目前对血吸虫疫苗的研究主要集中在照射减弱活疫苗和可溶性抗原疫苗两方面。照射减弱活疫苗的研究历史较长,在制备和应用方面已积累了较成熟的经验,在实验室和动物实验上已取得了较好结果。但该种疫苗还存在以下有待解决的问题,(1)大量注射引起的局部炎症,(2)疫苗的安全性,如过敏反应,(3)疫苗的致病性,如灭活不完全造成的感染,(4)大规模制备疫苗时虫子的来源不足。为了避免上述缺点,人们正在积极从事合成的可溶性抗原疫苗的研究,此种疫苗的重要意义在于,可以采用人工大量合成。经人工合成的疫苗接种量小,可以减少异种蛋白的过敏反应,并可大量制备,且无灭活不完全造成的致病危险,有实际应用前景。
本发明的目的在于提供了一种可用于制备血吸虫疫苗的多肽,其优点在于用其制成的疫苗接种量小,可以减少异种蛋白的过敏反应,无灭活不完全造成的致病危险,并可以采用人工大量合成。
本发明所涉及的有机化合物-多肽,即是血吸虫可溶性抗原疫苗多肽,其特征在于化学结构如下所示:

实施例1
Boc-Val-Gly-OBzl
2.6g Boc-Val溶于THF中,加入少量DMF,加入1.62g HOBt搅拌溶解,冰水浴冷却,加入2.5g DCC搅拌溶解10分钟,将2.11g Gly-OBzl·HCl溶于DMF中,加入1.14-1.18mLNMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,过滤,除去溶剂,固体用乙酸乙酯溶解,然后用5%KHSO4,5%NaCl,5%NaHCO3,5%NaCl水溶液依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩得固体3.96g,收率88.4%,在乙酸乙酯--石油醚(30-60)中重结晶,熔点59-61℃,[α]D 29=-20°(C=1.0,甲醇)。
Val-Gly-OBzl·HCl
3.5g Boc-Val-Gly-OBzl溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,搅拌到脱保护完全,有黏稠状物析出,浓缩得油状物2.5g,收率87%。
Boc-Alla-Val-Gly-OBzl
1.6g Boc-Ala溶于THF中,加入少量DMF,加入1.2g HOBt搅拌溶解,冰水浴冷却,加入1.77g DCC搅拌溶解10分钟,将2.5g Val-Gly-OBzl·HCl溶于DMF中,加入NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,过滤,除去溶剂,固体用乙酸乙酯溶解,然后用5%KHSO4,5%NaCl,5%NaHCO3,5%NaCl水溶液依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩得白色固体3g,收率83%,熔点139-141℃,[α]D 18=-64°(C=0.5,甲醇)。
Ala-Val-Gly-OBzl·HCl
2.5g Boc-Ala-Val-Gly-OBzl溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,有固体物析出,过滤得固体物1.91g,收率90%,熔点166-168℃[α]D 18=-40°(C=0.5,甲醇)。

1.1g Boc-Leu-Ala-Val-Gly-OBzl溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,有固体物析出,浓缩得固体物0.89g,收率92%,熔点196-198℃,[α]D 18=-52°(C=0.5,甲醇)。
Boc-Lys(CBZ)-Leu-Ala-Val-Gly-OBzl

Lys(CBZ)-Leu-Ala-Val-Gly-OBzl·HCl
0.85g Boc-Lys(CBZ)-Leu-Ala-Val-Gly-OBzl溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,浓缩,甲醇-乙醚重结晶,得松黏透明固体物,用4∶1乙酸乙酯∶石油醚(30-60)为洗脱剂经硅胶柱层析得固体0.7g,收率89.4%,熔点192-194℃,[α]D 18=-40°(C=0.5,甲醇)。
Boc-Thr(Bzl)-Gly-OMe
1.85g Boc-Thr(Bzl)溶于THF中,加入少量DMF,加入0.81g HOBt搅拌溶解,冰水浴冷却,加入1.24g DCC搅拌溶解10分钟,将0.625g Gly-OMe·HCl溶于DMF中,加入NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,除去溶剂,固体用乙酸乙酯溶解,然后用5%KHSO4,5%NaCl,5%NaHCO3,5%NaCl水溶液依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩,残渣用乙酸乙酯--石油醚(30-60)重结晶得白色固体1.2g,浓缩母液又得0.81g,收率88%,熔点60-62℃,[α]D 29=+34°(C=1.0,甲醇)。
Thr(Bzl)-Gly-OMe·HCl
