Abstract
本发明提供一种复合微生物发酵膨化饲料的制备方法,依次包括如下步骤:(1)粉碎:饲料物料粉碎过筛,混匀后得饲料粉末;(2)膨化:饲料粉末经双轴调质器蒸汽加热后,经膨化机挤压膨化得膨化饲料;(3)冷却:膨化饲料经风冷机冷却至45℃后出料,得湿饲料;(4)接种:喷洒接种复合微生物发酵剂混合液至湿饲料;(5)密封发酵:密封包装,恒温发酵,得复合微生物发酵膨化饲料。本发明将膨化后的湿态饲料作为有益微生物的发酵基质,膨化挤压作用使蛋白质结构变为松散状态,容易被微生物和消化酶作用生成小肽、胨类物质,提高饲料蛋白质的消化吸收率,所得发酵膨化饲料的营养含量满足水产动物的需求。

技术领域
本发明属于生物发酵饲料制备领域,尤其涉及一种复合微生物发酵膨化饲料的制备方法。
背景技术
随着水产养殖业规模化、集约化和产业化程度的不断提升,养殖过程中有害污染物的积累严重破坏养殖水体生态环境,导致水产养殖动物应激不断增加、免疫力下降,各种养殖疾病频繁发生,严重制约水产养殖业的可持续健康发展。
在水产养殖过程通常使用物理方法、化学药物法、抗生素法和微生物法等进行养殖水体环境的改善和病害的控制。物理方法使用具有吸附能力的物质,如活性硅藻土、沸石粉等,将亚硝酸根吸附在其结构中,这种方法见效快,但是药品用量大,容易引起重金属残留超标,而且该法并不能根本消除氨氮和亚硝酸盐,故容易反弹;化学方法主要使用二氧化氯、臭氧、双氧水、二氯异氰脲酸等氧化物,氨氮和亚硝酸盐遇强氧化剂时会改变氮的价态变成无毒的硝酸根离子,但化学氧化物是对鱼类刺激较大,降低鱼类摄食量,破坏水体微生态系统;而抗生素的长期使用也带来了一系列负面影响,如病原细菌的耐药性产生,鱼体抗生素的残留超标,破坏鱼类肠道微生物平衡等;微生物发酵技术应用于水产绿色高效养殖是一种新兴方法,常用的有益微生物如枯草芽孢杆菌、凝结芽孢杆菌、乳酸菌、光合细菌等,微生物的繁殖消耗水体中饲料的残渣、排泄物,能减轻养殖自身污染、改良池塘底部环境,提高鱼类的免疫力。
近年来,随着高密度水产养殖的发展,膨化饲料得到广泛的推广,因为膨化饲料的利用率比沉水性颗粒饲料高,对水体污染相对减轻,但膨化饲料生产过程中温度达到120℃以上,对热敏性的物质破坏性大,如微生物、酶制剂和维生素等。虽然膨化饲料的后喷涂工艺、微胶囊包埋技术的应用,提高了膨化饲料的品质,但这些工艺还存在一些不足之处,如后喷涂工艺存在喷涂不均匀,烘干后的饲料中微生物处理休眠状态,在鱼体内需要一定活化时间,导致在肠道的滞留时间短,对肠道微生态体系改善作用受到限制;微胶囊技术也能减少饲料的中热敏物质及微生物的损失,但其高成本带来较大的局限性;还有专利(如申请号201910717478.9)将干燥的膨化饲料与发酵剂、水混合,发酵一定时间制备发酵膨化饲料,但这种方法的工艺复杂、生产效率低、加工能耗高。
发明内容
为克服现有技术存在的缺陷和不足,本发明的目的在于提供一种复合微生物发酵膨化饲料的制备方法。
本发明采取的技术方案为:
一种复合微生物发酵膨化饲料的制备方法,依次包括如下步骤:
(1)粉碎:饲料物料粉碎过筛,混匀后得饲料粉末;
(2)膨化:饲料粉末经双轴调质器蒸汽加热后,经膨化机挤压膨化得膨化饲料;
(3)冷却:膨化饲料经风冷机冷却至40℃~45℃后出料,得湿饲料;
(4)接种:喷洒接种复合微生物发酵剂混合液至湿饲料;
