Abstract
以含薯蓣皂甙的薯蓣属植物为原料,用黑曲霉菌发酵,可联合生产薯蓣皂素和柠檬酸两项产品。通过发酵,薯蓣皂素的分子结构保持不变,并使薯蓣皂素的产率成倍提高;经过发酵,将淀粉转为柠檬酸进入液相,使投入水解过程的物料减少,酸、溶剂和能量的消耗降低,可使每吨薯蓣皂素的生产成本降低约3万元。
Description
本发明是一种由薯蓣属植物提取薯蓣皂素的科学发酵方法。
薯蓣皂素是生产多种甾体药物的前体,全世界生产的甾体激素,60%以上取材于薯蓣皂素。世界上最大生产国是墨西哥,年产量约500吨,原料品种为菊叶薯蓣和多花薯蓣。其次是我国,原料品种为盾叶薯蓣和穿龙薯蓣,其它如危地马拉、波多黎哥、印度等国都有一定量的生产。预计1985年世界年产量(不包括我国)将增长为2400吨,平均每年增长速度为10%。
早期,生产薯蓣皂素的方法是直接酸水解法-将薯蓣片放入水解罐中,用一定浓度的硫酸或盐酸催化水解,然后用有机溶剂从水解物中浸提薯蓣皂素,糖和淀粉留在废渣废液中。这种方法水解不彻底,皂素得率低,一般达到2%左右。
若经预发酵处理,可使产量提高17~48%,然而,现行的预发酵处理是将薯蓣片泼水加湿,成堆堆放,自然发酵,或称为自酵发酵。有的在池中浸泡发酵,有的加入外源酶促进发酵。上述方法简便易行但不科学,名曰自酶发酵,实则杂菌发酵,空气和水中的任何菌种都可参加,往往造成霉变。穿龙、盾叶、福州和蜀葵叶等薯蓣根茎,发霉后皂素得量高,但熔点降低,经分离可得到表-丝兰皂甙元和丝兰皂甙元酮,未经发霉的则没有。这两种衍生物的出现,使产品质量不合要求,同时使产量下降。特别是气候条件对自酶发酵影响大,发酵条件和发酵程度很难控制。某皂素厂因一场突降的大雨使三、四十吨正在发酵的黄姜片全部霉坏,失去提取价值而被迫倒掉。自酶发酵不仅难于有效地促进水解提高得率,而且经常由于局部霉变,使薯蓣皂素结构改变,转变为其他衍生物,降低产品熔点,拉大熔距,造成更大损失。因而,绝大多数厂家的皂素得率仍然在2~2.2%。
为寻求提高皂素得率和利用淀粉的途径,Rothock,J.W.等曾用土曲霉MF-118菌株酶解D.barbasco薯蓣皂素产率未超出直接酸水解法水平;四川生物所曾进行过预发酵提高薯蓣皂素产率及淀粉综合利用方面的研究,并提出“盾叶薯蓣在预发酵过程中有种能影响皂甙转化为皂甙元的酶存在,当适合这种酶催化的条件存在时,其酶活性强,就能促进皂素产率的提高。反之,当这种酶活性受到破坏后,虽然按预发酵条件处理,都不能提高皂素产率”,因而,“不经预发酵,当酒精发酵时的高温、高压蒸料,就起到了杀灭皂甙酶的作用,故不能提高皂素产率。”亦即酒母在发酵过程中产生的酶,不能使皂甙降解为次级甙,因而不能取代“自酶发酵”;一年后,陕西省轻工业研究所将盾叶薯蓣粉碎后即行蒸煮、糖化,接入1300#酵母进行酒精发酵,每百斤薯蓣生产96度酒精12.65斤(淀粉出酒率27.3%),生产皂素0.78斤(原料中皂素含量0.9至1.5%)。
他们只着眼于利用淀粉,对酵母发酵能否提高皂素得率未加以探讨。本发明为根治“自酶发酵”的弊病,筛选了几种纯种黑曲霉菌,进行了多次发酵实验,然后取直接酸水解法提取的总皂甙元,和纯种黑曲霉菌发酵后,又经酸水解而提取的总皂甙元,与薯预皂素标样对照,进行薄层分析,得到了两对色斑。第一对色斑Rf=0.45,与标样的Rf值完全一致,可见这种色斑就是薯蓣皂素的色斑。第二对色斑小而色浅,Rf=0.85,这是共存的其他皂甙元或其衍生物的色斑,除此以外未见其他明显色斑。
将发酵和未发酵的两类总皂甙元样品,分次经氧化铝干柱吸附后,先后用乙醚和苯洗脱,得到两个洗脱峰,分别收集产物,测定红外光谱。结果,这两类样品的乙醚洗脱物的红外光谱是相同的ν〈`;film;max`〉cm-1;1057(C3-OH和△5),986,926<904,868(25R螺留烷),与萨德勒商品谱图S-092对照可判定为薯蓣皂素;质谱M/Z:414(M+),139(基峰)282,300,经库检索也可判定为薯蓣皂素。这两类样品的苯洗脱物的红外光谱也相同,它们是同一衍生物。
