, Blei oder Kunststoff, gegebenenfalls mit Innenkaschie-rung, oder in anderen Abgabebehältern oder druckbetätigten Behältern abgepackt werden.
Die erfindungsgemässen Zahnpflegemittel können mit organischen oberflächenaktiven Stoffen versetzt werden, um eine bessere prophylaktische Wirkung zu erzielen und eine gründliche und vollständige Dispersion des Zahnstein verhindernden Bestandteils in der Mundhöhle zu verbessern und die Zahnpflegemittel kosmetisch besser zu gestalten. Vorzugsweise sind die oberflächenaktiven Substanzen anionisch, nichtionisch oder ampholytisch, wobei vorzugsweise ein Tensid mit reinigenden Eigenschaften verwendet wird, das auch das Schäumen verbessert. Beispiele für anionische Tenside sind wasserlösliche Salze höherer Fettsäuremono-glyceridmonosulfate, wie das Natriumsalz des monosulfa-

tierten Monoglycerids von hydrierten Kokosnussölfettsäu-ren, höhere Alkylsulfate wie Natriumlaurylsulfat, Alkylaryl-sulfonate, wie Natriumdodecylbenzolsulfonat, höhere Alkyl-sulfoacetate, höhere Fettsäureester von 1,2-Dihydroxypro-pansulfonat und die im wesentlichen gesättigten höheren aliphatischen Acylamide niederer aliphatischer Aminocarbon-säureverbindungen, die beispielsweise 12 bis 16 Kohlenstoffatome im Fettsäure-Alkyl- oder Acylrest enthalten, wie beispielsweise N-Lauroylsarcosin und die Natrium-, Kalium-und Ethanolaminsalze von N-Lauroyl, N-Myristoyl- oder N-Palmitoylsarcosin, die im wesentlichen frei von Seifen oder ähnlichen höheren Fettsäureprodukten sein sollen. Der Einsatz dieser Sarconisate in den erfindungsgemässen Mundpflegemitteln ist besonders vorteilhaft, da diese eine verlängerte und deutliche Wirkung hinsichtlich der Inhibierung einer Säurebildung in der Mundhöhle zeigen, und zwar aufgrund des Aufbrechens der Kohlenhydrate neben einer Verringerung der Löslichkeit des Zahnschmelzes in sauren Lösungen.
Beispiele von wasserlöslichen nichtionischen Tensiden sind Kondensationsprodukte von Ethylenoxid mit den verschiedensten reaktiven Wasserstoff enthaltenden Verbindungen, die mit diesen reagieren und dadurch lange hydrophobe Ketten haben, wie beispielsweise aliphatische Ketten mit 12 bis 20 Kohlenstoffatomen, wobei die Kondensationsprodukte (Ethoxamere) hydrophile Polyoxyethyleneinheiten enthalten, wie Kondensationsprodukte von Poly(ethylenoxid) mit Fettsäuren, Fettalkoholen und Fettamiden, mehrwertigen Alkoholen, wie Sorbitanmonostearat und Polypropylenoxid (wie Pluronic).
Die erfindungsgemässen Zahnpflegemittel können noch weitere Zusätze enthalten, wie Weissmachungsmittel, Konservierungsstoffe, Silikone, Chlorophyllverbindungen, andere Zahnstein verhindernde Mittel und/oder Ammoniak enthaltende Verbindungen, wie Harnstoff, Diammoniumphos-phat und deren Mischungen. Diese Zusätze werden gegebenenfalls in solchen Mengen eingesetzt, dass sie die gewünschten Eigenschaften nicht wesentlich benachteiligen.
Ferner können übliche Aromastoffe und Süssungsmittel verwendet werden, wie beispielsweise Aromaöle auf Basis von Spearmint, Pfefferminz, Wintergrün, Sassafras, Klee, Salbei, Eukalyptus, Majoran, Zimt, Limone und Orange und Methylsalicylat. Geeignete Süssungsmittel sind u.a. Su-crose, Lactose, Maltose, Dextrose, Lävulose, Sorbit, Xylit, d-Tryptophan, Dihydrochalcone, Natriumcyclamat, Peril-lartin, Aspartylphenylalanin-methylester, Saccharin und dergleichen. Die Aroma- und Süssungsmittel sind im allgemeinen in Mengen von etwa 0,1 bis 5 Gew. % oder mehr vorhanden.
Die erfindungsgemässen Mundpflegemittel wie Mundspülmittel oder Zahnpflegemittel mit einem Gehalt an den beschriebenen Polyphosphaten und der Inhibitor-Kombination in einer Menge zur wirksamen Verhinderung einer Zahnsteinbildung werden vorzugsweise regelmässig auf den Zahnschmelz beispielsweise jeden zweiten oder dritten Tag oder vorzugsweise 1 bis 3 mal täglich bei einem pH-Wert von etwa 4,5 bis etwa 10 und im allgemeinen von etwa 5,5 bis 9 und vorzugsweise von etwa 6 bis 8 mindestens 2 bis 8 Wochen oder mehr aufgebracht.
Die erfindungsgemässen Zusammensetzungen können auch in Pastillen oder Kaugummi oder anderen Produkten eingesetzt werden, indem man beispielsweise diese in eine warme Gum-Basis einrührt oder die Aussenfläche einer Gum-Basis beschichtet, wobei als Gum-Basis Jeluton, Kautschuk, Latex, Vinylitharze und dergleichen verwendet werden, und zwar vorzugsweise mit üblichen Weichmachern oder Weichstellungsmitteln, Zucker oder anderen Süssungs-

