Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von 3,8-Dialkylxanthinen, die fuer die Verwendung in der Humanmedizin und Veterinaermedizin fuer therapeutische Zwecke zu Praeparaten verarbeitet werden. Durch das erfindungsgemaesse Verfahren werden 3,8-Dialkylxanthine der allgemeinen Formel und deren physiologisch vertraegliche Salze hergestellt, worin R hoch 1Aethyl, n-Propyl, n-Butyl, Isobutyl, n-Pentyl, 2-Methylbutyl, 3-Methylbutyl, 2,2-Dimethylpropyl, Cyclopropyl, Cyclobutan, Cyclopentyl oder Cyclohexylmethyl und R hoch 2Methyl, Aethyl, n-Propyl, Isopropyl, Cyclopropyl, n-Butyl, Isobutyl, Tertiaerbutyl oder Cyclobutyl bedeutet, wobei R hoch 1Aethyl ist, wenn R hoch 2Methyl bedeutet.

Anwendungsgebiet der Erfindung
Die durch das erfindungsgemäße Verfahren hergestellten 3,8-Dialkylxanthine können für die Verwendung in der Humanmedizin und Veterinärmedizin zu Präparaten zur Behandlung von Erkrankungen der Luftwege und der Herzgefäße verarbeitet werden.
Theophyllin und verschiedene' Salze desselben werden bei der Behandlung chronischer Erkrankungen, die die Luftwege verstopfen, und von Herzerkrankungen verwendet. Die therapeutischen liaupteffekte von Theophyllin sind die, daß es die
ί glätte Bronchienmuskulator entspannt und die Herzmuskulatur stimuliert. ·
Ziel der Erfindung ist die Herstellung neuer besserer Verbindungen für die Behandlung chronischer/ die Luftwege verstopfender Erkrankungen und von Herzgefäßerkrankungen.
Darlegung des Wesens der Erfindung
Die Aufgabe der Erfindung ist ein Verfahren zur Herstellung von Xanthinderivaten, die wirksamer als Theophyllin sind. Diese Verbindungen sind 3,8-Dialky!xanthine der allgemeinen Formel

und deren physiologisch verträgliche Salze, worin R Äthyl, n-Propyl, η-Butyl, Isobutyl, n-Pentyl, 2-Methylbutyl, 3-Methylbutyl, 2,2-Dimethylpropyl, Cyclopropyl, Cyclpbutyl, Cyclopentyl oder Cyclohexylmethyl bedeutet und R2 Methyl, Äthyl, n-Propyl, Isopropyl, Cyclopropyl, η-Butyl, Isobutyl, Tertiärbutyl oder Cyclobutyl bedeutet, wobei R Äthyl ist, wenn R2 Methyl bedeutet. Diese Verbindungen besitzen gegenüber Theophyllin eine bessere bronchienerweiternde Wirkung und Herzgefäßwirkung.
Die Erfindung.ist auch darauf gerichtet, daß Verbindungen, Vielehe sich strukturell von 'der Formel (1) ableiten, nach Verabreichung an lebende Organismen darin zu einer Verbindung der Formel (1) umgewandelt werden und in dieser strukturellen Form ihre Wirkungen zeigen. Dies ist ein weite-
rer Aspekt der Erfindung.
D.ie Erfindung betrifft auch pharmazeutisch verträgliche Salze von Verbindungen der Formel (1) mit pharmazeutisch verträglichen Basen. Unter dem Begriff "pharmazeutisch verträgliche Salze" versteht man Salze, deren Kationen relativ unschädlich für den tierischen Organismus sind, wenn sie in therapeutischen Dosen verwendet werden, so daß die vorteilhaften pharmakologischen Eigenschaften der Grundverbindun- gen der allgemeinen Formel (1) nicht durch Nebenwirkungen, die jenen Kationen zuzuschreiben sind, aufgehoben werden. Geeignete Salze sind beispielsweise die Alkalimetallsalze, wie Natrium- und Kaliumsalze, und Ammoniumsalze und Salze von Aminen, die in der Technik als pharmazeutisch verträglieh bekannt sind, wie von Glycin, Äthylendiamin, Cholin, Diäthanolamin, Triäthanolamin, Octadecylamin, Diäthylamin, " Triäthylamin, i-Ainino^-propanol-^-amino^- (hydroxymethyl)-propan-1,3-diol und i-(3,4-Dihydroxyphenyl)-2-isopropylaminoäthanol.
