Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af hidtil ukendte bisindolalkaloider samt farmaceutisk acceptable salte heraf, nærmere betegnet aminosyrederivater, herunder peptidderivater af vinblastin.
5 De ved fremgangsmåden ifølge opfindelsen fremstillede forbindelser kan inkorporeres i farmaceutiske præparater og tjene som antitumormidler, navnlig til behandling af maligne tumorer hos mennesker.

Eksempler på sådanne alkaloider er vincaleukoblastin (US patentskrift 3.097.137), leurocristin eller vineristin
og leurosidin (US patentskrift 3.205.220), vinglycinat (BE
30 patentskrift 659.112) og vindesin (BE patentskrift 813.168).
Sidstnævnte forbindelse vindes ved kemisk modifikation af 1 2 naturligt vinblastin (formel I, R = OCH^, R = COCH^), der kan vindes ved ekstraktion fra blade af Catharanthus roseus. Vinblastin, vineristin og vindesin er tilgængelig i 35 handelen til brug i humanterapien, navnlig til behandling af leukæmi og visse faste tumorer.

Disse lægemidler har imidlertid vist sig at have væ- j sentlige ugunstige bivirkninger. Vineristin har neurotoxiske virkninger og vinblastin menes at have en kraftig skadelig virkning for knoglemarven, dvs. hæmatopoietisk toxicitet.
5 Virkningsmekanismen af disse lægemidler menes at minde om den mekanisme der er postuleret for den antimitotiske virkning af colchicin. I sådanne tilfælde ville disse stoffer virke ved inhibering af polymerisationen af tobulin til dannelse af mikrotubuler og påfølgende standsning af cellede-10 lingsforløbet i den mitotiske fase.

Udnyttelse af 1:1 complekser af tubulin med antitumo-rale bisindoi-alkaloider er beskrevet i BE patentskrift 554.053. I nogle tilfælde er der angivet lavere toxicitet og mere effektiv kemoterapeutisk aktivitet end for de fire alka-15 loiders vedkommende.
Forskellige andre kemiske modifikationer af vinblastin- molekylet er blevet afprøvet. En af cbisse modifikationer, 1 2 vinglycinat-sulfat (formel I,sulfat, R = OCH^/ R = COCH^NfCH^)2? Cancer Research 1967, 27, 221-227) er også af-20 prøvet klinisk, men har alment vist sig ikke at være overlegne over vinblastin eller vineristin.
BE patentskrift 813.168 angiver vindesin- (formel I, 1 2 R = NH2' R = H) og vinblastin-karboxamidderivater. Senere raporter tyder på at disse forbindelser, trods den almene 25 nytte af vindesin eller 3-karboxamid-4-0-desacetylvinblastin, ikke er effektive til behandling af mus induceret med murin 1210 leukæmi (C.J. Barnett et al., J. Med. Chem. 21, 88, 1978). Vindesin sælges for tiden på det europæiske marked (bl.a. i Frankrig og Tyskland) og er under anmeldelse til FDA-registre-30 ring i USA.
Aminosyrederivater af vinblastin og andre bisindol-alkaloider er foreslået generelt i BE patentskrift 813.168.
Der er imidlertid ikke angivet nogen specifikke aminosyrederivater og følgelig heller ikke nogen fysisk-kemiske beskrivel-35 ser, ingen specifikke fremstillingsmåder og ingen specifikke fysisk-kemiske egenskaber. Det må således antages at sådanne forbindelser faktisk ikke er blevet syntetiseret og/eller ik- 3

