Disclosure of Invention
Aiming at the defects of the prior art, the invention aims to provide a skin care composition containing lactobacillus/algae extract fermentation product, and a preparation method and application thereof.
In order to achieve the aim of the invention, the invention adopts the following technical scheme:
In a first aspect, the present invention provides a skin care composition comprising a lactobacillus/algae extract fermentation product, the skin care composition comprising a lactobacillus/algae extract fermentation product, a chlorella composition, a plant extract, a lipid, an emulsifier, and a humectant.
The lactobacillus/algae extract fermentation product contains alpha-hydroxy acid (AHA) such as lactic acid and other bioactive substances, can help balance the microbiota of skin, accelerate the generation of new cells, strengthen the natural barrier of skin, protect the skin from free radicals, improve the resistance of the skin to external stimulus and pollution, have excellent effects of resisting oxidation, moisturizing, resisting inflammation and maintaining the balance of skin microorganisms, can help the skin to keep moisture, improve the moisturizing capability of the skin, make the skin look more moist and smooth, and can improve the permeability and effect of other active ingredients in a formula, and the plant extract can reduce skin inflammation and has potential benefits for improving sensitive skin or inflammatory skin diseases (such as acne).
The invention creatively discovers that the lactobacillus/algae extract fermentation product, the chlorella composition, the plant extract and the grease have remarkable effects of resisting oxidation, moisturizing, resisting inflammation and maintaining the balance of microorganisms on the surface of skin when being used together, and the four have a certain synergistic effect on the effects, so that the invention is very suitable for skin care products, is mild and does not irritate, and has good stability.
Preferably, the skin care composition containing lactobacillus/algae extract fermentation product comprises 0.5-20 parts of lactobacillus/algae extract fermentation product, 1-10 parts of chlorella composition, 1-15 parts of plant extract, 1-10 parts of grease, 1-10 parts of emulsifier and 1-10 parts of humectant.
The lactobacillus/algae extract fermentation product may be added in an amount of 0.8 parts, 1 part, 2 parts, 5 parts, 7 parts, 9 parts, 10 parts, 13 parts, 15 parts, 17 parts, 19 parts, or the like in the skin care composition containing the lactobacillus/algae extract fermentation product of the present invention;
The adding amount of the chlorella composition can be 2 parts, 3 parts, 4 parts, 5 parts, 6 parts, 7 parts, 8 parts or 9 parts, etc.;
the plant composition may be added in an amount of 2 parts, 3 parts, 4 parts, 5 parts, 6 parts, 7 parts, 8 parts, 9 parts, 10 parts, 11 parts, 12 parts, 13 parts, 14 parts, or the like;
The addition amount of the emulsifier can be 2 parts, 3 parts, 4 parts, 5 parts, 6 parts, 7 parts, 8 parts or 9 parts, etc.;
The humectant may be added in an amount of 2 parts, 3 parts, 4 parts, 5 parts, 6 parts, 7 parts, 8 parts, 9 parts, or the like.
Preferably, the skin care composition containing lactobacillus/algae extract fermentation product comprises 3-15 parts of lactobacillus/algae extract fermentation product, 2-9 parts of chlorella composition, 2-12 parts of plant extract, 2-10 parts of grease, 3-9 parts of emulsifier and 2-9 parts of humectant.
Preferably, the skin care composition containing lactobacillus/algae extract fermentation product comprises 5-12 parts of lactobacillus/algae extract fermentation product, 3-8 parts of chlorella composition, 3-10 parts of plant extract, 3-9 parts of grease, 3-7 parts of emulsifier and 2-8 parts of humectant.
Preferably, the chlorella composition comprises an extract of nannochloropsis oculata and an extract of nannochloropsis oculata.
Preferably, the mass ratio of the nannochloropsis oculata extract to the polar nannochloropsis extract is 1 (0.5-10), for example, 1:0.8, 1:1, 1:2, 1:3, 1:4, 1:5, 1:6, 1:7, 1:8 or 1:9, etc.
Compared with other chlorella extracts, the chlorella oculopsis extract and the very small chlorella oculopsis extract are matched with other components in the composition, so that the composition can be quickly absorbed by skin and forms a layer of protective film on the surface layer of the skin, thereby reducing water evaporation and playing a better role in moisture preservation.
Preferably, the plant extract comprises any one or a combination of at least two of a compact Luo Mushe/stem extract, a coconut fruit extract, a chamomile extract, an aloe vera leaf extract, a tea leaf extract, a calamus root extract, or a myrrh resin extract.
Preferably, the plant extract is a combination of coconut fruit extract, calamus root extract and myrrh resin extract.
As a preferred technical scheme of the invention, the coconut fruit extract, the calamus root extract and the myrrh resin extract are used in a matched mode, can inhibit the synthesis of inflammatory factors and slow down the release of the inflammatory factors, play an anti-inflammatory role, are natural and non-irritating, can promote the growth of dominant bacteria on the surface of the skin, and maintain the skin micro-ecology.
Preferably, the mass ratio of the coconut fruit extract, the calamus root extract and the myrrh resin extract is (3-5): 2-4): 1-5.

