The α-adrenoceptor blocking effect of the test compound is expressed as the molar concentration of the compound required for 50% inhibition of contraction before the addition of norepinephrine (i.e., IC 50 value). Po 15 minutách, 30 minutách, 1 hodině a 2 hodinách se reakce zastaví tak, že se do každého vzorku přidá 0,7 M vodný roztok fluoridu sodného (0,5 ml) jakožto inhibitoru esterasy. Ke směsi se přidá 0,1 M fosfátový pufr (pH 7,6) a chlorid sodný (asi 1 g) a vzniklá směs se extrahuje diethyletherem (5 ml) . Diethyletherová fáze se pak zahustí pod proudem dusíku. Odparek se rozpustí v mobilní fázi (300 μΐ) , 10 μΐ roztoku se injikuje do kapalinového chromatografu s vysokou rozlišovací schopností a testovaná sloučenina a sloučenina A se stanoví za následujících podmínek. Výsledky jsou uvedeny v tabulce 3.-27(R)-1-(3-hydroxypropyl)-5-[2-[[2-[2-(2,2,2-trifluoroethoxy)phenoxy]ethyl]amino]propyl]-1H-indole-7-carboxamide, (Compound A) and the internal standard (R)-3-chloro-1-(3-hydroxypropyl)-5-[2-[[2-[2-(2,2,2-trifluoroethoxy)phenoxy]ethyl]amino]propyl]-1H-indole-7-carboxamide (1 μg) and the mixture was incubated at 37° C. After 15 minutes, 30 minutes, 1 hour and 2 hours, the reaction was stopped by adding 0.7 M aqueous sodium fluoride solution (0.5 ml) to each sample as an esterase inhibitor. 0.1 M phosphate buffer (pH 7.6) and sodium chloride (about 1 g) were added to the mixture, and the resulting mixture was extracted with diethyl ether (5 ml). The diethyl ether phase was then concentrated under a stream of nitrogen. The residue was dissolved in the mobile phase (300 μΐ), 10 μΐ of the solution was injected into a high-performance liquid chromatograph, and the test compound and compound A were determined under the following conditions. The results are shown in Table 3.

(R)-3-[7-carbamoyl-5-[2-[[2-(2-ethoxyphenoxy)ethyl]amino]propyl]-1-indol-1-yl]propylpivalate hydrochloride was suspended in 0.5% aqueous methylcellulose to a concentration of 100 mg/ml and administered orally to rats at a dose of 1000 mg/kg. No rats died 24 hours after administration.
