Den foreliggende opfindelse angår et monoklonalt anti-humant IgG-antistof, HG2-25, som angivet i krav 1, samt en fremgangsmåde til fremstilling af dette antistof, som angivet i krav 2.
5 Ved allergiske sygdomme forårsaget af antigen-antistofreaktioner har undersøgelsen og identifikationen af allergenet gjort det muligt at diagnosticere og klassificere sygdommene og lægge en terapeutisk plan.
Det monoklonale antistof ifølge opfindelsen anvendes som et diagnostisk reagens ved identifikation af et allergen for allergiske sygdomme, såsom 10 eksogen bronkial astma, atopisk dermatitis, immunkomplekssygdomme o.s.v., og til belysning af mekanismen for forekomsten af de allergiske sygdomme.
Γnogle få årtier har man erkendt betydningen af måling af immunoglobulin G (IgG) over for et allergen i humant serum (J.W. Younginger et 15 al., J. Cl i n. Invest. 52, 1268 (1973), A.K. Sobotka et al, J. Immunol.
117, 84 (1976)). I den seneste tid er en radioallergosorbenttest (herefter omtalt som RAST) blevet anvendt til måling af specifikt IgE over for et allergen i humant serum (N.F. Adkinson, Jr., American Society for Microbiology, Washington, D.C., p590-602(1976)). Anvendelsen af RAST er 20 imidlertid mislykket ved en måling af humant allergenspecifikt IgG
bortset fra visse tilfælde, der er beskrevet i nogle få rapporter (Shimizu et al., J. Immunol. Methods, 19, 317(1978)). Det formodes, at en hovedgrund til det mislykkede resultat er, at ved RAST er bindingsniveauet for et ikke-specifikt antistof i normalt serum højere end for et 25 specifikt. Det er lykkedes for Hamilton et al. at reducere det ikke-spe-cifikke bindingsniveau ved at anvende 125I-protein A adskilt fra Staphylococcus aureus i stedet for kanin anti-humant IgG-antistof (J.
Immunol. 122, 1073(1979)). Det fra Staphylococcus aureus adskilte protein A bindes specifikt til Fc-stedet på humant IgG af underklasse 1, 2 30 og 4, men ikke humant IgG3. Selv om protein A bindes til IgAx og IgA2, der hører til underklassen af Immunoglobulin A (IgA), er det sandsynligt, at der ikke er nogen speciel relation mellem IgA-underklas-se og protein A (Buruda et al. J. Immunol., Vol. 123, 1457(1979)). Jo-hanson og Inganas har bemærket, at Immunoglobulin E (herefter omtalt som 35 IgE) adskilt fra serumet, der indeholder polyklonalt IgE, har en affinitet for protein A, men dets bindingssted er ikke Fc fragmentet, som er IgG's bindingssted (Immunological Rev. 41, 248 (1978)).
Opfindelsens formål er derfor at udvikle en ny fremgangsmåde til DK 166503 B1 2 måling af humant allergenspecifikt IgG ved hjælp af det monoklonale an-ti-humant IgG-antistof, der kan bindes til specifikke antigeniske determinanter for humant IgG med samme affinitet. Endvidere kan en stor mængde monoklonalt antistof produceres i bughulen på mus. Allergenspecifikt 5 humant IgG vil kunne måles med det monoklonale antistof, som fortrinsvis bindes til det allergenspecifikke humane IgG, men bindes mindre til ik-ke-specifikt humant IgG.
Det monoklonale anti-humant IgG-antistof, der tilvejebringes ved opfindelsen, har følgende karakteristika: 10 1) Det genkender CH2-området i humant IgG, 2) det har en høj affinitet for humant IgG og IgG-underklasserne,
IgGi, IgG2l IgG3 og IgG4-, 3) det bindes mere præferentielt til det allergenspecifikke IgG end til det ikke-specifikke IgG, 15 4) det har næsten ingen affinitet for det frie humane IgG i form af en opløsning, og det kan anvendes til måling af allergen-specifik IgG og immunkompleks ved RAST eller ELISA (enzymlinked immunosorbent assay) og til belysning af mekanismen for forekomsten af allergiske sygdomme ved hjælp af oven-20 stående karakteristika. Det monoklonale antistof har lille ikke-specifik affinitet i normalt serum og kan også anvendes til måling af det allergenspecifikke IgG, hvor de kommercielle allergenskiver eller allergen-fikserede bærere (f.eks. rørbelægninger, perler og lignende), der fremstilles til måling af IgE, kan anvendes.

