Opfindelsen angår en fremgangsmåde til påvisning og/eller bestemmelse af et biologisk stof, fortrinsvis et antigen eller et hapten, i en prøve, ved hvilken man sammenblander prøven med en kendt mængde af et kob-5 lingsprodukt mellem det biologiske stof og et enzym og en kendt mængde af et antistof, som er aktivt overfor det biologiske stof, og efter reaktionen bestemmer enzymaktiviteten som et mål for mængden af det biologiske stof i prøven 10 Indenfor medicinsk praksis og forskning er der til stadighed behov for hurtige, nøjagtige kvantitative bestemmelser af biologiske stoffer i yderst lave koncentrationer. Tilstedeværelsen af lægemidler eller narkotica i legemsvæsker, såsom spyt, blod eller urin, skal kunne 15 bestemmes i meget små mængder med tilfredsstillende nøjagtighed. Endvidere er det ved medicinske diagnoser ofte vigtigt at kende tilstedeværelsen af forskellige stoffer, som syntetiseres naturligt af legemet, eller som er indført. Disse stoffer omfatter hormoner, både steroider og 20 polypeptider, prostaglandiner og toksiner samt andre materialer, som kan indgå i legemsfunktionen.
Til opfyldelse af disse behov er der blevet anvist en række metoder til analyse for spormængder af materialer. En sædvanlig anvendt fremgangsmåde til isola-25 tion og påvisning af stoffer i biologisk fluidum er tyndtlagschromatografi (TLC), f.eks. kombineret med massespektroskopi eller gasfasechromatografi. Imidlertid har TCL flere mangler, idet denne fremgangsmåde er langsom, kan lide af interferens fra mange forskellige ma-30 terialer og har manglende pålidelighed. Mangelen på tilfredsstillende alternativer har således fremkaldt en intens forskning for at finde forbedrede fremgangsmåder til adskillelse og identifikation.
Et alternativ til TLC har været radioaktivitets-35 immun-undersøgelse (radioimmunoassay, RIA). Ved denne fremgangsmåde anvendes antistoffer for specifikke hapte-ner eller antigener. Ved blanding af et antistof med opløsninger af haptenet eller antigenet og med en radio- 2 149808 aktiv hapten- eller antigen-analog, vil den radioaktive analoge hindres i at hindes til antistoffet i en udstrækning, som er direkte afhængig af koncentrationen af haptenet eller antigenet i opløsningen. Ved derefter 5 at separere og bestemme den frie radioaktive analoge fra den til antistof bundne radioaktive analoge, kan man indirekte bestemme mængden af hapten eller antigen i den oprindelige opløsning. Imidlertid kan anvendelsen af radioisotoper ved en sådan bestemmelse være potentielt 10 sundhedsskadeligt, og den instrumentation, som sædvanligvis er nødvendig til radioaktivitets-immun-undersøgelse, er forholdsvis kompliceret og er sædvanligvis for kostbar til at mindre hospitaler og læger rutinemæssigt kan foretage f.eks. analyse af en patients blod eller urin.
15 Enzym-immun-bestemmelse afhjælper de ovennævnte ulemper og har endvidere den særlige fordel at give mulighed for forstærkning af den målte parameter.
Principielt erstattes ved denne fremgangsmåde den radioaktive analoge til det biologiske stof med et enzym-20 mærket biologisk stof (hapten eller antigen). Sådanne modificerede enzymmolekyler bevarer deres enzymatiske aktivitet, og det enzym-mærkede biologiske stof vil konkurrerer med den ukendte mængde af frit biologisk stof i systemet med hensyn til antistof-kompleksdannelse.
25 Komplekserne kan separeres (jf. britisk patentbeskrivelse nr. 1.348.935) ved udnyttelse af deres uopløselighed i visse tilfælde, og aktiviteten af det uopløselige stof eller den del, der forbliver i opløsning, anvendes som et mål for den oprindeligt tilstedeværende mængde anti-30 gen. Det samme princip kan anvendes til et omvendt system, hvor der anvendes enzym-mærkede antistoffer, hvis den umodificerede form af det samme antistof i en biologisk væske skal bestemmes.
