Die Erfindung betrifft ein Verfahren zur Herstellung von Aminopropanol-Derivaten von 1,4:3,6-Dianhydro-hexit-nitraten, die zur Vorbeugung und Behandlung von Herz-iCreialauferkrankungen geeignet ainde
In der europäischen Patentpublication 167 008 wurden erstmals 1,4:3,6-Dianhydro-hexit-Derivate beschrieben, die eine substituierte З-Атіло-2-propanol-Seitenkette enthalten. Bekanntlich stellt diese funktioneile Gruppe das typische Strukturmerkmal einer blasse sogenannter ß-Rezeptorenblocker dar. Sämtliche derzeit in der Therapie eingesetzten Vertreter dieses Wir^stofftyps enthalten als basischen Rest entweder eine Iaopropylamino- oder eine uert.-Butylamino-Gruppe, Uach den bisher vorliegenden Befunden scheinen beide genannten Amine-das Optimum der synthetischen Bemühungen auf dem Gebiet der ß-antagoniatischen Wirkungen darzustellen. Die in EP 1б7 .008 vorbeschriebenen Verbindungen enthalten als AmIn Komponente geradkettige oder verzweigte Alkyl- und Dialkylamine bzw. solche, in denen der Aminstickstoff Bestandteil eines Purin-Ringsystems ist. Beim Austausch dieser bekannten лтіп-Reste gegen die in der vorliegenden Erfindung geoffenbarten tritt eine in keiner V/eise vorhersehbare und somit völlig unerwartete deutlich positive qualitative und quantitative Beeinflussung der pharmakologiseilen Wirkung auf. Dieser Befund steht im »Viderspruch zur vorherrschenden
Lehrmeinung, wonach die oben erwähnten, verzweigten, sekundären ITiederalkylaminreste die bevorzugte funktionelle Gruppe bei den Aryloxypropanolaminen darstellt.
Es muß deshalb als überraschend angesehen werden, daß mit den neuen enindungsgemäß hergestellten Verbindungen sowohl eine Steigerung, als auch eine LiOdifizierung der therapeutischen Wirksamkeit erzielt werden kann. Dies gestattet einerseits eine Verringerung der Dosis, so daß bei vergleichbarer Toxizität die therapeutische Breite vergrößert und damit die Arzneimittelsicherheit erhöht wird. Andererseits erfolgt auch eine Verschiebung der '.i'irkungsquälitat, so daß eine Ausweitung der Indikationsgebiete und somit der Anwendu-igsbreite ermöglicht wird. Die erfindungsgemäß hergestellten Verbindungen sind nicht nur wirksamer als Betarezeptoren, sie haben auch eine unerwartete Wirksamkeit als Vasodilatoren.
Der Erfindung liege die Aufgabe zugrunde, ein Verfahren zur Herstellung neuer Aminopropanol-Derivate von 1, 4:3,6--Dianhydro" hexit-nitraten zur Verfugung zu stellen.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer ^minopropanol-Derivate von 1,4:3>6-Dianhydrohejcit-nitraten der allgemeinen Pormel I

1,4:3,6-Dianhydro-hexite besitzen vier chirale Zentren, nämlich die Kohlenstoffatome 2,3,4 und 5. Sie stellen deshalb optisch aktive Verbindungen dar. Die zur Herstellung der erfindungsgemäßen Verbindungen eingesetzten Ausgangsprodukte liegen in optisch reiner Form, als D-Isomannid, L-Isoidid und D-Isosorbid vor. besitzt im Kohlenstoffatom 2 ebenfalls ein Asymmetriezentrum, was der Anlaß zur Existenz von R- und S-konfigurierter, enantiomerer Formen dieser Seitenkette ist. Aus diesem Grund treten die erfindungsgemäßen Verbindungen der allgemeinen Formel I als Diastereoisomere auf. Sowohl die Diastereoisomerengemische, als auch die getrennten, konfi-
gurativ einheitlichen Komponenten sind Gegenstand der vorliegenden Erfindung.
Die Verbindungen der allgemeinen Formel I können in an sich bekannter Weise, auf verschiedenen Wegen hergestellt werden. Einige dieser Wege sind in dem nachfolgenden Formelschema dargestellt.
Danach sind die erfindungsgemäßen Verbindungen der allgemeinen Formel I, worin X die oben angegebene Bedeutung besitzt, z.B. dadurch erhältlich, daß man ein Isohexid-nitrat der Formel II entweder direkt, durch Umsetzung mit einem 2,3-Epoxy-1-halogen-propan, oder indirekt, durch Umsetzung mit einem Allylhalogenid, über die Zwischenstufe III und nachfolgende Oxidation, in ein O-(2,3-Epoxypropyl)-isohexidnitrat der Formel IV überführt und dieses durch Reaktion mit HX, wobei X die angegebene Bedeutung besitzt, zum gewünschten Produkt I umsetzt.
