R3 1 3 hvor R , R og X har den ovenfor anførte betydning, i nærværelse af et opløsningsmiddel omlejres ved henstand ved stuetemperatur eller ved let opvarmning.
Det er kendt, at 3- og 2-cephemderivater med den almene formel I
El ίΤΎ hvor R1 betegner et eventuelt substitueret aminoradikal, og R4- be= tegner hydrogen eller R som ovenfor defineret, og hvor de stiplede linier viser en fakultativ binding til dannelse af 3-cephem- eller 2-cephem-isomere, har gunstig antibakteriel virk= ning mod både grampositive og gramnegative bakterier. Aktiviteten vides at være større end de fleste antibakterielle forbindelsers, uden at de dog forårsager uønskede kontraindikationer, når de skal administreres til mennesker, som de mere almindeligt tilgængelige antibakterielle forbindelser, f.eks. penicillin, gør det. Hidtil har de kendte metoder til fremstilling af sådanne cephemderivater 3 147143 imidlertid i almindelighed vist sig at være ufordelagtige ud fra et industrielt synspunkt, idet det opnåelige udbytte var lavt og krævede temmelig komplicerede isolations- og rensningsforanstaltninger Den totale fremstillingsmåde var derfor urimelig kostbar, således at produktet økonomisk ikke kunne konkurrere med de kommercielt tilgængelige antibakterielle forbindelser med mindre fysiologisk virkning.
Det har været overvejet at fremstille cephemderivater ud fra cepha= losporin C. Imidlertid må anvendelsen af et sådant udgangsmateriale betragtes som uønskelig, idet det er ret kostbart at fremstille.
Det har senere været udbredt omtalt, at cephamderivater kunne fremstilles ud fra mindre kostbare og lettere tilgængelige penam= forbindelser. Imidlertid resulterer de omhandlede processer alminde= ligvis i lave udbytter og almindeligvis et urent produkt, og det var derfor ikke muligt at fremstille et produkt, som er økonomisk konkurrencedygtigt med de tidligere kendte antibakterielle forbin= delser.
USA patentskrift nr. 3.725.397 angår omdannelse af 63-acylamido-pe-nicillansyre-l-oxid-estere, som ikke er halogenerede, til 73-acylamido- 3-methylcephemcarboxylsyrederivater i nærværelse af en katalysator og i et opløsningsmiddel, som opvarmes kraftigt.
USA patentskrift nr. 3.725.399 angår omdannelse af ββ-acylamido-penicillansyre-l-oxidestere, som heller ikke er halogenderivater, til 73-acylamido-3-methylcephem-carboxylsyrederivater i nærværelse af bestemte katalysatorer og ved det anvendte opløsningsmiddels kogetemperatur .
USA patentskrift nr. 3.862.181 angår omdannelse af forbindelser indeholdende en beskyttet aminogruppe, nemlig 1-oxider af en Schiff'sk base af 6-aminopenicillansyrederivater, som ikke er halogenerede, i nærværelse af en katalysator, og i eksemplerne er der illustreret kogning under tilbagesvaling i toluen med p-toluensulfonsyre som katalysator og lignende kraftige betingelser.
USA patentskrift nr. 3.843.637 angår ej heller halogenerede derivater, men omlejring af 6-acylamidopenicillansyre-sulfoxider eller salte deraf ved kraftig opvarmning i et svagt basisk organisk opløsningsmiddelsystem 4 U7143 i nærværelse af en katalysator. Af eksemplerne fremgår det, at der fås meget lave udbytter.
Fremgangsmåden ifølge den foreliggende opfindelse adskiller sig fra de processer, der er beskrevet i den kendte teknik ved, at den kan udføres under yderst skånsomme betingelser på forbindelser, der indeholder en ubeskyttet aminogruppe i 6-stillingen.
Renamnomenklaturen for penicillinerne er beskrevet af Sheehan, Henery-Logan og Johnson i J. Am. Chem. Soc., 75*3293» fodnote 2 (1953) og den er overført til cephalosporinerne af Morin, Jackson, Elynn og Roeske i J. Am. Chem. Soc., 84*3400 (1962). Ifølge disse nomenklatursystemer refererer "penam" og "cepham" til nedenstående mættede ringstrukturer: -CH ^<j!H2 0H|-pifSx0E2 0 = C-1--ch2 0 = C- penam cepham medens "penem" og "cephem" refererer til de samme ringstrukturer med en dobbeltbinding.