mittein oder Kohlehydraten wie Glycose, Sorbit und dergleichen.
Beispiel A
Um die Wirkung der Speichelenzyme auf die Verhinderung einer HAP-Bildung durch Natriumhexametaphosphat (HAP) und Tetranatriumpyrophosphat (TSPP) zu zeigen, wurde die HAP-Bildung in vitro titrimetrisch mittels einer statischen pH-Wertbestimmung gemessen. Es wurden eine 0,1 m CaCl2- und eine 0,1 m NaH2P04-Vorratslösung in car-bonatfreiem, entsalzten, destillierten Wasser frisch hergestellt zu 23 ml C02freiem, entsalzten, destillierten Wasser wurden 1,0 ml der Phosphatausgangslösung und 1,0 ml einer wässrigen Lösung von 1 x 10~4 des zu untersuchenden Zahnstein verhindernden Mittels gegeben und anschliessend 1,0 ml der CaCk-Ausgangslösung zugesetzt, um die Reaktion auszulösen. Die Umsetzung wurde unter Stickstoff bei einem pH-Wert von 7,4 durchgeführt. Der Verbrauch an 0,1 n NaOH wurde automatisch aufgezeichnet, woraus sich die erforderliche Zeit für die Kristallbildung ergab. Die folgende Tabelle A zeigt die Messergebnisse der zeitlichen Verhinderung des Kristallwachstums in Stunden bei Verwendung von Natriumhexametaphosphat (HMP) und Tetranatriumpyrophosphat (TSPP) als Zahnstein verhinderndes Mittel.

Tabelle A zeigt, dass beide Verbindungen in Wasser die HAP-Bildung deutlich verzögern. Jedoch wird die Wirksamkeit dieser Komponenten drastisch verringert, wenn Speichel vorhanden ist, was sich durch die kürzere Inhibierungszeit zeigt. Diese Verringerung der Wirksamkeit beruht auf der enzymatischen Hydrolyse von P-O-P-Bindungen.
Bei einer Inkubation dieser Substanzen mit Pyrophosphatase und alkalischer Phosphatase wird die Verzögerung deutlich verhindert, was die Ansprechbarkeit der P-O-P-Bindungen gegenüber der Hydrolyse durch Phosphatase zeigt.
Beispiel 1
Um die Stabilisierung von Tetranatriumpyrophosphat (TSPP), Natriumtripolyphosphat (STPP) und Natriumhexametaphosphat (HMP) gegenüber einer enzymatischen Hydrolyse in Gegenwart von Inhibitoren zu zeigen, wurde die enzymatische Hydrolyse in einer 0,1 m Morpholinpropan-sulfonsäure/NaOH-Pufferlösung mit einem pH-Wert von 7,0 durchgeführt, die 1,3 mg/ml TSPP enthielt. Es wurden neben einem Vergleichsversuch erfindungsgemässe Inhibitoren bis zu einer Endkonzentration von 1000 ppm Fluoridionen von Natriumfluorid und 0,5% des Natriumsalzes von hydrolysiertem Methoxyethylen/Maleinsäureanhydridcopo-lymeren im Verhältnis von 1:1 mit einem Molekulargewicht von 70 000 (Gantrez S-97 in pharmazeutischer Reinheit) zugesetzt. Anschliessend wurden saure Phosphatase, alkalische Phosphatase und anorganische Pyrophosphatase in Mengen gleicher Aktivitäten zugesetzt, um eine gesamte Phosphatase Aktivität von 0,3 Einheiten/ml zu erhalten. Proben der Testlösungen wurden genommen und der Gesamtwert der zur Verfügung stehenden Orthophosphatase in jeder Probe nach 3 Stunden Hydrolyse in 4 n HCl bei 100 °C bestimmt. Die

Reaktionsmischungen wurden bei 37 °C unter Schütteln inkubiert, und es wurden jeweils gleiche Anteile im Verlaufe von mindestens 90 Minuten zur Bestimmung von Orthophosphat entnommen. Die folgende Tabelle I zeigt die Ergebnisse an Orthophosphat in Prozent, welches aufgrund der Hydrolyse der Polyphosphate freigesetzt wurde.