In the invention, the emulsifier uses the combination of the PEG-8C 12-20 acid ester and the potassium cetyl phosphate, so that the skin care composition and other components can be better emulsified, and the stability of the system is more excellent.
Preferably, the mass ratio of the PEG-8 ester of C12-20 acid to the potassium cetyl phosphate is 1 (0.1-1), and can be, for example, 1:0.1, 1:0.2, 1:0.3, 1:0.4, 1:0.5, 1:0.6, 1:0.7, 1:0.8, or 1:0.9, etc.

In a second aspect, the present invention provides a method for preparing a skin care composition comprising lactobacillus/algae extract fermentation product as described in the first aspect, said method comprising:
(1) Mixing an emulsifier, a humectant, grease and deionized water, and carrying out high-pressure microjet treatment to obtain a vesicle solution;
(2) Mixing the vesicle solution obtained in the step (1) with a chlorella composition, lactobacillus/algae extract fermentation products and plant extracts, and carrying out high-pressure micro-jet treatment to obtain the skin care composition containing the lactobacillus/algae extract fermentation products.
Preferably, the temperature of the mixing in step (1) is 45-55 ℃, e.g. 46 ℃, 48 ℃, 50 ℃, 52 ℃, 54 ℃ or the like.
Preferably, the high pressure micro-jet treatment in step (1) has a pressure of 200-300MPa and a jet velocity of 500-700m/s, for example, the pressure may be 210MPa, 230MPa, 250MPa, 270MPa or 290MPa, etc., and the jet velocity may be 520m/s, 550m/s, 580m/s, 600m/s, 620m/s, 650m/s or 680m/s, etc.
Preferably, the mixing of step (1) further comprises adding a skin conditioning agent and/or preservative.
Preferably, the temperature of the mixing in step (2) is 40-50 ℃, e.g. 41 ℃, 43 ℃, 45 ℃, 47 ℃, 49 ℃ or the like.
Preferably, the pressure of the high-pressure micro-jet treatment in the step (2) is 200-300MPa, the jet velocity is 500-700m/s, for example, the pressure can be 210MPa, 230MPa, 250MPa, 270MPa or 290MPa, etc., and the jet velocity can be 520m/s, 550m/s, 580m/s, 600m/s, 620m/s, 650m/s or 680m/s, etc.

In a third aspect, the present invention provides the use of a skin care composition comprising a lactobacillus/algae extract fermentation product as described in the first aspect for the preparation of a cosmetic.

Preferably, the skin care composition containing lactobacillus/algae extract fermentation product is 0.1-15% by mass in the cosmetic, for example, may be 0.5%, 0.8%, 1%, 2%, 3%, 4%, 5%, 6%, 7%, 8%, 9%, 10%, 11%, 12%, 13% or 14% etc.

Compared with the prior art, the invention has the following beneficial effects:
(1) The invention creatively discovers that the lactobacillus/algae extract fermentation product, the chlorella composition, the plant extract and the grease have remarkable effects of resisting oxidation, moisturizing, resisting inflammation and maintaining the balance of microorganisms on the surface of skin when being used together, and the four have a certain synergistic effect on the effects, so that the invention is very suitable for skin care products, is mild and does not irritate, and has good stability.
(2) The preparation method of the skin care composition containing lactobacillus/algae extract fermentation product provided by the invention has the advantages of simple operation steps, low cost and easiness in industrial mass production.

The preparation method of the skin care composition containing the chlorella vulgaris comprises the following steps:
(1) Mixing an emulsifier, a humectant, grease, a skin conditioning agent, a preservative and deionized water at 50 ℃, and uniformly carrying out high-pressure micro-jet treatment to obtain a vesicle solution, wherein the pressure of the high-pressure micro-jet treatment is 250MPa, and the jet speed is 500m/s;
(2) Mixing the vesicle solution obtained in the step (1) with the chlorella composition, the lactobacillus/algae extract fermentation product and the plant extract at 40 ℃, and uniformly carrying out high-pressure microjet treatment to obtain the skin care composition containing the lactobacillus/algae extract fermentation product, wherein the pressure of the high-pressure microjet treatment is 300MPa, and the jet speed is 650m/s.