Med de ved fremgangsmåden ifølge den foreliggende opfindelse fremstillede cephamderivater kan der fremstilles et cephemderivat med den almene formel I

R4 hvor har den ovenfor anførte betydning. Egnede acylsubstitu-tionsgrupper omfatter aliphatiske acylradikaler og acylradikaler indeholdende en aromatisk eller heterocyclisk ring. Egnede aliphatiske acylradikaler omfatter mættede eller umættede alkanoylradikaler, 5 U7U3 som kan være forgrenede, eller som kan indeholde en cyclisk ring, f.eks. de aliphatiske acylradikaler, f.eks. formyl, acetyl, propionyl, butyryl, isobutyryl, valeryl, isovaleryl, piva= loyl, acryloyl, crotonoyl, 2-methyl-acryloyl, cyclopentylcarbonyl, cyclohexylcarbonyl, cycloheptylcarbonyl, cyclohexylacetyl, cyclo= heptylacetyl, cyclohexylpropionyl, cycloheptylpropionyl, dihydroben= zoyl, 2,4,6-cycloheptatrienylacetyl eller dihydrophenylacetyl, og mættede eller umættede alkanoylradikaler indeholdende oxygen eller svovl, f.eks. methoxyacetyl, methylthioacetyl, 2-propenylthioacetyl, cyclohexylthioacetyl, cyclohexyloxyacetyl, dihydrophenoxyacetyl, dihydrophenylthioacetyl, cyclopentyloxycarbonyl, cyelohexyloxycar= bonyl, dihydrophenoxycarbonyl eller cycloheptyloxycarbonyl. Egnede acylradikaler indeholdende en aromatisk ring omfatter f.eks. aryl= radikaler (f.eks. benzoyl, toluqyl, naphthoyl, a-methylnaphthoyl, phthaloyl.eller tetrahydronaphthoyl) eller aralkanoylradikaler (f.eks. phenylacetyl, phenylpropionyl, phenylbutyryl, tolylacetyl, xylylacetyl, naphthylacetyl eller tetrahydronaphthylacetyl). Car= bonatomet i alkylgruppen i de ovenfor anførte aralkanoylradikaler kan være erstattet med et oxygen- eller svovlatom, f.eks. phenoxy= acetyl, benzoylxocarbonyl, xylyloxycarbonyl, naphthoxycarbonyl, phenoxycarbonyl, 2-phenoxypropionyl eller 2-phenoxybutyryl. 10 ml eddikesyre sættes til 3,50 g 6-(2-phenoxyacetamido)-2,2-di= methylpenam-3-carboxylsyre, og til denne blanding sættes 100 mg natriumwolframat-dihydrat. Blandingen tilsættes 1,2 ml 30fo1 s hydro= genperoxid under afkøling i isbad og omrøres i 1 time.

1,0 g 6-(2-phenoxyacetamido)-2,2-dimethylpenam-3-carboxylsyre suspen= deres i 3 ml iseddikesyre, og til denne suspension sættes 0,01 g natriummolybdat. Til den omrørte blanding sættes dråbevis 0,7 ml 30$’s hydrogenperoxid under afkøling i et isbad i løbet af 5 minut= ter. 1 Time senere isoleres bundfaldet ved filtrering, vaskes med vand og tørres, hvorved der fås 0,85 g farveløse krystaller af 6-(2-phenoxyacetamido)-2,2-dimethylpenam-3-carboxylsyre-l-oxid; smel= tepunkt 166,5°C.

Reak= tionsblandingen inddampes til tørhed under reduceret tryk, og remanensen opløses i 100 ml ethylacetat. Opløsningen vaskes med 5$‘s vandig natriumcarbonatopløsning tre gange og derefter med vand, tørres over magnesiumsulfat og inddampes, hvorved der fås 4,41 g remanens. Eksempel B 2.
