Charakteristik der bekannten technischen Lösungen Characteristic of the known technical solutions
For the preparation of granules containing active ingredients or drug mixtures with poor water solubility, these active ingredients or Vi'irkatoffmischungen are commonly processed together with readily water-soluble excipients, such as urea, succinic acid, ammonium chloride, etc. The readily water-soluble excipients promote the wettability of the resulting granules and cause better solubility of the incorporated Yiirkstoffe or Wirkst off mixtures.

The simultaneous use of the active ingredients and water-soluble excipients with the properties described leads to powder mixtures of low cohesion, the granulation of which is difficult. Above all, these difficulties are manifested by the fact that, on addition of the binder solutions or dispersion dispersions customarily used, Yerklumpungen occur and / or the resulting granules have poor water solubility. Although in DD-W? I43 213 a method for granulation is described, which leads by the use of specific binders to granules and in particular tablets with good disintegratability · Bs has been shown, however, that this method is not suitable for the aforementioned purpose and thereby the same difficulties as in the commonly used Granulation occur, in particular clumping in the addition of the binder solution or dispersion, poor wetting of the substance mixture to be granulated, uneven distribution of the binder, losses of binder due to spraying the binder solution or dispersion, a poor drying behavior of the fresh granules, Nachhärteeffekte the Granules, to list only the serious disadvantages. According to the generally accepted view on agglomerate formation (eg Leuenberger, Pharmac.Acta Helvetiae ^ 7, 72 (1982), Dittgen et al., Pharmacie 2Ü 2 37 (1980)), the main causes of most of the difficulties mentioned are the lack of wetting of the Powder mixture through the binder solution or dispersion. Köhler (Dissertation Wr. 4386 ETH Zurich 1969, p. I60) found that one can avoid these difficulties very well by using water vapor as Granulierflüssigkeit, due to its relaxed surface, a good wetting and agglomeration against 1 over weakly cohesive and complicated powder blending
The disadvantage of this in principle achievable granule erving jf ahrens exists for the aforementioned Anv.'endungszvieck above all in that due to the high temperature of the water vapor between 370 and 400 K usually thermolabile agents are damaged.
Ziel der Erfindung N > Object of the invention N >
The object of the invention is a process for granulating weakly cohesive powder mixtures, which does not have the stated disadvantages of the aforementioned processes. In particular, the process should be suitable for the disadvantages caused by conventional processes and granulation binders with respect to the wetting of the powder to be granulated, the solubility of the finished granules and the post-curing of the same to avoid, wherein the applied technology must not cause any thermal stress on the ent, held active ingredients.
A process has been developed for the granulation of low-cohesive powder mixtures, which is based on the use of a binder of a film former and salts of weak acids. The film formers are carboxyl- or carboxyvinyl-containing copolymers of acrylates and / or celluloses, which have a temperature of between 275 and 380 K, but preferably between 290 and 320 K, with between 275 and 380 K, preferably between 290 and 320 K, warm aqueous liquid are swellable.
The swelling capacity of the binder, which is required in a variety of practical requirements in different ways, but for the agglomeration in the granulation is absolutely necessary, can be adjusted and amplified from the application point of view many times desired by the addition of salts of weak acids.
Salts of organic acids are suitable for this purpose, which are harmless, physiological and environmentally harmless. These are salts of weak organic acids which react alkaline in aqueous solution, preferably the alkali metal and / or alkaline earth metal salts of carbonic, tartaric and citric acids. For granule forming various devices and technologies are known. Wet granulation is most widespread, using a suitable binder solution or dispersion or a solvent for agglomeration of the powder particles contained in the powder mixture to be granulated. Typical granulation methods are, for example, the kneader, VUr 'belschichtsprüh-, plate and high-speed mixer. It is characteristic of the process according to the invention that these conventional granulation processes can be maintained if the binder is added in solid, preferably finely dispersed form to the powder mixture to be granulated and then granulated with an aqueous liquid.
It is furthermore characteristic of the granulation process according to the invention that during the granulation process both the temperature of the powder mixture and that of the granulation liquid is between 275 and 380, but preferably between 290 and 320 K, whereby the active substances contained in the powder mixture do not thermally or only slightly be charged.
In particular, weakly cohesive powder mixtures which are obtained can be granulated in this way if hydrophobic and poorly water-soluble active ingredients have to be processed together with readily water-soluble auxiliaries, such as urea, succinic acid, ammonium chloride, etc., in order to achieve the solubility of the product from a practical point of view To achieve active ingredient. It is characteristic of the process according to the invention that mixtures of this kind are present in a wide mixing ratio.
liältnis zwischen Wirk- und Hilfsstoffen granulieren lassen, daß aber in den Granulaten die beabsichtigte verbesserte Löslichkeit der Wirkstoffe weiterhin gegeben bzw. in vielen Fällen noch weiter verbessert ist.granularity between active ingredients and excipients, but that in the granules the intended improved solubility of the active ingredients continues to exist or in many cases is even further improved.
Thus, the granulation method according to the invention is preferably also suitable for improving the solubility of the active ingredients and, if appropriate, adjusting it to a specific application.