ke er blevet afprøvet for antimitotisk styrke, navnlig i betragtning af en angivelse i skriftet side 3, linje 10 ff., at "anti-neoplastisk aktivitet synes at være begrænset til meget specifikke strukturer, og chancerne for at vinde mere 5 aktive stoffer ved modifikation af disse strukturer synes at være tilsvarende svage".
Det har nu vist sig at de ved fremgangsmåden ifølge den foreliggende opfindelse fremstillede forbindelser har evne til i væsentlig grad at forsinke døden af mus der er podet intravenøst med P 388 og L 1210 leukæmier, og de udviser væsentlige fordele i forhold til de tidligere beskrevne vinbla-stinanaloger. Da Vinca-alkaloider generelt ikke udviser nogen aktivitet mod L 1210 leukæmi, er de resultater der er opnået med de omhandlede nye forbindelser i høj grad uventede og 15 overraskende aktiviteter. Virkninger på forsøgsfremkaldt P 388 tumorer har overraskende også vist sig i høj grad bedre end virkningerne af reference-Vinca-alkaloiderne. Der er iagttaget talrige totale afhjælpninger deraf. Forbindelserne udviser yderligere andre vigtige og uventede fordele i sam-20 menligning med vinblastin og kendte analoger, navnlig vinde-sin. Især er den iagttagede toxicitet generelt lavere end den tilsvarende toxicitet af vindesin eller vineristin.

5 i den almene formel II angiver en ester afledet af en hvilken som helst af de naturligt forekommende aminosyrer og deres optiske isomerer med D-konfiguration, nemlig glycin, alanin, valin, leucin, isoleucin, serin, threonin, asparaginsyre, glutaminsyre, asparagin, glutamin, arginin, lysin, cyste-10 in, cystin, methionin, fenylalanin, tyrosin, tryptofan, pro-lin, histidin, hydroxylysin eller hydroxyprolin.
Forbindelserne kan betegnes som 3-dekarbometoxy-0-4-desacetyl-vinblastin-3-karboxamid. De vil imidlertid i nærværende beskrivelse fortrinsvis blive betegnet scan N-(vin-15 blastin-23oyl)-aminosyrederivater (således at man altså udelader prefixet "dekarbometoxy").
Særligt foretrukne aminosyreesterforbindelser med den almene formel II er sådanne hvor R er et hydrogenatom og aminosyredelene er afledede grene til de følgende aminosyrer: 20 L- eller D-tryptofan, leucin, isoleucin, valin, alanin og fenylalanin. L-tryptofan, L-isoleucin og L-alanin har -særlig betydning.
Særligt foretrukne peptidderivater er sådanne ’med den almene formel II, hvor n er tallet 2 og aminosyreresterne er 25 afledet af de foretrukne naturlige aminosyrer som er angivet ovenfor, og i en hvilken som helst rækkefølge.
For så vidt angår forbindelsernes farmakologiske egenskaber må det ventes at små variationer i strukturen af peptidskelettet vil give forbindelser med sammenlignelig styrke.
30 Særlig vil tilstedeværelse af ikke-naturlige a-aminosyrer (fx norleucin, N-monosubstituerede α-aminosyrer eller α,α-dialkyl-aminosyrer) give forbindelser med tilsvarende aktivitet mod forskellige tumorer, og fremstilling af sådanne forbindelser skal anses for at indgå i den foreliggende opfin-35 delses rammer.

Fremgangsmåden ifølge opfindelsen er ejendommelig ved det i krav 1's kendetegnende del anførte.
Når aminosyre- eller peptiddelen indeholder en funk-3 tionel sidekæde R kan det være nødvendigt at beskytte de 5 funktionelle grupper i henhold til velkendte metoder der bruges i peptidkemien. Dette er specielt tilfældet for lysin og cystein. Beskyttelsen kan alt efter arten af aminosyren opnås ved hjælp af en trifluoracetyl-, benzyl-, t-butyl-, benzyl-oxykarbonyl-, t-butoxykarbonyl- eller tritylgruppe kondense- i — — j 10 ret med den funktionelle gruppe. Andre velkendte beskyttende grupper der bruges almindeligt i peptidkemien kan også bruges med held.
De omhandlede forbindelser kan fremstilles ud fra vin-blastin ved hydrazinolyse efterfulgt af nitrosering og omsæt- 15 ning med aminosyreesteren eller peptidet i overensstemmelse med nedenstående reaktionsskema: .sA NaW02.> r* J . I CH-OH HCl ^

