Opfindelsen angår en hidtil ukendt β-amylase, en fremgangsmåde til dets fremstilling samt præparater indeholdende β-amylasen sammen med andre enzymer.
5 Som β-amylase betegnes sædvanligvis de exo-virkende maltogene amylaser, som katalyserer hydrolysen af 1,4-a-glucosid bindinger i amylose, amylopektin og beslægtede glucosepolymerer. Maltoseenheder fjernes trin for trin fra de ikke-reducerende kædeender, indtil molekylet er nedbrudt eller, hvis det drejer 10 sig om amylopektin, indtil et grenpunkt nås. Den frigivne maltose har den β-anomere form, heraf betegnelsen β-amylase.
β-formen af maltose vil i vandige opløsninger isomeri-sere spontant til en blanding af a- og β-formerne. Langvarig reaktion af β-amylase på amylopektin eller delvist nedbrudt 15 amylopektin fører til dannelsen af β-grænsedextrin, d.v.s.
materiale, som ikke er påvirkelig ved yderligere hydrolyse med β-amylase.
β-amylaser kan anvendes til fremstilling af maltose-holdige sirupper, der kan bruges i konfekture-, bageri- og 20 bryggeriproduktion.
Ved fremstillingen af højmaltosesirupper anvendes β-amylaser almindeligvis sammen med afgreningsenzymer for at forøge maltoseindholdet i produktet ved hydrolyse af 1,6-a-glucoside bindingerne i β-grænsedextrinerne.
25 β-amylaser er blevet isoleret fra forskellige planter og mikroorganismer (W.M. Fogarty og C.T. Kelly: Progess in Industrial Microbiology, bind 15, p. 112-115, 1979). Disse β-amylaser er kendetegnet ved temperaturoptimer i området fra 40°C til 65°C og pH-optimum i området fra 4,5 til 7.
30 Afgreningsenzymer udviser oftest et pH-optimum i området fra 3,5 til 6 og er således mere aktive i en sur opløsning. Ved kombineret anvendelse af β-amylaser og afgreningsenzymer er det valgte pH sædvanligvis et kompromis mellem pH-optimet af hvert af de valgte enzymer, eller enzymerne anvendes 35 successivt.

Ved en fremgangsmåde til fremstilling af maltose, hvori stivelse i en vandig opløsning hydrolyseres ved hjælp af en β-amylase, er det endvidere en fordel at anvende temperaturer på eller over 60°C for at hæmme retrogradering og for at undgå 5 mikrobiel infektion.
Plånte-β-amylaserne har yderligere den ulempe, at der til fremstilling deraf kræves store mængder råmateriale og et stort energiforbrug til ekstrahering og forarbejdning, hvorimod mikrobielle β-amylaser kan produceres i stor målestok med 10 relativt lave omkostninger.
Dansk patent nr. 153.799 beskriver en maltogen amylase, som er tilstrækkeligt thermostabil til anvendelse ved temperaturer i området fra 60 til 65°C. Imidlertid ligger dette enzyms pH-optimum ved pH 4,5 til 5,5. Ydermere hydrolyserer enzymet 15 ifølge det nævnte danske patent maltotriose, hvilket resulterer i fremstilling af glucose.
I højmaltosesirupper bør tilstedeværelsen af glucose undgås, og anvendelsen af den omtalte maltogene amylase kræver derfor påfølgende fjernelse af den fremkomne glucose til et 20 akceptabelt niveau.
Der er derfor stadig et behov for et effektivt mikrobielt β-amylaseprodukt, som er tilstrækkeligt thermostabilt til at kunne anvendes ved 60 - 70°C i perioder af længere varighed til at muliggøre hydrolyse af stivelsen på en rentabel måde, som 25 er tilstrækkeligt stabil ved pH-værdier under 5 til opnåelse af samtidig hydrolyse af stivelse med β-amylasen og et afgrenings-enzym under optimale forhold, og som under produktionen af højmaltosesirup ikke producerer for store mængder glucose, som i nogle tilfælde bagefter skal fjernes fra siruppen.
30 Det er den foreliggende opfindelses formål at tilveje bringe en hidtil ukendt mikrobiel β-amylase, som har en høj temperaturstabilitet, et pH-optimum ved pH-værdier under 5, og som producerer minimale mængder glucose.
Den foreliggende opfindelse bygger på den opdagelse, at 35 en hidtil ukendt mikrobiel extracellulær β-amylase (21 - 51 β-amylase), der har disse egenskaber, kan fremstilles fra en nyligt isoleret Bacillus stamme NCIB 11608.