Det har nu vist sig, at covalent binding af biotin 35 (vitamin H) til antistofmolekylet fører til et opløseligt biotin-mærket kompleks, hvilket muliggør bekvem separering af alle antistofformer, indbefattet alle enzym-mærkede og ikke-mærkéde biologiske stof-antistof-komplekser.

Separationen kan derefter udføres ved anvendelse af det biotin-specifikke receptor-protein avidin, som immobili-seres i en uopløselig form. Den meget kraftige affinitet mellem avidin og biotin, som nærmer sig karakter af en 5 covalent binding, resulterer i uopløseliggørelse af alle antistofformer og muliggør som følge heraf en særdeles effektiv og nem fjernelse af alle biotin-mærkede antistoffer og komplekser, der er dannet med sådanne antistoffer.
10 Ifølge en udførelsesform for fremgangsmåden i- følge opfindelsen tilvejebringes således en fremgangsmåde til påvisning og/eller bestemmelse af et biologisk stof i en prøve, hvilken fremgangsmåde er ejendommelig ved det i krav l's kendetegnende del anførte.
15 Ved fremgangsmåden ifølge opfindelsen kan påvises og/eller bestemmes et hvilket som helst biologisk stof, for hvilket der kan fremstilles et passende antistof med tilfredsstillende specificitet og affinitet til det biologiske stof. Nyere litteratur indeholder et stigende 20 antal beskrivelser af antistoffer for stadig flere forskellige biologiske stoffer. Forbindelser, for hvilke der kan tilvejebringes antistoffer varierer fra simple phe-nylalkylaminer, f.eks. amphetamin, til polymerer:::med meget høje molekylvægte, f.eks. proteiner.
25 De biologiske stoffer, som kan påvises og/eller bestemmes ved fremgangsmåden ifølge opfindelsen kan opdeles i tre forskellige kategorier afhængigt af deres biologiske forhold til antistoffet. Den første kategori er antigener, som, når de indføres i blodstrømmen af et 30 hvirveldyr, resulterer i dannelse af antistoffer. Den anden kategori er haptener, som, når de bindes til en antigenbærer, og den haptenbundne antigenbærer indføres i blodstrømmen af et hvirveldyr, giver dannelse af antistoffer, som er specifikke for haptenet. Den tredie ka-35 tegori af biologiske stoffer indbefatter de, som har naturligt forekommende antistoffer i en levende organisme, hvor antistofferne kan isoleres i en form, som er 149908 4 specifik for det biologiske stof.
Den vigtigste gruppe biologiske stoffer med hensyn til fremgangsmåden ifølge opfindelsen er stofferne fra anden kategori, haptenerne. Fremgangsmåde til frem-5 stilling af antistoffer for de tre forskellige kategorier af biologiske stoffer er kendte.
Udvælgelse af enzymet til anvendelse ved fremgangsmåden ifølge opfindelsen er afhængig af forskellige forhold. Det skal således have potentielt reaktionsdygti-10 ge grupper, til hvilke det biologiske stof kan kobles uden at enzymaktiviteten ødelægges, og det må ikke forekomme naturligt i væsentlig grad i den type væv, som skal analyseres for det pågældende biologiske stof. Endvidere skal enzymet have forholdsvis lang holdbarhed, 15 høj’ specifik aktivitet og være i stand til let at kunne bestemmes, f.eks. ved hjælp af et spektrophotometer med synligt lys.
Eksempler på enzymer, som sædvanligvis kan anvendes ved fremgangsmåden ifølge opfindelsen, er malat-20 dehydrogenase, staphylococ-nuclease, delta-5-ketosteroidm isomerase, fra gær stammende dehydrogenase, fra gær stammende glykose-6-phosphat-dehydrogenase, a-glycero-phosphatdehydrogenase, triosephosphatisomerase og ræd-dike-peroxidase, mere foretrukket alkalin-phosphatase, 25 asparaginase, glykoseoxidase, β-galactosidase og ribo-nuclease. Sædvanligvis foretrækkes det at rense enzymet, f.eks. ved dialyse mod saltvand, før anvendelsen.