Et dyr, f.eks. en mus, en rotte osv., immuniseres ved intraperito-neal administrering af immunoglobulin fra humant serum sammen med komplet Freund's adjuvans. Efter ca. 3 uger administreres en yderligere DK 166503 B1 3 mængde immunoglubolin fra humant serum (en mængde til yderligere stimulering) intraperitonealt sammen med en alunadjuvans. Efter 3 dage fjernes milten og omdannes til en cellesuspension for at frembringe antistofproducerende celler til cellefusion.
5 2. Fremstilling af Myelomaceller.
En myelomacellelinie, der mangler hypoxanthin»guanin*phosphoribo-syltransferase, f.eks. myelomacelle P3-NS-1/1-AG-4-1 (herefter omtalt som NS-l/Ag) af mus, BALB/C (Proc. Natl. Acad. Sci., U.S.A., 76, 4061 10 (1979)) holdes i et medium f.eks. RPMI-1640 medium indeholdende 15% kal- vefosterserum (FCS). Væksten af NS-l/Ag inhiberes i et hypoxanthin*ami-nopterin-thymidinmedium (HAT medium).
3. Fremstilling af hybridoma.
15 De under 1. fremstillede miltceller og de under 2. fremstillede myelomaceller NS-l/Ag sammensmeltes i nærvær af polyethylenglykol (PEG) ved fremgangsmåden ifølge Oi og Herzenberg (Selected Methods in Cellular Immunology, 351-372 (1980), San Francisco, W, H, Freeman and Co.). De resulterende hybridceller suspenderes i HAT mediet og inokuleres i hver 20 fordybning i vævskulturplader. På dette punkt er det hensigtsmæssigt at tilsætte en lille mængde af thymocyterne eller peritonealperkolerings-cellerne fra mus, BALB/C, der forefandtes som fødeceller. På 1., 2., 3., 5., 8. og 11. dagen efter inkubationens begyndelse erstattes halvdelen af HAT-mediet med nyt medium og 14, 15 og 16 dage efter med et medium, 25 der kun indeholder hypoxanthin*thymidin (HT medium).
4. Undersøgelse af hybridomer, der producerer anti-humant immunoglobulin antistof.
(1) Undersøgelse ved passiv hæmagglutinering under anvendelse af 30 fåreerythrocyter sensi bi 1iseretmed humant immunoglobulin.
Fåreerythrocyter vaskes med chrom (III) chlorid og sensibiliseres med humant IgG ved chromchloridmetoden (E.R. Gold og Η. H. Fudenberg, J. Immunol., 99, 859 (1976)). På samme måde tilvejebringes fåreerythrocyter sensibiliseret med Fc-fragment og F(ab')2 fra humant immunoglobulin og 35 humant myelomaprotein. Antistofaktivitet i supernatanten fra dyrkningsmediet undersøges med ovenstående sens i bil serede erythrocyter ved hjælp af passiv hæmagglutinering.
(2) Undersøgelse ved hjælp af ELISA

Humant immunoglobulin fikseres på en polystyrenplade med 0,05 M carbonatpuffer (pH 9,1) og gives anledning til at reagere med en supernatant fra hybridomamediet, efter at de resterende bindingssteder på polystyrenpladen er mættet med okseserumalbumin. Efter vask bringes pladen 5 til at reagere med antimuseimmunoglobul i nantistof, som er konjugeret med en peroxidase og ikke har nogen krydsaffinitet i forhold til humant immunoglobulin, og gives efter vask lejlighed til at reagere med substratet (hydrogenperoxid). Reaktionen afbrydes med et natriumazid, hvorefter absorbansen måles.