(5)密封发酵:密封包装,恒温发酵,得复合微生物发酵膨化饲料。
优选的,步骤(1)中所述粉碎包括粗粉碎和超微粉碎,所述粗粉碎所得粉末细度为20目筛通过率93%~99%,优选为98%;所述超微粉碎所得粉末细度为80目筛通过率为75%~90%,优选为85%。
优选的,步骤(2)所述蒸汽加热为调节双轴调质器内含水量为20%~30%,通入蒸汽提高饲料粉末温度至90℃~110℃,保温。
优选的,步骤(2)所述挤压膨化为调节膨化机膨胀倍数为1.5~1.7倍,膨化腔中料温为120℃~135℃,出料模板孔径为2mm~6mm。
优选的,步骤(4)所述复合微生物发酵剂混合液为复合微生物发酵剂与40℃~60℃温水混合液,所述湿饲料含水量为25%~35%。
优选的,步骤(5)中所述恒温发酵为35℃~40℃下恒温发酵。
优选的,上述复合微生物发酵剂的质量组成为:复合微生物发酵液1~5份、复合酶制剂0.1~0.5份、维生素预混料0.2~0.6份,葡萄糖1~3份。
优选的,上述复合微生物发酵液由凝结芽孢杆菌发酵液、枯草芽孢杆菌发酵液、植物乳杆菌发酵液按1~3:1~3:2~5比例复合;所述复合酶制剂含中性蛋白酶3~6万IU/g、植酸酶1~3万IU/g、木聚糖酶1~3万IU/g。
优选的,上述饲料物料的组成为:鱼粉、豆粕、菜籽粕、速爆大豆、棉粕、高筋面粉、麦麸、豆油、油糠、磷酸二氢钙、微量元素预混料。
优选的,上述微量元素选自:CoCl2·6H2O、FeSO4·7H2O、ZnSO4·7H2O、CuSO4·5H2O、MnSO4·H2O的至少一种。
优选的,上述饲料物料的质量组成为:鱼粉8~16份、豆粕15~25份、菜籽粕10~25份、速爆大豆4~10份、棉粕4~10份、高筋面粉7~16份、麦麸3~8份、豆油1~3份、油糠6~16份、磷酸二氢钙1.5~2份、微量元素预混料1~3份。
本发明的有益效果为:
(1)本发明将膨化后的湿态饲料作为有益微生物的发酵基质,膨化挤压作用使蛋白质结构变为松散状态,容易被微生物和消化酶作用生成小肽、胨类物质,提高饲料蛋白质的消化吸收率,所得发酵膨化饲料的营养含量满足水产动物的需求,可以直接投喂;省去了传统颗粒饲料投喂前拌和微生物的工序,这工序操作繁琐、效果不稳定。
(2)与普通膨化饲料相比,本发明饲料经过体外预消化发酵处理,膨化的湿态饲料不需烘干处理,成本低;本发明采用补料高密度发酵培养微生物,确保发酵膨化饲料中微生物含量高,活性代谢产物丰富,有效调理动物肠道微生态稳定,提高动物的抗病能力,减少养殖的抗生素使用,提高水产品的食品安全性;净化养殖水体环境,减少养殖排水对水环境的污染。
(3)与粉状生物发酵饲料相比,本发明饲料的高温膨化消除原料中杂菌,提高发酵饲料的生物安全性,膨化使纤维形成的空隙有利于有益微生物的生长,发酵膨化饲料的耐水性更高,浮于水面,更易观察鱼的采食情况,营养物质不容易溶解到水中,饲料利用率更高,减少饲料对水体的污染。

具体实施方式
为了使本发明的发明目的、技术方案及其技术效果更加清晰,以下结合具体实施方式,对本发明进行进一步详细说明。应当理解的是,本说明书中描述的具体实施方式仅仅是为了解释本发明,并非为了限定本发明。
方便比较起见,本发明实施例或对比例中微量元素为:每千克饲料含有0.3mg的CoCl2·6H2O、120.1mg的FeSO4·7H2O、300.5mg的ZnSO4·7H2O、20.5mg的CuSO4·5H2O、50.8mg的MnSO4·H2O。