Fremstillingen af de enzym-mærkede biologiske stoffer til anvendelse ved fremgangsmåden ifølge opfin-30 delsen kan foregå på flere kendte måder. Nogle biologiske stoffer kan allerede indeholde grupper, som kan krydsbindes med reaktive grupper på overfladen af enzymet, medens andre stoffer skal forsynes med sådanne grupper ved organisk kemiske omsætninger. Det skal fremhæves, at 35 hverken de oprindelige immunologiske egenskaber af det biologiske stof eller aktiviteten af enzymet må ændres væsentligt ved denne proces. De grupper i enzymet, som er specielt egnet til koblingsreaktioner, er amino- og 5 149908 carboxyl-grupper. Hvis dét modificerede eller ikke-modi-ficerede biologiske stof også indeholder sådanne grupper, kan koblingen f.eks. udføres ved reaktioner, der er kendte fra peptidsynteser. Endvidere kan sådanne stoffer, som f.eks. glutaraldehyd, difluordinitrodiphenylsulphon, 5 toluendiisocyanat, di- og tri-chlor-s-triazin og andre anvendes ved koblingsreaktionen.
Eksempler på kobling af biologiske stoffer til enzymer er f.eks. beskrevet i L.A.Steinberger, Immuno-cytochemistry, Prentice Hall, New Jersey, (1974).

Academic Press, New York, 1967. De omtalte fremgangsmåder anvendes til fremstilling af conjugerede for immu-15 nisering, men de kan også anvendes til fremstilling af enzym-mærkede biologiske stoffer, som er afgørende ved fremgangsmåden ifølge opfindelsen.
Biotin-mærket antistof fremstilles sædvanligvis ved omsætning af et biotinderivat, fiieks. et biotinester-20 derivat, sådom N-hydroxysuccinimidesteren af biotin, med antistoffet. Biotinesterderivatet opløses i et polært, aprotisk opløsningsmiddel,.f.eks. dimethylformamid, og sættes derefter i et 20 til 300 molært overskud til antistoffet i 0,01 M til 1,0 M, fortrinsvis 0,05 M til 0,5 M, 25 mest foretrukket 0,1 M phosphatpuffer, f.eks. kalium-phosphatpuffer ved en pH-værdi på 6,5 til 8,5, fortrinsvis 7,5. Efter sammenblanding af reaktanterne tillades reaktionen at forløbe ved en temperatur på fra 2-10°C, fortrinsvis ved 4°C, i tilstrækkelig tid til at den 30 kan afsluttes. Sædvanligvis tager dette ca. 10 timer.
Efter afslutning af reaktionen kan det biotin-mærkede antistof separeres fra reaktionsblandingen ved kendte fremgangsmåder, f.eks. ved gelpermeationschromatografi1 på f.eks. en krydsbundet dextran.
35 Uopløseliggjort avidin, dvs. avidin, der er im- mobiliseret ved tilknytning til et fast bærestof, kan fremstilles ved en fremgangsmåde, der er kendt fra prak- 6 149908 sis eller fra litteraturen, f.eks. ved covalent binding med makromolekylære uopløselige bærere, såsom agarose, polystyren, polyacrylamid, nylon, krydsbundet dextran eller filterpapir, eller ved fysisk kobling til uopløse-5 lige bærere, såsom glasperler eller plastikgenstande, eller til indersiden af reagensglas fremstillet af enten plastik eller glas eller til mikrotiter-plader.
Specielle eksempler på koblinger af haptener og andre biologiske molekyler til agarose og polyacrylami-10 der er beskrevet af Cuatrecasas i J.Biol. Chem., 245, 3059-3065, (1970). W.B.Jacoby og M.Wilcheck, Methods in Enzymology, bind 34, Academic Press, New York 1974.
