Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von Xanthin-Derivaten fuer die Anwendung als Arzneimittel. Ziel der Erfindung ist die Bereitstellung von neuen Verbindungen mit wertvollen pharmakologischen, insbesondere antiallergischen und bronchodilatorischen Eigenschaften. Erfindungsgemaess werden neue Xanthin-Derivate der allgemeinen Formel I hergestellt, worin beispielsweise bedeuten: R1 Wasserstoffatom oder Alkylradikal mit 1 bis 5 Kohlenstoffatomen; R2 gerades oder verzweigtes Alkylradikal mit bis zu 6 Kohlenstoffatomen u. a.; R3 Wasserstoffatom oder Alkyl-Hydroxyalkyl, u. a.; Y ist Wasserstoff- oder Halogenatom, Alkyl- oder Alkoxy-Radikal u. a.; Z ein Methylenradikal u. a.; A ein Amin-Rest; X eine Einfachbindung, ein Sauerstoffatom u. a. Formel I

Verfahren zur Herstellung von neuen Xanthin-Derivaten
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Xanthin-Derivaten mit wertvollen pharmakologiscnen Eigenschaften, insbesondere mit anti-allergischer und bronchodilatorischer Wirkung·
Die erfindungsgemäß hergestellten Verbindungen werden angewandt als Arzneimittel, beispielsweise für die Behandlung von Erkrankungen, bei denen es notwendig ist, die Antigen-Antikörper-Reaktlon zu inhibieren, sowie von Erkrankungen, bei denen eine bronchodilatorisehe Wirkung erwünscht ist, wie asthmatische Atemnot und chronische bronchopneumatische Verstopfungen·
Es sind keine Angaben bezüglich Verbindungen mit anti-allergischer und bronchodilatorischer Wirkung bekannt·
Es sind auch keine Angaben über Verfahren zur Herstellung von Xanthin-Derivaten bekannt.
Ziel der Erfindung ist die Bereitstellung von neuen Verbindungen mit wertvollen pharmakologischen, insbesondere antiallergischen und bronchodilatorisehen Eigenschaften, geringer Toxizität und langer Wirkungsdauer·

Die Kondensation wird vorzugsweise in einem Lösungsmittel, ausgewählt unter den Alkoholen mit bis zu 5 Kohlenstoffatomen, wie 2·. B· Methanol, Ethanol, Propanol oder Butanol, durchgeführt· Es ist vorteilhaft, bei einer Temperatur zwischen'64 0C und 130 0C und in Anwesenheit eines Akzeptors für die im Verlauf der Reaktion gebildete Wasserstoffsäure su arbeiten« Dieser Akzeptor kann ausgewählt werden unter den Alkalicarbo« naten wie Natriumcarbonat und Kaliumcarbonat, den tertiären Aminen wie Triethylamin, oder es kann auch ein Überschuß des Amin-Derivats der Formel III für die Reaktion verwendet werden·

Die Kondensation wird vorzugsweise in einem zweckmäßigen Lösungsmittel wie z. B. Dimethylformamid bei einer Temperatur zwischen 80 0C und 120 °C in Anwesenheit eines Akzeptors für die im Verlauf der Reaktion gebildete Wasserstoffsäure durchgeführt· Dieser Akzeptor kann u» a· ein Alkalicarbonat wie z· B· Natrium- oder Kaliumcarbonat sein.
Die neuen Derivate der allgemeinen Formel I können in ihre Ädditionssalze mit Säuren umgewandelt werden, deren Herstellung ebenfalls einen Teil der Erfindung darstellt« Als für die Bildung dieser Salze verwendbare Säuren kann man z«. B« aus der Mineralgruppe nennen? Chlorwasserstoffsäure, Bromwasserstoff säure, Schwefelsäure, Phosphorsäure und aus der organischen Gruppe Essigsäure, Propionsäure, Maleinsäure, fumarsäure, Weinsäure, Zitronensäure, Oxalsäure, Benzo©säure, Methansulfonsäure und Isethioneäure»
Diese neuen Derivate können nach physikalischen Methoden wie Kristallisation oder Chromatographie, sowie nach chemischen Methoden wie Bildung der Additionssalze mit Säuren und Zersetzung dieser Salze mit alkalischen Mitteln, gereinigt werden· \