Tabelle I zeigt, dass nach 90 Minuten Inkubation in Gegenwart von Enzym mehr als 95% des zur Verfügung stehenden Orthophosphat von den Polyphosphaten bei Abwesenheit von Inhibitoren freigesetzt wird. Mit Inhibitoren wird die Hydrolyse der P-O-P-Bindungen im Pyrophosphat Tripolyphosphat und Hexametaphosphat um 41 %, bzw. 39%, bzw. '57% verringert. In diesem Zusammenhang ist es bemerkenswert, dass die Enzymaktivitäten bei diesem Versuch mindestens 2 bis 3 mal grösser sind als sie übhcherweise im Speichel auftreten. Diese Werte zeigen, dass die erfin-dungsgemässe Kombination von Inhibitoren deutlich die en-zymatische Hydrolyse von linearen Polyphosphaten verringert.

Claims (15)
1. Mundpflegemittel mit einem Gehalt eines oral verträglichen Trägers und einer Zahnstein verhindernden Komponente, dadurch gekennzeichnet, dass das Mundpflegemittel ein oder mehrere lineare Metaphosphatsalze oder Polyphos-phatsalze in einer den Zahnstein wirksam verhindernden Menge und als Kombinations-Inhibitor gegen eine enzyma-tische Hydrolyse dieser Komponente im Speichel einen Fluoridionen-Lieferanten in einer ausreichenden Menge, um 25 bis 2000 ppm Fluoridionen zu liefern, und 0,05 bis 3 Gew.% eines synthetischen anionischen linearen Carboxy-gruppen aufweisenden Polymeren mit einem Molekulargewicht von 1000 bis 1 000 000 enthält.
2. Mundpflegemittel nach Anspruch 1, dadurch gekennzeichnet, dass es ein wasserlösliches Alkalipyrophosphat, -tripolyphosphat oder -hexametaphosphat oder eine Mischung dieser enthält.
3. Mundpflegemittel nach Anspruch 1 und 2, dadurch gekennzeichnet, dass es etwa 0,1 bis 7 Gew.% an Polyphos-phatsalzen enthält.
4. Mundpflegemittel nach Anspruch 1 bis 3, dadurch gekennzeichnet, dass es ein Dialkalidihydrogenpyrophosphat, Trialkalimonohydrogenpyrophosphat oder Tetraalkalipyro-phosphat oder Mischungen dieser enthält.
5. Mundpflegemittel nach Anspruch 2 oder 4, dadurch gekennzeichnet, dass das Alkali in den Phosphaten unabhängig voneinander Natrium oder Kalium ist.
6. Mundpflegemittel nach Anspruch 1 bis 5, dadurch gekennzeichnet, dass es Tetranatriumpyrophosphat enthält.
7. Mundpflegemittel nach Anspruch 1 bis 6, dadurch gekennzeichnet, dass der Fluoridionen-Lieferant Natriumfluo-rid ist.
8. Mundpflegemittel nach Anspruch 1 bis 7, dadurch gekennzeichnet, dass das lineare, Carboxygruppen aufweisende Polymere ein Copolymeres von Vinylmethylether und Maleinsäure oder -anhydrid ist.
9. Mundpflegemittel nach Anspruch 8, dadurch gekennzeichnet, dass das Copolymere ein Molekulargewicht von etwa 30 000 bis etwa 500 000 hat.
10. Mundpflegemittel nach Anspruch 8, dadurch gekennzeichnet, dass das Copolymere ein Molekulargewicht von etwa 70 000 hat.
11. Mundpflegemittel nach Anspruch 1 bis 10, dadurch gekennzeichnet, dass das Carboxygruppen aufweisende Polymere ein wasserlösliches Alkali- oder Ammoniumsalz ist.
12. Mundpflegemittel nach Anspruch 1 bis 11, dadurch gekennzeichnet, dass es etwa 2 bis 6 Gew.% Polyphosphat-salz oder -salze, eine hinreichende Menge eines Fluoridio-nen-Lieferanten enthält, um 800 ppm bis 1500 ppm Fluoridionen zu liefern und 0,1 bis 2 Gew.% Carboxygruppen aufweisendes Polymer enthält.
13. Mundpflegemittel nach Anspruch 1 bis 12, dadurch gekennzeichnet, dass es als Zahnpasta, Zahngel oder Zahncreme vorliegt und einen pH-Wert von etwa 4,5 bis 9 hat und ein dentalverträgliches Poliermaterial enthält.
14. Mundpflegemittel nach Anspruch 13, dadurch gekennzeichnet, dass das Poliermaterial Kieselsäure ist.
15. Mundpflegemittel nach Anspruch 1 bis 12, dadurch gekennzeichnet, dass es als Mundwasser vorliegt und einen pH-Wert von 4,5 bis 9 hat.
