Description

  • Verfahren zur Darstellung von optisch aktiven Phenylpropanolmethylaminen Es wurde gefunden, daß man durch Spaltung von Phenylpropanolmethylamin mit Hilfe von 1-Weinsäure als erstes Kristallisationsprodukt das 1-weinsaure Salz des 1-Phenylpropanolmethy lamins erhält.
  • Diese -Spaltung ist von technischer Bedeutung, da die physiologische Wirkung des 1-Phenylpropanolmethylamins bekanntlich eine bedeutend bessere ist als die des racemischen und des d-Phenylpropanolmethylamins. 1-Phenylpropanolmethylamin ist identisch mit dem natürlichen 1-Ephedrin und soll ebenso wie das Naturprodukt Verwendung finden.
  • Es war nicht vorauszusehen, daß die Löslichkeit des 1-weinsauren 1-Phenylpropanolmethylamins eine geringere ist als die des d-weinsauren 1-Phenylpropanolmethylamins.
  • Die Spaltung kann in geeigneten Lösungsmitteln, wie Wasser und Alkohol, z. B. Methylalkohol, Äthylalkohol und Propylalkohol, vorgenommen werden.
  • Beispiel. Sog rac.Phenylpropanolmethylamin und Sog 1-Weinsäure werden in t25 ccm Äthylalkohol gelöst und die Lösung zur Kristallisation abgestellt. Nach einiger Zeit beginnt erst langsam, dann schneller die Lösung zu kristallisieren. Nach beendeter Kristallisation saugt man von der Mutterlauge ab und wäscht mit wenig Alkohol nach. Die erhaltene Verbindung besteht zum größten Teil aus 1-weinsaurem 1-Phenylpropanolmethylamin. Durch Umkristallisieren aus Alkohol erhält man ein Salz mit einem Schmelzpunkt von 148°. Das aus dem 1-Tartrat hergestellte Hydrochlorid zeigt einen Schmelzpunkt von 215 bis 216° und eine optische Drehung von etwa ;4°.

  1. PATENTANSPRUCH: Verfahren zur Darstellung von optisch aktivenPhenylpropanolmethylaminen, darin bestehend, daß man synthetisches racemisches Phenylpropanolmethylamin mit 1-Weinsäure in seine optisch aktiven Komponenten zerlegt.
