Opfindelsen angår hidtil ukendte gastrin-analoge heptapeptidderivater med den almene formel I
HOOC-(CH2)3-CO-Ala-Tyr-Gly-Trp-X-Asp-Phe-NH2, (I) 5 i hvilken X står for leucin eller methionin, til anvendelse som diagnostika.
Der er blevet beskrevet talrige syntetiske peptider fra det C-terminale område af gastrin, der på lignende måde som det naturlige hormon gastrin bevirker 10 stimulation af maven med hensyn til syreproduktion og volumenudskillelse. Blandt disse syntetiske peptider udmærker sig pentapeptidet med formlen IV
Boc-fl-Ala-Trp-Met-Asp-Phe-Ni^, (IV) 15 i hvilken Boc er en tert.butyloxycarbonylrest (The lancet 1966,s. 993). Pentapeptidet udviser den stærkeste stimulerende virkning i sammenligning med andre syntetiske peptider.
Dette pentapeptid IV er også allerede sammenlignet 20 med et heptapeptid, der kun adskiller sig fra den her omhandlede forbindelse med formlen I med X = Met ved, at glutaroylresten mangler. Det udviser en biologisk virkning, der kan sammenlignes med peptidet IVs.
Derimod har det her omhandlede peptid med formlen I, 25 i hvilken X står for methionin, en 1,3 gange så stor biologisk virkning som peptidet med formlen IV, det her omhandlede peptid med formlen I, i hvilken X = leucin, endda en 2,3 gange så’ stor virkning som peptidet med formlen IV, beregnet på molærbasis.

De her omhandlede hidtil ukendte peptider er diagnostika. De er fri for ikke-fysiologiske komponenter, stimulerer mavesaftproduktionen og kan derfor anvendes 10 som diagnostika til undersøgelse af mavesækkens funktion.
Anvendelsen af de her omhandlede peptider ved mave-funktionsdiagnostik sker på i og for sig kendt måde, ved titrimetrisk analyse af den udskilte syre efter indgivelse af prøveforbindelsen, jf. L. Demling, "Der kranke 15 Magen", Muncheh 1970, side 64-84, se især side 67-68.
Peptiderne indgives herved subcutant eller intravenøst. I undtagelsestilfælde kan peptiderne også indgives ikke-pa-ranteralt.
Ved parenteral indgivelse anvendes en 0,001 til 20 0,1%'s opløsning i vand, eventuelt med tilsætning af fysiologiske mængder natriumchlorid, eller i en vandig pufferopløsning. Som puffer kan for eksempel anvendes 1/15 molær phosphat-puffer (primært kaliumphosphat og sekundært natriumphosphat ifølge SørenS.en og Clark) med 25 pH-værdier på 5,0 til 7,5. Disse opløsninger tjener til subcutan og intramuskulær anvendelse.
Til nasal anvendelse anvendes ca. 0,1%'s vandige opløsninger, 0,1 til 3%'s olieholdige suspensioner eller et 0,1 til 3%'s, tørt pulverpræparat med for eksempel 30 lactose eller mannitol som bærer.
Fremstillingen af de her omhandlede peptider illustreres ved de følgende eksempler.
Eksempler
Til renhedsprøvning af de i det følgende fremsti1-35 lede forbindelser anvendes tyndtlagschromatografi med følgende løbemiddelsysterner: 1) tert.-butanol/pyridin/petroleumsether, 1:1:8 2) methanol/vand 80:20 3) methylethylketon/pyridin/vand/iseddikesyre 70:15:15:2 4) butanol/eddikesyre/vand 2:1:1