Die Reaktion der Verbindung der Formel IV mit HX wird durchgeführt entweder in einem geeigneten inerten Lösungsmittel oder ohne Lösungsmittel. Geeignete Lösungsmittel schließen polare protische Lösungsmittel, wie niedere Alkohole, offenkettige oder zyklische Ether oder aprotische Lösungsmittel ein. Bevorzugte Lösungsmittel sind Methanol, Ethanol, 1-Propanol, 2-Propanol, 1-Butanol, Tetrahydrofuran, Dioxan, Dimethoxyethan, Diethoxyethan, Acetonitril, DMF und DMSO. Methanol, Ethanol, 1-Propanol und 2-Propanol werden besonders bevorzugt. Die Reaktion findet bei einer Temperatur zwischen etwa 0" und etwa 150eC statt, vorzugsweise zwischen Raumtemperatur und der Rückflußtemperatur der entsprechenden Reaktionsmischung. Die Zeit zur Vervollständigung der Reaktion ist natürlich von der angewandeten Temperatur abhängig und kann variieren von 1 Stunde bis 48 Stunden. Besonders wenn bei Raumtemperatur ausgeführtj läuft die Reaktion etwa 20 bis etwa 30 Stunden und wenn sie bei Rückflußtemperatur ausgeführt wird etwa 1 bis 6 Stunden.
Das Verhältnis der Verbindung IV zu HX kann ein stoichiometrisches Verhältnis wie auch einen Überschuß von jeder der Komponenten von zwischen etwa 1.5:1 und etwa 1:5 einschließen. Vorzugsweise wird ein beinahe stoichiometrisches Verhältnis benutzt.
Eine weitere Möglichkeit zur Herstellung der erfindungsgemäßen Verbindungen besteht in der Umsetzung eines Isohexids der Formel V zu einem 0-(2,3-Epoxypropyl)-isohexid der Formel VII, wobei diese Umsetzung sowohl direkt, mittels eines 2,3-Epoxy-1-halogen-propans oder in 2 Stufen, durch Reaktion von V zunächst mit einem Allylhalogenid unter Bildung des Allylethers VI und nachfolgende Oxidation zu VII erfolgen kann. Danach setzt man VII mit HX um, wobei X die angegebene Bedeutung besitzt, unter Bildung eines 0-(3-Amino-2-hydroxypropyl)-isohexids VIII und überführt dieses in die gewünschte Verbindung I durch Veresterung mit Salpetersäure.
Schließlich führt ein weiterer Syntheseweg über die O-Allylisohexide VI, die wie vorstehend angeführt erhältlich sind, indem man diese mit Salpetersäure zu den O-Allyl-isohexidnitraten III verestert und diese dann, wie oben beschrieben, in die Endprodukte I umwandelt.
Die Nitratesterbildung, die die Schritte von VI bis III und von VIII bis I im Reaktionsschema umfaßtfkann durchgeführt werden indem man eine Verbindung VI oder VIII reagieren läßt respektive mit einem Nitratester bildenden Reagenz wie rauchende Salpetersäure oder eine Mischung von rauchender Salpetersäure und konzentrierter Schwefelsäure oder eine Mischung von 65 Prozent Salpetersäure und Essigsäureanhydrid unter niedriger Temperatur in Anwesenheit oder Abwesenheit eines inerten Lösungsmittels. Die Reaktionstemperaturen liegen im allgemeinen im Bereich zwischen etwa -60eC und Raumtemperatur, vorzugsweise zwischen etwa 0"C und -20eC Das molare Verhältnis von Reaktanden liegt im Bereich von etwa 1 zu etwa 10.

Die hier aufgeführten Umsetzungen erfolgen ausnahmslos in Analogie zu bekannten Methoden und können deshalb von jedem Fachmann ohne weitere Erläuterungen nachvollzogen werden. Deshalb erübrigen sich an dieser Stelle Angaben zu Lösungsmitteln, Reaktionsbedingungen und -zeiten sowie zu Aufarbeitung und Isolierung der Produkte.
