Abstract
本发明提出了一种米氮平片剂及其制备方法,由片芯和包衣层组成,片芯包括以下原辅料:米氮平20~45份、乳糖120~240份、羧甲基淀粉钠40~80份、微晶纤维素30~60份、低取代羟丙基纤维素15~30份、硬脂酸镁1~3份与微粉硅胶1~3份;包衣层为含有二氧化钛与聚乙二醇的聚合物包衣层。制备方法,包括以下步骤:1)将片芯的原辅料进行过筛备用;2)将片芯原料进行湿法制粒得到片芯;3)将片芯进行包衣,得到米氮平片剂。该片剂溶出度具有粒度依赖性,药物释放速度稳定,能够避免药物释放速度波动造成副作用的问题。
Description
技术领域
本发明属于抗抑郁药物制剂技术领域,具体涉及一种米氮平片剂及其制备方法。
背景技术
米氮平(Mirtazapine)是众所周知的用于治疗抑郁症的药物,具有四环结构,属于哌嗪-氮卓类化合物。作为一种抗抑郁药,米氮平自成一类。米氮平是中枢突触前膜α2受体拮抗剂,可以增强肾上腺素能的神经传导。它同时阻断中枢的5-HT2和5-HT3受体,米氮平的二种旋光对映体都具有抗抑郁活性,左旋体阻断α2和5-HT2受体,右旋体阻断5-HT3受体。米氮平的抗组织胺受体(H1)特性起着镇静作用。该药有较好的耐受性,几乎无抗胆碱能作用,对心血管系统无影响。米氮平适用于抑郁症,对症状如快感缺乏,精神运动性抑制,睡眠欠佳以及体重减轻均有疗效。也可用于其它症状如:对事物丧失兴趣,自杀观念以及情绪波动,用药后一至二周后起效,其治疗剂量对心血管系统无影响。
米氮平片口服后,其活性成分米氮平很快被吸收(生物利用度约为50%),约2小时后血浆浓度达到高峰,约85%与血浆蛋白结合,平均半衰期为20~40小时;偶见长达65小时。清除半衰期的大小正适合于将服用方式定为每日一次。血药浓度在服药三至四天后达到稳志,此后将无体内聚积现象发生。在所推荐的剂量范围内,“米氮平的药代动力学形式为线性”。米氮平大多被代谢并在服药后几天内通过尿液和粪便排出体外。其主要生化方式为脱甲基及氧化反应在,随后是结合反应。脱甲后的代谢产物与原化合物一样仍具药理活性。肝肾功能不良可引起米氮平清除率降低。
中国专利CN101129341公开了米氮平口腔崩解片另一种制备方法,包含米氮平、崩解剂、填充剂和其他赋形剂的药物配方及其制备方法。然而,现有米氮平制剂仍然存在释放均匀度差、溶出速度慢等问题。
发明内容
本发明提出一种米氮平片剂,该片剂溶出度具有粒度依赖性,药物释放速度稳定,能够避免药物释放速度波动造成副作用的问题。
本发明的技术方案是这样实现的:
一种米氮平片剂,由片芯和包衣层组成,按照重量份数计算,片芯包括以下原辅料:米氮平20~45份、乳糖120~240份、羧甲基淀粉钠40~80份、微晶纤维素30~60份、低取代羟丙基纤维素15~30份、硬脂酸镁1~3份与微粉硅胶1~3份;包衣层为含有二氧化钛与聚乙二醇的聚合物包衣层。
作为优选,本发明一些实施例中,所述聚合物为羟丙甲纤维素、羟乙基纤维素、羟乙基甲基纤维素、羧甲基纤维素钠、羟丙基纤维素、聚乙烯吡咯烷酮、异丁烯酸二甲氨基乙酯-丙烯酸甲酯共聚物、丙烯酸乙酯-异丁烯酸甲酯共聚物、甲基纤维素和乙基纤维素的一种或一种以上。
