The invention relates to a process for the preparation of Lö-Oehydro-na-cyanomethyl ^ ß-hydroxysteroidderivaten from 15-dehydro-17-ketosteroids.
ITβ-hydroxysteroid derivatives are pharmacologically interesting steroid compounds or also intermediates for the synthesis of pharmacologically highly potent steroid products, e.g. 17a-cyanomethyl-17β-hydroxyestra-4,9-dien-3-one, such as "dienogest" or α-cyanomethyl---hydroxy-IS-alkylm-gonadiene-S-one or also 1 7a-cyanomethyl-17β -hydroxy-13-alkyl-4,9,1-i-gonatrien-3-one derivatives, which can be used advantageously for the treatment of endocrinopathies and for reproductive control in human and veterinary medicine because of their specific hormonal or antihormonal effects is the good compatibility of these compounds in use, which cause hardly any unwanted side effects even at elevated dosage and thus represent a desirable enrichment of the range of pharmacologically interesting steroid drugs compared to the known 17a-ethynyl-17ß-hydroxysteroids.
The introduction of a further double bond in these 17a-cyano-methyl-17ß-hydroxysteroids in the position C 2 to C 6 of the steroid backbone leads to a significant increase in activity, with a number of these new derivatives show particularly favorable dissociation of the characteristic partial effects.
Charakteristik des bekannten Standes der TechnikCharacteristic of the known state of the art
The synthesis of 17a-cyanomethyl-17β-hydroxysteroids has been described in a number of patents. Thereafter, 17-ketosteroids are converted in a multistage synthesis to the steroid 17β-spiro-1 ', 2'-oxiranes, which are then reacted with alkali cyanide to give the 17a-cyanomethyl-17β-hydroxysteroid derivatives.
Nach K.Ponsold u.a. DD-PS 132497 is 3-methoxy-13-alkylgona-2,5 (10) -diene-17ß-spiro-1 ', 2'-oxirane (prepared according to DD-PS 80023) by reaction with alkali metal cyanide and nachfoglende enol ester hydrolysis in the 17a-cyanomethyl-17β-hydroxy-13-alkylgon-5 (10) -en-3-one. An improvement of this synthesis has been reported by K.Ponsold et al. in DD-WP 160418, according to which the volatile starting material of the aforementioned method (DD-WP 132497) by 3,3-dimethoxy-13-alkyl-gona-5 (10) -en-17-one is replaced, which then in the same way with trimethylsulfonium iodide, alkali metal cyanide and subsequent Ketalspaltung in the Ua-cyanomethyl-ITß-hydroxy-IS-alkylgon-BIKH-en-S-on is transferred. Although the individual stages of this synthesis proceed with high yields, the overall economy is burdened by the use of expensive reagents and the high workload caused by the complex Zwwischenisolierungen. The main disadvantage of this procedure, however, is the high environmental impact associated with this synthesis, which is caused by the use of
- Trimethylsulfoniumjodid, was die Entsorgung der Abluft und des Abwassers erfordert,Trimethylsulfonium iodide, which requires disposal of the waste air and waste water,
- Alkalicyanid, einem Gift der Abt. 1 des Giftgesetzes, was besondere Anforderungen an die Verarbeitung im technischen Maßstab stellt und eine spezielle Entsorgung der Abluft und der Abwasser erfordert.- Alkali cyanide, a poison of Dept. 1 of the law on poisoning, which places special demands on the processing on an industrial scale and requires a special disposal of the exhaust air and waste water.
The in situ metallation of the acetonitrile with lithium alkyls or Lithiumdialkylamiden to LiCH 2 CN and its subsequent reaction in a one-pot process with 15-dehydro-17-ketosteroids to the IS-dehydro- ^ a-cyanomethyl-17ß-hydroxysteroids in inert organic solvents such as aliphatic or aromatic hydrocarbons, preferably pentane, hexane, heptane, benzene, toluene ethers, preferably diethyl ether, diisopropyl ether, tetrahydrofuran, oioxane, anisole, dimethoxyethane, diethoxyethane, etc., or in tertiary amines, such as triethylamine, diisopropylethylamine, pyridine, tetraalkylethy! endiaminu.a. are used, or these solvents are also used as a mixture.
According to the procedure of the invention, 1 to 5 moles of acetonitrile and 1 to 5 moles of lithium alkyl or lithium dialkylamide are employed per mole of 15-denydro-17-ketosteroid, the process being such that the acetonitrile is reacted at low temperatures with the employed lithium derivatives in situ in the LiCH 2 CN and this is reacted with the 15-dehydro-17-ketosteroid to 15-dehydro-1 7a-cyanomethyl-17ß-hydroxysteroid.
This procedure also implies that the in situ preparation of the LiCH 2 CN occurs in the presence of the 15-dehydro-17-ketosteroid and the follow-up reaction with the 15-dehydro-17-ketosteroid to the IS-dehydro-cyanomethyl-β-hydroxysteroid derivative nearly approaches can run at the same time.
Vorteilhaft werden diese Reaktionen bei tiefen Temperaturen durchgeführt, d.h. in Temperaturbereichen unter +O0C, bei denen die Reaktion des LiCH2CN mit Acetonitril zum ß-KetopropionitrilThis procedure also implies that the reactions of the alkyllithium with the acetonitrile can also be carried out in the presence of lithium halides, ie that the separation of the Li-HaI is not required before the further use of the alkyllithium b2w. the presence of lithium halide does not interfere with the performance of the subsequent reactions. Advantageously, these reactions are carried out at low temperatures, ie in temperature ranges below + O 0 C, in which the reaction of LiCH 2 CN with acetonitrile to ß-Ketopropionitril

The work-up of the reaction mixtures can be carried out by adding water, the reaction products can be isolated to give protective groups such as ketals or enol ethers, inter alia. These aqueous workups are advantageously carried out at temperatures <10 ° C in pH ranges> pH 6, wherein the pH adjustment can also be effected by additions of salts such as NH 4 Cl, NH 4 OAc, NaH 2 PO 4 or acids.
However, the workup can also be carried out so that the decomposition of the reaction mixtures, i. the destruction of the excess lithium organyl with the addition of water and acids or dilute acids and is carried out at pH ranges <pH 6 and this optionally present in the molecule existing protective groups such as ketals, enol ethers u. a.

As acetic acid, oxalic acid, p-toluenesulfonic acid, inter alia, are used.
In the work up can be generally moved so that the decomposition of the excess lithium organyl started at low temperatures, and the acid hydrolysis may then be conducted at temperatures up to 50 0 C.
The isolation of the reaction products can be carried out by extraction or filling, it is advantageous, however, to distill as far as possible from the reaction mixture under vacuum, the solvent mixture and to isolate the case, or by the addition of an alcohol or ketone in high purity crystals.