Läßt man die Verbindungen II bzw. V mit racemischen Halogenepoxypropanen reagieren, so resultieren daraus Diastereoisomerenpaare von IV bzw. VII. Das Gleiche ist der Fall, wenn die entsprechenden Allylether III bzw. VI epoxydiert werden. Setzt man hingegen enantiomerenreine Epoxide mit II bzw. V um, so erhält man jeweils nur ein einheitliches diastereoisomeres Produkt IV bzw. VII. Dementsprechend sind auch alle weiteren Folgereaktionen, von IV zu I bzw. von VII über VIII zu I sowohl mit den Diastereoisomerengemischen, als auch mit einheitlichen Diastereoisomeren durchführbar. Außerdem kann im Falle des Vorliegens von Diastereoisomerengemischen sowohl auf jeder der Zwischenstufen IV, VII und VIII, als auch auf der Stufe der erfindungsgemäßen Verbindungen I eine Trennung in die beiden optisch einheitlichen Komponenten erfolgen. Dabei können die üblichen Trenntechniken, wie z.B. Kristallisation, Destillation, chromatografische Verfahren usw., zur Anwendung kommen.
Nach beendeter umsetzung werden die Reaktionsprodukte der allgemeinen Formel I auf übliche Weise isoliert, gereinigt und gegebenenfalls in das Salz einer anorganischen oder organischen Säure, vorzugsweise in ein pharmazeutisch akzeptables Salz überführt. Beispiele für derartige Salze sind Hydrochloride, Hydrobromide, Nitrate, Sulfate, Phosphate, Acetate, Oxalate, Maleate, Fumarate, Tartrate, Lactate, Maleinate, Malonate, Citrate, Salicylate, Methansulfonate, Benzolsulfonate, Toluolsulfonate und Naphthalin-
35 sulfonate.
Diese oder andere Salze der neuen Verbindungen, wie z.B. Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freie Base in ein Salz überführt, dieses abtrennt und gegebenenfalls umkristallisiert oder anderweitig reinigt und aus dem Salz wiederum die Base freisetzt.
Die als Ausgangsprodukte eingesetzten Verbindungen der allgemeinen Formel II und V sind seit langem bekannt. Die Zwischenprodukte III, IV, VI und VII finden bereits in der EP 167 008 Erwähnung. Lediglich die Verbindungen VIII stellen neue Zwischenprodukte auf dem Weg zu den erf indungs'gemäßen Strukturen der allgemeinen Formel I dar.
Die erfindungsgemäßen Verbindungen zeichnen sich überraschenderweise durch ein breites pharmakologisches Wirkungsspektrum aus und stellen deshalb wertvolle Arzneistoffe dar. Sie besitzen insbesondere eine ausgeprägte kardiovaskuläre Wirkung. An isolierten Venenstreifen des Hundes wirken sie
in geringerer Konzentration gegen die K+-induzierte Kontraktion als an Aortenstreifen. An isolierten Meerschweinchenherzen steigern sie den Koronarfluß dosisabhängig. An narkotisierten Ratten verhindern sie den Lypressin-Koronarkrampf dosisabhängig und steigern die Überlebensrate von Ratten nach einem akuten Herzinfarkt signifikant. An normotonen, wachen Hunden senken sie nach oraler Verabreichung den systolischen Blutdruck für die Dauer von 6-8 Stunden. An narkotisierten, thorakotomierten Hunden mit künstlicher Beatmung vermindern sie den arteriellen Mitteldruck, den linksventrikulären systolischen, den linksventrikulären enddiastolischen Druck sowie den Druck in der Arteria pulmonalis und reduzieren den peripheren Gefäßwiderstand. Im Gegensatz zu den bekannten organischen Nitratestern führen die erfindungsgemäßen Verbindungen auch an spontanhypertonen Ratten zu einer langdauernden Senkung des systolischen Blutdrucks.
Ein derartiges Wirkungsprofil ist von keiner der vorbeschriebenen Substanzen mit ähnlicher Struktur bekannt. Die hier vorgestellten, neuen Verbindungen stellen deshalb eine unerwartete Bereicherung der Pharmazie dar. Sie eignen sich zur Vorbeugung und Behandlung von Herz- und Kreislauferkrankungen, insbesondere des Bluthochdrucks, und auch für den Einsatz bei Angina pectoris und Lungenhochdruck.
Zur Erläuterung der vielfältigen pharmakologischen Wirkungen werden nachfolgend einige der an den Beispielen 17, 19 und 20 gewonnenen Daten mitgeteilt.
Bei der Ermittlung der orientierenden Toxizität der Verbindungen nach Beispiel 17, 19 und 20 vertrugen Mäuse 500 mg/kg peroral ohne klinische Symptome und ohne Todesfolgen.