Opfindelsen forklares nærmere i det følgende under henvisning til tegningen, hvori fig. 1 illustrerer det maltogene enzyms relative aktivitet i forhold til temperatur, og 5 fig. 2 illustrerer relativ aktivitet i forhold til pH.
Ifølge et første aspekt tilvejebringer den foreliggende opfindelse en hidtil ukendt thermostabil β-amylase, som er ejendommelig ved, at den har temperaturoptimum målt ved 30 10 minutters reaktionstid i acetatpuffer (0,1M) ved pH 4,5 på ca 70°C, pH optimum ved ca. 60° i området mellem 4 og 5 og kan fremstilles ved dyrkning af Bacillusstammen NCIB 11608 i et passende næringsmedium og efterfølgende isolering af den dannede β-amylase med de ovennævnte egenskaber fra kulturvæsken.
15 Ifølge et andet aspekt tilvejebringer den foreliggende opfindelse en fremgangsmåde til fremstilling af den ovenfor omtalte thermostabile β-amylase, hvilken fremgangsmåde er ejendommelig ved at Bacillusstamme NCIB 11608 eller varianter eller mutanter deraf med i det væsentlige samme egenskaber som 20 denne dyrkes i et egnet næringsmedium indeholdende carbon- og nitrogenkilder og uorganiske salte, efterfulgt af udvinding af β-amylaseenzymet fra dyrkningsmediet.
Forsøg har vist, at den nye β-amylase er egnet til produktion af maltose og højmaltosesirupper. sådanne produkter er 25 af betydelig interesse i forbindelse med produktionen af visse konfekturevarer p.g.a. lav vandoptagelse, lav viskositet, god varmestabilitet og mild, ikke for sød maltosesrnag.
Den industrielle fremgangsmåde til fremstilling af maltosesirupper omfatter forflydigelse af stivelse, derpå 30 forsukring med et maltoseproducerende enzym, og eventuelt med et enzym, der spalter 1,6-grenpunkterne i amylopektin, f.eks. pullu-lanase eller isoamylase.
Ifølge yderligere et aspekt tilvejebringer den foreliggende opfindelse et enzympræparat, som er ejendommeligt ved at 35 det omfatter en β-amylase ifølge krav 1 i kombination med mindst et afgreningsenzym.

Ved anvendelse af det nye enzympræparat er det muligt at udføre den ovennævnte forsukring af stivelsen under optimale pH-betingelser, hvorved udbyttet af maltose øges, og mængden af tilsat enzym kan mindskes.
5 21-51 β-amylase hydrolyserer amylopektin, glycogen og amylose, idet der kun frigives β-maltose.
Fra forgrenede polysaccharider, såsom amylopektin og delvist hydrolyseret amylopektin danner 21 - 51 β-amylase β-grænsedextriner, som kan hydrolyseres med glucoamylase, isoamy-10 lase, pullulanase eller lignende.
21-51 maltogen amylase adskiller sig fra andre Bacillus β-amylaser i følgende henseender: 1. det er stabilt i puffer ved 70°C, 2. det er stabilt i et pH-område fra 4,0 til 5,0, hvilket 15 betyder, at det kan anvendes i forbindelse med afgreningsenzymer såsom isoamylase og pullulanase.
3. glucose produceres kun i ubetydelige mængder.
Den mikroorganisme, der er i stand til at producere β-20 amylasen ifølge den foreliggende opfindelse, blev udvalgt under hensyntagen til dens evne til at vokse på et agarsubstrat fremstillet som følger: trypton (10 g), amylopektin (CPC snowflake 10 g), Bacto agar (40 g) og deioniseret vand (1000 ml) blandes aseptisk ved 25 55°C med en tilsvarende mængde saltopløsning med følgende s ammensætning: (NH^) 2 SO^ : 0,04% vægtprocent

Den isolerede mikroorganisme blev deponeret hos National Collection of Industrial Bacteria (NCIB), Torry Research Station, Aberdeen, Scotland, d. 15. marts 1983 og fik tildelt nr. 35 NCIB 11608. NCIB, der er et internationalt deponeringsinstitut,

5 Taxonomi Morfologi
Stavformede bakterier med en diameter på 0,6 til 0,8 pm.
10 Sporerne er af oval eller cylindrisk form, anbragt centralt til subterminalt, og forårsager ikke opsvulmen af sporehusene.
Biokemiske reaktioner 15 gramfarve: positiv, anaerob vækst: negativ, catalase: positiv, VP reaktion: negativ, nitratreduktion: positiv, 20 lecithinase: negativ, vækst i 3,5% NaCl: negativ, hydrolyse af: stivelse: positiv, casein: svagt positiv, 25 syreproduktion fra: glucose: positiv, arabinose: negativ, xylose: negativ, mannitol: positiv, 30 anvendelse af: citrat: negativ, propionat: negativ, og vækst ved 50°C negativ.
35 Bacillus NCIB 11608 vokser ikke på almindeligt anvendte medier med en pH-værdi over 6. De optimale vækstbetingelser for denne bakterie er pH 4,8 til 5,8 og en temperatur på 30 til 37°C.