Der anvendes de i peptidkemien sædvanlige forkortelser; dicyclohexylcarbodiimid = DCC; dimethylformamid = DMF. Z betyder benzyloxycarbonyl.
Eksempel 1.
g Glutaroyl-Ala-Tyr-Gly-Trp-Leu-Asp-Phe-NH2 a) Z-Tyr(But)-Gly-OCH3 74 g (0,2 mol) Z-Tyr(OBu1^-OH og 54 g (0,4 mol) 1-hydroxy-benzotriazol opløses i 300 ml tetrahydrofuran, der afkø-^0 les til 5°C, og en opløsning af 42 g (0,2 mol) DCC i 50 ml tetrahydrofuran sættes til. Derpå efteromrøres i 1 time ved 0°C og 1 time ved stuetemperatur. Så skilles det udskilte dicyclohexylurinstof fra ved sugning. Til den tilbageblivende opløsning sættes ved 0°C en opløsning ^ af 25 g (0,2 mol) glycin-methylester-hydrochlorid og 27 ml (0,2 mol) N-ethylmorpholin i 150 ml tetrahydrofuran. Derpå efteromrøres i endnu 2 timer ved stuetemperatur, så afdestilieres opløsningsmidlet i vakuum ved stuetemperatur, remanensen optages i ethylacetat og ethylace-2q tatopløsningen rystes med 0,2 N svovlsyre, mættet natri-umhydrogencarbonatopløsning og vand. Efter tørring af ethylacetatopløsningen over magnesiumsulfat fås slutproduktet ved afdampning af opløsningsmidlet i vakuum som en svagt gullig olie. Produktet kan ikke adskilles ved 2g tyndtlagschromatografi i systemerne 1, 2, 3 og 4.
Udbytte: 64 g (73% af teoretisk) b) H-Tyr(But)-Gly-OCH3, HC1 60 g Z-Tyr(Bu^)-Gly-OMe i 1 liter methanol hydrogeneres katalytisk ved en pH-værdi på 3 ved hjælp af 10 g 10%'s 30 Pd/BaSO4 under tilsætning af 1 N methanolisk HC1. Efter fraskillelse af katalysatoren afdestilleres opløsningsmidlet. Den tilbageblivende harpiks bliver fast ved udrivning med ether. Udbytte 45 g, Smp. fra 97°C under opskumning. [a]20 = + 27,0° (c = 1 i methanol).

Til en opløsning af 29 g (0,13 mol) Z-Ala-OH og 35 g 1-hydroxybenzotriazol i 240 ml DMF sættes ved 5°C 29 g DCC. Der omrøres 1 time ved 0°C og 1 time ved stuetempera-5 tur, det udfældede dicyclohexylurinstof filtreres fra, og der tilsættes en opløsning af 44,8 g (0,13 mol) Η-Tyr (Bu1*) -Gly-OCH^, HC1 og 16,5 ml N-ethylmorpholin i 300 ml DMF. Efter en reaktionstid på 3 timer ved stuetemperatur afdestilleres opløsningsmidlet i vakuum, re-10 manensen optages i ethylacetat, og ethylacetatopløsningen mættes med 1 N HC1. Der vaskes med natriumhydrogencar-bonat og vand, tørres over natriumsulfat og inddampes i vakuum. Det halvfaste produkt bliver fast ved digerering med ether.
15 Udbytte: 46,5 g (70%). Smp. 107°C.

d) Z-Ala-Tyr(Bufc)-Gly-OH
44,6 g methylester fra c) i 150 ml dioxan og 89 ml 1 N natriumhydroxydopløsning omrøres i 1 time ved stuetemperatur. Der neutraliseres med 1 N HC1, inddampes i vakuum 25 til 1/3 af det oprindelige volumen og pH-værdien indstilles til 3 med 1 N HC1. Den udfældede olie optages i 700 ml ether. Til den over MgS04 tørrede opløsning sættes 250 ml petroleumsether. Bundfaldet udkrystalliserer ved 0°C i løbet af nogle dage.

83 g Z-Trp-Leu-OMe, fremstillet ifølge J. Org. Chem 31 (1966), s. 3400, omrøres i en blanding af 500 ml dioxan

5 147733 og 100 ml 2 N NaOH i 100 minutter. pH-værdien indstilles til 6 med 1 N HC1, der inddampes, tilsættes noget vand, og pH-værdien indstilles til 3 med 1 N HC1. Det udfældede produkt omkrystalliseres fra methanol/vand.

Til en opløsning af 23,8 g (52 mmol) Z-Trp-Leu-OH,.
19,5 g (42 mmol) Η-Asp (OBu*") -Phe-N^iHCl, fremstillet ifølge J. Chem. Soc. (C) 1966, s. 555, 12 g N-hydroxy-succinimid og 6,65 ml N-ethylmorpholin i 400 ml tetra-hydrofuran sættes ved -5°C under omrøring en opløsning af 10,8 g DCC i 18 ml tetrahydrofuran. Under omrøring afkø- 1 5 les frivilligt til stuetemperatur. Efter henstand natten over frafiltreres det udfældede dicyclohexylurinstof, og opløsningen inddampes til tørhed i vakuum. Den tilbageblivende olie mættes med 5%'s citronsyre. Den digereres med natriumhydrogencarbonat og vand, tørres i vakuum 2 Ω over P2°5 og udkoges med lidt ethanol.
Udbytte: 31,5 g, smp. 212°C, [a]20 =-36,7° (c = 1 i DMF)

g) H-Trp-Leu-Asp(OBu1^ -Phe-NH2,HCl 7,68 g Z-Tetrapeptid fra f) hydrogeneres katalytisk i 350 ml methanol analogt med eksempel lb). Efter sædvanlig oparbejdning fås 6,5 g af et amorft produkt.