Reaktionsfølgens første trin består fortrinsvis i at man sætter vandfrit hydrazin i overskud til en opløsning af vinblastinbase i vandfrit metanol. Opløsningen opvarmes under en inert atmosfære eller Ar) i 12-30 timer ved en tempera-5 tur mellem ca. 30 og 70°C. Fortrinsvis holdes temperaturen på ca. 60°C, hvorved reaktionstiden vil være ca. 24 timer.
Det resulterende hydrazid af 4-O-desacetyl-vinblastin-1 2 23-oinsyre (formel I, R = NH-N^, R = H) isoleredes derefter ved tilsætning af vand, ekstraktion med et vandublandbart op-10 løsningsmiddel såsom metylenklorid og koncentration under nedsat tryk. Forbindelsen kan renses yderligere ved søjlekromatografi, fortrinsvis på neutralt silika.
I næste trin omdannes hydrazidgruppen i det modificerede vinblastin til en acylazidgruppe. Denne omdannelse sker 15 bedst ved at man sætter natriumnitrit til hydrazidet opløst i en vandig-sur metanolblanding. Syren i denne blanding kan fx være saltsyre. Reaktionstemperaturen holdes mellem ca. 0°C og 5°C.
Efter ekstraktion med et vandublandbart, aprotisk op-20 løsningsmiddel, fortrinsvis kloroform eller metylenklorid, fraskilles den organiske fase og koncentreres partielt.
Acylazidet isoleres i almindelighed ikke men sættes direkte til den ønskede aminosyreester eller polypeptid, eller et beskyttet derivat deraf, opløst i et passende opløsningsmid-25 del såsom metylenklorid.
Den mængde aminosyre som skal bruges er omkring 1-5 ækvivalenter i forhold til vinblastinkarboxazidet.
Reaktionsblandingen holdes typisk mellem ca. -3°C og +10°C i 8-72 timer og hyppigst ca. 15 timer. Overvågning af 30 reaktionen opnås bedst ved tyndlagskromatografi. Efter reaktionens fuldførelse kan opløsningsmidlet fjernes under nedsat tryk og det resulterende produkt omdannes til et sulfatsalt eller et andet passende salt afledet fra en mineralsk eller organisk syre, ved krystallisation fra en metanolisk opløs-35 ning af den tilsvarende syre. Den rene forbindelse kan isoleres og renses ved konventionel teknik såsom krystallisation eller kromatografi.
Eventuelt kan det resulterende 4-0-desacetyl-modifice-rede vinblastin genacyleres enten direkte til dannelse af 2

8 et vinblastinderivat II hvor R betegner COCH^ (J. Med.Chem.
22, 391, 1979) eller ved dannelse af 3,4-diacetoxyderivatet efterfulgt af selektiv hydrolyse af 3-acetoxygruppen i stilling 3. Hydroxygruppen i stilling 4 kan også forestres med 5 andre aktiverede syrederivater indeholdende 1-9 kulstofatomer.
De ved fremgangsmåden fremstillede forbindelser kan indgå i farmaceutiske præparater, navnlig til brug ved behandling af cancer hos mennesker, idet sådanne præparater indeholder et eller flere af de omhandlede bisindolalkaloider, 10 fortrinsvis sammen med en farmaceutisk bærer.
Forbindelserne udviser særlig bemærkelsesværdige anti-tumoir-egenskaber, der med held kan udnyttes i behandlingen a‘f kræft hos- .mennesker. j
Forbindelser kan fx være nyttige når de bruges til be-15 handling af L· 1-ilO, P 388, gliomer, lymfosarcomer og andre leukæmier eller maligne tumorer. I humanmedicinen kan de være nyttige til behandling af Hodgkins’ syge og mod andre faste tumorer som er tilgængelige for behandling med vinblastin, vineristin eller vindesin. De omhandlede forbindelser er også 20 nyttige i veterinærmedicinen til behandling af tumorer hos dyr.
Der kan også kame andre terapeutiske anvendelser i betragtning for de omhandlede hidtil ukendte forbindelser, idet disse ligesom vinblastin kan bruges til behandling af nogle 25 former af arthritis (US patentskrift 4.208.411) eller som vineristin, der er påvist at være aktiv til behandling af psoriasis (US patentskrift 3.749.784).
Til behandling af eksperimentelt fremkaldte maligne tilstande hos dyr, er de kemoterapeutiske virkninger afprø-30 vet ved hjælp af de tilsvarende sulfatsalte.
Ved disse forsøg blev DBA 2 hunmus (stammen Charles 4
River France) podet intravenøst med 10 leukæmiceller vun det fra 7 dage gamle P 388 eller L 1210 leukæmi-ascites. Dag 0 er dagen for podning af tumorcellerne.
35 Den til afprøvning værende forbindelse (i sulfatform) injiceres derefter intravenøst som en opløsning i fysiologisk saltopløsning (NaCl 9 o/oo) enten ved hjælp af et skema med en enkelt injektion (dag 1) eller med tre injektioner (dag 1, 9