Disse fremgangsmåder kan principielt også anvendes til fremstilling af (a) avidin-fast bærer, (b) hapten-prote-15 inconjugerede og (c) biotin-antistof-conjugerede. Sædvanligvis aktiveres enten avidin eller bæreren før den covalente binding finder sted, men sædvanligvis aktiveres bæreren før den covalente binding. Ved en anden form for fremgangsmåden ifølge opfindelsen anvendes der som 20 bærer agarose, da dette har en udmærket evne til kobling af avidin og bevarelse af evne til binding af biotin.
Mest foretrukket anvendes benzoquinon-aktiveret agarose, da det er nemt at fremstille, og på grund af dets manglende evne til ikke-specifik absorption af enzym. Ved 25 andre former for fremgangsmåden ifølge opfindelsen kobles avidin til nylønringe eller -stænger, eller til indersiden af polystyren-reagensglas.
En skematisk fremstilling af bestemmelsen er vist i fig. 1. Til den beholder, hvor reaktionen skal finde 30 sted, sættes ved stuetemperatur nær fysiologisk pH-værdi, fortrinsvis ved pH-værdi på 7 efter hinanden med et minimalt tidsrum mellem tilsætningerne: frie biologiske stoffer, f.eks. indeholdt i en serumprøve, en vandig opløsning af enzym-mærket biologisk stof og en tilstrækkelig 35 mængde forud mærket antistof til at neutralisere (dvs. danne kompleks med) 50%-80% af det enzym-mærkede biologiske stof. Disse tre komponenter blandes og netop uop-løseliggjort avidin tilsættes til reaktionsblandingen 149908 7 til frembringelse af et heterogent system, og blandingen rystes til der opnås et forud bestemt ligevægtspunkt.
Den anvendte mængde avidin er sædvanligvis flere hundrede gange større end den teoretisk krævede. Den faste fase 5 fjerner det frie antistof samt antistof bundet til det biologiske stof. Den overstående væske indeholder enzym-mærket biologisk stof i en mængde, der er direkte proportional med indholdet af frit biologisk stof, medens den udfældede faste fase indeholder enzym-mærket biolo-10 gisk stof i en mængde, der er omvendt proportional med mængden af frit biologisk stof. Enten den udfældede faste fase eller den overstående væske kan undersøges for enzymatisk aktivitet til bestemmelse af biologisk stof i den ukendte prøve.
15 Målingen af enzymaktivitet af den faste og/eller væskeformige fase af reaktionsblandingen fra fremgangsmåden ifølge opfindelsen kan udføres ved kendte fremgangsmåder. Se f.eks. H.U.Bergmeyer, Method for Enzymatic Analysis, Academic Press, New York (1965). Analysen af 20 den omhyggeligt separerede og skyllede faste fase kan finde sted ved fjernelse af den overstående væske ved f.eks. frasugning.
De forskellige former i hvilke reagenserne ifølge opfindelsen kan anvendes er mangfoldige. F.eks. kan det 25 enzym-mærkede biologiske stof være frysetørret eller opløst i en puffer. Endvidere kan anvendes en fast bærer, f.eks. en papirstrimmel imprægneret med det enzym-mærkede, biologiske stof.
Opfindelsen angår endvidere et analysesæt til 30 anvendelse ved fremgangsmåden, hvilket sæt er ejendommeligt ved det i krav 6’s kendetegnende del anførte.
Eventuelt kan sættet også indeholde de nødvendige hjælpemidler til fremstilling af seriefortyndinger af prøven, på hvilken der skal udføres en kvantitativ ber -35 stemmelse, såsom reagensglas, pipetter og flasker med fortyndingsmiddel.

Opfindelsen angår også et biotin-mærket antistof til anvendelse ved fremgangsmåden og til inkorporering i analysesættet ifølge opfindelsen.
Opfindelsen belyses nærmere ved hjælp af de føl-5 gende eksempler.
Eksempel 1
Kobling af digoxin til enzymer.
Digoxin blev koblet til enzym på følgende måde: 3 10 H-digoxin (250 yCi/mg) i ethanol blev omsat med et lille molært overskud natriummetaperiodat i to timer.
En portion på 50 nmol H-digoxin blev sat til 5 nmol enzym i 1 ml natriumacetatpuffer (200 mM, pH-værdi 7).