• Pharmazeutisch verträgliche Salze können durch Umsetzung stöchiometrischer Mengen einer Verbindung der Formel (1) . und der geeigneten Base, d.h. einer Base, wie sie oben be-. schrieben wurde, beispielsweise bei erhöhter Temperatur mit oder ohne ein geeignetes Lösungsmittel hergestellt werden, worauf vorzugsweise Umkristallisation des so gebildeten Salzes aus einem geeigneten Lösungsmittel, wie beispielsweise einem hydroxylhaltigen Lösungsmittel, erfolgt.
In der klinischen Praxis werden die Verbindungen nach der Erfindung normalerweise oral, rektal, nasal, sublingual, durch Injektion oder durch Inhalation in der Form eines pharmazeutischen: Präparates verabreicht, das den aktiven Bestandteil in der Form der ursprünglichen Verbindung oder gegebenenfalls in der Form eines pharmazeutisch verträglichen Salzes derselben in Verbindung mit einem pharmazeutisch verträglichen Träger enthält, welcher ein festes, halbfestes oder flüssiges Verdünnungsmittel oder eine ein-

nehmbare Kapsel sein kann, und solche Präparate stellen einen weiteren Aspekt der Erfindung dar. Gewöhnlich macht die aktive Substanz 0,1 bis 99 Gewichts-% des Präparates aus, wie beispielsweise 0,5 bis 20 Gewichts-% für Präparate für die Injektion und 0,1 bis 50 Gewichts-% für Präparate für orale Verabreichung.
Um pharmazeutische Präparate in der Form von Dosierungseinheiten für orale Anwendung mit einer Verbindung nach der Erfindung herzustellen, kann der Wirkstoff mit einem festen pulverförmigen Trägermaterial, wie beispielsweise Lactose, Saccharose, Sorbit, Mannit, einer Stärke, wie Kartoffelstärke, Maisstärke, Amylopectin, Lamin.arienpulver oder Zitruspulpepülver, einem Cellulosederivat, Polyvinylpyrrolidon oder.Gelatine vermischt werden und kann auch Schmiermittel,
wie Magnesium- oder Calciumstearat oder ein Carbowax ^ oder andere Polyäthylenglycolwachse enthalten und unter Bildung von Tabletten oder Drageekernen verpreßt werden. Wenn Dragees erforderlich sind, können die Kerne beispielsweise mit konzentrierten Zuckerlösungen, die Gummi arabicum, Talkum und/oder Titandioxid enthalten können, oder alternativ mit einem filmbildenden Mittel, das in leicht flüchtigen organischen Lösungsmitteln oder einem anderen geeigneten Lösungsmittel oder Gemischen organischer Lösungsmittel gelöst ist, beschichtet werden. Farbstoffe können zu diesen Überzügen zugesetzt werden, wie beispielsweise, um zwischen unterschiedlichen Gehalten des Wirkstoffes zu unterscheiden. Für die Herstellung weicher Gelatinekapseln (perlförmiger geschlossener Kapseln), die aus Gelatine und beispielsweise Glycerin als Weichmacher bestehen, oder zur Herstellung ähnlicher geschlossener Kapseln kann der Wirkstoff mit einem Carbowax ^ oder einem geeigneten Öl, wie beispielsweise Sesamöl, Olivenöl oder Erdnußöl, vermischt werden. Harte Gelatinekapseln können Granulate'des Wirkstoffes mit festen, pulverförmigen Trägermaterialien, wie Lactose, Saccharose, Sorbit, Mannit, Stärken (wie beispielsweise Kartoffelstär-
- ke, Maisstärke oder Amylopectin), Cellulosederivaten, Polyvinylpyrrolidon oder Gelatine, und außerdem auch Magnesium-
stearat oder Stearinsäure als Schmiermittel enthalten.
Eine Verbindung nach der Erfindung kann auch zu einer Dosierungsform mit verzögerter Wirkstoffabgabe unter Verwendung geeigneter Bindemittel verarbeitet werden. Es können unterschiedliche Methoden für die verzögerte Wirkstoffabgabe benutzt werden, wie Diffusionsverfahren und Ionenaustausch. Methoden unter Verwendung des Diffusionsverfahrens können beispielhalber durch Produkte erläutert werden, die beschichtete Granalien oder Teilchen, ein in ein Grundmaterial, eingebettetes Arzneimittel oder mäßig lösliche Formen einschließen.