复合微生物发酵液中凝结芽孢杆菌发酵液、枯草芽孢杆菌发酵液、植物乳杆菌发酵液的制备如下:
凝结芽孢杆菌发酵液制备:
一级培养:将试管斜面凝结芽孢杆菌菌种接种于一级培养基(蛋白胨10.0g,牛肉膏8.0g,酵母膏5.0g,柠檬酸氢二铵2.0g,葡萄糖25.0g,乙酸钠5.0g,磷酸氢二钾2.0g,硫酸镁0.58g,硫酸锰0.25g,蒸馏水1000mL,调节pH值6.0~6.5;灭菌温度121℃下灭菌15min),1000mL三角瓶的装液量为150mL,培养温度40℃,摇床转速200rpm,培养时间18h,得到一级培养液。
二级高密度培养:将一级培养液按用量2%接种到二级培养基(大豆蛋白胨20g/L,酵母膏5g/L,豆粕粉20g/L,玉米浆干粉10g/L,葡萄糖20g/L,硫酸镁1g/L,硫酸锰0.18g/L,磷酸氢二钾3g/L,碳酸钙15g/L,氯化钠4g/L,调节pH值6.5~7.0;灭菌温度121℃下培养15min。),发酵罐的初始装液量为50%,培养温度40℃,搅拌转速150rpm,溶氧值维持30%以上,发酵到第20h后开始连续补加质量浓度20%无菌葡萄糖溶液,补加速度15mL/L/h,到第30h时停止补料,发酵全过程缓慢滴加质量浓度18%氨水维持pH值6.5~7.0,得到凝结芽孢杆菌发酵液,活菌数2.2×1010cfu/g。
枯草芽孢杆菌发酵液制备:
一级培养:将试管斜面枯草芽孢杆菌菌种接种于一级培养基(蛋白胨10.0g,酵母膏5.0g,葡萄糖15.0g,磷酸氢二钾2.0g,硫酸镁0.5g,蒸馏水1000mL,调节pH值6.0~6.5,灭菌温度121℃下灭菌15min。),1000mL三角瓶的装液量为150mL,培养温度37℃,摇床转速200rpm,培养时间18h,得到一级培养液。
二级高密度培养:将一级培养液按用量2%接种到二级培养基(大豆蛋白胨5g/L,豆粕粉30g/L,玉米浆干粉10g/L,玉米粉25g/L,硫酸镁1.2g/L,磷酸氢二钾3g/L,氯化钠4g/L,淀粉酶1g/L,调节pH值6.5~7.0。培养基加热到90℃时保温30min,然后升温到121℃,保温15min灭菌。),发酵罐的初始装液量为50%,培养温度37℃,搅拌转速150rpm,溶氧值维持30%以上,发酵第18h后开始连续补加质量浓度20%无菌葡萄糖溶液,补料速度15mL/L/h,到第26h时停止补料,发酵全过程维持pH值6.5~7.0,得到枯草芽孢杆菌发酵液,活菌数1.8×1010cfu/g。
植物乳杆菌发酵液的制备:
一级培养:将试管半固体植物乳杆菌菌种接种于一级培养基(蛋白胨10.0g、牛肉膏10.0g、酵母膏5.0g、柠檬酸氢二铵2.0g、葡萄糖20.0g、吐温80 1.0mL、乙酸钠5.0g、磷酸氢二钾2.0g、硫酸镁0.58g、硫酸锰0.25g、水1000mL,面pH值6.2~6.6,灭菌温度115℃下培养15min。),100mL三角瓶的装液量为80mL,培养温度37℃,厌氧培养时间24h,得到一级培养液。