Example 12
This example provides a skin care composition comprising lactobacillus/algae extract fermentation product which differs from example 1 only in that the skin conditioning agent does not include lecithin, the reduced amount of which is distributed in parts ratio into isononyl isononanoate, ascorbic acid and squalane, the remaining components and the added amounts being in accordance with example 1, the preparation method being referred to example 1.
Example 13
This example provides a skin care composition comprising lactobacillus/algae extract fermentation product which differs from example 1 only in that squalane is not included in the skin conditioner, the reduced amounts thereof are distributed in parts ratio to isononyl isononanoate, ascorbic acid and lecithin, the remaining components and added amounts are the same as in example 1, and the preparation method is referred to in example 1.
Comparative example 1
This comparative example provides a skin care composition comprising lactobacillus/algae extract fermentation product, which differs from example 1 only in that the formulation does not include chlorella composition, the reduced amount of which is distributed to each component of lactobacillus/algae extract fermentation product, plant extract and fat in parts ratio, and the remaining components and added amounts are the same as in example 1, and the preparation method is referred to in example 1.
Comparative example 2
This comparative example provides a skin care composition comprising lactobacillus/algae extract fermentation product, which differs from example 1 only in that the formulation does not include plant extract, the reduced amount of which is distributed in parts ratio to the components of lactobacillus/algae extract fermentation product, chlorella composition and fat, the remaining components and the added amounts are the same as in example 1, and the preparation method is referred to in example 1.
Comparative example 3
This comparative example provides a skin care composition comprising lactobacillus/algae extract fermentation product, which is different from example 1 only in that no fat is included in the formulation, the reduced amount of which is allocated to each component of lactobacillus/algae extract fermentation product, chlorella composition and plant extract in parts ratio, and the remaining components and added amounts are identical to example 1, and the preparation method is referred to example 1.
Comparative example 4
This comparative example provides a skin care composition differing from example 1 only in that the formulation does not include lactobacillus/algae extract fermentation product, the reduced amount of which is apportioned to the components of the chlorella composition, plant extract and grease in parts, the remaining components and added amounts being identical to example 1, the preparation method being as described in example 1.
Experimental example 1 erythrocyte hemolysis experiment
The erythrocyte hemolysis test (Red blood cell haemolysis test, RBC) is one of the alternative methods for the rabbit eye-stimulating test, and the basic principle is to evaluate the damage of chemicals to eye tissue cells by measuring the amount of cellular hemoglobin and the degree of denaturation. RBC experiments can be used to evaluate eye irritation of cosmetics.
The samples of examples 1-13 were subjected to erythrocyte hemolysis experiments. The experiment adopts the RBC experiment test method and the judgment standard of the European alternative method verification center, the specific judgment standard is shown in the table 2, wherein HD 50 is the sample concentration when 50% of red blood cells are hemolyzed, DI is the protein denaturation index, and L/D is the ratio of HD 50 to DI.

From the results of the irritation of the samples in Table 2, the compositions prepared in examples 1-13 of the present invention were classified as non-irritating, indicating that the skin care compositions of the present invention all have mild and non-irritating characteristics.
Test example 2 antioxidant efficacy study-DPPH free radical scavenging experiment
Taking 1g of samples of examples 1-13 and comparative examples 1-4, adding 2mL of absolute ethyl alcohol, adding 2mL of 60 mu mol/L DPPH solution after complete dissolution, mixing uniformly, standing for 30min, zeroing by absolute ethyl alcohol, measuring absorbance at 517nm as Ai, mixing 2mL of absolute ethyl alcohol solvent and 2mL of DPPH solution uniformly to measure absorbance as Ac, dissolving 1g of composition matrix, adding 2mL of absolute ethyl alcohol solvent, mixing uniformly, and measuring absorbance as Aj.
The radical scavenging rate was calculated as follows.

Wherein, the Aj reaction sample itself contributes to absorbance, the Ai sample acts on the absorbance value after DPPH, and Ac is the absorption of DPPH itself at the measurement wavelength.
The DPPH free radical scavenging rate reflects the antioxidant capacity to a certain extent, and the higher the scavenging rate is, the better the antioxidant degree is, and the better the anti-aging effect is, so the anti-aging effect can be judged through the research of the DPPH free radical scavenging capacity of the composition. The results are shown in table 3 (5 replicates per group of samples, data presented as average).