Brausepulver können hergestellt werden, indem man den Wirkstoff mit ungiftigen Carbonaten oder Hydrogencarbonaten, wie beispielsweise Natrium-, Kalium- oder.Calciumcarbonaten oder -hydrogencarbonaten, wie Calciumcarbonat, Kaliumcarbonat und Kaliumhydrogencarbonat, festen ungiftigen Säuren, wie Weinsäure, Ascorbinsäure und Zitronensäure, und beispielsweise Aromastoff vermischt.
Flüssige Präparate für. orale Verabreichung können in der Form von Elixieren, Sirupen oder Suspensionen vorliegen, wie beispielsweise als Lösungen mit einem Gehalt von etwa 0,1 bis 20 Gewichts-% Wirkstoff, Zucker und einem Gemisch von Äthanol, Wasser, Glycerin, Propylenglycol und gegebenenfalls Aromastoff, Saccharin und/oder Carboxymethylcellulose als Dispergiermittel.
Für parenterale Verabreichung durch Injektion können Präparate eine wäßrige Lösung oder Suspension der Wirkstoffe nach der Erfindung, gegebenenfalls in. einer Konzentration von 0,5 bis 10 %, und gegebenenfalls auch ein Stabilisierungsmittel und/öder 'Puffersubstanzen in wäßriger Lösung enthalten. Dosierungseinheiten der Lösung können vorteilhafterweise in Ampullen eingeschlossen werden.
Die Dotierung, mit der die Wirkstoffe verabreicht werden,
!können in einem weiten Bereich variieren und hängen von verschiedenen Faktoren ab/ wie beispielsweise den individuellen Erfordernissen eines jeden Patienten. Ein geeigneter • oraler Dosierungsbereich liegt bei 50 bis 1000 mg ein- bis viermal täglich. Ein geeigneter Dosierungsbereich bei parenteraler Verabreichung liegt bei 20 bis 500 mg.
Die pharmazeutischen Präparate, die die Wirkstoffe enthalten, können zweckmäßig so bereitet werden, daß sie Dosierungen innerhalb dieser Bereiche entweder als einzelne Dosierungseinheiten oder als mehrere Dosierungseinheiten ergeben.

Beispiel 1 · ·
Herstellung von 3,7-Dihydro-3-äthyl-8-methyl-1H-purin-2,6-dion VI, D 4202 20
a) Herstellung von 6-Amino-1~äthyl-2,4-(1H, 3H)-pyrimidindion II
Zu einer Lösung von 127,5 g (1,5 Mol) Cyanoessigsäure und 200 ml Essigsäureanhydrid wurden 120 g (1,36- Mol) Äthylharnstoff zugesetzt. Die Lösung wurde 2 Stunden bei 60 bis 70° C gerührt. Nach dem Kühlen wurden weiße Kristalle abfiltriert und mit Äthanol gewaschen. Ausbeute 153 g (74 '%) (I). Diese wurde in 11 heißem Wasser gerührt, und 160 ml 2N NaOH wurden in Portionen derart zugesetzt, daß die Lösung die ganze Zeit über basisch war. Das Reaktionsgemisch wurde 20 Minuten, unter Rückfluß erhitzt und dann mit 5 N HCl neutralisiert. Nach dem Kühlen wurden weiße Kristalle abfiltriert. Ausbeute.100 g (65 %) (II) NMR: (DMSOd5) 3H, 35-1,20 t; 2H, 3,97 m; 2H, 7,03 s; 1H, 4,77 s; 1H, 10,50 s.
b.) Herstellung von 6-Amino-1-äthyl-5-nitroso-2,4-(1H, 3H)-pyrimidindion III
era
Zu- TOO g (0,65 Mol) 6-Amino-1-äthyl-2,4-(1H, 3H) -pyrimidindion (II), gelöst in 1 1 heißem Wasser, wurden 145 ml 5 N HCl und 50 g NaNO3 (0,72 Mol) gelöst in Wasser zugesetzt. Nach dem Kühlen wurden die roten'Kristalle abfiltriert und mit Wasser gewaschen. Ausbeute 97,7 g (83 %) (III) NMR: (DMSOd6) 3H, 1,27 t; 2H, 4,03 q; 1H, 9,30 b; 1H7 11,63 b; 1H, 13,50 s.
c) Herstellung von Sjö-Diamino-T-äthyl-^^-(1H, 3H)-pyrimidindion IV
Zu einer Suspension von 97 g 6-Amino-1-äthyl-5-nitroso-2,4-(1H, 3H)-pyrimidindion (III) wurden 240 g Natriumdithionit in Ante'ilen zugesetzt. Die schwach roten Kristalle wurden abfiltriert und mit Wasser gewaschen. Ausbeute 89 g (99 %) • IV. NMR: (DMSOd6) 3H, 1,23 t; 2H, 4,00 q; 2H, 2,50 - 3,70; 2H, 6,30 s. ' .
d) Herstellung von 3,7-Dihydro-3-äthyl-8-methyl-1H-purin-2,6-dion VI
Eine Lösung von 82 g 5/6-Diamino-1-äthyl-2,4-(1H, 3H)-pyrimidindion (IV) in 200 ml Essigsäure wurde 2 Stunden unter Rückfluß erhitzt. Die heiße Lösung wurde zu 1 .1 Aceton zugegeben. Die erhaltenen Kristalle wurden abfiltriert. Ausbeute 97,4 g V.
