Verfahren zur Herstellung von Peptiden
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Peptiden, das dadurch gekennzeichnet ist, daß man
entweder
a) ein Peptid oder dessen Derivate mit mindestens einer
freien Aminogruppe mit Isobornyloxycarbonylchlorid in Gegenwart eines säurebindenden Mittels bei einem
pH-Wert zwischen 6 und lh oder mit einem Isobornyloxycarbonyl~aktivester
umsetzt oder
b) eine N-Isobornyloxycarbonyl-aminosäure oder ein
N-Isobornyloxycarbonyl-peptid, die keine freien
Aminogruppen enthalten, mit einer Aminosäure, deren Aminogruppe frei und deren Carboxylgruppe durch Salzbildung
oder eine Schutzgruppe blockiert ist, oder mit einem Peptid, das mindestens eine freie Arninogruppe enthält,
nach in der Peptidchemio üblichen Kondensationsniethoden
umsetzt
und gegebenenfalls den Isobornyloxycarbonyl-reiit durch Behandlung
mit einer starken Säure abspaltet.

Die nach dem erfindungsgemäßen Verfahren verwendete Isobornyloxycarbonyl-Schutzgruppe,
die zum Urethantyp gehört, läßt sich» obwohl sie sich von einem sekundären Alkohol ableitet,
überraschend glatt mit Säuren wie z.B. Trifluoressigsäure oder
Chlorwasserstoff in Eisessig· unter milden Bedingungen abspalten.
Die bisher am häufigsten benutzten Schutzgruppen für Amino-
bzw. Iminofunktiorien von Aminosäuren zählen ebenfalls zum
Urethantyp,lassen sich aber in vielen Fällen, zumals wenn sie
sich von einem primären oder sekundären Alkohol ableiten wie die Carbobenzoxy-, p-Phenylazocarbobenzoxy-, Diisopropyline fchoxycarbonyl-,
CyclopentyIoxycarbonyl-, Cyclohexyloxycarbonyl-,
2-Methyl-C3rclohexyloxycarbonyl-Gruppe nur unter härteren
Bedingungen mit sehr starken Säuren wie Bromwasserstoff, wobei
häufig zusätzlich erwärmt werden muß, abspalten (vgl. I?. Schröder und K. Lübke, "The Peptides", New York and London,

599 bis 6Oh). Die p-Methoxy-carbobenzoxy-, tert.-Butyloxycarbonyl-,
tert.-Amylox3rcarbonyl- und 1-Adamantyloxycarbonyl-Gruppen
kann man mit schwächeren Säuren wie Trifluoressigsäure abspalten,
ihre Einführung in Aminosäuren ist aber durch den Umstand wesentlich erschwert, daß ihre Chloride bei Raumtemperatur
instabil sind (s. Experientia, Band 25, I969, Seiten 576 bis
578 sowie Bulletin Chem. Soc. Japan, Band h2, 19<$9, Seiten
809 bis 811) und man im allgemeinen auf die schwerer zugänglichen
Azide oder Aktivester angewiesen ist. Lediglich 1-Adamantyloxy-carbonylchlorid
ist einigermaßen beständig, setzt sich aber mit zahlreichen Aminosäuren nur in schlechten Ausbeuten
um (s. J. Araer. Chem. Soc. Band 88, I966, Seiten 1088
bis 1992), und außerdem ist das als Ausgangsmaterial dienende 1-Adamantanol relativ schwer zugänglich. Bei der p-Methoxycarbobenzoxy-Gruppe
stört weiterhin die Instabilität gegen
katalytische Hydrierung, so daß die oft erstrebte selektive Abspaltung der Carbobenzoxy-Gruppe, die mit katalytisch