2g Boc-Thr(Bzl)-Gly-OMe溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,浓缩,残渣用石油醚(30-60)重结晶得白色固体1.6g,收率96%,熔点135-137℃,[α]D 18=-8°(C=0.5,甲醇)。
Boc-Ser(Bzl)-Thr(Bzl)-Gly-OMe
1.1g Boc-Ser(Bzl)溶于THF中,加入少量DMF,加入0.486g HOBt搅拌溶解,冰水浴冷却,加入0.74g DCC搅拌溶解10分钟,将1.0g Thr(Bzl)-Gly-OMe·HCl溶于DMF中,加入NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,除去溶剂,固体用乙酸乙酯溶解,然后用稀酸,水,稀碱,水依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩得白色固体1.67g,收率95%,熔点81-84℃,[α]D 18=+4°(C=0.5,甲醇)。
Boc-Ser(Bzl)-Thr(Bzl)-Gly-OH
1.5g Boc-Ser(Bzl)-Thr(Bzl)-Gly-OMe溶于适量甲醇中,加入2mL 2N的KOH溶液,室温下搅拌4小时,用10%KHSO4中和到pH=3,过滤除去残渣,浓缩得白色固体1.36g,收率93%,熔点53-55℃,[α]D 18=+28°(C=0.5,甲醇)。
Boc-Ser(Bzl)-Thr(Bzl)-Gly-Lys(CBZ)-Leu-Ala-Val-Gly-OBzl
230mg Boc-Ser(Bzl)-Thr(Bzl)-Gly-OH溶于THF中,加入少量DMF,加入70mgHOBt搅拌溶解,冰水浴冷却,加入92mg DCC搅拌溶解10分钟,将300mg Lys(CBZ)-Leu-Ala-Val-Gly-OBzl·HCl溶于DMF中,加入NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,浓缩除去溶剂,固体用乙酸乙酯溶解,然后用稀酸,水,稀碱,水依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩得白色固体420mg,收率85%,熔点208-212℃,[α]D 29=-10°(C=0.2,甲醇)。
Ser(Bzl)-Thr(Bzl)-Gly-Lys(CBZ)-Leu-Ala-Val-Gly-OBzl·HCl
300mg Boc-Ser(Bzl)-Thr(Bzl)-Gly-Lys(CBZ)-Leu-Ala-Val-Gly-OBzl溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,浓缩得油状物,加入甲醇溶解,过滤,浓缩,加入乙酸乙酯得白色固体250mg,收率90%,熔点192-194℃,[α]D 18=-40°(C=0.2,甲醇)。
Boc-Lys(CBZ)-Gly-OMe
4.5g Boc-Lys(CBZ)溶于THF中,加入少量DMF,加入1.62g HOBt搅拌溶解,冰水浴冷却,加入2.5g DCC搅拌溶解10分钟,将1.25g Gly-OMe·HCl溶于DMF中,滴加NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,除去溶剂,固体用乙酸乙酯溶解,然后用5%KHSO4,5%NaCl,5%NaHCO3,5%NaCl水溶液依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩,残渣用乙酸乙酯-石油醚(30-60)重结晶得白色固体4.1g,收率91%,熔点76-78℃,[α]D 29=-6°(C=1.0,甲醇)。
Lys(CBZ)-Gly-OMe·HCl
2.1g Boc-Lys(CBZ)-Gly-OMe溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,过滤得固体,用甲醇-乙酸乙酯重结晶得白色固体1.65g,收率92%,熔点157-159℃,[α]D 29=30°(C=1.0,甲醇)。
Boc-Leu-Lys(CBZ)-Gly-OMe
0.81g Boc-Leu溶于THF中,加入少量DMF,加入0.473g HOBt搅拌溶解,冰水浴冷却,加入0.742g DCC搅拌溶解10分钟,将1.16g Lys(CBZ)-Gly-OMe·HCl溶于THF和DMF中,滴加NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,除去溶剂,固体用乙酸乙酯溶解,然后用稀酸,水,稀碱,水依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩得白色固体1.6g,收率95%,熔点78-80℃,[α]D 29=-38°(C=1.0,甲醇)。
Leu-Lys(CBZ)-Gly-OMe·HCl