2 og 3). MST (middeloverlevelsestid), dvs. den dag hvor halvdelen af dyrene er døde, beregnes efter 30. dag.
Værdien ILS (Increased Life Span, dvs. øget livslængde) beregnes i overensstemmelse med følgende formel: s * ils -fåHSfgtix 1001 -100
Antallet af overlevende mus efter 30. og 60. dag angives også.
Når doserne er for toxiske, kan ILS-procenten blive negativ, dvs. således at ubehandlede mus overlever længere 10 end de der er blevet injiceret med det antitumorale stof.
Under visse omstændigheder kan der iagttages nogen variabilitet af ILS-værdierne, i afhængighed med musenes oprindelse (DBA2 Frankrig eller USA).
De fundne resultater sammenlignedes med dem der opnås 15 med vinblastin (VLB) samt med vindesin (VDS) og vineristin (VCR) i tabel 1. Den farmakologiske overlegenhed af de ved fremgangsmåden ifølge opfindelsen fremstillede forbindelser fremgår også af tabellerne II-XII.
I tabel II gengives der resultater opnået med visse 20 derivater af naturligt leuein.
Følgende forbindelser er afprøvet: VLE = ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-leucinat VLM = metyl-N-(0-4-desacetylvinblastin-23-oyl)-L-leucinat.
I tabel III er der angivet resultater opnået med D-25 leueinderivater, nemlig ætyl-N-(O-4-desacetylvinblastin-23- oyl)-D-leucinat (VDLE).
I tabel IV er der vist resultater opnået med L-trypto-fanderivater, nemlig: V-Trypt-E = ætyl-N-(0-4-desacetylvinblastin-23-qyl) -L-tryptofa-30 nat, MST beregnet for 30 og 60 dage.

Stofferne indgives intravenøst i løbet af 3 injektioner. Dyrene aflives på 23. dag efter podningen. Vægten af 10 den primære tumor måles og udtrykkes i g; de pulmonære metastaser beregnes og deres diameter er målt. Metastaternes vægt er Bedømt ud fra den antagelse at de er kugler med en tæthed på 1; antallet af mus uden metastaser er også angivet.