erregtem Wasserstoff leicht entfernbar ist, nicht möglich ist. Das zur Einführung der tert.-Butyloxycarbonylgruppe verwendete
tert.-Butyloxycarbonylazid ist überdies physiologisch
gefährlich (vgl. Chimia, Band 23, I9.69, S. 298).
Die neue Amino- bzw. Iminoschutzgruppe besitzt diese Nachteile nicht, da sie in sehr hohen Ausbeuten mittels des
leich zugänglichen und bei Raumtemperatur sehr stabilen Tsobornyloxycarbonylchlorids einführbar ist. Auch ist sie gegen
kataly-tische Hydrierung stabil und bei Raumptemeratur mit Trifluoressigsäure oder Chlorwasserstoff leich abspaltbar.
Die Tierstellung der Isobornyloxycarbonyl-Peptide erfolgt durch Umsetzung von Peptiden bzw. deren Derivaten, die wenigstens eine
oder mehrere freie Amino- oder Iminogruppen aufweisen, mit dem sehr stabilen Isobornyloxvcarbonylchlorid, das man entweder
vor Beginn oder im Verlauf der Acylierung zusetzt. Man arbeitet in Gegenwart eines säurebindenden M-jttels wie Alkali- oder
Erdalkalihydroxyden, -carbonaten, - acetaten, -amiden, -alkoholaten oder tert. organische Aminen, wodurch eine freie
Carboxylgruppe salzartig gebunden und freiwerdende Salzsäure abgefangen werden. So verwendet man z.B. Natriumtrydroxyd,
Kai iuinhydroxyd, Lithiumhydroxyd, Bariumhydroxyd, Natrium-
oder Kaliumcarbonat, -amid, -acetat, tert. Amine wie Triethylamin, Tripropylamin, Tributylamin, N-Methyl-morpholin,
N-Äthylinorpholin. Man arbeitet bei - I5 i>±s +50 C, bevorzugt
O bis 25 C, und einem pII-Tvrert von 6 bis 1U, bevorzugt bei
pH 8 bis 10. Es kann racemisches oder optisch aktives Isobornyloxycarbonylchlorid
(D- oder L-Form) verwendet werden; man erhält die Chloride leicht aus D.L-Isoborneol bzw. D- und L~Isoborneol
(s. J. Chein. Soc. (London) Band 91

1^78 bis 1^79) mit Phosgen in an sich bekannter Weise.
Die neue Schutzgruppe kann ebenso mit Hilfe eines D.L-, D-
oder L-Isobornyloxycarboiiyl-aktivesters wie z.B. 2.4.5-

Trichlorphenyl- oder N-Hydroxysuccinimidesters eingeführt werden. Die benötigten Isobornyloxycarbonylaktivester sind
z.B. aus Isobornyloxycarbonylchlorid und der aktivierenden Komponente wie Trichlorphenol oder N-Hydroxysuecinimid leicht
zugänglich.
Als Lösungsmittel kommen bei der Einführung der Schutzgruppe 'Wasser, Dioxan, Tetrahydrofuran, Äther,Dimethylformamid,
Dimethylacetamid, Pyridin und andere, in der Peptidchemio gebräuchliche Lösungsmittel,allein oder in Mischung miteinander,
in Frage.
Das erfindungsgemäße Verfahren -erlaubt die Verwendung von N-Iso·
bornyloxycarbonyl-Aminosäuren, die sich von allen in natürlich
vorkommenden Peptiden anzutreffenden oder synthetisch zugänglich
enjjO - oderuz-Amino- oder Iminosäuren in ihrer L-,D- oder
racemischen Form ableiten können, z.B. die Verwendung von neutralen N*' -Isobornyloxycarbonyl-Aminosäuren wie z.B.
No^ -Isobornyloxycarbonyl-glycin, -alanin, -valin, norvalin, -leucin,
-isoleucin, -norleucin, -aminobuttersäure, -isovalin, -aminoeyclohexancarbonsäure, -phenylalanin, -C-phenylglycin
asparagin, -glutamin, -methionin und Diisobornyloxycarbonylcystin, ferner von N1^ -Isobornyloxycarbonyl-Atninosäuren
wie z.B. N-Isobornyloxycarbonyl-ß-alanin, -Tf-aminobuttersäure,
und - C-aminocapronsäure, weiterhin von N-Isobornyloxycarbonyl-Aminosäuren
mit funktionellen Gruppen in den Seitenketten, die geschützt sein können, wie z.B. N -Isobornyloxycarbonylserin,
N«^ - Isobornyloxyearbonyl-O-tert.-butyl-serin, N1-^-Isobornyloxycarbonyl-threonin,
N01' -Isobornyloxycarbonyl-tyrosin,

in denen die andere Aminogruppe mit einem zweiten Isobornyloxycarbonyl-rest oder mit einer