上述结果表明,黑曲霉菌发酵未引起薯蓣皂素分子结构的改变,衍生物是原料中固有的,而不是发酵中产生的。
用未经自酶发酵的一般生产原料,经蒸煮破坏了自身存在的各种酶以后,直接用黑曲霉菌接种发酵,再经酸水解提取薯蓣皂素,其得率都在4~5%,使产量提高了100%以上。
另外,本发明采用薯蓣属植物为原料,经过黑曲霉菌一次发酵,可以在一套生产流水线上同时生产两种产品-薯蓣皂素和柠檬酸。在薯蓣属植物的根茎中,除含有各种皂甙和纤维素外,还含有大量的淀粉,在原有的自酶发酵中,或者分解为水和CO2跑掉,或者霉坏,或者随废酸液排掉,既使能从酸液中进一步利用淀粉,也必须先行中和,再经二次发酵才能转为其他产品。而本发明则是,在黑曲霉菌作用下,只需一次发酵,一边使淀粉水解并转变为柠檬酸,一边使薯蓣皂素分子中的糖链部分酶解,变为次级甙和糖。经过发酵(如附图流程图所示),可分别从发酵液中提取柠檬酸,从滤渣中提取薯蓣皂素。经过湖北进出口商品检验局检验,提取的薯蓣皂素和柠檬酸均达到出口标准。
可推荐的实施条件如下:
1.将5~15克薯蓣粉放入250ml锥瓶中,加50ml水,包好瓶口,用1.15~1.5kg/cm2的直接蒸汽灭菌,并糊化20~40分钟,冷至40℃以下,接种后封好瓶口,放在摇床上控温30~40℃,发酵3~4天,取出过滤,对滤液测定柠檬酸的含量和纯度;对滤渣用4NH2SO4常压水解2~4小时,过滤并洗至近中性,烘乾,转入凯氏抽提器中,用沸程60~90℃的石油醚(或120#汽油)浸提2~6小时,然后浓缩,结晶,干燥,称重并测熔点、熔距。
也可采用如下的工艺条件。
2.首先将薯蓣片浸泡,磨浆后泵入发酵罐内,使原料∶水=7~13∶50。在搅拌下用蒸汽升温至120~150℃灭菌,糊化20~40分钟,待冷至40℃以下即可接种,控温28~42℃,发酵2~4天。搅拌速率为120~180转/分。以还原糖接近为零为发酵终点。发酵结束时,升温至65~75℃蒸煮料浆并趁热打入板框压滤机进行过滤。发酵液用CaOO3中和后过滤、洗糖,得白色粉末晶体即为柠檬酸钙,再经加工可得柠檬酸或其他盐类。滤渣加酸调浆后,在0.8~4kg/cm2的蒸汽压下,水解2~6小时,然后打入板框压滤机压滤并洗至近中性,经气流干燥至水份含量<5%时投入提取罐,用120#汽油浸提至净。在浓缩罐浓缩至汽油∶皂素=30~60∶1时转入结晶罐结晶、过滤、干燥后称重并测熔点、熔距。
由于黑曲霉菌发酵没有任何负反应,而且经过发酵和水解后的两次过滤洗涤,水解物中色素和其他杂质都很少,所以产品质量稳定,熔点、熔距一次结晶即达标准。
这里,给出三个实施例。
例1 取1份薯蓣粉,加5份水,装入250ml锥瓶中用纱布和牛皮纸封好瓶口,用1.2~1.5kg/cm2蒸汽消毒20分钟,冷至40℃以下,从冰箱保存的斜面上,取B826菌种一环,放入经过充分消毒的纯水中,用玻璃球击散使其悬浮,取悬浮液1ml由注射器注入锥瓶中,在摇床上发酵114小时。由此可得总糖转酸率95.0%,柠檬酸纯度98.9%,薯蓣皂素4.48%,收率88.4%。

例3 取11份薯蓣粉,加50份水,加少量液化酶,依同法消毒后接种B828黑曲霉菌悬浮液1ml,发酵4天。可得总糖转酸率90.7%,柠檬酸纯度71.1%,薯蓣皂素得率4.95%,收率97.6%。
Claims (3)
1、一种以含薯预皂甙的薯蓣属植物为原料联产薯蓣皂素和柠檬酸的新工艺,其特征是用黑曲霉菌发酵,提高皂素得率。
2、如权利要求1所述的新工艺,其特征是通过发酵薯蓣皂素的分子结构保持不变。
3、如权利要求1所述的新工艺,其特征是用一种原料只经一次发酵,就可同时得到薯蓣皂素和柠檬酸两项产品,生产中也可任取其一。