Gegenstand der Erfindung ist deshalb auch die Verwendung von Verbindungen der allgemeinen Formel I und ihrer Salze als Arzneimittel, insbesondere als solche zur Behandlung und Prophylaxe von Herz- und Kreislauferkrankungen. Dabei können die Arzneimittel mindestens eine Verbindung der allgemeinen Formel I/ gegebenenfalls in Form eines ihrer physiologisch verträglichen Säureadditionssalze/ als Wirkstoff enthalten und allein oder vermischt mit geeigneten Trägerstoffen verabreicht werden. Derartige Arzneimittel dürfen die erfindungsgemäßen Verbindungen oder deren Salze in einem Gehalt von 0,1 bis 99,9 % enthalten. Die Dosierung kann wie gewünscht gewählt werden und im Bereich von 1 mg bis 500 mg liegen.
Als pharmazeutische Darreichungsformen kommen alle, dem Fachmann geläufigen Formulierungen in Frage, wie z.B. Suppositorien, Pulver, Granulate, Tabletten, Kapseln, Suspensionen, Liquida, Injectabilia und transdermale Systeme. Zur Herstellung pharmazeutischer Darreichungsformen können feste, halbfeste oder flüssige Trägermaterialien oder Verdünnungsmittel eingesetzt werden. Darin sind eingeschlossen Corrigentien, Bindemittel, Gleitmittel, Emulgatoren usw.. Beispiele für derartige Mittel sind: Stärke, wie Kartoffel- und Getreidestärke, Zucker, wie Lactose, Sucrose, Glucose, Mannitol, Sorbitol, Cellulose, wie kristalline Cellulose, Methyl-cellulose, Calcium-carboxymethyl-cellulose, Natriumcarboxymethyl-cellulose und Hydroxypropyl-cellulose, anorganische Materialien wie Kaliumphosphat, Calciumsulfat, Calciumcarbonat und Talkum, Gelatine, Gummiarabicum, PoIyvinylpyrrolidon, oberflächenaktive Substanzen wie Fettsäureglyceride, Fettsäure-sorbitan-ester, Fettsäureester von Sucrose und Polyglycerol und andere.
Einige Beispiele für Arzneimittelformulierungen, unter Be-5 nutzung der erfindungsgemäßen Verbindungen, sind nachfolgend aufgeführt:

Die nachfolgenden Beispiele dienen zur Erläuterung der Erfindung. Sofern nicht anders vermerkt, handelt es sich bei den in den Beispielen aufgeführten Verbindungen jeweils um Diastereoisomerengemische. Die angegebenen Drehwerte [a]*° wurden bei einer Konzentration von с = 1 in Methanol gemessen

Die Herstellung der als Zwischenprodukte eingesetzten Allyl- und Epoxypropyl-ether der Isohexide und Isohexidnitrate ist bereits in der EP 167 008 beschrieben.
- 18 01 Beispiele
05 A. Umwandlung IV -*· I, bzw. VII -·· VIII
0,05 Mol des entsprechenden 0-(2,3-Epoxypropyl)-isohexids VII bzw. 0-(2,3-Epoxypropyl) -isohexid-nitrats IV werden mit 0,05 Mol des entsprechenden Amins in 100 ml Ethanol (oder 1-Propanol, oder 2-Propanol) 15-20 Stdn.
bei Raumtemperatur gerührt oder einige Stunden unter Rückfluß erhitzt. Nach dem Abdampfen des Lösungsmittels nimmt man den Rückstand in 50 ml 1 N Salzsäure auf und extrahiert 3mal mit je 30 ml Ether. Die wäßrige Phase wird nachfolgend mit verd. Natriumhydroxidlösung alkalisiert und 3mal mit je 100 ml Chloroform extrahiert. Der nach dem Trocknen und Einengen der Chloroformlösung verbleibende Rückstand wird entweder als Base gereinigt, z.B. durch Urakristallisation oder Säulenchromatografie oder in ein entsprechendes Salz überführt und als solches aus dem angegebenen Lösungsmittel umkristallisiert.
B. Umwandlung VI -» III
Zu einer Mischung aus 13 g 65-proz. Salpetersäure und 45 ml Essigsäureanhydrid tropft man unter Rühren, bei 0-5° C, 0.1 Mol O-Allyl-isohexid VI. Nachdem man weitere 15 Min. bei dieser Temperatur belassen hat, gießt man in Wasser und extrahiert das Reaktionsprodukt mit Dichlormethan. Die organische Phase wird am Rotationsverdampfer eingeengt. Der ölige Rückstand des O-Allyl-isohexidnitrats III wird ohne weitere Reinigung für die Folgestufe eingesetzt.