En blanding af 2,18 g 6-(2-phenoxyacetamido)-2,2-dimethylpenam-3= -carboxylsyre-l-oxid (fremstillet ifølge eksempel Al, 2 eller 3) og 1,00 g 5-methyl-l,3,4-thiadiazol-2-thiol i 100 ml tert.butanol opvarmes under tilbagesvaling i 25 timer. Reaktionsblandingen inddampes til tørhed under reduceret tryk, og til remanensen sættes ethylacetat. Der udskilles krystaller, som isoleres ved filtrering og omkrystalliseres af ethylacetat, hvorved der fås 1,60 g krystaller af 4-(5-methyl-l,3,4-thiadiazol-2-yl)dithio-3-(2-phenoxyacet= amido)-a-isopropenyl-2-oxazetidin-l-eddikesyre; smeltepunkt 142-144°C.
Eksempel B 3.
En blanding af 3,80 g methyl-6-(2-phenoxyacetamido)-2,2-dimethyl= penam-3-carboxylat-l-oxid og 1,67 g benzothiazol-2-thiol i 25 ml me= thylisobutylketon opvarmes under tilbagesvaling i 80 minutter. Reak= tionsblandingen inddampes til tørhed under reduceret tryk, og remanensen opløses i 20 ml ethylacetat under opvarmning. Efter afkøling udskilles krystaller, og de isoleres ved filtrering, og filtratet vaskes med 5$’s vandig natriumcarbonatopløsning og vand, 15 147143 tørres og inddampes. Bundfaldet blandes med de tidligere vundne krystaller og omkrystalliseres af ethylacetat, hvorved der fås 3,92 g krystalformigt methyl-4-(benzothiazol-2-yl)dithio-3-(2= -phenoxyacetamido)-oc-isopropenyl-2-oxoazetidin-l-acetat; smeltepunkt 146 - 147°C.
Eksempel B 4.
En blanding af 0,18 g methyl-6-(2-phenoxyacetamido)-2,2-dimethyl= penam-3-carboxylat-l-oxid og 0,08 g benzothiazol-2-thiol i 4 ml toluen opvarmes under tilbagesvaling i 4 timer. Reaktionsblandingen lades henstå, hvorefter der udskilles krystaller, som isoleres ved filtrering. Filtratet inddampes til tørhed, og remanensen opløses i 3 ml ethylacetat under opvarmning. En blanding af 0,180 g methyl-6-(2-phenoxyacetamido)-2,2-dimethyl= penam-3-carboxylat-l-oxid og 0,084 g benzothiazol-2-thiol i 4 ml benzen opvarmes under tilbagesvaling i 19 timer. Reaktionsblandingen behandles som beskrevet i eksempel B 4, hvorved der fås 185 mg kry= staller af methyl-4-(benzothiazol-2-yl)dithio-3-(2-phenoxyacetamido)= -a-isopropenyl-2-oxoazetidin-l-acetat; smeltepunkt 146 - 147°C.
Eksempel B 6.
En blanding af 10,9 g 6-(2-phenoxyacetamido)-2,2-dimethylpenam-3= -carboxylsyre-l-oxid og 5,01 g benzothiazol-2-thiol i 500 ml tert.bu= tanol koges under tilbagesvaling i 20 timer. Reaktionsblandingen inddampes til tørhed under reduceret tryk, og remanensen vaskes 16 147143 med etlier, hvorved der fås 11,95 g krystaller af 4-(benzothiazol= -2-yl)dithio-3-(2-phénoxyacetamido)-a-isopropenyl-2-oxoazetidin--l-eddi= kesyre; smeltepunkt 146 - 148°C.
Eksempel B 7.