Die Derivate der allgemeinen Formel I und ihre physiologisch tolerierbaren Salze besitzen interessante pharmakologische und therapeutische Eigenschaften, insbesondere anti-allergische und bronchodilatorische Eigenschaften, sowie inhibitorische Wirkungen der Phosphodiesterase· Ihre Toxizität ist gering und ihre DL1-Q, bestimmt an der Maus, ist höher als 100 mg/kg.auf intraperitonealem Weg und höher als 800 mg/kg auf oralem Weg«
Die bronchodilatatorische Aktivität wurde am Meerschweinchen untersucht, nach der Methode von H. Konzett und R. Rossler, Arch. Exp. ü· Pharnu* 195, 71 (1940). Man hat auf diese Weise beobachtet, daß die Derivate der vorliegenden Erfindung, injiziert auf intravenösem Weg in variablen Do~ sen von 1 bis 5 mg/kg, je nach Derivat, vollständig die durch intravenöse Verabreichung entweder von Histamin oder von Serotonin hervorgerufenen Bronchospasmen inhibieren sowie teilweise die Wirlaing von Acetylcholin und der "Slow
ν Reacting Substance" aufheben.
Im Tost von A. K. Armitage, Brit. J. Pharma· col. 17, 196 (1961) inhibieren die Derivate der vorliegenden Erfindung, auf oralem Weg in variablen Dosen von 0,5 bis 10 mg/kg verabreicht, je nach Verbindung, 50 % der am Meerschweinchen durch, ein 4 SSiges Histamin—Aerosol hervorgerufenen Wirkung. Bei einigen dieser Verbindungen ist die Wirkung nach 48 Stunden nach der oralen Verabreichung sehr beträchtlich·
Außerdem besitzen einige Verbindungen der Erfindung eine spezifische agonistische Wirkung gegenüber den zentralen und peripheren purinergischen Rezeptoren des Typs A1 und/oder A2, die zu einer spezifischen therapeutischen Wirkung führen kann·

Als nicht einschränkendes Beispiel kann angegeben werden, daß die orale Verabreichung von 5 mg/kg der Verbindung von Beispiel 1, 48 Stunden lang den durch ein Histamin-Aeroeol (4 %ig) hervorgerufenen Bronchospasmus inhibiert» Außerdem wurde eine Verringerung der anaphylaktisehen Hautreaktion bei der Ratte nach einmaliger Verabreichung von 20 mg/kg per os dieser Verbindung beobachtete
Diese Verbindung des Beispiels 1 ist bei den Rezeptoren A2 (IO 60 « 15/uM) stärker agonistisch als bei den Rezeptoren Af (10.50 >100/uM). -
Die oben beschriebenen pharmakologisehen Eigenschaften sowie die geringe Toxizität der Derivate der allgemeinen Formel I und ihrer physiologisch tolerierbaren Salze ermöglichen ihre Verwendung in der Therapeutik, insbesondere zur Behandlung aller Erkrankungen, bei denen es notwendig ist, die Antigen-Antikörper-Reaktion zu inhibieren, wie antoimmune9 allergische und inflammatorische Erkrankungen und insbesondere diejenigen, bei denen eine bronchodilatatorische Wirkung erwünscht ist» wie asthmatische Atemnot und chronische bronchopneumatische Verstopfungen, insbesondere in spastischer Form» Die lange Wirkungsdauer gestattet eine Krisenbehandlung auch dann, wenn die Grundbehandlung der asthmatischen Erkrankung einfach oder kompliziert ist*
Außerdem sind die spasmolytischen Eigenschaften eine Indikation zur Behandlung von nephretischen oder hepatitischen Koliken·
Die vorliegende Erfindung hat ebenfalls das Verfahren zur Herstellung der pharmazeutischen Zusammensetzungen zum Gegenstand, die als Wirkstoff ein Derivat der allgemeinen Formel I oder eines seiner physiologisch tolerierbaren

Salze, gemischt oder verbunden mit einem pharmazeutisch geeigneten Grundstoff enthalten· Sie betrifft insbesondere die Herstellung der Dosierungsformen, die 25 bis 250 mg Wirkstoff enthalten.
Die auf diese Weise erhaltenen pharmazeutischen Zusammensetzungen werden vorteilhafterweise in verschiedenen Dosierungsformen dargeboten, wie z. B· Tabletten, Dragees, Gele, Glosetten oder für eine sublinguale Verabreichung geeignete galenisehe Präparationen, Suppositorien, injizierbare oder trinkbare Lösungen sowie für eine Verabreichung als Aerosol angepaßte Formen· Sie können auf oralem, rektalem oder parenteralem Weg in Dosen von 25 bis 250 mg Wirkstoff, einbis zweimal pro Tag angewendet werden·
Die im folgenden gegebenen, nicht einschränkenden Beispiele veranschaulichen die Erfindung·