Den endelige koncentration af ethanol var sædvanligvis 1510-20%. 10-20mg natriumcyanborhydrid blev tilsat, og reaktionen fik lov at forløbe i tre dage ved 4°C. Mærket enzym blev separeret fra Reaktanterne ved gel-eksklusions chromatografi på SephadexG-50. Tabellen nedenfor viser graden af substitution med digoxin (beregnet ved hjælp 20 af radioaktivitetsmålinger) og bevarelsen af enzymatisk aktivitet for fire enzymer efter reaktion under de beskrevne betingelser.

Konjugering af H-digoxin med malat-dehydrogenase blev undersøgt grundigere (fig. 2). Oxideret digoxin blev omsat i forskellige koncentrationer (det molære 35 overskud varierede fra 5 til 50) med portioner af enzym, og bevarelsen af enzymspecifik aktivitet blev bestemt i hvert tilfælde. Endvidere blev graden af substitution af digoxin bestemt både ved tritiumtælling til bestem- 149903 9 melse af det absolutte antal haptener og ved RIA til bestemmelse af antallet af immunologisk reaktive digoxin-rester. Resultaterne viser, at malat-dehydrogenase er meget følsom for graden af mærkningen med digoxin, og at 5 kun 20% af de tilknyttede grupper er funktionelt til rådighed til at reagere med antistof.
Uopløseliggørelse af avidin.
Avidin blev koblet direkte til en fast bærer af 10 benzoquinon-aktiveret sepharose. Avidingelen blev fortyndet efter ønske med uomsat sepharose.
En 50%'s suspension af agarose i natriumphosphat-puffer (100 mM, pH-værdi = 8,0) blev i et forhold på 4:1 volumen/volumen sat til p-benzoquinon (250 m mo-:; 15 lær) i ethanol. Efter omrøring af suspensionen i en time ved stuetemperatur blev den aktiverede gel vasket ved filtrering under sugning ved anvendelse af lige store volumener efter hinanden af ethanol (20%), vandig natri-umchlorid (1,0 M), vand og natriumphosphat (pH-værdi 20 8, 100 m molær). Gelen (1 volumen) blev derefter sat til avidin (1 volumen, 10 mg/ml) opløst i natriumphosphat-puffer (pH 8, 100 mM) , og den resulterende suspen sion blev omrystet i 15 timer ved 4°C. Derefter blev den avidin-gel-conjugerede vasket efter hinanden med 5 25 volumener af hver af natriumacetat (0,1 M, pH-værdi 4,0) indeholdende natriumchlorid (500 mM) , natrium- bie arbonat (0,1 M, pH-værdi 9,0) indeholdende natriumchlorid (500 m M) og vand. Substitutionen af avidin på gelen var i området 4-5 mg/ml pakket gel. Den substi-30 tuerede avidin viste sig at have 100% af sin biotinbindingsevne.
Substitution af antistof med biotin.
Portioner af fåre-antidigoxin-antistof blev be-35 handlet i kaliumphosphat (0,1 M, pH-værdi 7,5) med forskellige mængder af N-hydroxysuccinimidesteren af biotin i dimethylformamid (DMF) svarende til et molært overskud andragende fra 20 til 300. Slutkoncen- 10 149908 trationen af DMF var 50%. Efter inkubation natten over ved 4°C blev det biotinbehandlede antistof separeret fra reaktanterne Ved gelpermeationschromatografi på Sephadex G-50. Titeren af det biotinbehandlede antistof blev be- 125 5 stemt ved anvendelse af I-digoxin og dextranovertruk-ket trækul. Sensibiliteten af det biotiniserede antistof overfor avidin-gel blev bestemt ved anvendelse af forud ioderet antistof og måling af nedbringelsen af 125 I med avidin-gel. Ved biotiniseringsniveauer over 10 3 biotinmolekyler pr. antis to fmolekyl bevarede antistoffet sin titer overfor digoxin og blev fuldstændig absorberet af avidin-gel. Det empirisk valgte antistof-biotin-præparat til enzym-immun-bestemmelsen opfyldte disse kriterier .