Das Amid (V) wurde in 300 ml 2N NaOH 1 Stunde unter Rückfluß erhitzt und dann mit 5N HCl neutralisiert. Die Kristal-Ie wurden abfiltriert und mit Wasser und Äthanol gewaschen. Ausbeute 57,8 g (62 %). NMR: (DMSOdg) 3H, 1,30 t; 2H, 4,10 g; 3H, 2,50 s; 1H, 10,97 s; 1H, 13,17 b.

Beispiel 2 .
Herstellung von 3,8-Diäthyl-3,7-dihydro-1H-purin-2,6-dion VII, D 4236
Eine Lösung von 5 g 5,6-Diamino-1-äthyl-2,4-(1H, 3H)-pyrimidindion (IV) in 25 ml Propionsäure wurde 2 Stunden unter Rückfluß erhitzt. Die heiße Lösung wurde zu 50 ml Aceton zugesetzt. Die erhaltenen Kristalle wurden abfiltriert. Ausbeute 5,4 g.
Das Amid wurde in 25 ml 2N NaOH 1 Stunde unter Rückfluß erhitzt und dann mit 5N HCl neutralisiert. Die Kristalle wurden abfiltriert und aus 40 ml Essigäsure umkristallisiert. Ausbeute 2,7 g (44%). NMRi (DMSOdg) 6H, 1,26 m; 2Hr 2,77 q; 2H, 4,03 q; 1H, 10,93 S;1H, 13,13 S.. ·
Herstellung von 3,7-Dihydro-8-äthyl-3-propyl-1H-purin-2,6-dion XIII,. D 4235 .
a.) Hersteilung von 6-Amino-1-propyl-2 ,4-(1K, 3H)-pyrimidindion IX
Zu einer Lösung von 47 g (0,55 Mol) Cyanoessigsäure und 100 ml Essigsäureanhydrid wurden 50 g (0,49 Mol) n-Propylharnstoff zugesetzt. Die Lösung wurde 1 Stunde bei 60 bis 70° C gerührt. Nach dem Kühlen wurden weiße Kristalle abfiltriert und mit Äthanol gewaschen. Ausbeute 56,2 g (68 %)' (VIII). Diese wurde in 100 ml heißem Wasser gerührt, und 60 ml 2N NaOH wurden portionsweise so zugegeben, daß die Lösung die ganze Zeit basisch blieb. Das Reaktionsgemisch wurde 20 Minuten unter Rückfluß erhitzt und dann mit 5N HCl neutralisiert. Nach dem Kühlen wurden weiße Kristalle abfiltriert. Ausbeute 34,3 g (61 %) (IX) NMR.
Ib) Herstellxing von 6-Amino-5-nitroso-1-propyl-2 ,4-(1H, 3H)-pyrimidindion X
Zu 34,3g (0,20 MoI)1 6-Amino-1-propyl-2,4-(1H, 3H)-pyrimidindion (IX), gelöst in 900 ml heißem Wasser, wurden 45 ml 5N HCl und 15 g NaNO9 (0,22 Mol), gelöst in Wasser, zugegeben. Nach dem Kühlen wurden die roten Kristalle abfiltriert und mit Wasser gewaschen. Ausbeute 33,3 g (83 %) (X) NMR.
c) Herstellung von 5,6-Diamino-1-propyl-2,4-(1H, 3H)-pyri-,midindion XI
33,3 g 6-Amino-5-nitroso-1-propyl-2,4-(1H, 3H)-pyrimidindion (X) wurden katalytisch in 800 ml DMF und in Gegenwart von 0,1 g PtO2 3 Stunden bei'Raumtemperatur und bei einem Druck von 200 kPa hydriert. Der Katalysator und die Kristalle wurden abfiltriert und mit Äthanol gewaschen. Ausbeute 29 g (93 %) (XI).
d) Herstellung von S^-Dihydro-S-äthyl-S-propyl-IH-purin-2,6-dion XIII
Eine Lösung von 2,5 g 5,6-Diamino-1-propyl-2,4-(1H, 3H)-pyrimidindion (XI) in 20 ml Propionsäure wurde 2 Stunden unter Rückfluß erhitzt. Die heiße Lösung wurde filtriert, und 50 ml Aceton wurden zugegeben. Die erhaltenen Kristalle wurden abfiltriert. Ausbeute 2,0 g (XII).