10 5. Kloning og udvælgelse af hybridomer
De hybridomer, der ved ovestående procedure 4. konstateres at have antistof-aktivitet, klones yderligere ved hjælp af grænsefortyndingsme-toden, der er velkendt inden for området. Antistofaktivi teten i en su-15 pernatant fra mediet, hvori klonen har vokset, bestemmes på samme måde som ved procedure 4. ovenfor, og en klon med høj antistofaktivitet udvælges.
6. Fremstilling af antirhumant immunoglobulin (IgG etc.) 20 Monoklonalt antistof
Den ved procedure 5. ovenfor valgte klon inkuberes i et passende medium f.eks. RPMI-1640-medium indeholdende 15% FCS, indtil cellekoncentrationen når maksimum, eller den udvalgte klon transplanteres i bughulen på mus, BALB/C, der forud er behandlet med pristan, og efter 3 uger 25 opsamles den akkumulerede ascites.
7. Isolering og rensning af det monoklonale antistof.
Det monoklonale antistof, hvorfra cellerne fjernes ved centrifugering efter inkubationen, kan let isoleres fra ascites eller supernatan-30 ten og renses ved hjælp af velkendte metoder f.eks. affinitetskromatografi med protein A Sepharose®.
Eksempel (1) Fremstilling af anti-humant IgG-antistof-producerende hybridoma 35 Til BALB/C mus (8-10 uger gamle) administreredes peritonealt 200 /ig
IgG fra humant serum (100 /il fysiologisk saltvand) sammen med 100 /il komplet Freund's adjuvant med henblik på immunisering. 21 dage efter administreredes 20 /ig humant IgG peritonealt som et alunsediment. 3 dage DK 166503 B1 5 efter fjernedes milten for at fremstille cellesuspensionen i RPMI-me-diet. RPMI-mediet (10 ml) indeholdende 108 miltceller blandedes med en suspension af 5 x 107 NS-1 myelomaceller i 10 ml RPMI-medium, og blandingen centrifugeredes ved 450 g i 10 minutter, og supernatanten 5 fjernedes. Derefter sattes 1 ml RPMI medium (RPMI-1640) indeholdende 50% PEG 4000 (Merck & Co.) langsomt til cellepillen under omrøring. Efter omrøring i yderligere 1 minut tilsattes 1 ml RPMI-opløsning i løbet af 1 minut. Efter tilsætning af yderligere 1 ml på samme måde og derefter 7 ml af samme opløsning i løbet af 3 minutter fjernedes supernatanten ved 10 centrifugering ved 400 g i 10 minutter. De resulterende celler suspenderedes i 20 ml RPMI-1640 medium indeholdende 15% kalvefosterserum (FCS), hvoraf 100 /il inokuleredes i hver fordybning i to vævskulturplader (Corning, 96-fordybninger) og inkuberedes ved 37°C i nærværelse af 7% carbondioxidgas.
15 Halvdelen af hvert medium erstattedes med HAT medium (RPMI-1640-me- dium suppleret med 10% FCS, 4 x 107 M aminopterin, 1,6 x 10 5 M thy-midin og 1 x 10~4 M hypoxanthin) 1, 2, 3, 5 og 8 dage efter, og dyrkning fortsattes. På dyrkningens 11., 13. og 14. dag erstattedes halvdelen af mediet med HT-medium (RPMI-medium indeholdende 10% FCS, 1,6 x 20 10’5 M thymidin og 1 x 10’4 M hypoxanthin).
Hybridomakolonier kom til syne i alle fordybninger 16 dage efter sammensmeltningsprocedurens start. De resulterende hybridomaer inkuberedes i RPMI-I640-medium indeholdende 15% FCS, og supernatanten undersøgtes som følger, hvis der var produceret specifikke antistoffer.
25 I hver fordybning i U-bunds-vævskul turpi ader inokuleres 5 /ti super- natantopløsningen, som skal undersøges, og 50 μ! phosphatforpufferet saltvand (PBS) indeholdende 10% FCS. Til hver fordybning i U-bundspla-derne sættes 50 μΐ 1% i suspension af fåreerythrocyter sensibiliseret med humant IgG, og der omrøres moderat.