Den antitumorale virkning vist i tabellerne I-XII bekræfter den uventede effektivitet af de ved fremgangsmåden ifølge opfindelsen fremstillede aminosyrederivater. De fleste af forbindelserne synes at være bedre end vindesin mod in-5 traperitonealt og intravenøst podede tumorer. Den exceptionel le aktivitet af disse forbindelser på L 1210 tumorer er påvist.
Det er værd at nævne at blandt de eksperimentelt fremkaldte tumorer der for tiden står til rådighed anses L 1210 10 leukæmi at være den eksperimentelle tumor, som er den mest signifikante med hensyn til valg af antitumor-lægemidler for mennesker.
I tabel XIII viser de antimetastatiske forsøg at VLB-Trypt-E-derivatet er langt bedre end vindesin. Effektivite-15 ten på en primær tumor er desuden sammenlignelig med effektiviteten af vinblastin eller vindesin.
Her bør særlig nævnes en fremragende virkning af ætyl-N-(desacetyl-0-4-vinblastin-23-oyl)-tryptofanat-sulfat, der giver en ILS efter 60 dage (P 388) på 457% med halvdelen 20 af musene overlevende yderligere. Den optimale dosis er ca.
60 mg. De aminosyrederivater som er afprøvet i amid- eller syreform udviser imidlertid kun svag eller ikke-signifikant aktivitet. Disse forbindelser er imidlertid nyttige som mellemprodukter til fremstilling af andre, meget aktive bisindol-25 alkaloider.
Generelt viser de ved fremgangsmåden ifølge opfindelsen fremstillede forbindelser sig væsentligt mindre giftige end de vinca-alkaloider der for tiden bruges i anti-cancer-terapien. Letaldosis udtrykt som LD^g for VLE er bestemt på 30 CD^ hunmus af stammen Charles River med en vægt på under 24 g. Litchfield og Wilcoxon's bedømmelsesmetode giver en LD^q på 32 mg/kg.
Tilsvarende doser for vinblastin og vindesin er henholdsvis 24 mg/kg og ca. 11 mg/kg. I modsætning til vinbla-35 stin og desacetylvinblastin er der konstateret fravær af hepa-tisk toxicitet ved doser på 20-40 mg/kg.
Den akutte toxicitet af V-Trp-E og VILE er også blevet bestemt på NMRI hunmus. De værdier som er opnået med 100,8 21

mg/kg og 17,7 mg/kg for henholdsvis V-Trp-E og VILE skal sammenlignes med de tilsvarende værdier på 27,4 og 13,8 mg/kg for henholdsvis vinblastin og vindesin. V-Trp-E er således tydeligt mindre giftig end VLB eller VDS.
5 Med henblik på terapeutisk anvendelse indgives de ved fremgangsmåden ifølge opfindelsen fremstillede forbindelser, eventuelt i lyofiliseret tilstand, fortrinsvis ad parenteral vej, opløst i et farmaceutisk acceptabelt bærestof enten i form af base eller i form af et farmaceutisk acceptabelt syre-10 additionssalt. Blandt syreadditionssaltene foretrækkes de u-giftige, farmaceutisk acceptable salte såsom salte med uorganiske syrer som hydrokloridet, fosfor- og svovlsyresalte, eller med organiske syrer såsom eddikesyre, propionsyre, ravsyre, vinsyre, oxalsyre, metansulfonsyre eller benzensulfon-15 syre. Fysiologisk saltopløsning eller andre saltopløsninger, fx pufret med et fosfat, er hensigtsmæssige opløsningsmidler.
I almindelighed kan forbindelserne bruges i humanterapien på analog måde med den teknik og under de begrænsninger som gælder for andre vinca-alkaloider. Foruden ad den ovennævnte par-20 enterale vej kan forbindelserne også indgives oralt. Den virksomme forbindelse (base eller syreadditionssalt), eventuelt mono- eller polyhydratiseret, fortyndes i et eller flere bærestoffer og inkorporeres som en enhedsdosis, der kan være en gelatinekapsel eller en tablet.
25 Det virksomme stof er i almindelighed i form af en en hedsdosis på omkring 2 mg til 900 mg i afhængighed af derivatet og det tilsigtede anvendelsesmønster. Totale doser pr. uge vil i almindelighed variere mellem ca. 0,1 og ca. 35 mg/kg.
De omhandlede hidtil ukendte forbindelser kan bruges 30 alene eller i kombination med andre carcinostatiske stoffer, fx forskellige alkyleringsmidler, anti-metaboliter såsom metotrexat, 5-fluoruracil, 6-merkaptopurin, 6-tioguanin, arabinosidet af cytosin, ellipticin og visse antibiotika såsom actinomycin D, daunorubicin, adriamycin og cis-diamino-35 diklorplatin.
Specielt kan flere af de omhandlede vinblastinderivater bruges i kombination.