anderen N-Schutzgruppe blockiert ist, wie z.B. N^'-Isobornyl-'
oxycarbonyl~N*~ -tert.-butyloxycarbonyl-lysin, N^-Benzyloxycarbonyl-No·-Isobornyloxycarbonyl-lysin
und N^-Isobornyloxycarbonyl-N"
-benzyl-oxycarbonyl-ornithin, des weiteren von N-Isobornyloxycarbonyl-aminodicarbonsäuren, die eine freie und
eine geschützte Carboxylgruppe aufweisen, wie z.B. N -Isobornyloxycarbonyl-glutaminsäure-^-teriv-butylester
oder N -Isobornyloxy-carbonyl-asparaginsäure-0^
- oder ß-benzylester, darüber hinaus von halogenhaltigen N-Isobornyloxycarbonyl-aminosäuren
wie z.B. N-Isobornyloxycarbonyl-ß-chloralanin, N-Isobornyloxycarbonyl-p-chlorphenylalanin,
N-Isobornyloxycarbonyl-jodgorgo- ™
säure und N-Isobornyloxycarbonyl-thyroxin, schließlich von mit
dem Isobornyloxycarbonyl-rest geschützten aromatischen Aminosäuren
oder Aminosulfonsäuren wie z.B.. N-Isobornyloxycarbonylp-aminobenzoesäure
oder N-Isobornyloxycarbonyl-p-aminosulfonsäure
und schließlich von N-Isobornyloxycarbonyl-Iminosäuren
Avie z.B. N-Isobornyloxycarbonyl-prolin, -hydroxy-prolin,
-azetidin-2-carbonsäure,-N-methylvalin, -N-methylleucin und
-N-methyl-ß-alanin.
Bei der unter b) erwähnten Arbeitsweise des erfindungsgemäßen
Verfahrens werden die in der Peptidchemie (vgl. Schröder und Lübke, The Peptides, Academic Press, New York und London, ™
1965 und 1966) bekannten Methoden verwendet, wobei, soweit
erforderlich, die gebräuchlichen Schutzgruppen verwendet
werden« Die wichtigsten Knüpfungsverfahren sind die Dicyclohexylcarbodiimid-,
die Aktivester- und Gemischte-Anhydrid-Methode.
Als N-Isobornyloxycarbonyl-peptide können alle Peptide, in denen
sämtliche Aminogruppen mit dem Isobornyloxycarbonyl-rest und außerdem mit anderen Schutzgruppen blockiert sind und in

denen eine Carboxylgruppe in freier Form vorliegt, eingesetzt werden, wobei andere funktioneile Gruppen in den Seitenketten
geschützt sein können·
Diese Isobornyloxycarbonyl-Aminosäuren bzw. -Peptide können mit
allen in natürlich vorkommenden Peptiden anzutreffenden oder
synthetisch zugänglichen 0^- oder ^-Amino- oder Iminosäuren in ihrer L-, D- oder racemischen Form einschließlich Diaminocarbon- und Aminodicarbonsäuren, in denen eine Aminogruppe frei vorliegt und die Carboxylgruppen durch Schutzgruppen oder Salzbildung
blockiert sind und weitere funktionelle Gruppen in den
Seitenketten geschützt sein können., umgesetzt werden, weiterhin mit Peptiden, die alle bereits erwähnten natürlich vorkommenden oder synthetisch zugängliche Aminosäuren enthalten können,
eine freie Aminogruppe aufweisen und in denen andere funktionelle Gruppen geschützt sein können.

Die erfindungsgemäßen Peptide besitzen dank der neuen N-Schutzgruppe
ausgezeichnete Löslichkeitseigenschaften in organischen Lösungsmitteln.
Nach dem erfindungsgemäßen Verfahren lassen sich Peptide jeder
beliebigen Zusammensetzung und Größe herstellen, falls keine
Einschränkung durch die Methode der Peptidsynthese besteht«

