Beschreibung
Die Erfindung betrifft ein neues mikrobiologisches Verfahren zur Herstellung von 6-Hydroxypyrazinca-rbonsäure ausgehend von 2-Cyanpyrazin.
6-Hydroxypyrazincarbonsäure kann beispielsweise zur Herstellung von Strukturanalogen des Tuber-kulostatikums Pyrazinamid (2-Pyrazincarbonsäureamid) (Römpps-Chemie-Lexikon, 1987, Band 5, S. 3411) oder zur Herstellung von 1,6-Dihydro-6-oxo-2-pyrazincarbonsäure-4-oxid, welches ein Wirkstoff zur Bekämpfung von Eimeria-Infektionen darstellt (Expérimental Parasitology, 1984, 57, S. 55-61) verwendet werden.
Bekannt ist ein mikrobiologisches Verfahren zur Herstellung von 6-Hydroxypyrazincarbonsäure ausgehend von Pyrazincarbonsäure mittels Mikroorganismen der Spezies Alcaligenes faecalis DSM 6269 (EP-A 529 653).
Dieses Verfahren hat jedoch den Nachteil, dass die Umsetzung mit einem Alkali- oder Ammoniumsalz der Pyrazincarbonsäure durchgeführt wird, wobei bei deren Aufarbeitung zur entsprechenden Säure beträchtliche Mengen an Salzen als Abfallprodukt zu Entsorgung anfallen.
Aufgabe der vorliegenden Erfindung war es, diese Nachteile zu beseitigen und ein einfaches, ökologisches und wirtschaftliches mikrobiologisches Verfahren zur Herstellung von 6-Hydroxypyrazincarbonsäure, ausgehend von 2-Cyanpyrazin, zur Verfügung zu stellen.
Diese Aufgabe wurde mit einem neuen Verfahren gemäss Patentanspruch 1 gelöst.
Erfindungsgemäss wird das Verfahren mit den Mikroorganismen der Spezies Alcaligenes faecalis (DSM 6335) und deren Deszendenten und Mutanten durchgeführt, wobei 2-Cyanpyrazin zu 6-Hydroxypyrazincarbonsäure biotransformiert wird und letzteres ins Medium akkumuliert wird.
Diese Mikroorganismen sind bereits in der EP-A 529 653 beschrieben und können mit Hilfe üblicher mikrobiologischer Techniken beispielsweise aus Kläranlagen mit 2-Cyanpyridin als Wachstumssubstrat selektioniert und isoliert werden. Diese Mikroorganismen wurden bei der Deutschen Sammlung für Mikroorganismen und Zellkulturen GmbH (DSM), Mascheroderweg 1 b, D-38124 Braunschweig am 31.1.1991 mit der Bezeichnung DSM 6335 hinterlegt.

Vor der eigentlichen Umsetzung werden diese Mikroorganismen üblicherweise kultiviert (angezogen) und die wirksamen Enzyme der Mikroorganismen zweckmässig mit 2-Cyanpyridin induziert.
Üblicherweise erfolgt die Kultivierung (Anzucht) und die Induktion mit 2-Cyanpyridin in einer Konzentration von 0,01 bis 20 Gew.-%, vorzugsweise in einer Konzentration von 0,1 bis 1 Gew.-%.
Anschliessend können die Mikroorganismen entweder vor der Substratzugabe (2-Cyanpyrazin) mittels üblichen Trennverfahren geerntet werden, oder das Substrat (2-Cyanpyrazin) kann direkt zu den Mikroorganismen zugegeben werden.
Für das eigentliche Verfahren wird dann zweckmässig die Zellsuspension auf eine optische Dichte bei 650 nm von 1 bis 100, vorzugsweise auf eine optische Dichte von 5 bis 80, eingestellt.
Als Medium können die in der Fachwelt üblichen angewendet werden, vorzugsweise wird das Medium verwendet, dessen Zusammensetzung in Tabelle 1 angegeben ist.
Das Substrat (2-Cyanpyrazin) kann zur Produktion von 6-Hydroxypyrazincarbonsäure einmalig oder kontinuierlich zugegeben werden.
Zweckmässig erfolgt die Substratzugabe so, dass die Substratkonzentration im Medium 20 Gew.-%, vorzugsweise so, dass die Substratkonzentration 10 Gew.-% nicht übersteigt. Üblicherweise erfolgt die Umsetzung von 2-Cyanpyrazin zur 6-Hydroxypyrazincarbonsäure mit ruhenden Zellen.
Der pH-Wert der Umsetzung liegt zweckmässig in einem Bereich von 4 bis 10, vorzugsweise in einem Bereich von 5 bis 9.
Zweckmässig wird die Umsetzung bei einer Temperatur von 10 bis 50°C, vorzugsweise bei einer Temperatur von 20 bis 40°C durchgeführt.
Nach einer üblichen Umsetzungsdauer von 1 bis 100 Stunden kann 6-Hydroxypyrazincarbonsäure beispielsweise durch Ansäuern der zellfreien Fermentationslösung, oder z.B. durch Ausfällen des Bariumsalzes von 6-Hydroxypyrazincarbonsäure (analog zur EP-A 529 653) isoliert werden.
Beispiel 1
Umsetzung von 2-Cvanpvrazin zu 6-HvdroxvDvrazincarbonsäure
Fermentation im 5L Massstab: Der Stamm Alcaligenes faecalis. DSM 6335 wurde auf A-N-Agarplat-ten enthaltend 0.2% 2-Cyanpyridin aufbewahrt. Die Vorkultur bestand aus 3 x 100 ml Kulturen gezogen im A-N-Medium (Tabelle 1) mit 2 g/L 2-Cyanpyridin und 1 g/L Hefeextrakt. Das Fermenter-Medium setzte sich wie folgt zusammen: A-N-Medium enthaltend 10 g/L Trypton, 5 g/L Fleischextract, 2 g/L 2-Cyan-pyridin und 1 ml/L Polypropylenglykol 2000.
Nach 6 Stunden Wachstum bei pH 7 und 30°C wurde während 24 Stunden ein Zusatzfeed bestehend aus 20 g/L 2-Cyanpyridin und 130 g/L Ammoniumacetat zum Fermenter-Medium geführt. Die Pumpleistung war ca. 5 ml Feed pro Liter Fermentervolumen und Stunde (600 ml Feed zu 5L Medium). Nach 30 Stunden Fermentationsdauer war die ODeso = 16. Im Medium war kein Cyanpyridin und keine