15 Gljicoseoxidase-digoxin-conjugerede med forskellige niveauer af substitution af digoxin blev titreret med forskellige fortyndinger af biotiniseret antistof og overskud af avidin-gel (fig. 3). Det var forud påvist, at enzymderivaterne ikke blev hæmmet af endog meget høje 20 koncentrationer af anti-digoxin-antistof. Dataene viser at ved fortyndinger på mellem 1:100 og lt 10-00 af stamopløsningen af antistof foreligger digoxin-enzym og antistof i omtrent støkiometriske mængder, således at i det væsentlige al enzymaktivitet udfældes af gelen.
25 Dataene viser også, at der med forøget digoxinsubstitution kræves mindre antistof til udfældning af 50% af enzymaktiviteten, hvilket viser, at immun-aktiviteten af den enzym-conjugerede forøges med substitution.
30 -Reaktionen mellem antidigoxin-antistof og digoxin.
Ved anvendelse af passende mængder glucoseoxidase-digoxin, biotinmærket antistof og overskydende avidin-gel fulgtes det tidsforløb af analysen, som krævedes til opnåelse af ligevægt mellem de tre komponenter.
35 Glucoseoxidase-digoxin og avidin-gel blev forud inkuberet ved stuetemperatur. Ved t = 0 blev tilsat biotinmærket antistof, og blandingen blev omrystet ved stuetemperatur. Med forskellige tidsintervaller blev portio- 11 149908 ner af forsøgsblandingen centrifugeret, og den overstående væske blev undersøgt for enzymaktivitet. Sædvanligvis opnåedes maksimal formindskelse af enzym-mærket digoxin forårsaget af antistof-avidin-gel-kompleks inden-5 for 90 minutter, medens binding af biotin-mærket antistof af avidin-gel i det væsentlige er fuldstændig indenfor 10 minutter.
Enzym-immun-bestemmelse for digoxin.
10 50 yl serum-digoxin, 50 yl glucoseoxidase-mærket digoxin, som indeholdt 1 ng/ml bundet digoxin, og 50 yl passende fortyndet biotin-mærket antidigoxin-antistof blev blandet i et reagensglas (fig. 1). Der blev straks tilsat en 50¾ vandig suspension af 50 yl avidin-sepha-15 rose, som forud var fortyndet i forholdet 1:10 med nativ sepharose. Alle analysereagenser var i phosphat-puffer (50 mM, ’ pH-værdi 7) indeholdende 0,1% bovin-seramalbumin. Reaktionsblandingen blev derefter inkuberet i 2 timer ved stuetemperatur under omrystning, hvor-20 efter den blev suspenderet i 5 ml iskold puffer indeholdende 0,1% bovinserumalbumin. Blandingen blev centrifugeret, og den overstående væske blev frasuget efterladende en gel-pellet. Efter vaskning af denne pellet blev tilsat 1,0 ml analysepuffer indeholdende glucose 25 (100 mM), o-dianisidin (0,1 mg/ml) og ræddike-per- oxidase (7,5 yg/ml) i natriumphosphatpuffer (100 mM, pH-værdi 6), og der blev omrystet i 1 time. Alternativt kan 0,1 mM, , 2,2-azino-bis-(3-ethylbenzthiozolin-sulphonsyre)(ABTS) anvendes i stedet for o-dianisidin 30 som enzymsubstrat.
Derefter blev blandingen afkølet, og den optiske tæthed af den overstående væske blev bestemt ved en bølgelængde på 450 nm, og værdierne for patientsera blev sammenlignet med en standardkurve fastlagt med kontrol-35 sera.
Fig. 4 viser virkningerne af tilsætninger af forskellige mængder fri digoxin til inkubationsblandingen (afsat som logarithmen af digoxinkoncentrationen i ng/ml 12 149908 mod % total enzymaktivitet på gelen). Forholdet mellem mængden af fri digoxin og enzymaktiviteten er lineær i området fra 0,15 til 10,00 ng/ml fri digoxin.
