Experimental Example 2 Study on Antioxidant Efficacy - DPPH Free Radical Scavenging Experiment

Take 1g of the sample of Example 1-13 and Comparative Example 1-4, add 2mL of anhydrous ethanol, add 2mL of 60μmol/L DPPH solution after complete dissolution, mix, let stand for 30min, adjust to zero with anhydrous ethanol, and measure the absorbance at 517nm as Ai; take 2mL of anhydrous ethanol solvent + 2mL DPPH solution and mix to determine the absorbance extreme Ac in the same way; take 1g of the matrix of the composition and dissolve it, add 2mL of anhydrous ethanol, then add 2mL of anhydrous ethanol solvent and mix to determine the absorbance as Aj.

The free radical scavenging rate was calculated according to the following formula.

Among them, Aj is the contribution of the reaction sample itself to the absorbance; Ai is the absorbance value of the sample after the action of DPPH; Ac is the absorption of DPPH itself at the measurement wavelength.

The DPPH free radical scavenging rate reflects its antioxidant capacity to a certain extent. The higher the scavenging rate, the better the antioxidant degree and the better the anti-aging effect. Therefore, the anti-aging effect of the composition can be judged by studying its ability to scavenge DPPH free radicals. The results are shown in Table 3 (each group of samples was measured 5 times in parallel, and the data are presented as average values).

It can be seen from Table 3 that the combination prepared in Examples 1-3 has a good scavenging effect on DPPH, with a scavenging rate of more than 86%. From the comparison between Example 1 and Comparative Examples 1-4, it can be seen that the lactobacillus/algae extract fermentation product, the spherical algae composition, the plant extract and the oil in the skin care composition of the present invention have a synergistic effect on the antioxidant effect. The synergistic combination of the four can make the antioxidant effect of the product more excellent, and the addition of skin conditioners and the selection of types have better antioxidant properties for the product.

Test Example 3

Propionibacterium acnes inhibition test:

The antibacterial effects of the samples prepared in Examples 1, 6-8 and Comparative Examples 1-4 on Propionibacterium acnes were measured. Preparation of the culture medium: 33 g of lecithin-Tween nutrient agar medium and 29.3 g of liquid thioglycollate medium were weighed respectively, added with 1 L of ultrapure water, stirred, heated and boiled until completely dissolved, and then packaged and sterilized at 121° C. for 30 min, cooled to 60° C., and kept warm in a water bath for later use.

Recovery of Propionibacterium acnes and preparation of bacterial suspension: Take freeze-dried powder of Propionibacterium acnes, melt the ampoule under an alcohol lamp, add 1mL of 0.85% saline, stir evenly, inoculate on 3 plates containing culture medium, and then culture under anaerobic conditions at 37°C for 3 days. Use an inoculation loop to scrape the slant colony, and dilute the bacterial suspension appropriately with saline to make its concentration 10 ⁶ CFU·mL ^-1 . Under sterile operation, take the above bacterial suspension and evenly spread it on the culture medium with a coating rod. Then use sterile tweezers to gently place the Oxford cup on the surface of the solidified culture medium, and gently press the Oxford cup (outer diameter 8mm) to make good contact. Add 200μL of liquid to the Oxford cup, and add deionized water as a blank control, culture in a 37°C incubator for 48h, take out, and measure the diameter of the effective inhibition zone of the test solution in each culture dish. Each group of experiments was repeated 3 times, and two samples were taken for testing each time.

Judgment criteria: inhibition zone diameter > 20 mm indicates significant inhibition, inhibition zone diameter 10-20 mm indicates moderate inhibition, and inhibition zone diameter < 10 mm indicates no inhibition. The results are shown in Table 4:

It can be seen from the data in Table 4 that the composition of the present invention has a good inhibitory effect on Propionibacterium acnes, and the products prepared in Comparative Examples 1-4 also have a certain inhibitory effect compared with the blank control group; and the three components of the lactobacillus/algae extract fermentation product, the sphaerocephala composition, and the plant extract in the composition have a certain synergistic effect on the above-mentioned effects, have a significant inhibitory effect on Propionibacterium acnes, effectively control the reproduction of Propionibacterium acnes, maintain the ecological balance of skin colonies, and the type of plant composition also has a great influence on the inhibitory effect of Propionibacterium acnes.

Test Example 4 Evaluation of Moisturizing Effect

The samples of Examples 1-5 and Comparative Examples 1-4 were evaluated for their long-lasting moisturizing effect and skin tightening effect:

90 trial subjects aged 20-45 were randomly selected and divided into 9 groups, with 10 in each group. The 9 groups of trial subjects used the samples obtained in Examples 1-5 and Comparative Examples 1-4, respectively, once in the morning and evening after cleansing, with the same usage amount, for 30 days, and no other skin care products were used during this period.

The skin moisture content was tested with a skin moisture tester Corneometer CM825 on the 0th day and the 30th day, respectively, to evaluate the skin moisture content before and after using the sample. The skin moisture content change rate = (skin moisture content after 30 days of use - skin moisture content before use) / skin moisture content before use × 100%; the test results are shown in Table 5.

As shown in Table 5, the skin care composition and lotion of the present invention have good long-lasting moisturizing effect, wherein the lactobacillus/algae extract fermentation product, the sphaerocephala composition, the plant extract and the oil promote each other in this effect, and have an unexpected synergistic effect. The type of sphaerocephala composition also has an important influence on the moisturizing property of the product.