Den maksimale væksttemperatur er for denne bakterie 45°C.
I de førnævnte forsøg anvendtes et standardmedium indeholdende lige store mængder af 1% trypton og BA-1 salte.

pH indstilles til 3,0 med 10 N H2S04 inden autoklavering.
pH er 4,8 til 5,2 i det færdigblandede trypton basis medium.
15 Alle nævnte biokemiske reaktioner og dyrkningsforsøg blev udført på dette substrat med eller uden tilsætning af 1% gærekstrakt.
Standarddyrkningsmediet til formering af NCIB 11608 indeholder 0,5% amylopektin og intet gærekstrakt. Vegetative 20 celler af NCIB 11608 indeholder ofte bobler, som i et fasekon-trastmikroskop fremstår som brydningskugler, og som ikke farves, når cellerne stænkes let med safranin eller methylenblåt.
De morfologiske og biokemiske egenskaber af Bacillus NCIB 11608 er meget lig dem, der kendetegner Bacillus megaterium; 25 men (3-amylasens egenskaber ligger meget langt fra, hvad der er beskrevet for B. megaterium β-amylase, der har temperaturoptimum ved ca. 50°C, pH optimum ved ca. pH 7.
30 Bestemmelse af enzymaktivitet
En β-amylaseenhed (βϋ) defineres som den mængde enzym, som under standardbetingelser (temperatur 60°C, pH 4,5 og reaktionstid 30 minutter) producerer reduktionssukker svarende til 1 μπιοί maltose/minut.

0,5 ml 2% stivelse i 0,1 M acetatpuffer inkuberes med 100 μΐ enzym opløst i deioniseret vand indeholdende 0,5 - 2 βϋ per ml. Reaktionen standses efter 30 minutter ved tilsætning af 0,3 ml 0,5 N NaOH, og blandingen fortyndes til et forhold på 1:10 5 med deioniseret vand.
Indholdet af reduktionssukker bestemmes derpå efter Somogyi metoden (Somogyi: J.Biol.Chem., 153, p. 375-80 (1944)).
Enzymfremstilling 10 En Bacillusstamme, der er i stand til frembringe β- amylasen ifølge den foreliggende opfindelse, opformeres oftest på et fast substrat inden dyrkning under aerobe betingelser i et egnet næringsmedium. Begge medier indeholder assimilerbare carbon- og nitrogenkilder foruden uorganiske salte, fortrinsvis 15 sammen med vækstfremmende næringsstoffer, såsom gærekstrakt.
Fermenteringen udføres typisk ved 30°C - 37°C og ved et pH på 5 -6 og holdes fortrinsvis automatisk konstant. Enzymet udskilles i mediet.
Den resulterende gæringsvæske kan befris for bakterie-20 celler og affaldsstoffer sammen med andre faste stoffer, f.eks. ved filtrering. Supernatanten indeholdende enzymet kan renses yderligere, f.eks. ved filtrering eller centrifugering, og derpå koncentreres som krævet, f.eks. ved ultrafiltrering eller i en inddamper under reduceret tryk til et koncentrat, som, om ønsket, 25 kan tørres, f.eks. ved lyophilisering eller forstøvningstørring.
Rensning af enzymet
Den maltogene amylase ifølge den foreliggende opfindelse kan renses fra en kontinuert gæringskulturvæske som følger: 30 250 liter dyrkningsmedium filtreres, og filtratet ultrafiltreres, sterilfiltreres, og frysetørres.
Pulveret opløses i 15 mM acetatpuffer, pH 5,0, og dialyseres mod 15 mM acetatpuffer, pH 5,0, indtil ledningsevnen er ca. 1 mS. Dialysatet sættes derefter på en kationbytter CM-35 sepharose® C1-6B, som er blevet bragt i ligevægt med den samme puffer. Amylasen passerer gennem søjlen, medens 60% af de resterende proteiner holdes tilbage af ion-bytteren.

pH i effluenten fra denne søjle indstilles til 4,0 med eddikesyre, og eluatet sættes derefter til en CM-sepharose® C1-6B søjle indstillet med 15 mM acetatpuffer med pH på 4,0. Under disse omstændigheder adsorberes amylasen af ion-bytteren. Enzymet 5 elueres derpå med acetatpuffer med pH 4,0 med stigende ionstyrke. Enzymaktiviteten i eluatet følger proteinindholdet i en symmetrisk top.
Enzymkemiske egenskaber
10 Den omhandlede β-amylases aktivitets afhængighed af pH
og temperatur blev bestemt ved den tidligere beskrevne metode under anvendelse af en reaktionsblanding, hvori pH og temperatur blev indstillet til forudbestemte værdier.
Der henvises igen til figurerne, hvor 15 fig. 1 grafisk illustrerer den relative aktivitet i forhold til temperatur (substrat 2% opløselig stivelse, pH 4,5 (0,1 M acetat), 30 minutter reaktionstid), og fig. 2 grafisk illustrerer den relative aktivitet i forhold til pH (temperatur 60°C, substrat 2% opløselig stivelse, 20 30 minutters reaktionstid, Mcllvaine puffer).
Det fremgår af figurerne, at 21 - 51 (3-amylase har et aktivitetsoptimum ved pH 4,5 på ca. 70°C, og at dets pH-optimum ligger i området fra 4,0 - 5,0. Mere end 60% af den maksimale aktivitet findes stadig ved 80°C.