En blanding af 1,14 g methyl-6-(2-phenoxyacetamido)-2,2-dimethylpe= nam-3-carboxylat-l-oxid og 620 mg benzylthiol i 20 ml methylisobu= tylketon koges under tilbagesvaling i 15 timer. Reaktionsblandingen inddampes under reduceret tryk, og remanensen opløses i ether, hvorefter der tilsættes petroleumsether. Det olieagtige lag fraskil= les ved dekantering, underkastes chromatografering på silicagel og elueres med chloroform. En blanding af 1,84 g methyl-6-(2-phenoxyacetamido)-2,2-dimethyl= penam-3-carboxylat-l-oxid og 0,552 g thiophenol i 80 ml benzen koges under tilbagesvaling i 30 timer. Reaktionsblandingen inddam= pes under reduceret tryk, og remanensen opløses i chloroform. Opløsningen vaskes tre gange med 5%*s vandig natriumcarbonatop= løsning og 1 gang med vand, tørres og inddampes. Remanensen under= kastes chromatografering på silicagel og elueres med 300 ml af en blanding af ether og petroleumsether i forholdet 1:3 og derpå med ether. Hver 50 ml af eluatet opsamles separat som fraktioner, og den femte fraktion inddampes, hvorved der fås 0,58 g methyl-4-phe= nyldithio-3-(2-phenoxyacetamido)-oc-isopropenyl-2-oxoazetidin-l-ace= tat i form af en farveløs olie. IR-Spektrum (flydende film): 1780, 1747 og 1690 cm-^.

Reaktionsblandingen inddampes, og remanensen underkastes cbromatografering på silicagel og elueres med chloroform. Hver 50 ml af eluatet opsamles separat som fraktioner, og den første og anden fraktion inddampes, hvorved der fås 0,405 g methyl-4-(thiazol-2-yl)dithio-3-(2-phenoxyacetamido)= -a-isopropenyl-2-oxoazetidin-l-acetat.i form af en farveløs olie. IR-Spektrum (flydende film): 1780, 1747 og 1687 cm-^.

En blanding af 0,736 g methyl-6-(2-phenoxyacetamido)-2,2-dimethyl= penam-3-carboxylat-l-oxid, 0,302 g benzoxazol-2-thiol og 20 ml benzen koges under tilbagesvaling i 19 timer. Reaktionsblandingen behandles på lignende måde som beskrevet i eksempel B 11, og den sjette fraktion inddampes, hvorved der fås methyl-4-(benzoxazol-2= -yl)dithio-3-(2-phenoxyacetamido)-cc-isopropenyl-2-oxoazetidin-l-acetat i form af en farveløs olie. En blanding af 0,368 g methyl-6-(2-phenoxyacetamido)-2,2-dimethyl= penam-3-carboxylat-l-oxid, 0,161 g quinolin-2-thiol og 10 ml methyl= isobutylketon koges under tilbagesvaling i 6 timer. Reaktions= blandingen inddampes under reduceret tryk, og remanensen under= kastes chromatografering på silicagel og elueres med chloroform.
Hver 50 ml af eluatet opsamles separat som fraktioner, og den ottende og niende fraktion inddampes. Remanensen underkastes chromatografering på silicagel og elueres med chloroform. Hver 30 ml af eluatet opsamles separat som fraktioner. 2,06 g 4-(benzothiazol-2-yl)dithio-3-(2-phenoxyacetamido)-a-iso= propenyl-2-oxoazetidin-l-eddikesyre (fremstillet ifølge eksempel B6) opløses i 20 ml tetrahydrofuran, og til den omrørte opløsning sættes dråbevis 3,2 g carbontetrachloridopløsning indeholdende 10 vægtprocent brom. Reaktionsblandingen omrøres i 1 time, hvorefter bundfaldet fraskilles ved filtrering. Filtratet inddampes under re= duceret tryk, og til remanensen sættes en ringe mængde ethylacetat. Ethylacetatopløsningen vaskes med mættet vandig natriumchloridop-løsning, tørres og inddampes. Eksempel D 3.