30 Efter henstand ved stuetemperatur i 2 timer vurderes det om hæmag- glutinering har fundet sted. Resultaterne vises i tabel I.

Anti-humant IgG-antistof-producerende hybridoma tilvejebragt under (1) suspenderedes i RPMI-1640 indeholdende 15% FCS, således at koncen-5 trationen var 5 celler per ml, og 100 μΐ af suspensionen per fordybning inokuleredes i 40 fordybninger i vævskulturplader (Cornig, 96 fordybninger). Den resulterende suspension fortyndedes til 1 celle per ml og inokuleredes i 32 fordybninger og fortyndedes yderligere til 0,5 celler per ml og inokuleredes i 24 fordybninger. 5 dage efter sattes 100 /il 10 RPMI-medium indeholdende 15% FCS til hver fordybning. Eksempelvis viste der sig for klon HG2-25's vedkommende hybridomakloner i 2 af 24 fordybninger (0,5 celler/ml) og 4 af 32 fordybninger (1 celle/ml) 10 dage efter dyrkningens start. Evnen til at producere antistof"i supernatanten for hver klon undersøgtes.
15 Resultat: Der iagttoges evne til at producere antistof i alle kloner.
(3) Fremstilling af anti-humant IgG-antistof.
En suspension af 5 x 106 celler af den ved ovenstående procedure tilvejebragte anti-humant IgG-antistofproducerende hybridoma i 0,5 ml 20 fysiologisk saltvand transplanteredes til bughulen på BALB/C mus, som på forhånd var behandlet med 0,5 ml pristan. 3 uger senere bekræftedes det, at musenes bug var udspilet, og acites opsamledes ved sugning, og cellerne fjernedes ved centrifugering.
25 (4) Isolering og rensning af anti-humant IgG-antistof.
Acites opsamlet under ovenstående trin udsaltedes med 45% vandig mættet ammoniumsulfat, hvorved der opnåedes et præcipitat, som opløstes i 0,1 M phosphatpuffer (pH 8,0) og dialyseredes med samme puffer. 20 mg gammafraktion opnået fra ascites ved udsaltning opløstes i 2 ml 0,1 M 30 phosphatpuffer (pH 8,0), og opløsningen adsorberedes på en søjle af protein A Sepharose® (Pharmacia AB). Søjlen elueredes med ca. 50 ml 0,1 M phosphatpuffer (pH 8,0), hvorved urenheder opnåedes, og derefter med ca.
100 ml 0,1 M citratpuffer (pH 5,0), hvorved det monoklonale anti-humant IgG-antistof, HG2-25, opnåedes.

Forsøg
Som vist i det følgende forsøg har det monoklonale anti-humant IgG-antistof ifølge opfindelsen, hvis affinitet for humant immunoglobolin DK 166503 B1 7 undersøgtes ved hjælp af agglutinering, høj affinitet for IgG og IgG-underklasserne, IgGu IgG2> IgG3 og IgG4. Det aggregerer imidlertid ikke humant IgM, IgA eller fåreerythrocyter overtrukket med humant serumalbumin. Det bindes til Fc fragmentet af IgG, men kun lidt til 5 F(ab')-fragmentet. Isotypen undersøgt ved hjælp af immunpræcipitering er '9G2a·
Det monoklonale anti-humant IgG-antistof mærkedes med radioaktivt i od (12 51), og det epitopi ske område bestemtes ved anvendelse af humant IgG-fragmenternes inhiberingsevne. Som resultat bekræftedes det, 10 at bindingen af HG2-25 inhiberedes af Fc-fragmentet, men ikke af pFc'.
Dette betyder at HG2-25 genkender CH2-området i humant IgG.