作为优选,本发明一些实施例中,所述包衣层为片芯重量的1.8%~2.4%。
本发明的另一个目的是提供一种米氮平片剂的制备方法,包括以下步骤:
1)将米氮平、硬脂酸镁、羧甲基淀粉钠、微粉硅胶、低取代羟丙纤维素、微晶纤维素、乳糖过筛备用;
2)取米氮平、羧甲基淀粉钠、乳糖、纯化水,并加入到湿法制粒机中制粒,并将制得的颗粒放到干燥机中干燥;并将干燥后的颗粒放到振动筛中进行整粒;
3)将步骤2)整粒得到的颗粒放入多向运动混合机中,并加入低取代羟丙纤维素、微晶纤维素、微粉硅胶与硬脂酸镁,混合、压片,得到片芯;
4)向步骤3)得到的片芯中加入包衣溶液,进行包衣,得到米氮平片剂。
作为优选,本发明一些实施例中,步骤2)中所述纯化水的重量份数为20~30份。
米氮平(1,2,3,4,10,14β-六氢-2-甲基吡唑并[2,1-a]吡啶并[2,3-c][2]苯并氮杂),是指包括化合物本身以及它的可药用盐类。可以按照van der Burg的美国专利No.4,062,848制备此化合物。
本发明米氮平包括以基本上与其它对映异构体分离的形式(即具有的对映体的纯度大于95%,并且最好大于99%)存在的米氮平的单个(R)和(S)对映异构体及其盐,以及包括外消旋混合物的任何比例的对映异构体的混合物。
由于米氮平水中溶解度较低,其粒度直接影响制剂的产品质量,本发明下所得米氮平剂质量稳定、制备操作简单、生产成本低,适于工业化大生产。
本发明中米氮平片不仅对抑郁症有良好的治疗效果,而且能够降低米氮平的不良反应发生率;并且本发明提供的米氮平片剂采用羧甲基淀粉钠、微粉硅胶以及微晶纤维素作为药物赋形剂,提高药物稳定性。包衣层为含有二氧化钛与聚乙二醇的聚合物包衣层,使其具有普通片剂所不具有的缓释性能,一定程度上能够控制米氮平在人体内稳定释放,从而使得米氮平片具有长效性,且能够避免传统米氮平片在人体释放速度波动而引起的副作用。
具体实施方式
实施例1
一种米氮平片剂,由片芯和包衣层组成,按照重量份数计算,片芯包括以下原辅料:米氮平30份、乳糖200份、羧甲基淀粉钠60份、微晶纤维素40份、低取代羟丙基纤维素20份、硬脂酸镁2份与微粉硅胶2份;包衣层为含有二氧化钛与聚乙二醇的聚合物包衣层。包衣层为片芯重量的2%。
制备方法,包括以下步骤:
1)将米氮平、硬脂酸镁、羧甲基淀粉钠、微粉硅胶、低取代羟丙纤维素、微晶纤维素、乳糖过筛备用;
2)取米氮平、羧甲基淀粉钠、乳糖、纯化水,并加入到湿法制粒机中制粒,并将制得的颗粒放到干燥机中干燥;并将干燥后的颗粒放到振动筛中进行整粒;
3)将步骤2)整粒得到的颗粒放入多向运动混合机中,并加入低取代羟丙纤维素、微晶纤维素、微粉硅胶与硬脂酸镁,混合、压片,得到片芯;
4)采用常规包衣设备、使用胃溶型欧巴代对制剂进行包衣。
实施例2
一种米氮平片剂,由片芯和包衣层组成,按照重量份数计算,片芯包括以下原辅料:米氮平20份、乳糖120份、羧甲基淀粉钠80份、微晶纤维素30份、低取代羟丙基纤维素150份、硬脂酸镁1份与微粉硅胶1份;包衣层为含有二氧化钛与聚乙二醇的聚合物包衣层。包衣层为片芯重量的1.8%。
制备方法,包括以下步骤:
1)将米氮平、硬脂酸镁、羧甲基淀粉钠、微粉硅胶、低取代羟丙纤维素、微晶纤维素、乳糖过筛备用;
2)取米氮平、羧甲基淀粉钠、乳糖、纯化水,并加入到湿法制粒机中制粒,并将制得的颗粒放到干燥机中干燥;并将干燥后的颗粒放到振动筛中进行整粒;
3)将步骤2)整粒得到的颗粒放入多向运动混合机中,并加入低取代羟丙纤维素、微晶纤维素、微粉硅胶与硬脂酸镁,混合、压片,得到片芯;
4)采用常规包衣设备、使用胃溶型欧巴代对制剂进行包衣。
实施例3
一种米氮平片剂,由片芯和包衣层组成,按照重量份数计算,片芯包括以下原辅料:米氮平45份、乳糖240份、羧甲基淀粉钠40份、微晶纤维素60份、低取代羟丙基纤维素30份、硬脂酸镁3份与微粉硅胶3份;包衣层为含有二氧化钛与聚乙二醇的聚合物包衣层。包衣层为片芯重量的2.4%。
制备方法,包括以下步骤:
1)将米氮平、硬脂酸镁、羧甲基淀粉钠、微粉硅胶、低取代羟丙纤维素、微晶纤维素、乳糖过筛备用;
2)取米氮平、羧甲基淀粉钠、乳糖、纯化水,并加入到湿法制粒机中制粒,并将制得的颗粒放到干燥机中干燥;并将干燥后的颗粒放到振动筛中进行整粒;
3)将步骤2)整粒得到的颗粒放入多向运动混合机中,并加入低取代羟丙纤维素、微晶纤维素、微粉硅胶与硬脂酸镁,混合、压片,得到片芯;
4)采用常规包衣设备、使用胃溶型欧巴代对制剂进行包衣。
试验例
释放度实验
按照释放度测定方法(中国药典2010版二部附录XD第一法),采用溶出度测试法第一法装置,以磷酸盐缓冲液为溶剂,依法操作。含量测定照高效液相色谱法(中国药典2010年二部附录VD)测定。

从释放度实验数据来看,本发明的米氮平片剂的药物释放可以持续24小时,且释放线性良好。

Claims (5)
1.一种米氮平片剂,其特征在于,由片芯和包衣层组成,按照重量份数计算,片芯包括以下原辅料:米氮平20~45份、乳糖120~240份、羧甲基淀粉钠40~80份、微晶纤维素30~60份、低取代羟丙基纤维素15~30份、硬脂酸镁1~3份与微粉硅胶1~3份;包衣层为含有二氧化钛与聚乙二醇的聚合物包衣层。
2.根据权利要求1所述的米氮平片剂,其特征在于,所述聚合物为羟丙甲纤维素、羟乙基纤维素、羟乙基甲基纤维素、羧甲基纤维素钠、羟丙基纤维素、聚乙烯吡咯烷酮、异丁烯酸二甲氨基乙酯-丙烯酸甲酯共聚物、丙烯酸乙酯-异丁烯酸甲酯共聚物、甲基纤维素和乙基纤维素的一种或一种以上。
3.根据权利要求1或2所述的米氮平片剂,其特征在于,所述包衣层为片芯重量的1.8%~2.4%。
4.如权利要求1所述的米氮平片剂的制备方法,其特征在于,包括以下步骤:
1)将米氮平、硬脂酸镁、羧甲基淀粉钠、微粉硅胶、低取代羟丙纤维素、微晶纤维素、乳糖过筛备用;
2)取米氮平、羧甲基淀粉钠、乳糖、纯化水,并加入到湿法制粒机中制粒,并将制得的颗粒放到干燥机中干燥;并将干燥后的颗粒放到振动筛中进行整粒;
3)将步骤2)整粒得到的颗粒放入多向运动混合机中,并加入低取代羟丙纤维素、微晶纤维素、微粉硅胶与硬脂酸镁,混合、压片,得到片芯;
4)向步骤3)得到的片芯中加入包衣溶液,进行包衣,得到米氮平片剂。
5.根据权利要求4所述的米氮平片剂的制备方法,其特征在于,步骤2)中所述纯化水的重量份数为20~30份。
