技术领域
本发明涉及宠物食品技术领域,特别涉及一种含NADH的组合物及其制备方法。
背景技术
根据犬猫运动和老年、体弱、术后、疾病康复身体机体特点,尤其是赛犬在大量运动后,机体处在高度缺氧状态时依靠糖酵解供给能量,乳酸是糖无氧酵解的产物,长时间剧烈运动使机体中乳酸积累过多会影响体内环境的相对稳定和体内的正常代谢过程。血乳酸水平可以反映有氧代谢能力、疲劳的产生和消除速度。
现阶段,全球宠物产业处于高速发展期,宠物食品、宠物营养品的发展百花盛开,但是目前市场上暂无专门针对宠物运动的营养品。
发明内容
本发明的目的是提供一种含NADH的组合物及其制备方法,解决上述现有技术问题中的一个或者多个。
一方面,本发明提供的一种含NADH的组合物,该组合物包括第一均质液,所述第一均质液包括NADH和二十八烷醇,并使用二十八烷醇包裹NADH,
其中:NADH与二十八烷醇的重量比为10:(4-10)。
其中:本发明通过犬猫运动过程中身体机能的变化特点,利用公司现有的制备稳定的原料NADH(烟酰胺腺嘌呤二核苷酸)作为营养配方核心产品,搭配相应功效原料,协同增效共奏快速提升体力,恢复身体机能,提高疲劳能力和运动能力,减缓运动损伤等作用。配方中NADH为烟酰胺腺嘌呤二核苷酸的还原态,其通过电子的传递,释放能量并被氧化磷酸化的过程利用,生产三磷酸腺苷,快速为宠物运动过程中补充机体能量。用二十八烷醇包裹保护NADH的同时,协同增效还原型的烟酰胺腺嘌呤二核苷酸对运动犬猫细胞的保护,降低宠物犬猫在运动过程中产生的氧化损伤,提升犬猫运动的机体修复能力。
在某些实施方案中,该组合物还包括第二均质液,所述第二均质液包括脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油和磷脂。
在某些实施方案中,该组合物还包括第三均质液,所述第三均质液包括枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水和单硬脂酸甘油酯。
在某些实施方案中,NADH、二十八烷醇和1,6-二磷酸果糖三钠盐的重量为:10:(4-10):(3.3-10)。
其中:本发明通过犬猫消化特点:其胃酸浓度高,肠道比较短,能够快速消化吸收肉食,相对于人类有更高的蛋白质需求,犬猫,尤其是猫能够在肝脏内通过糖异生作用从蛋白质代谢合成它们所需要的葡萄糖。通过NADH,1,6-二磷酸果糖三钠盐及二十八烷醇创造性的物料筛选和比例搭配,探寻出最佳的NADH肠溶包封和能量速补配方。

其中:本发明提供了科学合理的营养配方,具体如下:
合理搭配抗氧化剂辅酶Q10和维生素E,清除运动过程中游离自由基,激活犬猫细胞和细胞能量的营养,减少因长时间运动引起线粒体膜磷脂的减少所造成的疲劳感。同时兼具整个液体制剂的抗氧化剂,稳定产品质量。
1,6-二磷酸果糖三钠盐、磷脂、南极磷虾油为参与机体代谢和维持机体代谢的营养成分,维持肌肉功能,有助于长时间高强度运动中最佳运动表现的维持;
南极磷虾油中含有的虾青素,是一种红色类胡萝卜素和强大的抗氧化剂能够保障运动期间的机体修复,有效减缓和降低犬猫运动机体的氧化损伤,提升运动耐力,配以枸杞多糖,有效清除运动时的机体代谢产物,并增加糖原的储备,以达延缓疲劳发生和加速疲劳消除的作用。同时磷脂和南极磷虾油,用于液体制备乳化剂,稳定剂。
鸡油能够增加产品适口性,为各种宠物非常喜欢的营养补充剂,加入鸡油后可以增加液体制剂香气,一定比例的添加,令营养补充剂共同散发出犬猫喜欢味道,达到主动诱食的效果。但并不是越多诱食效果越好,在味道协调和体系稳定之间需要寻找最佳添加量;
鸡油与水,及配方中的营养元素不能共溶形成一个协调、稳定、均匀的液体体系。通过乳化剂的筛选,搭配,比例调式最终做成可保持18个月稳定的液体制剂。
单硬脂酸甘油酯:一种非离子表面活性剂,既有亲水又有亲油基团,具有润湿、乳化等多种功能。其是具有抗菌活性强和抗菌谱广的杀菌剂,可用于防腐保鲜;
磷脂是一种常用的带电两性表面活性剂;
两者作为乳化剂均能够在油和水之间形成稳定界面,使得油水结合形成稳定的分散体系。
南极磷虾油:其含有丰富的磷脂,在作为营养补充剂的同时,具有协同增效乳化效果的作用,根据三种乳化剂的特点可将配方物料经过试验测试后使用。与合适的溶剂进行均质分散处理,然后制定出单硬脂酸甘油酯、磷脂、南极磷虾油可做到最佳乳化剂,为持久稳定,均匀分散度好的宠物液体乳化剂。