800 mg carbontetrachloridopløsning indeholdende 10 vægtprocent brom sættes dråbevis under afkøling i isbad til en omrørt opløsning af 530 mg methyl-4-(benzothiazol-2-yl)dithio-3-(2-phenoxyacetamido)-a= -isopropenyl-2-oxozetidin-l-acetat (fremstillet ifølge eksempel B3-5) og 120 mg acetamid i 5 ml tetrahydrofuran. Blandingen filtre= res, og filtratet inddampes under reduceret tryk. Remanensen oplø= ses i ethylacetat, og opløsningen vaskes først med vand, derpå med en vandig opløsning af natriumcarbonat og igen med vand. Opløsnin= gen tørres og inddampes, hvorved der som remanens fås 430 mg olie= formigt methyl-6-(2-phenoxyacetamido)-2-methyl-2-brommethylpenam= -3-carboxylat. 0,96 g 2,2,2-trichlorethyl-4-(benzothiazol-2-yl)dithio-3-[2-(thio= phen-2-yl)acetamido]-cc-isopropenyl-2-oxoazetidin-l-acetat (frem= stillet ifølge eksempel B12) opløses i 15 ml methylenchlorid, og til denne opløsning sættes 0,09 g acetamid. Til den omrørte blanding sættes en carbontetrachloridopløsning indeholdende 10 vægtprocent brom under afkøling i isbad. Blandingen omrøres ved denne tempera= tur i 1 time, og bundfaldet isoleres ved filtrering. Filtratet vas= kes med mættet vandig natriumhydrogencarbonatopløsning og derefter med vand, tørres over magnesiumsulfat og inddampes, hvorved der som remanens fås 0,85 g olieformigt 2,2,2-trichlorethyl-6-[2-(thiophen= -2-yl)acetamido]2-methyl-2-brommethylpenam-3-carboxylat. Eksempel D 6.
800 mg carbontetrachloridopløsning indeholdende 10 vægtprocent brom sættes dråbevis til en blanding af 530 mg methyl-4-(benzothia= zol-2-yl)dithio-3~( 2-phenoxyacetamido )-oc-isopropenyl-2-oxoazetidin= -1-acetat, 120 mg acetamid og 5 ml tetrahydrofuran under afkøling i isbad. En blanding af 210 mg methyl-7-(2-phenoxyacetamido)-3-methyl-3-brom= cepham-4-carboxylat 1 5 ml acetonitril og 100 mg vandfrit natrium= acetat opvarmes under tilbagesvaling i 6 1/2 time. Reaktionsblan= dingen inddampes under reduceret tryk, og remanensen opløses i ethylacetat. Opløsningen vaskes med vand, tørres og inddampes. Remanensen krystalliseres af en blanding af ether og methanol, hvorved der fås 40 mg krystalformigt methyl-7-(2-phenoxyacetamido)= -3-methyl-3-cephem-4-carboxylat; smeltepunkt 124 - 125°C.
Eksempel G 2.
En blanding af 0,25 g methyl-7-(2-phenoxyacetamido)-3-methyl-3= -iodcepham-4-carboxylat i 10 ml tørt acetonitril og 45 mg natrium= acetat opvarmes under tilbagesvaling i 4 timer og inddampes. Rema= nensen blandes med vand og ekstraheres med chloroform. Chloroform= fasen vaskes med vandig natriumthiosulfatopløsning og derefter med vand, tørres og inddampes. Remanensen underkastes chromatografering på silicagel og elueres med chloroform, hvorved der fås 50 mg kry= stalformigt methyl-7-(2-phenoxyacetamido)-3-methyl-3-cephem-4-car= boxylat; smeltepunkt 124 - 125°C.
Eksempel G 3·
En opløsning af 860 mg 7-(2-phenoxyacetamido)-3-methyl-3-bromcepham=» -4-carboxylsyre i 20 ml tørt dichlorethan afkøles til en temperatur mellem -40 og -50°C, hvorefter der tilsættes pyridin og 4,7 g 10#'s dichlormethanopløsning af thionylchlorid. Blandingen omrøres ved en temperatur mellem -40 og -50°C i 1 time og derefter ved stuetemperatur natten over, hvorefter den hældes ud i isvand og omrøres i 30 minutter. Dichlorethanfasen ekstraheres med vandig natriumhydrogencarbonatopløsning, og den vandige fase syrnes ved tilsætning af phosphorsyre og ekstraheres med ethylacetat. Ethyl= acetatfasen vaskes med vand, tørres, behandles med aktivkul og inddampes, hvorved der fås 80 g krystaller af 7-(2-phenoxyacetami= do)-3-methyl-3-cephem-4-carboxylsyre; smeltepunkt 172 - 175°C.