Die Abspaltung der Isobornyloxycarbonylschutzgruppe gelingt
erfindungsgemäß durch Behandeln mit Säuren, wobei man,obwohl
sich die neue zum Urethantyp gehörende Schutzgruppe von einem sekundären Alkohol ableitet, überraschenderweise auch schwächere Säuren als HBr/Eisessig verwenden kann, wie z.B. die anorganischen Säuren HCl1 HF und H2SOk1 gegebenenfalls in organischen Lösungsmitteln," wie z.B. Eisessig, Essigester» Dioxan, Tetrahydrofuran, Nitromethan, Alkoholen oder Dimsthylacetamid oder z.B. die organischen Säuren Trifluoressigsäure, Trichloressigsäure oder Ameisensäure. können hierbei gleichzeitig als Lösungsmittel dienen. Die Möglichkeit,
die Isobornyloxycarbonylschutzgruppe unter diesen milden Bedingungen mit schwächeren Säuren als HBr abzuspalten
und die Beständigkeit gegen katalytisch« Hydrierung und Basen, erlaubt eine Differenzierung gegenüber anderen Schutzgruppen,
z.B· der Benzyloxycarbonylgruppe, die erst mit HBr entfernbar ist· So kann man einerseits die Isobornyloxycarbonylgruppe z.B.
mit HCl/Eisessig oder Trifluoressigsäure und andererseits
die Benzyloxycarbonylgruppe durch katalytische Hydrierung selektiv abspalten. Außerdem kann die Abspaltung der neuen
Schutzgruppen mit sauren Ionenaustauschern vorgenommen werden.
Die Verfahrensprodukte können als Therapeutika Verwendung finden
oder zur Herstellung anderer therapeutisch wertvoller Peptide, wie z.B. Oxytocin,. Vasopressin oder ACTH oder anderer
Therapeutika wie Sulfonamide als Zwischenprodukte eingesetzt werden.
Durch die nachstehenden Beispiele wird die Erfindung näher
erläutert. Es werden*die in der Peptidchemie üblichen Abkürzungen für die einzelnen Aminosäuren und Schutzgruppen
verwendet. Für die Isobornylcarbonylgruppe in der racemischen
Form wird die Abkürzung Ibc, für die in der D-Form D-Ibc und
die in der L-Form L-Ibc eingeführt.

Beispiel 1
Ibc-Pro-GIN-Ala-OH
Ibc-Pro-GIN-Ala-OH
Die Lösung von 2.15 g ( 5 mMol) H-Pro-GIN-AIa-OH.CF3COOH in
3 ecm Wasser und 1 ecm Dimethylacetamid wird mit 1,25 ecm (6mMol)
,Ibc-Cl versetzt. Unter Rühren tropft man anschließend 4 η
Natronlauge bei Raumtemperatur zu, wobei man den pH-¥ert konstant bei 10 hält. Nach dem Versuch von Λ ecm 4 η Natronlauge säuert
man mit Citronensäure an, extrahiert mit Essigester und wäscht die Essigester-Lösung mit Wasser, trocknet sie über Natriumsulfat
und dampft sie im Vakuum ein. Der Rückstand wird mit Äther verrieben. Ausbeute 1.75 g (71 /°)5 Schmp. 83
(Zersetzg.) fsL·/^ : -50.4° (c:- = 1 in Chloroform).

Die Lösung von 1.62 g (5.5 mMol) Ibc-OSu und 1.47 g (5 mMol)
H-Pro~Leu-Gly-NH2. i/2 HgO in 20 ecm Dimethylformamid wird
nach 3-stündigem Rühren bei Raumtemperatur im Hochvakuum eingedampft.
Den sirupartigen Rückstand verreibt man in Wasser, wobei er kristallisiert.
Ausbeute 2.3I g (quantitativ); Schmp. 117-1190ZQ(Zp2J -66,6°
(c = 1 in Chloroform).

ß) 1.^7 S (5mMol) H-Pro-Leu-Gly-NHg. I/2 H3O werden in 1 ecm
Wasser und 1 ecm Dimethylacetamid suspendiert. Nach.der Zugabe
von 1.25 ecm (6 mMol) Ibc-Cl tropft man unter Rühren bei
Raumtemperatur und pH 12 - 12.5 4 η Natronlauge zu, versetzt
mit 6 ecm Dimethylacetamid und säuert anschließend mit 0,5 m
Citronensäure-Lösung an. Das ausgeschiedene Öl nimmt man in Essigester auf und wäscht die Essigester-Lb'sung mit Wasser,
trocknet sie über Natriumsulfat und dampft sie im Vakuum ein. Der amorphe Rückstand kristallisiert beim Verreiben in Wasser. '
Ausbeute 1.65 g (71 $)5 Schmp. 118 - 120° ߣJ 22J -67.1°
(c = 1 in Chloroform). Das Produkt ist mit dem unter qQ erhaltenen
chromatographisch identisch.
H-Pro-Leu-Gly-NH2*CF COOH
1.00 g (2.15 mMol) IbC-PrO-LeU-GIy-NH2 wird in h ecm Trifluoressigsäure
gelöst. Nach 1 Stunde Stehenlassen bei Raumtemperatur engt man im Vakuum in der Kälte zum Öl ein und verreibt mit
50 ecm absol, Äther. Man dekantiert von den Kristallen, die man nochmals mit 50 ecm frischem absol. Äther behandelt und
schließlich über KOII und Pp0C im Hochvakuum trocknet.
Ausbeute 0.855 g (quantitativ). Zersetzung bei 95 - 100°. Die
Substanz ist mit H-Pro-Leu-Gly-NHg. CF-COOH dünnschichtchromatographisch
identisch (Rp= 0.21 i /

