Den foreliggende opfindelse angår en fremgangsmåde til rensning af et urent alkalimetalsalt af 7-(2,-thienylacetamido)cephalo-sporansyresalte.
I USA patent nr. 3.218.318 er beskrevet en ny klasse 7-acylamido-. cephaiosporansyreantibiotika. En af de mest værdifulde grupper af forbindelser som er beskrevet i ovennævnte patent er 7-(2*-thienylacetamido)-cephalosporansyresalte (nu kendt generelt som cephalothinsalte) med formlen: 2 142287

hvori M er en kation af et alkalimetal (f. eks. natrium, kalium eller lithium). Cephalothin anvendes nu viden om og er kommercielt tilgængelig i form af natriumsaltet..
Ved fremstillingen af de nævnte cephalothinsalte i stor målestok har det vist sig praktisk at udvinde natriumcephalothin fra vandige opløsninger heraf ved til den vandige opløsning i stort overskud (volumen) at tilsætte et organisk opløsningsmiddel, der er blandbart med vand, såsom isopropanol, hvilket bevirker udfældning af cephalothinsaltet.
Det således udfældede natriumsalt af cephalothin indeholder imidlertid en eller flere urenheder, hvilket giver en uønskværdig gul farvning af vandige pharmaceutiske opløsninger af saltet.
På nuværende tidspunkt kender man ikke den eksakte natur af disse urenheder, dog mener man, at urenheden er en polycarboxylsyre, der kan udfældes med protamin. Under alle omstændigheder fører udvindingen af cephalothinnatriumsaltet under anvendelse af ovennævnte procedure til samtidig udfældning af urenhederne. Denne udvindingsteknik er således ude af stand til at udvirke en separering af urenhederne.
Det er derfor et formål med den foreliggende opfindelse at tilvejebringe en ny og forbedret fremgangsmåde til rensning af cephalothinsalte, hvorved uønskede urenheder fjernes ved udfældning fra vandige opløsninger, hvoraf der opnås et salt, der i forøget omfang kan accepteres i pharmaceutisk henseende og som har forøget stabilitet.
Det har nu vist sig, at alkalimetalsalte af cephalothin selektivt kan udfældes fra vandige opløsninger deraf uden en samti- 3 142287 l ( dig udfældning af farveurenheder ved til den vandige opløsning at tilsætte visse alkalimetalsalte, hvorved cephalothinsaltet udfældes, medens farveurenhedeme forbliver i den vandige opløsning.
Det således opnåede cephalothinsalt er ikke blot i det væsentlige fri for disse urenheder, men er også ejendommelig ved en forbedret lagringsstabilitet.
Ved fremgangsmåden ifølge opfindelsen, som er ejendommelig ved det i kravets kendetegnende del anførte, sættes et ikke-toxisk alkalimetalsalt til en relativ koncentreret vandig opløsning af det urene cephalothinsalt til en selektiv udfældning af cephalo-thinsaltet uden samtidig udfældning af urenhederne. Koncentrationen af cephalothinsaltet i den vandige opløsning hvorfra cephalothinsaltet udfældes er ikke kritisk. Den nedre grænse for koncentrationen fastsættes ud fra et praktisk hensyn til det ønskede udbytte, mens den øvre grænse er den koncentration, ved hvilken opløsningen af cephalothinsaltet er mættet ved opløsningens temperatur. F. eks. danner natriumcephalothin ved en temperatur på 25° C en mættet opløsning, når der er opløst ca.