C. Umwandlung VIII -·· I
Zm einer Mischung aus M g 65-proz. Salpetersäure und 45 ml Essigsäureanhydrid tropft man unter Rühren, bei
0-5° С, 0,05 Mol (З-Атіпо-2-hydroxylpropyl)-isohexid VIII. Man läßt langsam auf Raumtemperatur erwärmen und hält das Reaktionsgemisch danach noch 5 Stdn. bei 50° C. Anschließend gießt man in Wasser, alkalisiert unter Kühlung mit Natriumcarbonat und extrahiert mit Dichlormethan. Die organische Phase wird mit Wasser gewaschen und am Rotationsverdampfer eingeengt. Das Reaktionsprodukt I wird entweder als Base gereinigt, z.B. durch Umkristallisation, Säulenchromatografie und dergl., oder
10 in ein geeignetes Salz überführt.

Claims (6)
- Patentansprüche1. Verfahren zur Herstellung eines Aminopropanol-Derivates eines 1,4:3,6-Dianhydro-hexitol-nitratesters der Formel(Dworin X bedeutetein Benzof1,4jdioxinyl-2-methylamin-Rest der Formelein Phenoxyethylamino-Rest der Formelworin R1 Wasserstoff, Hydroxy oder ein Сі-Сз-Alkoxy-Rest ist und der Substituent R^- in o-, m- oder p-Stellung stehen kann,
oder X einer der folgenden Reste ist:R2 , AО , -N S , -N N-R-worin r2 Wasserstoff oder ein unsubstituierter oder mono-, di- oder tri-s"ubstituierter Phenyl-Rest ist, wobei die Substituenten an dem Phenyl gleich oder verschieden sein können und ausgewählt sind unter C1-C3-Alkyl, С^-Сз-Аікоху, Halogen, Nitro und Trifluormethyl ,und worin r3 einen geradkettigen oder verzweigtkettigen C^-Cß Alkylrest, Wasserstoff,0
R6-C-, worin R6 Ci-Cy-Alkyl, Phenyl, PhenylJsubstituiertmit C^-C^Alky! oderI Iist,Benzyl, Benzhydryl, Pyridyl, Pyrimidinyl oder unsubstituiertes oder substituiertes Phenyl der Formeldarstelltworin r4 und r5 gleich oder verschieden sein können und jeweils Wasserstoff, Hydroxy , Ci-Cg-Alkyl,Ci-Cg-Alkoxy, Halogen, Nitro oder Trifluornethyl darstellen oder worin r4 und r5 zusammengenommen eine Methylendioxygruppe darstellen,oder eines pharmazeutisch annehmbaren Salzes davon, dadurch gekennzeichnet, daß man eine Verbindung der Formel IV л-0IVmit HX, worin X obige Bedeutung hat, umsetzt und gegebenenfalls das Säureadditionssalz herstellt,
oder indem man eine Verbindung der Formel VIIIVIIIworin X obige Bedeutung hat mit Salpetersäure umsetzt. - 2. Verfahren gemäß Anspruch 1, dadurch gekennzeichnet, daß man die Verbindung der Formel VIII herstellt durch Umsetzung einer Verbindung der Formel VIIHOVIImit HX, worin X obige Bedeutung hat.
- 3. Verfahren gemäß einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, daß die hergestellte Verbindung der Formel I eine Verbindung der Formel Ia ist(Ia)worin die beiden Substituenten in 2- und 5-Stellung in Ende—Stellung sich befinden und X die in Anspruch 1 angegebene Bedeutung besitzt.
- 4. Verfahren gemäß einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, daß die hergestellte Verbindung der Formel I eine Verbindung der Formel Ib ist(Ib)OHworin die beiden Substituenten in 2- und 5-Stellung in Exo-Stellung sich befinden und X die in Anspruch 1 angegebene Bedeutung besitzt.
- 5. Verfahren gemäß einem der Ansprüche 1 oder 2, dadurch
gekennzeichnet, daß die hergestellte Verbindung der
Formel I eine Verbindung der Formel Ic oder Id istO2N-OO2H-OOHIcIdworin der Substituent in 2-Stellung jeweils die Exo-Stellung und der Substituent in 5-Stellung jeweils die Endo-Stellung einnimmt und X die in Anspruch 1 angegebene Bedeutung besitzt. - 6. Verfahren gemäß einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, daß die hergestellte Verbindung der Formel I ein getrenntes Diastereomer von gleichförmiger Konfiguration oder ein Salz davon ist.