Ibc-OSu werden in 20 ecm Dimethylformamid k Stunden gerührt.
Nach dem Eindampfen im Hochvakuum wird der Rückstand in Äther/ Petroläther verrieben.
Ausbeute 5.3^ g (87 #) ; Schmp. 112 - 115° (Zersetzg.).

0.61 g (1 mMol) Ibc-Phe-Pro-Leu-Gly-NH» werden in 3 ecm Trifluoressxgsaure
gelöst. Nach 1 Stunde Stehenlassen bei Raumtemperatur engt man die Lösung im Vakuum in der Kälte ein
und verreibt den Rückstand mit absol. Äther. Die abgesaugten Kristalle wäscht man mit absol. Äther und trocknet sie über
KOII und P2 0C ini Hochvakuum.
Ausbeute Q.51 g ( 93 #) 5 Schmp-, 102 - 105° (Zersetzg.). Das
Produkt ist dünnschichtchromatographisch identisch mit H-Phe-Pro-Leu-Gly~NH2.CF CQOH„

Die Lösung von 1.22 g (2 raMol) Ibc-Phe-Pro-Leu-Gly-NHg in 10 ecm
HCl/Eisessig wird nach 2 Stunden Stiien bei Raumtemperatur mit
100 ecm absol. Äther versetzt. Man saugt die Kristalle ab,
wäscht sie mit absol. Xther und trocknet sie über KOH und P 0„.
Ausbeute O.76 g (81$|. Das Produkt ist chromatographisch mit
H-Phe-Pro-Leu-Gly-NH identisch. f
(ibe) -Insulin
120 mg (0,02 raMol) Rinderinsulin wrden mit 53 mg (0.i8mM-ol)
Ibc-OSu in 1 ecm Dimethylacetamid 2k Stunden gerührt. Die
Lösung wird dann mit Xther versetzt. Nach Verreiben wird vom Niederschlag dekantiert und der Rückstand noch zweimal mit
Äther behandelt.
Ausbeute 135 rag. Nach Behandlung mit Trifluoressigsäure wird
Insulin wieder freigesetzt» wie im Elektropherogramm erkennbar ist.
ä) Ibo-Gly-OSu
10,2 g (40 mMol) Ibc-Gly-OH und 5.53 g ( h8 mMol) N-Hydroxysuccinimid
in 80 ecm Essigester und 20 ecm Dimethylformamid
werden bei 0° mit 8.27 g ( ^O.mMol) Dicyclphexylcarbodiimid

versetzt. Nach 12-stündigem SMien bei O und 1-stündigem Stehen
bei Raumtemperatur kühlt man wieder auf O ab, saugt vom Harnstoff ab und dampft das FiItrat im Vakuum ein. Den Rückstand
behandelt man mit Methanol.
Ausbeute 10.5 S ( 75 $)1 Zersetzung bei ca. 100°.

Man rührt die Lösung von 1.47 S ( 5 mMol) H-Pro-Leu-Gly-NH^.

Die Lösung von 0,68 g (2.5 mMol) Ibc-Ala-OH und 0.45 g
( 2,75 mMol) 3-Hydroxy-4-oxo-3.4-dihydro-1.2.3-benzotriazin
in 12 ecm Dimethylformamid wird bei 0 mit 0,52 g Dicyclohexylcarbodiimid
versetzt. Nach 1 Stunde bei 0 und 1 Stunde bei Raumtemperatur wird abgesaugt und 0.74 g (2.5 mMol) H-Pro-Leu-GIy-NH2.
1/2 H2O in das Filtrat eingetragen. Man läßt 4 Stunden
bei Raumtemperatur reagieren, filtriert anschließend und dampft
das Filtrat im Hochvakuum ein. Den Rückstand behandelt man mit Äther. Ausbeute 1.32 g (quantitativ). Schmp. 95 - 98°
(Zersetzg, ). Die Substanz ist mit den unter <jj) und ß) erhaltenen
chromatogrfphisch identisch.