S1、按原料重量份数取各组分;
S2、将NADH与二十八烷醇进行均质,制得第一均质液;
S3、将脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油和磷脂进行均质,制得第二均质液;
S4、将枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水和单硬脂酸甘油酯进行均质,制得第三均质液;
S5、将所述第二均质液和第三均质液进行混合,制得脂质体液体;
S6、将所述第一均质液分散到脂质体液体中,制得所述含NADH的组合物。
在某些实施方案中,步骤S2包括:
将二十八烷醇加热到完全融化,将NADH加入到融化的二十八烷醇中,在4000r/min的转速下进行高速乳化均质,制得所述第一均质液,备用,现配现用。
在某些实施方案中,步骤S3包括:
将脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油、磷脂加入到高压均质设备中,通入氮气均质乳化,制得所述第二均质液。
在某些实施方案中,步骤S4包括:
将枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水加入到高压均质设备中,通入氮气均质乳化,均质过程中逐渐加入单硬脂酸甘油酯,制得所述第三均质液。
在某些实施方案中,步骤S5包括:
将所述第二均质液和第三均质液置于高压微射流设备中,通入氮气,得到均脂质体液体;
步骤S6包括:
将所述第一均质液通过乳化罐均匀分散到脂质体液体中,得到所述含NADH的组合物。
有益效果:本发明的一种含NADH的组合物以NADH为主要功效原料,配以身体机能代谢过程中的营养物质需求,配伍搭配,及抗氧化成分清除运动过程中自由基的产生,再利用枸杞多糖延缓运动疲劳及运动损伤的发生,可大大提升犬猫灵敏度,耐力,精力,缓解运动过程中自由基的伤害,缓解体力疲劳,速效补能恢复体力。利用制剂技术制备肠溶NADH,18个月稳定的脂质体溶液,营养吸收倍增,高效浓缩液体,减少食喂计量,及提升适口性和宠物食用依从性。

