Als Nachteil von THF-FU kann man eine Neurotoxizität ansehen, die nach der in der Literatur geäußerten
Meinung durch die Zerfallsprodukte des Präparats bedingt wird, die im Prozeß seines Metabolismus im
lebendigen Organismus gebildet werden (S. K. Germane, A. A. Kimenis, Experimentelle und klinische
Pharmakotherapie, Riga, 1970, H. 1, S. 85-92). Was FUDR anbetrifft, so wurde es wegen der Kompliziertheit
seiner Synthese und dementsprechend des Gestehungspreises zu keinem breit zugänglichen Präparat für
die praktische Medizin. Das bezieht sich auch auf FUDA, in dessen Synthese FUDRaIs Ausgangsprodukt
verwendet wird.
Das Ziel der Erfindung besteht in der Entwicklung einer neuen Verbindung, die eine antiblastische Wirkung
besitzt und weniger toxisch, jedoch wirksamer als bekannte Verbindungen solcher Wirkung wäre.
Erfindungsgemäß wird die neue Substanz l-(5-Fluoruracilyl-lJ^.S-di-O-acetyl-jff-D-glucofuranurono-o^-
lakton der Formel

2. Präparat mit antiblastischer Wirkung, das als
Wirkstoff HS-Fluoruracilyl-lH^-di-O-acetyl-jS-D-glucofuianurono-6,3-lakton
nach Anspruch 1 und einen Träger enthält.

Die Erfindung bezieht sich auf ein neues Derivat von 5-FluoruraciI, nämlich auf l-(5-Fluoruracilyl-l)-2,5-di-O-acetyl-jff-D-glucofuranurono-o^-lakton,
das eine antiblastische Wirkung besitzt.
Es ist eine große Zahl von chemischen Verbindungen bekannt, die eine geschwulsthemmende Wirkung besitzen,
darunter auch 5-Fluoruracylderivate, beispielsweise l-Carbamoyl-5-fluoruracil (japanische Auslegung
Nr. 148 365/75), l,3-Bis-(2-tetrahydropyranyl)-5-fluoruracil
(japanische Auslegung Nr. 77 072/77), 1-(2-Tetrahydrofuryl)-3-cyanoäthyl-5-fluoruracil
(japanische Auslegung Nr. 77 070/77) und 3-Aryl-5-fluoruracile (japanische Auslegung Nr. 91 880/77).
Die effektivsten unter allen bis jetzt bekannten Zytostatika unter den 5-Fluoruracilderivaten sind l-(2-Tetrahydrofuryl)-5-fluoruracil
(US-PS 36 35 946), nachstehend als THF-FU abgekürzt, 5-Fluor-2'-desoxyuridin FUDR (US-PS 29 49 415) sowie 5-Fluor-2'-desoxyuridin-5-carbonsäure
FUDA (J. Med. Chem., 12, 173 [1969]).

vorgeschlagen.
Diese Substanz wird im weiteren Text ais FUGA bezeichnet.
Es wurde von uns festgestellt, daß die genannte Substanz FUGA eine deutlich ausgeprägte antiblastische
Wirkung besitzt.
l-(5-Fluoruracilyl-l)-2,5-di-O-acetyl-j8-D-glucofuranurono-6,3-lacton
(FUGA) wird durch die Umsetzung von 2,4-Bis-(trimethylsilyl)-5-fluoruracil mit 1,2,5-Tri-O-acetyl-jS-D-glucofuranurono-ö.S-lakton
in Gegenwart von Zinntetrachlorid in protonenfreien Lösungsmitteln erhalten.

Die erhaltene Substanz FUGA stellt ein weißes, kristallines Pulver dar, das schwer wasserlöslich ist und in
protonenfreien Lösungsmitteln gut löslich ist. Die Ausgangsstoffe 2,4-Bis(trimethylsilyl)-5-fluoruracil und
1 ^,S-Tri-O-acetyl-jS-D-glucofuranurono-o.S-lakton sind
bekannt.
FUGA besitzt eine zystostatische Wirksamkeit auf einem breiten Spektrum von Transplantationstumoren
von Mäusen und Ratten: z. B. Leukämie P-388, Leukämie 1-5178, Adenokarzinom 755, Lewis-Lungenkarzinom,
Melanom B16. Die erfindungsgemäße Verbindung
ist um einige Male weniger toxisch als THF-FU und
besitzt insbesondere eine geringere Neurotoxizität Zum Unterschied von THF-FU setzt FUGA in einer
Dosis von 200 mg/kg die Köpertemperatur der Tiere praktisch nicht herab, während THF-FU die Körpertemperatur
in einer Dosis von 115 mg/kg um 3° und
mehr bei 50% der Tiere herabsetzt. Zum Unterschied von THF-FU vermindert die neue Substanz FUGA bei
der intravenösen Einfuhrung in Dosen bis 100 mg/kg den arteriellen Blutdruck nur unbedeutend um 5 bis
20 Torr in Versuchen an narkotisierten Katzen und Hunden und übt keinen wesentlichen Einfluß auf die
Atmungsamplitude und -frequenz aus.
Die antiblastische Wirkung von FUGA wurde in bezug auf Hämozytoblastose La und lymphatischer
Leukämie L-1210 untersucht Als Kriterium der zytostatischen
Wirksamkeit wird die Erhöhung der Überlebensdauer gegenüber nicht behandelten Kontrolltieren
bestimmt Die Hämozytoblastose La wurde Mäusen der Linie C57B1/6V in Form einer Milzsellensuspension in
einer Menge von 50 mg Gewebe pro Maus intraperitoneal implantiert. Die lymphatische Leukämie L1210
wurde Mäusen der Linie BDF1 in einer Menge von 106
Asziteszellen pro Maus intraperitoneal implantiert Die Behandlung wurde nach 24 Stunden begonnen. Die
Verbindungen wurden in destilliertem Wasser suspendiert und in Suspensionsform intraperitoneal und peroral
einmal täglich verabreicht. Die Ergebnisse sind in der Tabelle angeführt.