1.6g Boc-Leu-Lys(CBZ)-Gly-OMe溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,过滤得黏稠固体,干燥后重1.32g,收率93%,熔点77-80℃[α]D 18=-10°(C=0.6,甲醇)。
Boc-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OMe
0.89g Boc-Ser(Bzl)溶于THF中,加入少量DMF,加入0.41g HOBt搅拌溶解,冰水浴冷却,加入0.64g DCC搅拌溶解10分钟,将1.1g Leu-Lys(CBZ)-Gly-OMe·HCl溶于DMF中,滴加NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,除去溶剂,固体用乙酸乙酯溶解,然后用稀酸,水,稀碱,水依次洗涤,有机相用无水Na2SO4干燥,过滤,浓缩得白色固体1.3g,收率80%,熔点93-95℃,[α]D 18=-24°(C=0.5,甲醇)。
Boc-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OH
1.2g Boc-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OMe溶于适量甲醇中,加入2mL 2N的KOH溶液,室温下搅拌4小时,用10%KHSO4中和到pH=3,过滤除去残渣,浓缩得白色固体1.12g,收率95%,熔点84-86℃,[α]D 18=-36°(C=0.5,甲醇)。
Ser(Bzl)-Leu-Lys(CBZ)-Gly-OH·HCl
1.0g Boc-Ser-Leu-Lys(CBZ)-Gly-OH溶于乙酸乙酯中,加入4N HCl/乙酸乙酯溶液,室温下搅拌到脱保护完全,过滤,浓缩,残渣用乙酸乙酯--石油醚(30-60)重结晶得白色固体0.84g,收率92%,熔点123-125℃,[α]D 18=-36°(C=0.5,甲醇)。
Boc-Glu(OCHex)-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OH
0.52g Boc-Glu(OCHex)溶于THF中,加入少量DMF,加入0.22g HOBt搅拌溶解,冰水浴冷却,加入0.33g DCC搅拌溶解10分钟,将0.8g Ser(Bzl)-Leu-Lys(CBZ)-Gly-OH·HCl溶于DMF中,滴加NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,浓缩,柱层析分离得白色固体0.945g,收率84%,熔点79-80℃,[α]D 18=-24°(C=0.5,甲醇)。
Boc-Glu(OCHex)-Ser(Bzl)-Leu-Lys(CBZ)-Gly-Ser(Bzl)-Thr(Bzl)-Gly-
Lys(CBZ)-Leu-Ala-Val-Gly-OBzl
66mg Boc-Glu(OCHex)-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OH溶于DMF中,加入15mgHOBt搅拌溶解,冰水浴冷却,加入16mg DCC搅拌溶解10分钟,将70mg Ser(Bzl)-Thr(Bzl)-Gly-Lys(CBZ)-Leu-Ala-Val-Gly-OBzl·HCl溶于DMF中,滴加NMM调pH7.0-7.5,然后滴加到反应液中,搅拌过夜,滤掉DCU,浓缩,以20∶1∶0.4的氯仿∶甲醇∶冰乙酸为展开剂硅胶柱层析分离,浓缩后再以甲醇-水重结晶得白色固体110mg,收率89%,熔点212-215℃,[α]D 18=+5°(C=0.4,甲醇)。
H-Glu-Ser-Leu-Lys-Gly-Ser-Thr-Gly-Lys-Leu-Ala-Val-Gly-OH
在聚四氟乙烯肽脱保护装置中进行脱保护,向反应器中加入100mg Boc-Glu(OCHex)-Ser(Bzl)-Leu-Lys(CBZ)-Gly-Ser(Bzl)-Thr(Bzl)-Gly-Lys(CBZ)-Leu-Ala-yal-Gly-OBzl,1mL苯甲醚,1mL苯甲硫醚,在冰水浴冷却下放入5-8mL经无水三氟化钴干燥过的氟化氢,搅拌反应45分钟,减压抽干,用冷无水乙醚洗涤,得树脂及黏稠物,10%乙酸溶液溶解,过滤去掉残渣,浓缩至2mL左右,以5%乙酸为洗脱剂在Sephadex G-15柱上纯化,纯化两次,冷冻干燥得固体45mg,收率75%,[α]D 18=+30°(C=0.1,10%乙酸溶液)。
此肽还可以把原料Boc-Glu(OCHex)换成Boc-Glu(OBzl),不用把Boc-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OMe先皂化,而是先脱去Boc保护,然后合成Boc-Glu(OBzl)-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OMe,再皂化成Boc-Glu(OBzl)-Ser(Bzl)-Leu-Lys(CBZ)-Gly-OH继续接成十三肽。
下列多肽也可以用实施例1中给出的相似方法来合成和生产,