Das Amid (XII) wurde in 25 ml 2N NaOH 2 Stunden unter Rückfluß erhitzt und dann mit' 5N HCl neutralisiert. Die Kristalle wurden abfiltriert und aus 120 ml Äthanol umkristallisiert. Ausbeute 0,9 g .(68 %) (XIII) NMR: (DMSOdg) 3H, 0,97 q; 3H, 1,30 t; 2H,1,73 m;. 2H, 2,77 t;. 2H, 3,93 t, 1H, 10,90 s; 1H, 13,23 b. . ' 35

-21- 4Pharmakologische Versuche
Isolierte MeerschweinchenluftrÖhre
Meerschweinchen beiderlei Geschlechts mit einem Gewicht zwischen 150 und 250 g wurden mit einem Schlag auf den Kopf getötet und ausgeblutet. Die Luftröhre wurde entfernt und spiralig geschnitten und ergab ein oder zwei Präparate. Die Luftröhrenpräparate wurden in Organbädern befestigt, die Krebslösung enthielten, welche auf 37° C gehalten wurde und in die Carbogen (95 % O2 + 5 % CO2) eingeperlt wurde. Die isometrische Spannung, die hauptsächlich die Aktivität in . dem kreisförmigen Luftröhrenmuskel wiedergibt, wurde mit Hilfe eines Kraft-Verschiebungswandlers aufgezeichnet. Die Anfangsspannung wurde mit 0,5 g festgelegt, was die ungefähre Grundspannüng war, die während des Experimentes gehalten wurde. Bewertung von Entspannungseff'ekten erfolgte, wenn die Präparate sich durch die Zugabe von 0,1 μg Carbacholin je Milliliter des Bades zu einer stabilen Spannung kontrahiert hatten. EC^-Werte, d.h. molare Konzentrationen von Xanthinen, die erforderlich waren, um 50 % des maximalen Ansprechens-hervorzurufen, wurden aus logarithmischen Konzentrations-Ansprechkurven erhalten und verwendet, um die Stärke von Theophyllin im Vergleich mit jener des Testmittels zu berechnen. Nach dem Auswaschen der Mittel erlangte die Luftröhre ihren Grundtonus zurück, und man ließ sie wenigstens 15 Minuten sich stabilisieren, bevor die nächste Bewertung eines Mittels durchgeführt wurde. Zwischen zwei Bewertungen von Theophyllin wurde die Wirkung des Testmittels geprüft, und sein EC^-Wert wurdemitdem Mittel des vorausgehenden und des folgenden ECt-Q-Wertes von Theophyllin verglichen. In der Tabelle IV sind die Stärkeverhält— nisse erläutert. Theophyllin erhielt erhielt durch Definition den Wert 1, und ein größerer Wert als 1 zeigt, daß das Mittel stärker als Theophyllin ist.

- .1. Verfahren zur Herstellung von 3,8-Dialkylxanthinen der allgemeinen Formel0 HIl IHN C C-R2 (1)I "O=C C
N ^Noder physiologisch .-.verträgliche Salze derselben, worin R Äthyl, n-Propyl, η-Butyl, .Isobutyly n-Pentyl, 2-Methylbutylf 3-Methylbutyl, 2,/2-Dimethylpropyl, Cyclopropyl, Cyclobutyl, Cyclopentyl oder Cyclohexylmethyl bedeutet und R2 Methyl, Äthyl, n-Propyl, Isopropyl, Cyclopropylr η-Butyl, Isobutyl, Tertiärbutyl oder Cyclobutyl bedeutet, wobei R Äthyl ist, wenn R2 Methyl bedeutet, gekennzeichnet dadurch, daß mana) eine Verbindung der allgemeinen Formelmit einer Verbindung.der allgemeinen Formel R2-X, wo-. rin R und R2 die obige Bedeutung haben und X -COOHj, -CONH2 oder - OC-O-CO-R2 bedeutet, umsetzt und gegebenenfalls das erhaltene Produkt dehydratisiert oder b) eine Verbindung der allgemeinen Formel .