Forsøg 1
Karakterisering af det monoklonale antistof HG2-25 15 Supernatanten fra mediet indeholdende HG2-25-producerende hybridoma fortyndes med phosphatforpufferet saltvand indeholdende 10% FCS ved tofold fortynding, og 50 /il af hver anbragtes i hver fordybning i U-bunds-mikrotiterplader. Til hver fordybning sattes 50 μΐ 1% i suspension af fåreerythrocyter sensibiliseret med humant IgG. Der omrørtes med en pla-20 deblander, og reaktion fik lov til at finde sted ved stuetemperatur i to timer, hvorefter agglutineringsbilledet vurderedes. På samme måde udførtes prøven med fåreerythrocyter sensibiliseret med Fc-fragment og F(ab')2 af humant IgG og med humant myelomaprotein (IgG og dets underklasse, nemlig HCDj, IgG2, HCD3, IgG3 og IgGJ, og agglutine-25 ringsbilledet vurderedes. Som resultat bekræftedes agglutineringsbilledet, der gav formodning om høj affinitet med IgG, Fc-fragment og HCD1}
IgG2, HCD3 og IgG4 hørende til underklassen. Tabel 2 viser hæmag-glutineringstiteren.

Forsøg 2 35 Mærkning af det monoklonale antistof HG2-25 med iod (iodogenmetode) I et glasrør anbragtes 50 μΐ opløsning af 40 μΐ/ml 1,3,4,6-tetra-chlor-3a,6a-diphenylglycoluryl (I0D0-GEN, Pierce Chemical Co.) i dichlormethan, og der tørredes ved stuetemperatur under nitrogenatmosfæ- DK 166503 Bl 9 re. Til røret sattes en opløsning af HG2-25 antistof (30 /ig/15 μΐ) opløst i 15 μΐ 0,1 M phosphatpuffer (pH 7,4), hvorefter der tilsattes 0,5 mCi/5 μΐ radioaktivt natriumiodid (Na125I), og blandingen rystedes moderat i 15 minutter ved stuetemperatur. Den resulterende reaktions-5 blanding ledtes igennem en søjle (1,6 x 50 cm) af Sephadex® G-25 (Pharmacia AB),, og elueredes med phosphatforpufferet saltvand. Den først elu-erede fraktion opsamledes og gav 1Z5I-mærket monoklonalt antistof HG2-25. Kromatogrammet (fig. 1) på en søjle (1,6 x 56 cm) af Sepharose® 6B indikerer, at produktet er rent, indeholder næsten ikke agglutinat.

Forsøg 3
Genkendelsessted for monoklonalt antistof HG2-25 I hver fordybning i en mikrotiterplade fremstillet af polystyren (Immulon 2 plade, Dynatech Co.) anbragtes 100 μΐ IgG opløsning (2 Mg/ml) 15 i 0,05 M carbonatpuffer (pH 9,1), og pladen henstilledes natten over ved 4°C. Derefter vaskedes fordybningerne 3 gange med 0,1 M phosphatpuffer-opløsning (pH 7,4) indeholdende 0,05% Tween® 20 og 0,01% natriumazid og blokeredes med 0,1 M phosphatpufferopløsning indeholdende 1 mg/ml humant serumalbumin. Efter vask med samme pufferopløsning anbragtes hver opløs-20 ning af humant IgG, F(ab')2, Fc, tlmFc og pF'c (5 μg/ml i 0,1 M phosphatpufferopløsning) i ovenstående plade sammen med 125I-mærket HG2-25-antistof, og pladen henstilledes ved stuetemperatur i 16 timer og vaskedes så på samme måde som nævnt ovenfor. Det mærkede antistof HG2-25, der blev bundet specifikt til IgG fikseret på væggene, frigjordes 25 fra IgG med 10% eddikesyre, hvis radioaktivitet måltes. Som vist i tabel 3 inhiberes specifik binding af det monoklonale antistof HG2-25 i høj grad (96%) af intakt humant IgG. Endvidere inhiberede Fc-fragmentet HG2-25 99%, mens F(ab')2 og pFc' næsten ikke gav nogen inhibering.