Gæringen blev udført i en BIOFLO® (New Brunswick,
Canada) fermentor med et arbejdsvolumen på 1 liter.
Gæringen blev påbegyndt med 100 ml af det ovenfor beskrevne podemateriale og substratdoseringen påbegyndt efter 24 ...... 30 timer ved 30°C.
pH blev indstillet til 5,5 ± 0,2 med 3% svovlsyre og temperaturen blev holdt på 30°C ± 0,2.
Beluftning: 0,5 liter/liter substrat/minut
Fortyndingshastighed: D = 0,090 ± 0,005 t.
35 24 timer efter podning var kulturen vokset ud og kontinuert kørsel startede, og allerede 24 timer senere var kulturen i ligevægt.

Over ca. 7 døgn blev der over tøris opsamlet 4450 ml kulturvæske, som blev centrifugeret ved 2300 rpm, 45 min. 4376 ml centrifugat blev koncentreret i et ultrafiltrerings-laboratoriemodul fra Sartorius med G-600 membran til 1200 ml 5 efter udskylning af apparatet. Dette koncentrat blev yderligere inddampet til 200 ml på en Buehi rotationsfordamper, 3 timer, 45°C. Dette materiale var udgangsmateriale for adskillelse af (3-amylasen fra de andre aktiviteter.

Adskillelse af β-amylaseaktiviteten fra pullulanase- og a-, amylaseaktiviteterne_
Et præparat, fremstillet som angivet ovenfor, opløstes 15 i 0,05 M acetat pH 4,0 og sattes på en ionbytningssøjle med CM-sepharose-CL-6B. Under disse betingelser bindes såvel pullulanase- som a-amylaseaktiviteterne, og β-amylasen løber lige igennem søjlen.
Fraktionerne analyseredes med stivelse og pullulan som 20 substrat, og aktivitetstyperne bestemtes ved tyndtlagskromatografi.
De fraktioner, der ikke gav reaktion med pullulan, og som hydrolyserede stivelse til maltose og ikke andet, samledes og anvendtes til analyse af β-amylasens egenskaber.
25 ' Eksempel 2
Forsukringssubstrater blev fremstillet ved at genopløse 30 en 7 DE (dextrinækvivalent) forstøvningstørret maltodextrin i deioniseret vand og tilnærme til ca. 30% tørstof (DS). Forsukringseksperimenterne blev udført i standardiaboratorie-batch-reaktorer på 500 ml. Substratprøver blev opvarmet til 55°C eller 60°C, pH blev indstillet til en begyndelsesværdi på 5,5, og 35 5 βϋ eller 20 j3U/g DS blev derpå tilsat. Prøver blev udtaget efter 4, 24, 48, 96 og 168 timer og opvarmet i kogende vand i 10 minutter for at inaktivere enzymet. Efter afkøling blev prøverne

filtreret og behandlet med mixed-bed ionbytterharpiks (Bio-Rad® AG 501 x 8 (D)) for at fjerne aske og opløselig N før analyse ved HPLC og gelkromatografi.
Fremgangsmåden blev gentaget med henholdsvis blandinger 5 af 5 eller 20 βυ/g DS af B-amylasen ifølge opfindelsen og 1 pullulanaseenhed (PNU)/g DS pullulanase og 100 isoamylaseenheder (IU)/g DS.
Resultaterne fremgår af tabel 1, hvor DPn angiver polymeriseringsgraden for produktet, d.v.s. DP1 er 10 monosaccharid(glucose) o.s.v..

Fremgangsmåde til fremstilling af β-amylaseenzymet ifølge krav 1, kendetegnet ved, at Bacillusstamme NCIB 11608 eller en variant eller mutant deraf med i det væsentlige samme egenskaber som denne dyrkes i et passende næringsmedium indeholdende carbon- og nitrogenkilder og uorganiske 25 salte, efterfulgt af udvinding af β-amylaseenzymet fra dyrkningsmediet.
- 3. Enzympræparat, kendetegnet ved, at det omfatter en β-amylase ifølge krav 1 i forbindelse med et eller 30 flere afgreningsenzymer.