二级培养:将一级培养液按用量3%接种到二级培养基(蛋白胨10.0g/L、牛肉膏5.0g/L、大豆蛋白15g/L、柠檬酸氢二铵2.0g/L、葡萄糖30.0g/L、乙酸钠5.0g/L、磷酸氢二钾2.0g/L、硫酸镁0.58g/L、硫酸锰0.25g/L,调节pH值5.5~6.0,灭菌温度115℃下灭菌15min。),发酵罐的装液量为80%,培养温度37℃,厌氧发酵36h,每2h搅拌1次,每次1min,发酵全过程用质量浓度15%氨水补料维持pH值6.5~7.0,得到植物乳杆菌发酵液,活菌数3×109cfu/g。
实施例1:一种复合微生物发酵膨化饲料的制备方法,依次包括如下步骤:
(1)粗粉碎:分别将鱼粉8份、豆粕24份、菜籽粕16份、速爆大豆6份、棉粕10份、麦麸3份,粉碎至20目筛通过率98%;然后与高筋面粉13.3份、豆油3份、油糠10份、磷酸二氢钙2份、微量元素预混料1份,混合均匀得饲料粗粉末;
(2)超微粉碎:将饲料粗粉末经超微粉碎至80目筛的通过率85%,得饲料细粉末;
(3)膨化:将饲料细粉末输送到双轴调质器内,调节器含水量为22%,通入蒸汽提高饲料细粉末温度到98℃,保温5min,然后输送到膨化机中,膨化机中料温120℃,膨化出料模板孔径2mm;
(4)冷却:采用网带式风冷机冷却,网带上饲料堆积厚度为10cm,冷却时间6min,出料温度为45℃,得湿饲料;
(5)搅拌接种:将复合微生物发酵液0.15份、复合酶制剂0.3份、维生素预混料0.2份、葡萄糖3份溶于10份45℃的温度,并边搅拌边喷洒至湿饲料,得含水量为30%的膨化饲料;
(6)密封发酵:将膨化饲料装入内附塑料薄膜袋的编织袋,热熔薄膜袋封口,恒温35℃发酵1天,得复合微生物发酵膨化饲料。
实施例2:一种复合微生物发酵膨化饲料的制备方法,依次包括如下步骤:
(1)粗粉碎:分别将鱼粉12份、豆粕15份、菜籽粕20份、速爆大豆6份、棉粕10份、麦麸6.6份,粉碎至20目筛通过率98%;然后与高筋面粉13.4份、豆油3份、油糠10份、磷酸二氢钙2份、微量元素预混料1份,混合均匀得饲料粗粉末;
(2)超微粉碎:将饲料粗粉末经超微粉碎至80目筛的通过率85%,得饲料细粉末;
(3)膨化:将饲料细粉末输送到双轴调质器内,调节器含水量为28%,通入蒸汽提高饲料细粉末温度到98℃,保温5min,然后输送到膨化机中,膨化机中料温130℃,膨化出料模板孔径2mm;
(4)冷却:采用网带式风冷机冷却,网带上饲料堆积厚度为10cm,冷却时间10min,出料温度为45℃,得湿饲料;
(5)搅拌接种:将复合微生物发酵液0.3份、复合酶制剂0.5份、维生素预混料0.2份、葡萄糖3份溶于20份45℃的温度,并边搅拌边喷洒至湿饲料,得含水量为33%的膨化饲料;
(6)密封发酵:将膨化饲料装入内附塑料薄膜袋的编织袋,热熔薄膜袋封口,恒温45℃发酵1天,得复合微生物发酵膨化饲料。
对比例1:一种复合微生物发酵膨化饲料的制备方法,依次包括如下步骤:
(1)粗粉碎:分别将鱼粉12份、豆粕15份、菜籽粕20份、速爆大豆6份、棉粕10份、麦麸6.6份,粉碎至20目筛通过率98%;然后与高筋面粉13.4份、豆油3份、油糠10份、磷酸二氢钙2份、微量元素预混料1份、复合微生物发酵液0.3份、复合酶制剂0.5份、维生素预混料0.2份、葡萄糖3份,混合均匀得饲料粗粉末;
(2)超微粉碎:将饲料粗粉末经超微粉碎至80目筛的通过率85%,得饲料细粉末;
(3)膨化:将饲料细粉末输送到双轴调质器内,调节器含水量为28%,通入蒸汽提高饲料细粉末温度到98℃,保温5min,然后输送到膨化机中,膨化机中料温130℃,膨化出料模板孔径2mm;