Nogle eksempler tjener til nærmere belysning af fremgangsmåden ifølge opfindelsen.
Almen metode til fremstilling af N-(desacetyl-O-4-vinblastin- 23-oyl)-aminosyrederivater -3 5 1 g (1,3 x 10 M) 3-dekarbometoxy-0-4-desacetylvinbla- stin-3-karboxhydrazid opløses i 23 ml vandfri metanol og

74 ml IN HC1. Opløsningen afkøles derefter til -10 C og der j tilsættes 207 g natriumnitrit i en portion. j
Blandingen holdes i 10-30 minutter på 0°C. Efter over-10 vågning ved tyndlagskromatografi (t.l.c.) reguleres blandingens pH-værdi til 8,5 ved -10°C ved tilsætning af en mættet opløsning af natriumbikarbonat.
Den alkaliske opløsning ekstraheres ved 0°C med et rumfang metylenklorid der er det samme som opløsningens eget 15 rumfang, det sker ved 0°C og indtil der er konstateret negativ Meyer-reaktion på den vandige fase. De organiske faser forenes, tørres over Na^SO^ og filtreres ved 0°C.
1,43 x 10 (1,1 ækvivalent) af den ønskede aminosyre- ester tilsættes og opløsningen koncentreres under nedsat tryk 20 indtil der er vundet et rumfang på ca. 4 ml. Denne opløsning holdes på 4°C i 24-48 timer. Reaktionens fremadskriden overvåges ved tyndlagskromatografi. Opløsningsmidlet fjernes fuldstændigt og der vindes 1,05 g tør forbindelse der viser sig som en enkelt plet ved t.l.c.
25 Rensning
Det ovennævnte tørrede produkt renses i en silikagel-kolonne idet elueringsmidlet er en blanding af æter og ammo-niakmættet metanol, 96% + 4%.
Der opsamles fraktioner på 10 ml og de afprøves ved 30 t.l.c. Som fremkaldelsesmiddel anvendtes ninhydrin (amino syrer) eller cerisalt (alkaloider).
Når alle aminosyrerne har undergået eluering (ninhydrin-) bruges der yderligere 100 ml af elueringsmidlet før der ændres til en blanding af 92% og 8% af opløsningsmidlerne.

Identiske fraktioner forenes, inddampes til tørhed (rotavapor), opløses i me ty lenklori d, tørres over Na2SO^, filtreres og inddampes til tørhed. Det dannede skum er amino-syrederivatet af det dimere alkaloid, hvis fysisk-kemiske 5 egenskaber vil blive bestemt på portioner deraf. Det tilbageværende stof omdannes direkte til sulfat. Udbytter af basen af de forskellige forbindelser er kun retningsgivende og kan forbedres.
Fremstilling af sulfatet 10 Den amorfe fase (skum) opløses i den 20-dobbelte vægt mængde ætanol.
Til denne opløsning sættes der meget langsomt og under hurtig omrøring 2 ækvivalenter svovlsyre som en opløsning bestående af 2% svovlsyre og 98% vandig ætanol (0,484 ækviva- 15 lenter/liter) . Efter at der er tilsat 2 ækvivalenter samt efter 1/2 times omrøring sker der koncentrering under nedsat tryk. Ved tilsætning af svovlsyreæter under hurtig omrøring udfælder sulfatet af den oprindelige forbindelse sig. Efter filtrering og tørring under nedsat tryk ved 10°C vindes det 20 ønskede sulfatderivat, rede til brug.
Nummereringen ved beskrivelsen af NMR-spektrene i de følgende eksempler er inspireret af forslag der er givet af Le Men og Taylor for derivater af aspidospermidintypen (Ex-perientia 21, 508, 1965).
25 Eksempel 1
Vinblastin-base (VLB)
Til en opløsning af 1,5 g VLB-sulfat (1,65 x 10 ^M) i 15 ml destilleret vand sættes der successivt under kraftig omrøring 15 ml metylenklorid og 1,5 ml koncentreret ammoniak.
30 Efter 5 minutter dekanteres blandingen og den vandige fase ekstraheres yderligere med 3 x 15 ml metylenklorid. De forenede organiske faser vaskes med 2 x 40 ml deioniseret vand, tørres over Na2SO^ og inddampes til tørhed på rotavapor. Der vindes 1,32 g VLB-base (99%).

