Abstract
本发明提供了一种溶葡萄球菌复合酶杀菌纱布。该纱布为含溶葡萄球菌酶和另一种溶菌酶的复方新型杀菌纱布,作用物体表面2~10分钟可迅速杀灭大肠杆菌、金黄色葡萄球菌、绿脓杆菌、白色念珠菌等,杀灭率达99.9%,并且对创口和粘膜无任何刺激,有很高的临床使用价值。

本发明涉及医用生物敷料,具体涉及一种溶葡萄球菌复合酶(高科复合溶菌酶漱口水)杀菌纱布,
目前,在临床上使用的敷料,如医用纱布,一般有以下几种:①普通型无菌纱布,常用作为包扎材料;②抗粘型纱布,由于经过特殊处理,可防止伤口粘连,但无杀菌作用;③抗菌型纱布,因添加了抗生素,有一定的杀菌作用,但对一些大面积的伤口,杀菌作用差,并因长期使用,易产生抗药菌株。因此,临床上尚无有效的杀菌纱布。
本发明的目的在于克服上述足之处,设计一种不易产生耐药性,具有高效、广谱杀菌作用的纱布。
本发明提供了一种溶葡萄球菌复合酶杀菌纱布(溶葡萄球菌复合酶杀菌纱布),该纱布是由溶葡萄球菌复合酶及药用辅料与纱布组成的,其中溶葡萄球菌复合酶杀菌纱布原液为溶葡萄球菌酶(1ysostaphin)0.008~0.01%和溶菌酶(lysozyme)10~20%;辅料为磷酸钾7~15%、氯化钠0.5~1%、抑菌肽2~3%和蒸馏水60~80%。
由于溶葡萄球菌复合酶中的一种成分溶葡萄球菌酶是一种革兰氏阳性菌的溶菌素。组成该类革兰氏阳性菌(例如金黄色葡萄球菌)细胞壁肽聚糖的四肽侧链的氨基酸,依次为L-丙氨酸,D-谷氨酸,L-赖氨酸,D-丙氨酸;而首位的L-丙氨酸通过一个酰胺键与胞壁酸相连,该聚糖链四肽侧链第3位的L-赖氨酸,通过五肽(五个甘氨酸)交联桥联结到相邻聚糖链四肽侧链D-丙氨酸羧基上。由此纵横交叉,左右联结构成十分坚韧的三维立体多孔结构,并聚合成较厚的肽聚糖层。溶葡萄球菌酶可切断肽聚糖中的Gly-Gly键,而溶葡萄球菌复合酶中另一种生物酶可切断N-乙酰胞壁酸和N-乙酰葡萄糖胺之间的键,从而使杀菌活性更强,达到溶解并杀死细菌的目的,并可避免细菌耐药性的产生。
由于溶葡萄球菌复合酶独特的杀菌机理,使其不同于一般抗生素,常用抗生素是抑制细菌生长,而该酶则是裂解细菌杀死细菌。上海市华山医院抗生素研究所、第二军医大学长海医院检验科、上海市第六人民医院检验科、上海市第九人民医院检验科等单位曾将临床各科收集到的400多株细菌进行了溶葡萄球菌复合酶的抑杀菌实验。实验结果表明:溶葡萄球菌复合酶对革兰氏阳性菌,如临床上常见的金黄色葡萄球菌,表皮葡萄球菌、肺炎双球菌、D群肠球菌、四联球菌、产单核李斯特菌、链球菌以及胃幽门螺旋杆菌、大肠杆菌、绿脓杆菌等均有明显的抑杀作用。
实验还证明溶葡萄球菌酶对耐药性菌株,如目前医学临床上最感棘手的耐甲氧西林金黄色葡萄球菌,耐药性肺炎双球菌、嗜麦芽窄食单孢菌的效果更优于常用抗生素。
本发明的溶葡萄球菌复合酶杀菌纱布,经上海市预防医学研究院检测毒性、皮肤刺激杀菌试验及稳定性试验结果如下:
一、急性经口毒试验:
试验目的:检测短时间经口染毒时的毒性,其结果可作为毒性分级和确定亚慢性毒性等试验剂量的依据。
(一)材料与方法:
1、受试样品:
性状:白色纱布
配制方法:将样品挤出液作为原液,取5g原蒸馏水至20ml,充分混匀后作为受试样品。
2、实验动物:昆明种小鼠,体重18-22克,由上海医科大学实验动物部提供,合格证号:02-22-1。
实验动物饲养室温度20±2℃,相对湿度40-70%,动物房合格证号:02-28,小鼠饲养料,由苏杭实验动物科技发展公司提供。
3、剂量和分组:将动物分为雌雄各一组,每组10只,剂量为5g/kg体重。
4、试验步骤:动物禁食16小时后,根据不同体重,用灌胃针一次经口灌服受试样品,给样容量按0.2ml/l0g体重。观察给样后动物的症状及死亡情况,观察期为二周。
5、LD50计算方法:用最大剂量限量法得出LD50,并确定急性经口毒性分级。
(三)试验结果;
给样后,动物未见中毒症状和死亡。