Dimethylformamid werden bei 0° C 0.62 g (3 mMol) Dicyclohexylcarbodiimid
zugegeben. Nach 1 Stunde bei 0 C und 1 Stunde bei Raumtemperatur saugt man vom Harnstoff ab und

in 30 ecm Dimetliylf ormaraid. Bei 0° C fügt man 0.83 ecm (6 mMol)
Triäthylamin zu und rührt dann 2 Stunden bei Raumtemperatur.
Nach dem Eindampfen im Hochvakuum verreibt man den Rückstand mit Essigester, saugt das kristallisierte Produkt ab, löst es
in Methanol/Wasser und rühr.t 30 Minuten mit Dowex 50. Man
filtriert vom Ionenaustauscher ab, wäscht mit Methanol/Wasser, dampft das Filtrat im Vakuum ein und trocknet den Rückstand
im Hochvakuum über PO. Das Rohprodukt wird aus Isopropanol
umkristallisiert. Ausbeute I.98 g (72 $). Schmelzpunkt 207 2090

Dimethylformamid. In das Filtrat trägt man I.3U g (1 mMol)
H-Tyr-Ser-Met-Glu(OtBu)~IIis-Phe~Arg~Try~Gly-OH (Liebig's Ann.
Chem. Band 726 (19^9), Seite 177) ein, fügt bei 0° C 0.11 ecm
(1 mMol) N-Methylmorpholin zu und rührt 12 .Stunden bei Raumtemperatur,
Dann dampft man im Hochvakuum ein, verreibt den Rückstand mit Essigester/Äther, saugt ab und verreibt mit dem
Gemisch aus 20 ecm Dimethylformamid, 1O ecm Methanol und 20 ecm
Essigester. Schließlich wird mit !Isopropanol ausgekocht. Ausbeute
1.0 1 g (66 $,). Schmelzpunkt 238° C (Zersetzung).

12.5 g (O.5 mMol) H-Lys(Boc)-Pro-Val-Gly-Lys(Boc)-Lys(Boc)-Arg-Arg~Pro-Val-Lys(Boc)-Val-Tyr-NH
-Trihydrochlorid oder - Tritosylat (Chem. Ber. _9_7_, ( 196*0 ι 1197) werden in 15 ecm reinem
Dimethylacetamid gelöst. Nach Zugabe von 270 mg (2 mMol) 1-Hydroxy-benzotriazol
fügt man bei Raumtemperatur,1/3 der Lo- "
sung von 65O mg (ca. 3 mMol) Dicyclohexylcarbodiimid in 2 ecm
Dimethylacetamdd zu. Nach einer halben Stunde wird ein weiteres
Drittel, nach einer Stunde das letzte Drittel zugegeben. Man rührt noch 2-3 Stunden bei Raumtempei^atur weiter und fällt
dann das rohe, noch geschützte Corticotropin-(1-23)-amid mit 150 ecm Äther aus. Ausbeute 2,0 g.
Zur Abspaltung der Schutzgruppen löst man in 20 ecm 90-ρτοζ.
Trifluoressigsäure, fällt nach einer Stunde Stehen bei Raumtemperatur
mit 150 ecm Äther und reinigt das Reaktionsprodukt
in bekannter Weise durch Chromatographie an Carboxymethylcellulose unter Elution mit wässrigem Ammonacetat oder Essig- λ
säure steigender Molarität.

a) Isobornyloxycarbonylchlorid
Zur Lösung von 100 g Phosgen in 35O ecm trockenem Benzol
wird unter Rühren bei -5 bis +5° G die Lösung von 28 g (181,5 mMol) D.L-, D- oder L-IsoborneoX und 25 ecm Pyridin
in 400 ecm trockenem Benzol zugetropft· Nach einstündigeia Nachrühren bei 20 C wird abgesaugt, der Filterrückstand
mit Benzol gründlich gewaschen und das Filtrat zweimal mit eiskaltem Wasser gewaschen, über Natriumsulfat getrocknet
und im Vakuum eingedampft. Der ölige Rückstand wird nach Versetzen mit etwas Petroläther nochmals im Vakuum
eingedampft und im Hochvakuum kurz getrocknet.
Ausbeute 39»^ 8 Öl (quantitativ)·
Das Öl siedet bei 77° - 78° C / 0,4 - 0,45 Torr.
Ibc-, D-Ibc- und L-Ibc-Chlorid zeigen identische IR-Spektren,