3-Dekarbometoxy-Q-4-desacetylvinblastin-3-karbohydrazid (VLH) -3
Til en opløsning af 1 g vinblastin-base (1,23 x 10 M), opløst i 7 ml vandfri ætanol, sættes der 14 ml vandfrit hydra-5 zin og 7 ml vandfri ætanol. Reaktionsblandingen opvarmes der efter til 60°C i 24 timer.
Efter afkøling tilsættes der 28 ml saltmættet vand og der foretages ekstraktion med samme rumfang metylenklorid indtil der konstateres negativ Meyer-reaktion på den syrnede 10 vandige fase. De forenede organiske faser tørres over MgSO^ og inddampes til tørhed under nedsat tryk.
Hydrazidet, der vindes i et udbytte på 88% (0,706 g) gav en enkelt plet ved t.l.c.
Eksempel 2 15 Ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-leucinat (VLE) -4
Til en opløsning af 275 mg (3,58 x 10 M) O-4-desace-tylvinblastinhydrazid i 7 ml vandfri metanol og 20,5 ml IN HCl tilsættes der efter afkøling af opløsningen til 0°C 58 mg natriumnitrit. Efter omrøring i 25 minutter re-20 guleres pH til 8,5 ved tilsætning af en passende mængde 5%s opløsning af NaHCO^. Det dannede azid ekstraheres med metylenklorid. Den organiske fase tørres over MgSO^ og filtreres.
Der tilsættes 68,4 mg ætyl-L-leucinat (4,29 x 10 ^M) og opløsningen koncentreres under nedsat tryk til ca. 4 ml. Opløshin-25 gen henstår i køleskab i 24 timer.
Når reaktionen er fuldført tilsættes der 50 ml metylen-klorid og opløsningen vaskes nogle gange med rumfang af de-ioniseret vand som er lig med opløsningens rumfang, og en gang med en NaCl-mættet opløsning.
30 De forenede organiske faser tørres over MgSO^, filtre res og inddampes til tørhed.
På denne måde vindes 300 mg råprodukt hvortil der sættes 0,035 g i opløsning i 1 ml vandfri metanol.
Det dannede salt udfældes med æter og bundfaldet vaskes 35 10 gange med 50 ml vandfri svovlsyreæter.
På denne måde vindes der 183 mg (57%) produkt som er i det væsentlige tørt og som ikke indeholder noget ætylleucinat.

Efter frigivelse af basen, kan den underkastes silika-gel-kromatografi (10 g SiC>2) og eluering med 5.Q ml æter/am-moniakmættet metanol (92/8) og derefter 250 ml æter/ammoniak-mættet metanol (85/15). VLE vindes i det andet eluat. På den-5 ne måde opsamles der 49% VLE, dvs. 152 mg base.

Eksempel 23 Ætyl-N-(vinblastin-23-oyl)-L-vallnat 100 mg ætyl-N-(0-4-desacetylvinblastin-23-oyl-L-vali-25 nat omsættes med en blanding af 2,5 ml pyridin og 2,5 ml eddikesyreanhydrid under en inert atmosfære og omrøring i 24 timer. Efter tilsætning af metanol og koncentrering under nedsat tryk opløses produktet i diklormetan og vaskes med vand mættet med NaCl. Den organiske fase tørres over Na2SO^ 30 og koncentreres under nedsat tryk. Produktet renses ved t.l.c. med anvendelse som elueringsmiddel af en blanding af isopro-panol/ætylacetat/cyklohexan 40:20:40. På denne måde vindes der 29 mg ætyl-N-(vinblastin-23-oyl)-L-valinat.