230-250 mg/ml vand. De bedste resultater opnås normalt, når man anvender en vandig opløsning, der indeholder mindst 10 vægt-# cephalothinsalt.
Som alkalimetalsalt med samme kation som cephalothinsaltet kan anvendes et hvilket som helst ikke-toxisk salt* Man foretrækker normalt at anvende alkalimetalsalte af syrer, der har en pK_, der er mindre end ca. 5* Foretrukne alkalimetalsalte er alkalimetalsalte af organiske carboxylsyrer repræsenteret af eddikesyre, citronsyre, mælkesyre, fumarsyre, gluconsyre og glutarsyre. Alkalimetalsalte af myresyre kan også anvendes, hvis man sørger for at fjerne myresyre, der optræder som biprodukt, fra det separerede alkalimetalcephalothin. Alkalimetalhalogenider, såsom alkalimetal-bromider og -chlorider kan anvendes ved fremgangsmåden ifølge opfindelsen. Dog foretrækkes anvendelsen af sådanne halogenidsalte ikke, da koncentrerede opløsninger heraf er korroderende. Kationen af alkalimetalsaltet svarer til kationen af cephalothinsaltet for at undgå indføring af fremmede kationer i systemet.
Det er normalt ønskværdigt at anvende tilstrækkelige mængder alkalimetalsalt til udfældning af næsten al cephalothin fra opløsningen. På grund heraf foretrækkes det at anvende alkali- 4 142257 metalsalte i mængder af størrelsesordenen 0,5-8,0 vægtdele pr. vægtdel cephalothinsalt indeholdt i den vandige opløsning. Som det imidlertid vil være klart for fagmanden, kan man anvende overskud af alkalimetalsalt, selvom der ikke er nogen fordel ved at anvende sådanne mængder i overskud i forhold til de mængder, der danner en mættet opløsning af alkalimetalsalt i den vandige opløsning.
Alkalimetalsalte teyn sættes til den vandige opløsning af cephalothinsalt på en hvilken som helst måde. F. eks. kan saltet anvendes som fast stof eller som en koncentreret opløsning i et væskeformigt hæremedium såsom vand eller en lavere alkanol (f. eks. ethanol, isopropanol, butanol etc.). For at undgå udbyttebegrænsende fortynding med vand og for at muliggøre en rensende og steriliserende filtrering kan alkalimetalsaltet anvendes i form af en opløsning i en alkanol. F. eks. foretrækkes det oftest at anvende en opløsning af et alkalimetalsalt i en alkanol indeholdende 20-60 vægt-# af alkalimetalsaltet.
Efter tilsætning af alkalimetalsaltet opnås der et bundfald af cephalothinsaltet, der kan adskilles fra den vandige moderlud indeholdende farveurenheder i opløsning på sædvanlig måde som f. eks. ved filtrering og/eller centrifugering.
Ved fremgangsmåden ifølge opfindelsen opnås således et produkt i udbytter på 80-90 #, hvilke produkter indeholder en betydeligt reduceret mængde urenheder. Desuden viser de opnåede produkter en signifikant forbedret stabilitet med hensyn til dannelse af nye urenheder ved lagring i vandig opløsning.
Følgende eksempler illustrerer nærmere fremgangsmåden ifølge opfindelsen.
EKSEMPEL 1
Natriumsaltet af cephalothin med et urenhedsindhold på 18 mg/g opløses i vand til dannelse af en opløsning indeholdende 100 g af saltet pr. 400 ml vand. Den fremkomne opløsning er en klar opløsning med gul farvning.