(四)结论:按急性经口毒分级,属实际无毒级。
二 、皮肤刺激试验
目的:观察受试物对动物一次接触皮肤后可能产生的炎性反应。
1、样品性状:白色纱布
2、受试样品处理:将样品剪成2.5×2.5cm2小块作为受试样品。
3、受试动物:新西兰种家兔,体重2.0-3.0kg,由上海医科大学实验动物部提供,合格证号:02-52-1。
4、测试依据:消毒技术规范第三版(卫生部1999.11)
5、试验方法:试验前将动物背部两侧毛剪掉,去毛范围左右各约3×3cm2,选左侧完好皮肤划出2.5×2.5cm2试验区,右侧皮肤作对照。
将受试样品贴敷在试验区皮肤上,用一层油纸,胶布固定,贴敷4小时后,除去覆盖物,用温水清洗除去残留物,并于除去受试样品后1、24、48小时分别观察皮肤反应按消毒技术规范第三版(卫生部1999.11)皮肤反应积分和刺激强度评价。

4、评价:
该样品经检测,作用2分钟对大肠杆菌、金黄色葡萄球菌、白色念珠菌均有杀菌作用;样品经54℃水浴二周后,再次检测,作用2分钟对大肠杆菌仍有杀菌作用;无菌试验检测结果符合GB15980-1995。
本发明的另一目的是提供了上述溶葡萄球菌复合酶杀菌纱布的制备方法,该方法是在无菌室中,按处方量将溶葡萄球菌酶(从美国Sigma公司购买),溶菌酶,磷酸钾,氯化钠,按比例依次加入洁净的玻璃或不锈钢盛具中,轻轻搅拌,充分混和均匀后,再加入所示比例的抑菌肽和蒸馏水,充分混匀,即为原液。将无菌纱布置于原液中,浸透,静置10分钟,将纱布包装于无菌塑
本发明的溶葡萄球菌复合酶杀菌纱布为含溶葡萄球菌酶和另一种溶菌酶、增效剂、稳定剂辅酶复方新型杀菌纱布,作用物体表面2-10分钟可迅速杀灭大肠杆菌、金黄色葡萄球菌、绿脓杆菌、白色念珠菌等,杀灭率达99.9%。广泛适用于烧伤及灼伤皮肤创口的敷贴和手术中的腹腔及肠道粘膜消毒等,具有对创口和粘膜无任何刺激,杀菌效果好使用方便等特点,有很高的临床使用价值。
溶葡球菌复合酶杀菌纱布在使用过程中,由于其作用成分为酶制剂,是一类蛋白质,不会对皮肤产生任何刺激,溶葡球菌酶有一定的组织修复作用,可促进伤口的愈合。因其作用机理是直接溶解细菌细胞壁而致细菌死亡,不易产生耐药性菌株。又因该酶的杀菌活性高,又有促进组织粘膜生长的作用可减轻病人的痛苦。
实例1、每制造1片溶葡球菌复合酶杀菌纱布的配制方法(每片杀菌纱布需原液10克)。分别将溶葡萄球菌酶0.0008克,溶菌酶2克,磷酸钾1.4克,氯化钠0.1克,混匀再加入抑菌肽0.2克和蒸馏水6.3ml,混合均匀,放入一片无菌纱布静置10分钟,包装封口。
实例2、分别将溶葡萄球菌酶0.00085克,溶菌酶1.5克,磷酸钾6.3克,氯化钠0.1克,混匀再加入抑菌肽0.2克和蒸馏水6.9ml,混合均匀,放入一片无菌纱布静置10分钟,包装封口。
实例3、分别将溶葡萄球菌酶0.0009克,溶菌酶1.5克,磷酸钾1.3克,氯化钠0.1克,混匀再加入抑菌肽0.2克和蒸馏水6.9ml,混合均匀,放入一片无菌纱布静置10分钟,包装封口。
Claims (2)
1、一种溶葡萄球菌复合酶杀菌纱布,其特征在于该纱布是由溶葡球菌复合酶及药用辅料与纱布组成的,其中溶葡球菌复合酶以重量百分比计为溶葡萄球菌酶0.008~0.01%和溶菌酶10~20%;辅料以重量百分比计为磷酸钾7~15%、氯化钠0.5~1%、抑菌肽2~3%和蒸馏水60~80%。
2、一种如权利要求1所述的溶葡球菌复合酶杀菌纱布的制备方法,其特征在于该方法是在无菌室中,按处方量将溶葡萄球菌酶,溶菌酶,磷酸钾,氯化钠,按比例依次加入洁净的玻璃或不锈钢盛具中,轻轻搅拌,充分混和均匀后,再加入所示比例的抑菌肽和蒸馏水,充分混匀,即为原液,将无菌纱布置于原液中,浸透,静置10分钟,将纱布包装于无菌塑料袋中,封口,入库,室温保存即可。