h) Z-Ala-Tyr (Bu1") -Gly-Trp-Leu-Asp (OBut) -Phe-NH2 0,5 g (1 mmol) Z-Ala-Tyr(Bufc)-Gly-OH, 0,64 g (1 mmol) 35 H-Trp-Leu-Asp(OBut)-Phe-NH2,HCl, 0,27 g (2 mmol) 1-hydro- xybenzotriazol og 0,13 ml N-ethylmorpholin opløses i 10 ml 6

147733 DMF, dertil sættes ved 0°C 0,24 g DCC, og der omrøres i 15 timer ved stuetemperatur. Det udfældede dicyclohexyl- urinstof filtreres fra, og opløsningen inddampes i vakuum.
Remanensen optages i ethylacetat og ekstraheres ved under 5 5°C med 10%'s citronsyre, hvorefter der vaskes med mættet natriumhydrogencarbonat og vand. Efter tørring af ethyl- acetatopløsningen over MgSO^ inddampes. Derved udfældes 450 mg af Z-heptapeptidet. Yderligere 193 mg lader sig udvinde af moderluden ved fældning med ether. Smp. 192°C, 10 [a]20= -27,8° (c = 1 i DMF)

15 i) H-Ala-Tyr(But)-Gly-Trp-Leu-Asp(OBut)-Phe-NH2,HC1 4 g Z-heptapeptid fra h) hydrogeneres katalytisk i 1,3 liter methanol analogt med eksempel lb. Efter afdestillering af opløsningsmidlet digereres remanensen med ether, og der opnås 2,8 g med et smp. på 199-200°C (dekompone-ring).
k) Glutaroyl-Ala-Tyr (Ru*1) -Gly-Trp-Leu-Asp (OBu^) -Phe-NH2
Til en opløsning af 8,8 g (12 mmol) af H-heptapeptid-25 hydrochloridet fra i) i 50 ml DMF sættes ved 0°C i løbet af 40 minutter portionsvis under omrøring 2,3 g (20 mmol) glutarsyreanhydrid og 2,56 ml N-ethylmorpholin. Der omrøres i 5 timer ved 0°C med påfølgende henstand i 16 timer ved 4°C, opløsningsmidlet afdestilleres i vakuum, og rema-nensen digereres med ether.
Udbytte: 8,8 g, smp. 230-232°C (dekomponering).

Glutaroyl-Ala-Tyr-Gly-Trp-Met-Asp-Phe-NI^ a) Z-Ala-Tyr(But)-Gly-Trp-Met-Asp(OBut)-Phe-NH2 17,7 g Z-Ala-Tyr(But)-Gly-OH, fremstillet ifølge eksem- pel ld), 23 g H-Trp-Met-Asp (OBu*") -Phe-NH2, fremstillet ifølge J. Chem. Soc. (C), 1967, s. 2410, og 9,6 g 1-hydro- xybenzotriazol opløses i 120 ml DMF. Ved 0°C tilsættes 8g DCC, og der omrøres natten over ved stuetemperatur.
Efter frafiltrering af dicyclohexylurinstoffet inddampes
2q opløsningen i vakuum, remanensen udrives med vand, 0,2 N

b) H-Ala-Tyr(Bufc)-Gly-Trp-Met-Asp(OBufc)-Phe-NH2 37 g af Z-heptapeptidet fra a) hydrogeneres katalytisk i 30 400 ml DMF under tilsætning af 15 ml cyclohexylamin og rigeligt Pd. Efter at fraspaltningen af Z-gruppen er sket, filtreres katalysatoren fra, opløsningen inddampes til tørhed i vakuum, og remanensen udrives med ether. For at rense produktet yderligere fældes fra DMF/ether/petroleums-ether.
Udbytte: 24,3 g, smp. 209°C (dekomponering), ta]20= -22,5° (c = 1 i DMF)

Til en opløsning af 12 g H-heptapeptidet fra b) i 60 ml DMF sættes portionsvis under omrøring ved 0°C i løbet af 40 minutter 3 g glutarsyreanhydrid og 4,3 ml N-ethyl-5 morpholin. Der omrøres endnu 5 timer ved 0°C med påfølgende henstand i 16 timer ved 4°C. Så afdestilleres opløsningsmidlet i vakuum, og remanensen digereres med ether.
Udbytte: 10,3 g, smp. 231°C (dekomponering).

d) Glutaroyl-Ala-Tyr-Gly-Trp-Met-Asp-Phe-NH2 10,1 g af det beskyttede glutaroylheptapeptid fra c) oplø-^ ses i 50 ml 90%'s trifluoreddikesyre. Efter 75 minutters henstand ved stuetemperatur sidestilleres trifluoreddi-kesyren i vakuum, remanensen digereres med 10 ml ethanol, og derpå tilsættes 100 ml ether.