Sil denne opløsning sættes 100 g formalet natriumchlorid. Opløsningen omrøres og krystallisation observeres ved fuldstændig tilsætning af natriumchlorid. Ben fremkomne suspension henstår herefter i ca. 30 min for at sikre fuldstændig opløsning af natriumchlorid.
Be således formede krystaller opsamles ved filtrering og vaskes med en vandig opløsning af ethanol. Produktet, natriumcephalothin, tørres under vakuum i adskillige timer og har herefter et urenhedsindhold på 2,0 mg/g. Udbyttet er 90
En prøve på 100 g af den rensede natriumcephalothin behandles dernæst med 15 ml vand til dannelse af en pasta, der henligger til tørring i den almindelige rumatmosfære natten over.
Den tørrede pasta formales og sigtes igennem en 30 mesh-sigte. Analyse af produktet indikerer, at der ikke er sket nogen signi-fikant ændring af urenhedsindholdet ved tilsætning af vand.

En prøve på 100 g af natriumcephalothin med et urenhedsindhold på 18,0 mg/g opløses i 400 ml vand til dannelse af en gul opløsning.
Berefter tilsættes en opløsning indeholdende 55 vægt-j6 natriumlactat i ethanol til den vandige opløsning af natriumcephalothin. Efter tilsætning af ca. 100 g opløsning begynder udfældningen. Hatrium-cephalothinkrystalleme separeres dernæst og vaskes ifølge fremgangsmåden beskrevet i eksempel 1, og de viser sig at have et urenhedsindhold på ca. 3,0 mg/g.
De i tabel 1 anførte data demonstrerer, at natriumcephalothin, som er renset ved fremgangsmåden ifølge opfindelsen, er mere stabilt end natriumcephalothin, der er udfældet med isopropanol.

Ved bestemmelse af stabiliteten af produkterne behandlet ifølge den foreliggende opfindelse opløses prøver af natriumcephalothin udvundet med natriumlactat under anvendelse af fremgangsmåden beskrevet i eksempel 2 og prøver udvundet under anvendelse af isopropanol, i sterilt vand og lagres i 48 timer, enten ved stuetemperatur (25° 0) eller ved 5° C. Lagringen udførtes med to koncentrationer af natriumcephalothin, nemlig med 0,54 M svarende til ampul-betingelse (d.v.s. 1 g salt i 4 ml fortyndingsmiddel) og i en koncentration på 0,01 M svarende til intravenøs indgivelse (d.v.s* 4 g salt i 1000 ml fortyndingsmiddel).
Por hvert eksempel bestemmes urenhedsindholdet ved hjælp af et protamin-nephelometrisk forsøg. I denne metode udfældes urenhederne fra en cephalothinsaltopløsning ved tilsætning af protamin. Pen fremkomne uklarhed sammenlignes så med den uklarhed, der opnås på lignende måde fra opløsninger af cephalothinsalte indeholdende kendte mængder af urenheder. Ved prøven, der anvendes i dette eksempel, tilsættes 0,5 ml af en 0,1 % opløsning af protaminsulfat langsomt med en kontrolleret hastighed til en prøve-opløsning af 0,005 M cephalothinsalt og 0,01 M natriumphosphat ved pH
7,0, og den fremkomne uklarhed måles på konventionel måde.'
Resultaterne af lagringsforsøgene er vist i Tabel I.

Som det ses af tabel 1 opnår man ved fremgangsmåden ifølge den foreliggende opfindelse et cephalothinsalt med en forbedret stabilitet, især hvor den vandige opløsning af saltet lagres under køleskabsbetingelser (d.v.s. ved 5°C).
EKSEMPEL 3
Et kaliumsalt af cephalothin med et urenhedsindhold på ca. 15 mg/g opløses i vand til dannelse af en opløsning indeholdende 25 vægt-% af cephalothinsaltet.
Til denne opløsning sættes ca. 85 g kaliumacetatkrystaller. Den fremkomne blanding omrøres i ca. 10 minutter og henstår dernæst for at sikre fuldstændig opløsning af kaliumlactatet.
Herefter separeres de dannede krystaller i den vandige opløsning ved filtrering og vaskes med vandig ethanol. Produktet behandles under vakuum og viser sig at være kaliumcephalothin med et urenhedsindhold på ca. 3,0 mg/g.
EKSEMPEL 4 '
Til en opløsning indeholdende 26 vægt-°/o natriumcephalothin i vand tilsættes 105 g natriumacetat i form af en mættet opløsning af natriumacetat i vand.
Natriumcephalothinkrystaller dannes som resultat af tilsætningen og disse separeres som beskrevet i eksempel 1, vaskes og tørres. Watriumc ephalo thine t viser sig at have et urenheds indhold af samme størrelse som de rensede salte i de foregående eksempler.
EKSEMPEL 5
Fremgangsmåden ifølge eksempel 4 gentages, dog tilsættes natriumcitrat. Natriumcephalothin separeres fra den vandige opløsning og man finder igen, at det udfældede natriumcehphalothin har et urenhedsindhold af samme størrelsesorden som de rensede salte i de foregående eksempler.