下面通过实施方式对本发明进行进一步详细的说明。
实施例1
一种含NADH的组合物,包括以下重量的组分:
其制备方法包括以下步骤:
S1、按原料重量份数取各组分;
S2、将二十八烷醇加热到完全融化,将NADH加入到融化的二十八烷醇中,在4000r/min的转速下进行高速乳化均质,制得第一均质液,备用,现配现用;
S3、将脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油、磷脂加入到高压均质设备中,通入氮气均质乳化,制得第二均质液;
S4、将枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水加入到高压均质设备中,通入氮气均质乳化,均质过程中逐渐加入单硬脂酸甘油酯,制得第三均质液;
S5、将第二均质液和第三均质液置于高压微射流设备中,通入氮气,得到均脂质体液体;
S6、将第一均质液通过乳化罐均匀分散到脂质体液体中,得到含NADH的组合物。
其中:NADH产生于糖酵解和细胞呼吸作用中的柠檬酸循环。NADH分子是线粒体中能量产生链中的控制标志物。NADH是细胞能量代谢所必需的辅酶,参与糖酵解、三羧酸循环、氧化磷酸化,放出质子和电子,提供能量合成ATP;
生命现象的各种活动和细胞更新,整个生命的结构维持和平衡,都需能量才能进行,而NADH在生物过程中起电子传递作用;
NADH活化多酶系统,促进核酸、蛋白、多糖的合成及代谢,增加物质运转和调节控制,改善代谢功能均有密切关系,所以对细胞的再生、修复和保护具有物质基础。NADH在维持细胞生长、分化和能量代谢中起重要作用。
二十八醇:(1)增强耐力、精力和体力。(2)提高肌肉力量(肌肉机能);改善肌肉疼痛,减少肌肉摩擦。(3)缩短肌神经反应时间,提高反应敏锐性。(4)强化心脏机能。(5)提高基础代谢率。(6)增强对高山等反应的抵抗性。(7)降低收缩期血压。(8)提高氧的输送能力,减少需氧量。增加体力、耐力和精力;提高肌肉耐力;提高反应敏锐性,缩短反应时间;增加登高动力,提高能量代谢率,消除肌肉痉挛;增强包括心肌在内的肌肉功能,降低收缩期血压,提高基础代谢率,刺激性激素,促进脂肪分解。
辅酶Q10是一种脂溶性抗氧化剂,能激活人体细胞和细胞能量的营养,抗疲劳效果显著。
维生素E可以抑制自由基介导的膜脂蛋白氧化损伤,清除游离自由基,减少因长时间运动引起线粒体膜磷脂的减少所造成的疲劳感。
枸杞多糖:可明显提高运动小鼠血浆、肌组织SOD活性及肝组织GSH-Px的活性,从而有利于活性氧的清除,延缓运动性疲劳及运动损伤的发生,显著增加小鼠LDH活力,有效清除运动时的机体代谢产物,并增加糖原的储备,为机体提供更多的能量从而延缓疲劳发生和加速疲劳消除。
1,6-二磷酸果糖三钠盐是葡萄糖代谢过程中的一种重要的中间产物,对多种代谢酶有调节作用。可激活糖酵解中限速酶—磷酸果糖激酶及丙酮酸激酶活性,加速磷酸烯酮式丙酮酸向丙酮酸转化,增加细胞内能量。能直接进入细胞内作为能量底物供给能量,通过激活糖酵解的同时抑制糖异生而促进糖的利用并抑制糖原分解,促进糖原合成。还能抑制甘油利用,促进脂肪酸甘油三酯、磷脂的合成。与细胞膜相互作用改变膜离子通透性,能有效防止平滑肌细胞、神经元、血小板、的“钙超载”,提高激活Na+-K+ATP酶的活性,防止细胞。
南极磷虾油作为一种新型海洋Omega-3多不饱和脂肪酸来源,是EPA、DHA丰富的天然来源,磷脂型ω-3脂肪酸比甘油三酯型ω-3脂肪酸具有更佳的组织吸收率和生物利用度。能够改善轻度膝骨关节炎患者疼痛。其含有的磷脂(磷脂酰胆碱、磷脂酰丝氨酸和磷脂酸)具有合成代谢特性,可以参与维持肌肉功能,有助于长时间高强度运动中最佳运动表现的维持。南极磷虾油中含有的虾青素,是一种红色类胡萝卜素和强大的抗氧化剂,可抑制大强度运动后氧化应激。磷脂型DHA、EPA可促进骨骼肌糖代谢相关蛋白葡萄糖转运蛋白4、葡萄糖-6-磷酸脱氢酶基因和脂肪氧化分解关键酶肉碱棕榈酰转移酶I、酰基辅酶A氧化酶基因表达增加;还可通过促进线粒体呼吸链复合物I、Ⅱ、Ⅲ、Ⅳ和三羧酸循环相关酶mRNA表达,提升骨骼肌产能速率,进而提高跑步成绩。补充南极磷虾油可改善长跑运动员的血液黏度,这是提升耐力运动员代谢能力的重要体现。服用Omega-3多不饱和脂肪酸可通过改善血脂代谢和降低循环脂浓度,改善血液黏度并调节血液流变;Omega-3多不饱和脂肪酸还通过改变红细胞膜磷脂成分改善红细胞功能,包括改善红细胞形态和红细胞变形能力等。
磷脂对活化细胞,维持新陈代谢,基础代谢及荷尔蒙的均衡分泌,增强人体的免疫力和再生力,都能发挥重大的作用。另外,磷脂还具有促进脂肪代谢,防止脂肪肝,降低血清胆固醇、改善血液循环、预防心血管疾病的作用。
鸡油能够改善适口性,鸡油中的成分物质,比较容易与空气中的氧气发生反应而导致变质,食用之后容易对人体健康产生不利影响。要求选用十天内新鲜熬制的鸡油。
单硬脂酸甘油酯用作乳化剂。

一种含NADH的组合物,包括以下重量的组分:

S3、将脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油、磷脂加入到高压均质设备中,通入氮气均质乳化,制得第二均质液;

S6、将第一均质液通过乳化罐均匀分散到脂质体液体中,得到含NADH的组合物。

在实施例1配方的基础上,去掉鸡油,其他条件保持不变。具体为:

S3、将脂溶性原料辅酶Q10、维生素E、南极磷虾油、磷脂加入到高压均质设备中,通入氮气均质乳化,制得第二均质液;
S4、将枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水加入到高压均质设备中,通入氮气均质乳化,均质过程中逐渐加入单硬脂酸甘油酯,制得第三均质液;
S5、将第二均质液和第三均质液置于高压微射流设备中,通入氮气,得到均脂质体液体;
S6、将第一均质液通过乳化罐均匀分散到脂质体液体中,得到含NADH的组合物。
性能测试
1、对实施例1的脂质体液体进行稳定性考察
A、4℃下稳定性考察,结果如表1所示:

2、适口性考察
A、对实施例1的含NADH的组合物进行单碗试验,结果如表4所示:

3、功效性
选用40只10-12岁年龄的金毛,体重在25-30公斤,公母各半,随机分为2组,每组10只公,10只母。分别为营养组和空白对照组。
营养组提前5天喂食实施例1的含NADH的组合物(营养液),每10公斤体重喂食10mg营养液,对照组除营养液外,其他食物与营养组完全一致。
喂食5天营养液后,最后一天完成营养品饲喂后,即让40只(营养液组标识红色,对照组标识绿色)犬持续跑步运动10公里,记录犬跑完10公里所用时间,并在跑前和跑完后3分钟内测试其心率。观察犬身体状态。结果如表6所示:

综上所述:本发明的含NADH的组合物具有以下优点:
1、本发明通过试验设计和验证,合理搭配出稳定好的油、水、颗粒共存的混悬均质溶液乳化剂配方及制剂工艺,成功建立18个月稳定混悬液体体系;
2、本发明解决了原辅料及液体适口性不好的固有问题,最终产品具有较好的宠物犬猫适口性;
3、本发明的产品可以让宠物达到速效补能的目的,能够有效激发宠物运动和比赛过程中的耐力、精力及其反应灵敏性,缓解运动过程中自由基的伤害,缓解体力疲劳,减少运动炎症产生。
以上表述仅为本发明的优选方式,应当指出,对本领域的普通技术人员来说,在不脱离本发明创造构思的前提下,还可以做出若干变形和改进,这些也应视为发明的保护范围之内。
Claims (10)
1.一种用于宠物能量速补和缓解疲劳的NADH组合物,其特征在于,该组合物包括第一均质液,所述第一均质液包括NADH和二十八烷醇,并使用二十八烷醇包裹NADH,
其中:NADH与二十八烷醇的重量比为10:(4-10)。
2.根据权利要求1所述的NADH组合物,其特征在于,该组合物还包括第二均质液,所述第二均质液包括脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油和磷脂。
3.根据权利要求2所述的NADH组合物,其特征在于,该组合物还包括第三均质液,所述第三均质液包括枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水和单硬脂酸甘油酯。
4.根据权利要求3所述的NADH组合物,其特征在于,NADH、二十八烷醇和1,6-二磷酸果糖三钠盐的重量比为:10:(4-10):(3.3-10)。
6.一种制备权利要求1至5中任一权利要求所述的NADH组合物的方法,其特征在于,包括以下步骤:
S1、按原料重量份数取各组分;
S2、将NADH与二十八烷醇进行均质,制得第一均质液;
S3、将脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油和磷脂进行均质,制得第二均质液;
S4、将枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水和单硬脂酸甘油酯进行均质,制得第三均质液;
S5、将所述第二均质液和第三均质液进行混合,制得脂质体液体;
S6、将所述第一均质液分散到脂质体液体中,制得所述NADH组合物。
7.根据权利要求6所述的NADH组合物的制备方法,其特征在于,步骤S2包括:
将二十八烷醇加热到完全融化,将NADH加入到融化的二十八烷醇中,在4000r/min的转速下进行高速乳化均质,制得所述第一均质液,备用,现配现用。
8.根据权利要求6所述的NADH组合物的制备方法,其特征在于,步骤S3包括:
将脂溶性原料辅酶Q10、维生素E、鸡油、南极磷虾油、磷脂加入到高压均质设备中,通入氮气均质乳化,制得所述第二均质液。
9.根据权利要求6所述的NADH组合物的制备方法,其特征在于,步骤S4包括:
将枸杞多糖糖浆、1,6-二磷酸果糖三钠盐、水加入到高压均质设备中,通入氮气均质乳化,均质过程中逐渐加入单硬脂酸甘油酯,制得所述第三均质液。
10.根据权利要求6所述的NADH组合物的制备方法,其特征在于,步骤S5包括:
将所述第二均质液和第三均质液置于高压微射流设备中,通入氮气,得到均脂质体液体;
步骤S6包括:
将所述第一均质液通过乳化罐均匀分散到脂质体液体中,得到所述含NADH的组合物。