1. Analogifremgangsmåde til fremstilling af vinblastinderi-vater med den almene formel f^-f} ^ co9ch_ II rOO CH3oJO^O—-0R3 J I f\ R O CH3 / ' i II % 4 HO |-(-NH-CH-C-)nOR 2 5 hvor R er et hydrogenatom eller en C2-9 alkanoylgruppe, fortrinsvis en acetylgruppe, R^ er et hydrogenatom, en lige eller grenet C^_g alkylgruppe, hydroxy-C-^_g-alkylgruppe, aminohydro-xy-C^_g-alkylgruppe, karboxy-C^_g-alkylgruppe, amido-C^_g-al-kylgruppe, amino-C1_g-alkylgruppe, guanidin-C1_g-alkylgruppe, 10 tiol-C^_g-alkylgruppe, cysteinyl-metylgruppe, metyltioætyl-gruppe, benzylgruppe, hydroxybenzylgruppe eller en gruppe CE0~ CH„- -S 2 N_S' 2 I I eller II | Η H 3 eller hvor R sammen med det tilknyttede kulstofatom og amido-nitrogenatomet danner en azol- eller hydroxyazolring, n er 15 et helt tal 1-6 og R4 er en lige eller grenet C^_g alkylgruppe eller en benzylgruppe, eller farmaceutisk acceptable syre- DK 160614 B 38 : additionssalte deraf med mineralske eller organiske syrer, kendetegnet ved at man omsætter vinblastin med vandfrit hydrazin til dannelse af 0-4-desacetyl-3-vinblastin-3-karboxhydrazid, omsætter det således dannede hydrazid med et 5 nitroseringsmiddel i surt medium til dannelse af det tilsvarende acylazid og omsætter acylazidet med 1-5 ækvivalenter tilsvarende aminosyreester eller peptid, hvor eventuelle reaktive sidegrupper om nødvendigt er beskyttet, hvorpå sådanne beskyttelsesgrupper fjernes og det fremkomne produkt om øn-10 sket omdannes til et syreadditionssalt deraf.
2. Fremgangsmåde ifølge krav 1, kendetegnet ved at azidet af 0-4-desacetylvinblastin-23-oesyre omsættes med 1-5 ækvivalenter af den ønskede aminosyreester eller peptid, hvilket peptid indeholder 2-6 aminosyrer,
3. Fremgangsmåde ifølge krav 1 eller 2 til fremstilling af ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-tryptofanat, kendetegnet ved at 0-4-desacetyl-3-dekarbometoxy-vinblastin-3-karboxhydrazid omsættes med et nitroseringsmiddel i surt medium, hvorefter det på denne måde vundne acyl-20 azid omsættes med ætyl-L-tryptofanat.
4. Fremgangsmåde ifølge et hvilket som helst af kravene 1-3 til fremstilling af ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-tryptofanat, kendetegnet ved at azidet af 0-4-desacetylvinblastin-23-oesyre omsættes med 1-5 ækvivalenter 25 ætyl-L-tryptofanat.
5. Fremgangsmåde ifølge krav 3 eller 4, kendetegnet ved at den dannede forbindelse omsættes med svovlsyre til fremstilling af ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-tryptofanatsulfat, hvor sulfatmængden er ækvimolær med 3. tryptofanatmængden.
6. Fremgangsmåde ifølge krav 1, kendetegnet ved at den bruges til fremstilling af ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-leucinat, metyl-N-(0-4-desacetylvinblastin-23-oyl)-L-leucinat, 35 n-butyl-N-(0-4-desacetylvinblastin-23-oyl)-L-leucinat, octyl-N-(0-4-desacetylvinblastin-23-oyl)-L-leucinat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-D-leucinat, DK 160614 B ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-glutamat, ætyl-N-(0-4-desacetyl-vinblastin-23-oyl)-L-serinat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-tryptofanat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-D-tryptofanat, 5 metyl-N-(0-4-desacetylvinblastin-23-oyl)-L-tryptofanat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-alaninat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-fenylalaninat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-isoleucinat, metyl-N-(0-4-desacetylvinblastin-23-oyl)-L-isoleucinat, 10 ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-valinat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-tyrosinat, ætyl-N-(0-4-desacetylvinblastin-23-oyl)-L-valinyl-L-trypto-fanat, ætyl-N-(vinblastin-23-oyl)-L-valinat 15 eller farmaceutisk acceptable syreadditionssalte deraf med mineralske eller organiske syrer.
