The methyl-protected compound is best prepared by a synthesis as illustrated in Preparation 1 below. For this purpose, 3-methoxybenzenethiol and o-bromo-4-methoxyacetophenone in the presence of a strong base at relatively low temperaure to oi- (3-methoxyphenylthio) -4-methoxyacetophenon, and this compound is then by ring closure with a suitable ring closing agent, such as Polyphosphoric acid, converted at a correspondingly high temperature in the desired starting material.
The acylation according to the invention is a Friedel-Crafts acylation which is carried out in the usual way using aluminum chloride or aluminum bromide, preferably aluminum chloride, as the acylation catalyst.
The acylation is usually carried out in a solvent, and any inert organic solvent which is not significantly attacked under the reaction conditions can be used for this purpose. As such solvents are, for example, halogenated solvents such as dichloromethane, 1,2-dichloroethane or chloroform, or aromatic solvents such as benzene or chlorobenzene. Preferably, a halogenated solvent is used, such as in particular dichloromethane.
Toluene is quite easily acylated under the conditions of a Friedel-Crafts acylation, so that when using such a solvent, the toluene must be removed as completely as possible from the protected starting material in an earlier stage of the process in order to avoid a waste of acylating agent.
The acylations may be carried out at temperatures of about -30 0 C to 100 0 C, preferably being worked at about ambient temperature, namely at temperatures in the range of about 15 to 30 C C.

4 Benzoesäure, worin R Chlor oder Brom ist.4 benzoic acid, wherein R is chlorine or bromine.
4 Bei den bevorzugten Acylierungsmitteln bedeutet R Chlor.4 In the preferred acylating agents R is chlorine.
Specific examples of particularly preferred acylating agents are 4- (2-piperidinoethoxy) benzoyl chloride, 4- (2- (3-methylpyrrolidino) ethoxy) benzoyl chloride or 4- (2-pyrrolidinoethoxy) benzoyl chloride.
The examples below show that the acyl chloride to be used as the acylating agent can be prepared by reacting a corresponding carboxylic acid with a suitable chlorinating agent such as thionyl chloride. Care must be taken, however, that any excess of chlorinating agent is removed from the acyl chloride. It is best to form the acyl chloride in the reaction mixture and distill off the excess chlorinating agent under vacuum.
Conveniently, stoichiometric amounts of benzothiophene and acylating agent are used. If desired, a slight excess of one of the two reactants may also be added to ensure that the other reactant is completely consumed.
Preferably, a large excess of the acylation catalyst is used, for example an excess of about 2 to 12 moles, preferably about 5 to 10 moles, per mole of product.
The acylation proceeds rapidly. Reaction times of about 15 minutes to a few hours give the desired acylated intermediate already in high yield. If desired, longer reaction times can be used, but usually no further advantage. The lower the reaction temperature, the longer the reaction times.
238653 1238653 1
After completion of the stage of the acylation, the demethylation stage is started by addition of the respective sulfur compound (methionine or a compound of the general formula X-S-Y already mentioned) to the reaction mixture.
The sulfur compounds to be used are preferably alkylthiols, such as methanethiol, ethanethiol (preferred sulfur compound), isopropanethiol or butanethiol, dialkylsulphides, such as diethylsulfide, butyl-s-butylsulfide, ethylpropylsulfide, butylisopropylsulfide, dimethylsulfide or methylethylsulfide, benzenethiol, methionine or alkylphenylsulfides, such as methyl phenyl silane, ethyl phenyl sulfide or butyl phenyl sulfide.
Demethylation proceeds best with the use of a substantial excess of sulfur compound, namely an excess of about 4 to 10 moles of sulfur compound per mole of benzothiophene used as starting material.
The process, albeit less effectively, can also be carried out with a lower amount of sulfur compound, namely an amount of sulfur compound of about 2 or 3 moles per mole of starting material. Further, in this method, it is also possible to work with a small amount of sulfur compound, for example, an amount of sulfur compound of 2 or 3 moles per mole of the starting material, while still adding about 1 to 3 moles of an alkali metal halide such as sodium and potassium to improve the yield - or lithium chloride, iodide or bromide added.
A corresponding effect for sodium iodide is shown in Tetrahedron Letters 22 (1981) 4239 to 4240.
Kurze Reaktionszeiten im Bereich vonThe demethylation reaction goes well at about ambient temperature, namely at a temperature between 15 et un wa <ä 3O 0 C, and such operation is preferred. If desired, however, the stage of demethylation can also be carried out at temperatures between about -3O 0 C and about 50 0 C. Short reaction times in the range of

The product obtained after demethylation is recovered and isolated in a conventional manner. The complex of the acylation catalyst is normally decomposed by the addition of water, with the addition of a dilute aqueous acid being particularly suitable. The desired product often precipitates or can be extracted in the usual way with an organic solvent. The isolation of the desired product is further described in the examples below.
The products obtained in the process according to the invention can be obtained either in the form of the free bases or as acid addition salts, as is customary in the synthesis of amine-containing products. The compounds may be isolated, for example, in the form of salts of inorganic or organic acids, such as hydrobromic acid, hydriodic acid, sulfonic acids including naphthalenesulfonic acid, methanesulfonic acid and toluenesulfonic acid, sulfuric acid, nitric acid, phosphoric acid, tartaric acid, pyrosulfuric acid, metaphosphoric acid, succinic acid, formic acid, phthalic acid or lactic acid, wherein the salts of hydrochloric acid, citric acid, benzoic acid, maleic acid, acetic acid or propionic acid are preferred. By using dilute hydrochloric acid to decompose the catalyst complex, the product can be isolated, for example, in the form of the hydrochloride.

Reinigung von 6-Hydroxy-2-{4-hydroxyphenyl)-3-^4-(2-piperidinoethoxy)benzoyl/benzo/b/thiophen-HydrochloridPurification of 6-hydroxy-2- {4-hydroxyphenyl) -3- ^ 4- (2-piperidinoethoxy) benzoyl / benzo / b / thiophene hydrochloride
Man gibt 200 g des nach Beispiel 11 erhaltenen rohen 6-Hy-200 g of the crude 6-hydroxy obtained according to example 11 are
- - -"10-droxy-2- (4-hydroxyphenyl) -3- / 4- (2-piperidinoethoxy) benzoyl / benzo ^ b7-thiophene hydrochloride to 4400 ml of methanol and 60 ml of deionized water in a 5 liter flask." The slurry The residual solid is filtered off under vacuum using a filter aid, the reaction flask is equipped with a distillation head, and the solvent is distilled off until the remaining solution has a volume of about 1800 Then the heating mantle is removed and the solution is cooled very slowly with constant stirring overnight The resulting crystalline product is collected by vacuum filtration and the flask is washed with filtrate to recover the entire product The crystals are washed twice on the filter each 100 ml of cold methanol (below 0 0 C), and the washed product is under vacuum at 60 0 C dried, resulting in 140 g of dry product. - - -
The product is slurried in 3000 ml of methanol and 42 ml of water, heated to reflux temperature and then cooled very slowly. By subsequent filtration and drying of the product thus obtained in the manner described above, 121 g of highly pure product having a melting point of 259 to 260 0 C. is obtained.