6-Hydroxypicolinsäure nachweisbar (Totalvolumen war jetzt 5.5L). Zu diesem Zeitpunkt wurde 66 g 2-Cyanpyrazin (5 g/L) zum Fermenter gegeben. Nach weiteren 8 Stunden wurden nochmals 66 g 2-Cyan-pyrazin zum Fermenter gegeben. Nach weiteren 24 Stunden wurde die Fermentation abgebrochen. Die zellfreie Fermentationslösung wurde anschliessend auf 0°C gekühlt und mit konz. HCL bis zu einem pH von 1 angesäuert, um die 6-Hydroxypyrazincarbonsäure auszufällen. Insgesamt wurden 133 g Roh-6-Hydroxypyrazincarbonsäure isoliert, entsprechend einer Ausbeute von 76%, bezogen auf das eingesetzte Cyanpyrazin. Das ausgefällte Material enthielt neben 5-Hydroxypyrazincarbonsäure auch eine weitere, nicht untersuchte Substanz. Alcaliqenes faecalis Kie 31 besitzt eine gute Nitrilase-Aktivität. So konnten mit einer Zellsuspension (OD650 ca. 20) 100 g 2-Cyanpyrazin pro Liter innerhalb von 16 Stunden zur entsprechenden Säure hydrolysiert werden. Die Hydroxylierung der Pyrazincarbonsäure ist der limitierende Schritt der Biotransformation.
Beispiel 2
Isolierung von 6-Hvdroxvpvrazincarbonsäure aus 6-Hvdroxypvrazincarbonsäure-Bariumsalz
128 g (0.91 mol) Roh-6-Hydroxypyrazincarbonsäure wurden zu 4 L H2O gegeben und der pH mit 25% NH4OH auf 7.0 eingestellt. Die Lösung wurde mit 111 g (0.45 mol) BaCl2 • 2H2O versetzt, um die 6-Hydroxypyrazincarbonsäure auszufällen. Die Suspension wurde filtriert und der Rückstand mit 500 mL H2O gewaschen. Der Rückstand wurde zu 2 L H2O gegeben und mit 180 g (1.35 mol) (NH4)2S04 versetzt um Barium auszufällen. Die Suspension wurde 4 h gerührt und anschliessend filtriert (1. Filtration). Das 1. Filtrat wurde auf 0°C gekühlt und mit konz. HCl angesäuert, um 6-Hydroxypyrazincarbonsäure auszufällen. Der Rückstand der ersten Filtration wurde erneut in 2 L H2O aufgeschlämmt und mit 180 g (1.35 mol) (NH4)2SÛ4 versetzt, 4 h gerührt und filtriert (2. Filtration). Das 2. Filtrat wurde gleich behandelt wie das 1. Filtrat. Insgesamt konnten 64 g (0.45 mol) 6-Hydroxypyrazincarbonsäure entsprechend einer Ausbeute von 50% bezogen auf die eingesetzte Rohpyrazincarbonsäure isoliert werden.

Claims (4)
1. Mikrobiologisches Verfahren zur Herstellung von 6-Hydroxypyrazincarbonsäure, dadurch gekennzeichnet, dass man 2-Cyanpyrazin mit Mikroorganismen der Spezies Alcaiiaenes faecalis. DSM 6335, oder deren Mutanten zu 6-Hydroxypyrazincarbonsäure biotransformiert, wobei letzteres im Medium akkumuliert wird.
2. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass die wirksamen Enzyme der Mikroorganismen induziert werden.
3. Verfahren nach einem der Patentansprüche 1 und 2, dadurch gekennzeichnet, dass die Umsetzung unter einmaliger oder kontinuierlicher Substratzugabe erfolgt, so dass die Substratkonzentration 20 Gew.-% nicht übersteigt.
4. Verfahren nach einem der Patentansprüche 1 bis 3, dadurch gekennzeichnet, dass man die Umsetzung bei einem pH von 4 bis 10 und einer Temperatur von 10 bis 50°C durchführt.