Bine Suspension von 12,2 g 1-Methyl-3-isobutyl«»8<-broiaethylxanthin undS523 g 1-Bens3hydryl»pip@raz±n in 200 ml Ethanol wird unter Rückfluß erhitzt« Man beobachtet eine fortschreitende Auflösung und erhitzt daher 20 bis 24 Stunden lang» Man dampft dann bis zur Trockne ein und nimmt den Rückstand in einer 10 %igen Hatriumbicarbonat-Lösung auf· Man extrahiert mehrere Male mit Methylenchlorids trocknet über Natriumsulfat und konzentriert bis zur Srockne* Der ölige Rückstand wird durch Flash-Chromatographie über 1 kg Silicagel 0,04 - 0,063 mm gereinigt, indem mit Ethylacetat und anschließend mit einer .Ethylacetat-Methanol-Mischung (95/5) " eluiert wird·, Nach Fraktionierung Und Eindampfen des Eluats erhält man 14 g des reinen Produkts, das bei 200 bis 202 0C schmilzt.
Das 1-Methyl-3-isobutyl~8-bromethyl-xanthin als Ausgangsprodukt, das bei 210 0G schmilzt, wurde durch Bromierung mittels 48 %iger HBr des entsprechenden Methoxy-8-ethyl-Deriva.ts, das bei 1630C schmilzt, hergestellt, während dieses wiederum durch Cyclisierung von 1-Isobutyl-3-methyl-2,4-dioxo-1,2,3,4-tetrahydro-5-(3t-niethoxy-»propionamido)-6-aminopyrimidin (S1P = 200 °C) erhalten wurde« Dieses vorgenannte Produkt wurde durch Kondensation von 3 Methoxy-propionsäure mit 5,6-Diamino-1-isobutyl-3-methyl-2,4-dioxo-1,2,3*4-tetrahydropyrimidin hergestellt und dieses durch Reduktion des entsprechenden 5-Nitroso-Derivats (Fp = 228 0C) in Anwesenheit von Raney-Nickel als Katalysator unter einem Wasserstoff druck von 6 Atmosphären erhalten* Das letztgenannte Produkt schließlich wurde durch litrosierung von 1-Isobutyl-3-methyl-2,4-dioxo-6-amino-1,293?4-tetrahydro-pyrimidin mit HaNO2/CH3COOH hergestellt.

10 S 1-Methyl-3-isobutyl-8-^2-(4-diphenylmethyl-piperazinyl)-ethylJ7-xanthin und 37,6 g Kaliumcarbonat werden unter Rühren in 200 ml Dimethylformamid auf 100 0C gehalten· Man fügt schnell 10 ml Glycol-chlorhydrin hinzu und rührt 1 Stunde lang bei 110 0C* Dann setzt man schnell weitere 20 ml Glykolchlorhydrin hinzu, hält die Mischung 1 Stunde lang unter Rühren bei 110 0C und gibt dann 40 g Kaliumcarbonat, sowie noch 20 ml Glykolchlorhydrin zu. Die gesamte Mischung wird 3 Stunden 30 Minuten lang bei 110 0C und anschließend 72 Stunden lang bei Umgebungstemperatur gehalten· Anschließend wird die Mischung bis zur Trockne konzentriert und der Rückstand in einem Methylenchlorid-Wasser-Gemisch aufgenommen· Die organische Phase wird über Natriumsulfat getrocknet und dann bis zur Trockne eingedampft· Anschließend wird eine Chromatographie über 750 g Silicagel (0,04 0,063 mm) durchgeführt. Man eluiert zuerst mit Methylenchlorid, dann mit reinem Ethylacetat, dann mit einer Ethylacetat-Methanol-Mischung (95/5) und schließlich mit einer Ethylacetat-Methanol-Mischung ($0/10).

Man erhält 8,6 g der Base, die man in 50 ml Isopropanol löst* Dann gibt man ausreichend Chlorwasserstoff/Ether hinzu, um ein leicht saures Milieu zu erhalten· Man fällt das Hydrochlorid des gesuchten Produkts mit einem großen Überschuß an wasserfreiem Ether· Dann zentrifugiert man und wäscht mit Ether· Nach Trocknung bei 115 0C unter einem Druck von 0,6 mm Hg erhält man 8,7 g 1-Methyl-3-isobutyl-7-(2-hydroxy«-ethyl)- 8-£ 2-( 4-diphenylmethyi-piperazinyl )-ethyl .7-xanthin-Dihydrochlorid, Pp (Kapillare): 190 - 200 0G (Isopropanol/Ether), Ausbeute 71 %·
Auf die gleiche Art ind Weise wurde hergestellt:
1 -Me thyi-3-isobutyl- /-( 2,3-dihydroxy-propyl) -8-^" 2-( 4-di·» phenylmethyl-piperaiinyl)-ethyl^7-xanthin, dessen Dihydro» Chlorid bei 145 - V.:.'5 0C in der Kapillare schmilzt (Isopro panol/Ether)·