As can be seen from Table 3, the combination prepared in examples 1-3 has a better DPPH scavenging effect, the scavenging rate is more than 86%, and comparison of examples 1 and comparative examples 1-4 shows that the lactobacillus/algae extract fermentation product, the chlorella composition, the plant extract and the grease in the skin care composition have a synergistic effect on the antioxidation effect, and the synergistic cooperation of the four can lead to better antioxidation effect of the product, and the addition and the type selection of the skin conditioning agent have better antioxidation performance on the product.
Test example 3
Propionibacterium acnes inhibition assay:
The samples prepared in examples 1, 6-8 and comparative examples 1-4 were assayed for their bacteriostatic effect against propionibacterium acnes. Preparing culture medium, namely weighing 33g of lecithin-Tween nutrient agar culture medium and 29.3g of liquid thioglycolate culture medium, adding 1L of ultrapure water, stirring, heating, boiling to dissolve completely, sub-packaging, sterilizing at 121 ℃ for 30min under high pressure, cooling to 60 ℃, and preserving heat in a water bath kettle for later use.
Resuscitator and bacterial suspension of Propionibacterium acnes are prepared by taking freeze-dried powder of Propionibacterium acnes, fusing ampoule bottles under an alcohol lamp, adding 1mL of 0.85% physiological saline, stirring uniformly, inoculating on 3 plates containing culture medium, and culturing for 3 days under anaerobic and 37 ℃ conditions. The slant colonies were scraped with an inoculating loop, and the bacterial suspension was diluted with physiological saline to a concentration of 10 6CFU·mL-1. Under aseptic operation, the bacterial suspension is taken and uniformly smeared on a culture medium by a coating rod. The oxford cup is lightly placed on the surface of the solidified culture medium by using sterile forceps, and the oxford cup (with the outer diameter of 8 mm) is lightly pressed to ensure good contact. 200 mu L of the solution is added into an oxford cup, deionized water is added as a blank control, the solution is cultured for 48 hours in a 37 ℃ incubator and taken out, and the diameter of an effective inhibition zone of the test solution in each culture dish is measured. Each set of experiments was repeated 3 times, twice each time to perform the test.
The judgment standard is that the diameter of the inhibition zone is 20mm, the diameter of the inhibition zone is 10-20mm, the diameter of the inhibition zone is medium, the diameter of the inhibition zone is <10mm, the inhibition is not performed, and the results are shown in Table 4:

As shown in the data of Table 4, the composition provided by the invention has good inhibition effect on Propionibacterium acnes, the products prepared in comparative examples 1-4 also have a certain inhibition effect compared with a blank control group, and three components of lactobacillus/algae extract fermentation product, chlorella composition and plant extract in the composition have a certain synergistic effect on the effects, so that the composition has a remarkable inhibition effect on Propionibacterium acnes, effectively controls the propagation of Propionibacterium acnes, maintains the ecological balance of skin colonies, and the type selection of the plant composition has a great influence on the inhibition effect of Propionibacterium acnes.
Test example 4 evaluation of moisturizing Effect
The samples of examples 1 to 5 and comparative examples 1 to 4 were evaluated for long-lasting moisturizing effect and skin-tightening effect:
90 testers, 20-45 years old, were randomly selected and divided into 9 groups of 10. The samples obtained in examples 1-5 and comparative examples 1-4 were used by the 9 groups of testers, once a day after cleansing the face in the morning and evening, in the same amount for 30 days, during which no other skin care products were used.
Skin moisture content was measured with a skin moisture content tester Corneometer CM825 on day 0 and day 30, respectively, and the moisture content of the skin before and after the use of the samples was evaluated, and the skin moisture content change rate = (skin moisture content after 30 days of use-skin moisture content before use)/skin moisture content before use x 100%, and the test results are shown in table 5.

As can be seen from Table 5, the skin care compositions and emulsions of the present invention have a good long-lasting moisturizing effect, wherein the lactobacillus/algae extract fermentation product, the chlorella composition, the plant extract and the oil promote each other in the effect, and have unexpected synergy. And the choice of the type of the chlorella composition also has an important influence on the moisture retention of the product.
The applicant states that the present invention is illustrated by the above examples as a skin care composition comprising lactobacillus/algae extract fermentation product, and a method for preparing the same and application thereof, but the present invention is not limited to the above examples, i.e. it is not meant that the present invention must be practiced by relying on the above examples. It should be apparent to those skilled in the art that any modification of the present invention, equivalent substitution of raw materials for the product of the present invention, addition of auxiliary components, selection of specific modes, etc., falls within the scope of the present invention and the scope of disclosure.
The preferred embodiments of the present invention have been described in detail above, but the present invention is not limited to the specific details of the above embodiments, and various simple modifications can be made to the technical solution of the present invention within the scope of the technical concept of the present invention, and all the simple modifications belong to the protection scope of the present invention.