(4)烘干:进风温度125℃,干燥时间15min,得含水量为10%的干燥饲料;
(5)冷却:采用网带式风冷机冷却,网带上饲料堆积厚度为10cm,冷却时间12min,出料温度为40℃,得冷却饲料;
(6)密封发酵:将膨化饲料装入内附塑料薄膜袋的编织袋,热熔薄膜袋封口,得膨化饲料。
对比例2:一种复合微生物发酵膨化饲料的制备方法,依次包括如下步骤:
(1)粗粉碎:分别将鱼粉12份、豆粕15份、菜籽粕20份、速爆大豆6份、棉粕10份、麦麸6.6份,粉碎至20目筛通过率98%;然后与高筋面粉13.4份、豆油3份、油糠10份、磷酸二氢钙2份、微量元素预混料1份,混合均匀得饲料粗粉末;
(2)超微粉碎:将饲料粗粉末经超微粉碎至80目筛的通过率85%,得饲料细粉末;
(3)膨化:将饲料细粉末输送到双轴调质器内,调节器含水量为28%,通入蒸汽提高饲料细粉末温度到98℃,保温5min,然后输送到膨化机中,膨化机中料温130℃,膨化出料模板孔径2mm;
(4)烘干:进风温度125℃,干燥时间15min,得含水量为10%的干燥饲料;
(5)冷却:采用网带式风冷机冷却,网带上饲料堆积厚度为10cm,冷却时间12min,出料温度为40℃,得冷却饲料;
(6)搅拌接种:将复合微生物发酵液0.3份、复合酶制剂0.5份、维生素预混料0.2份、葡萄糖3份溶于20份45℃的温度,并边搅拌边喷洒至冷却饲料,得含水量为的膨化饲料;
(7)密封发酵:将膨化饲料装入内附塑料薄膜袋的编织袋,热熔薄膜袋封口,恒温45℃发酵1天,得膨化饲料。
将上述实施例1、2和对比例1、2制得的饲料进行如下检测,各检测及其依据的方法如下:
(1)水分检测:饲料中水分的测定(GB/T 6435);
(2)蛋白质检测:凯氏定氮法(GB/T 6432);
(3)小肽检测:大豆肽的检测(GB/T 22492)
(4)总酸检测:饲料酸化剂中柠檬酸、富马酸和乳酸的测定,高效液相色谱法(GB/T23877);
(5)菌落总数检测:食品安全国家标准,菌落总数测定(GB/T 4789)
(6)粗灰分检测:饲料中粗灰分的测定(GB/T 6438);
(7)浮水率测定:分别量取实施例1、2和对比例制备的饲料50粒,放入1000mL三角瓶,倒入500mL温度为30℃自来水,震动转速60rpm,观察2h后浮在水面的完整饲料颗粒数,记为m,计算其浮水率=m/50×100%。
经检测,实施例1、2和对比例制得的饲料检测结果如表1所示:

由表1可看出,实施例比对比例的膨化饲料的小肽、菌落总数含量高,由于微生物和酶制剂不耐热,膨化高温条件导致两者失去活性;而且微生物的繁殖需要饲料含水分均匀,对比例2的膨化饲料干燥至含水10%,再加水搅拌至含水量为33%时,由于其吸水性差,导致饲料发酵时底部饲料含水量过高,容易发酵,饲料容易烂、浮水性差。
试验例:
选择均重为约50g/条的斑点叉尾鮰240尾,用3%的食盐水溶液对试验鮰鱼苗消毒后,分为对照组和试验组,对照组投喂对比例1膨化饲料,试验组投喂实施例2发酵膨化饲料,每个组设3个重复,每个重复放养40尾,于300L水体(水族箱规格为70cm×70cm×80cm)中,预饲期7天饲喂对应的饲料,预饲期每天换水30%,正试饲养10天,正试期不换水,每2天补足水到原来水位。每天投喂3次,时间为6:00、12:00和18:30,以总鱼体重的3%--5%投喂。养殖过程保持水温26±1℃,24小时连续充气保持溶解氧6mg/L以上。