25 mMol Aminosäure und 6,25 ecm (30 mMal) Isobornyloxycarbonylchlorid
werden in 5 ecm Wasser und 5 ecm Dioxan unter
Rühren bei 20 bis 25 C tropfenweise mit h η Natronlauge
versetzt, wobei man anfangs den p„-Wert der Reaktionsmischung bei 8 bis 10, bevorzugt bei 10, hält. Nach dem
Anspringen der Acylierung sinkt unter Wärmeentwicklung

der p„-Wert rasch ab} man führt die Reaktion unter weiterem
Zutropfen von h η Natronlauge entweder bei p„ 10 oder bevorzugt
bei einem niedrigeren p„-Wert (7 bis 9) und tieferer
Temperatur (θ bis 10 c) zu Ende, wobei insgesamt ca. 12,5 bis 13i5 ecm h η Natronlauge bis zur Erreichung eines
konstanten p„-1iertes verbraucht werden. Entsteht ein in
Äther unlösliches Natriumsalz der entsprechenden Ibc-Aminosäure,
so entfernt man nun überschüssiges Isobornyloxycaxbonylchlorid
durch Waschen mit Äther. Nach dem Ansäuern mit verdünnter Citronensäure auf p„ 2 bis 3 nimmt
man die freigesetzte Ibc-Aminosäure in Essigester auf,
wäscht die Essigester-Lösung zweimal mit Wasser, trocknet sie über Natriumsulfat und dampft sie im Vakuum ein. Den ™
Rückstand verreibt man mit Petroläther· Durch Auflösen in
Äther oder gegebenenfalls in Petroläther und Zugabe von Dicycloliexylamin sind die Dxcycloliexylaminsalze der entsprechenden
Ibc—Aminosäuren zugänglich·
In der folgenden Tabelle sind die nach dieser Standardvorschrift hergestellten Ibc-Aminosäuren aufgeführt. Sie
wurden dünnschichtchromatographisch auf ihre Reinheit überprüft (Kieselgel F, Merck; Chloroform/Methanol 8 :

Die Lösung von 21,67 g (IOO mMol) D.L-Iscbornyloxycarbonylchlorid
und 15,0 g (13O mMol) N-Hydroxysuccinimid in 100 ecm
Dioxan wird bei 0° C unter Rühren mit 8,2 ecm (IOO mMol)
Pyridin in 65 ecm Dioxan tropfenweise versetzt. Nach fünfstündigem
Nachrühren und zwölfstündigem Stehenlassen bei Raumtemperatur wird abgesaugt, mit wenig Dioxan gewaschen
und das Filtrat im Vakuum eingedampft· Der ölige Rückstand wird in Methanol/Wasser kristallisiert.
Ausbeute 23,78 g (81 #) , Schmelzpunkt 95 - 97° C.

Claims (1)
- - 31 - HOE 70/PPatentanspruchVerfahren zur Herstellung von Peptiden, dadurch gekennzeichnet, daß man entwedera) ein Peptid oder dessen Derivate mit mindestens einer freien Aminogruppe mit Isoboriiyloxycarbonylchlorid in Gegenwart eines säurebindenden Mittels bei einem pH-Wert zwischen 6 und 14 oder mit einem Isobornyloxycarbonylaktive st er umsetzt oderb) eine N-Isobornyloxycarbonyl-aminosäure oder einN-Isobornyloxycarbonyl-peptid, die keine freien ™Aminogruppen enthalten, mit einer Aminosäure, deren Aminogruppe frei und deren Carboxylgruppe durch Salzbildung oder eine Schutzgruppe blockiert ist, oder mit einem Peptid, das mindestens eine freie Aminogruppe enthält, nach in der Peptidchemie üblichen Kondensationsmethoden umsetztund gegebenenfalls den Isobomylcarbonyl-Rest durch Behandlung mit einer starken Säure abspaltet.109843/18'β