5 Eksempel 2: Codein.
Til påvisning af den almene anvendelighed af den beskrevne enzym-immun-bestemmelse blev det narkotiske lægemiddel codein bestemt ved denne fremgangsmåde.
10 Kobling af codein til glucoseoxidase.
20 ymol codeinhemisuccinat opløst i 600 til tør, gendestilleret dimethylformamid blev sat til 200 ymol N-hydroxysuccinimid opløst i 300 yl tør gendestilleret dioxan og 20 ymol dicyclohexylcarbodiimid i 30 yl tør, 15 gendestilleret dioxan. Blandingen blev henstillet til reaktion ved stuetemperatur i 5-6 timer i et tætlukket reagensglas. Det således in situ fremstillede codein-hemisuccinat-N-hydroxysuccinimid blev anvendt uden rensning til kobling til enzymet. Til 5 mM enzym (glucose· 2o oxidase) opløst i 600-900 yl natriumacetatpuffer (100 mM, pH-værdi 7,0) blev sat 100-500 yl (fortrinsvis 300 yl) af den som overfor beskrevet fremstillede codein-N-hydroxysuccinimidester-opløsning. Efter 12-24 timer ved stuetemperatur blev enzymet (conjugeret med codein 25 samt eventuelt ikke-conjugeret enzym) separeret fra reaktanterne ved geleksklusionschromatografi. Graden af codeinsubstitution i forhold til giucoseoxidase blev bestemt ved radioaktivitets-immunbestemmelse. Som i tilfældet med dioxin varierer graden af substitution af codein 30 afhængigt af reaktionsbetingelserne. En substitution med 3-5 codeinmolekyler pr. enzymmolekyle (glucoseoxidase) viste sig at være ideel for den nærværende analysemetode.
Kobling af biotin til anti-codein-antistof.
35 Denne reaktion blev udført som beskrevet i eksem pel 1 for kobling af biotin til anti-digoxin-antistof.
Titeren af biotiniseret antistof var i det væsentlige 13 149908 uændret i forhold til antistof, der ikke var overført i et derivat. Antistof-præparater med 3 eller flere co-valentbundne biotinmolekyler kunne udfældes fuldstændigt med avidin i fast fase og viste sig at være ideelt egnecfe 5 for den nærværende bestemmelse.
Enzym-immunbestemmelse af codein.
Med passende, forudbestemte mængder af enzym-codein-conjugeret, biotiniseret anti-codein-antistof og 10 avidin i fast fase bestemtes den tid, som krævedes til opnåelse af ligevægt mellem de tre analysekomponenter. Sædvanligvis opnåede systemets ligevægt efter 45-60 minutters forløb.
Codeinstandard (50 μΐ 0,1-10.000 ng/ml opløsning), 15 50 μΐ. glucoseoxidase-codeinconjugeret (indeholdende 1-20 m mol codein) og passende fortyndet biotin-mærket anti-codein-antistof blev blandet i et reagensglas. 50 μΐ af en 50% vandig suspension af avidin i fast fase blev sat til ovennævnte blanding. Alle analysereagenser blev 20 fremstillet i 50 i molær phosphatpuffer, pH-værdi 7 indeholdende 0,1% bovinserumalbumin. Reaktionsblandingen blev efter inkubation i 2 timer ved stuetemperatur fortyndet med 5 ml iskold puffer indeholdende 0,1% bovinserumalbumin. Blandingen blev centrifugeret og den over-25 stående væske blev frasuget. Efter vaskning af den opnåede pellet blev tilsat 1 ml af analysepufferen indeholdende glucose (100 mM ) o-dianisidin (o,l mg/ml) og ræddike-peroxidase (7-5 μg/ml) i phosphatpuffer (100 mM , pH-værdi 6). Analyseblandingen blev in-30 kuberet i 30-90 minutter (fortrinsvis 60 minutter) ved stuetemperatur, afkølet i isbad, og den optiske tæthed ved 450 nm blev bestemt. På denne måde konstrueredes en standardkurve med forskellige codeinkoncentrationer (fig. 5) .