试验期间每2天定期采取水样,采取水样前用水样淌洗水样瓶和瓶塞3次,再在水箱下部相同水位处取100mL水样进行氨氮(NH3-N)浓度、亚硝酸盐氮浓度的测定;氨氮检测用钠氏试剂光度法;NO2 --N浓度采用N-(1-萘基)-乙二胺光度法测定,试验期间每个水族箱进行同样的管理。检测结果如图1、图2所示。
从图1、图2的数据分析,本发明生物发酵膨化饲料比普通膨化饲料能够降低养殖水体中的氨氮、亚硝酸盐的浓度,提高养殖水体的水质稳定性,减少频繁换水对动物的应激反应。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
Claims (10)
1.一种复合微生物发酵膨化饲料的制备方法,依次包括如下步骤:
(1)粉碎:饲料物料粉碎过筛,混匀后得饲料粉末;
(2)膨化:饲料粉末经双轴调质器蒸汽加热后,经膨化机挤压膨化得膨化饲料;
(3)冷却:膨化饲料经风冷机冷却至40℃~55℃后出料,得湿饲料;
(4)接种:喷洒接种复合微生物发酵剂混合液至湿饲料;
(5)密封发酵:密封包装,恒温发酵,得复合微生物发酵膨化饲料。
2.根据权利要求1所述的制备方法,其特征在于:步骤(1)中所述粉碎包括粗粉碎和超微粉碎,所述粗粉碎所得粉末细度为20目筛通过率93%~99%;所述超微粉碎所得粉末细度为80目筛通过率为75%~90%。
3.根据权利要求1所述的制备方法,其特征在于:步骤(2)所述蒸汽加热为调节双轴调质器内含水量为20%~30%,通入蒸汽提高饲料粉末温度至90℃~110℃,保温。
4.根据权利要求1所述的制备方法,其特征在于:步骤(2)所述挤压膨化为调节膨化机膨胀倍数为1.5~1.7倍,膨化腔中料温为120℃~135℃,出料模板孔径为2mm~6mm。
5.根据权利要求1所述的制备方法,其特征在于:步骤(4)所述复合微生物发酵剂混合液为复合微生物发酵剂与40℃~60℃温水混合液,所述湿饲料含水量为25%~35%。
6.根据权利要求1所述的制备方法,其特征在于:步骤(5)中所述恒温发酵为35℃~40℃下恒温发酵。
7.根据权利要求1或5所述的制备方法,其特征在于:所述复合微生物发酵剂的质量组成为:复合微生物发酵液1~5份、复合酶制剂0.1~0.5份、维生素预混料0.2~0.6份、葡萄糖1~3份。
8.根据权利要求7所述的制备方法,其特征在于:所述复合微生物发酵液由凝结芽孢杆菌发酵液、枯草芽孢杆菌发酵液、植物乳杆菌发酵液按体积比1~3:1~3:2~5比例复合;所述复合酶制剂含中性蛋白酶3~6万IU/g、植酸酶1~3万IU/g、木聚糖酶1~3万IU/g。
9.根据权利要求1~6任一项所述的制备方法,其特征在于:所述饲料物料的组成为:鱼粉、豆粕、菜籽粕、速爆大豆、棉粕、高筋面粉、麦麸、豆油、油糠、磷酸二氢钙、微量元素预混料。
10.根据权利要求1~6任一项所述的制备方法,其特征在于:所述饲料物料的质量组成为:鱼粉8~16份、豆粕15~25份、菜籽粕10~25份、速爆大豆4~10份、棉粕4~10份、高筋面粉7~16份、麦麸3~8份、豆油1~3份、油糠6~16份、磷酸二氢钙1.5~2份、微量元素预混料1~3份。