Forsøg 4
Affinitet for humant IgG under forskellige betingelser (1) Affinitet for humant IgG i opløsninger 10 1251 humant IgG mærket ved iodogenmetoden opløstes i 0,1 M phosphatpufferopløsning (pH 7,4) indeholdende 0,1% serumalbumin, 0,05% Tween 20 og 0,01% natriumazid (pufferopløsning B) så den indeholdt 0,04 jug/ml / Til en blanding af 50 μΐ af ovenstående opløsning og 50 μΐ mono-klonalt anti-humant IgG-antistofopløsning (fortyndet med pufferopløsning 15 B til 16 Mg/ml) sattes 50 μΐ 0,1-50 μο/ηιΐ humant IgG-opløsning, og blandingen henstilledes til reaktion i polystyrenrør i 16 timer.
Så tilsattes til 1 ml cyanogenbromid-aktiveret Sepharose®bærer (Pharmacia AB) 5 mg kanin antimuse IgG-antistof med en høj affinitet for IgG2a, en i sotype af det monoklonale anti-humane IgG-antistof, og det 20 fik lov til at reagere til frembringelse af et fikseret anti-muse Ig^g-antistof. Til hvert af ovenstående polystyrenrør sattes 0,1 ml af dette fikserede andet antistof (tilstrækkelig mængde til at binde alt det anvendte monoklonale anti-humant IgG-antistof), og det fik lov til at reagere 1 time ved stuetemperatur under omrøring. Efter vask 3 gange med 2 25 ml af pufferopløsning B måltes radioaktiviteten med en gammatæller. Affinitetkonstanten bestemtes ved Scatchardmetoden.
Resultat: Den ved ovenstående forsøg bestemte affinitetskonstant er 2,7 x 107. Fig. 2 viser en Scatchardafbildning.
30 (2) Affinitet med humant IgG, der specifikt bindes til aller- genskiven
Til en honningbigiftallergenskive af IgG · RAST (Phadebas IgG • RAST, Pharmacia AB) sattes 50 μΐ standardopløsning A indeholdende 5U/ml honningbigift-specifikt IgG, og arrangementet heristilledes i 3 ti-35 mer. 50 μΐ 1251-mærket HG2-25, hvis koncentration ændredes fra 0,2 μ Ci/ml til 10 μ Ci/ml, tilsattes, og arrangementet henstilledes ved 25°C i 16-18 timer. Efter vask 3 gange med samme vaskeopløsning måltes radioaktiviteten.

Resultat: Den ved ovenstående forsøg bestemte affinitetskonstant var 3,5 x 109. Fig. 3 viser en Scatchardafbildning.
(3) Affinitet for humant IgG, der ikke bindes specifikt til 5 mi krotiterplade I hver fordybning i mi kroti terp!ader fremstillet af polystyren (Immulon 2 plade, Dynatech Co.) anbragtes 50 μΐ 0,25 M carbonatpufferop-løsning (pH 9,1), hvori koncentrationen af humant IgG var 2 /ig/ml. Disse plader henstilledes ved 37°C i 2 timer, vaskedes med 0,1 phosphatpuffer 10 (pH 7,4) indeholdende 0,1% serumalbumin, 0,05% Tween 20 og 0,01% na-triumazid (pufferopløsning A) og henstilledes i pufferopløsning A ved 37°C i 1 time. Så sattes efter vask 2 gange med pufferopløsning A 50 μΐ 125I-mærket HG2-25 antistofopløsning (0,003-3 /iCi/ml) til hver fordybning. Efter henstand ved stuetemperatur i 16 timer vaskedes disse 15 plader 2 gange med pufferopløsning A. 125I-mærket HG2-25 antistof, der var bundet til humant IgG på pladerne, fjernedes fra pladerne med 10% eddikesyre, overførtes til rør for måling, og radioaktiviteten mål tes. Affinitetskonstanten (K) beregnedes ud fra måleresultaterne ved Scatchardmetoden og bekræftedes at være K= 1,5 x 109M 1.
Claims (5)
1. Monoklonalt anti-humant IgG-antistof, HG2-25, karakteriseret ved, at det 5 a) genkender CH2-området i humant IgG, b) udviser høj affinitet for humant IgG og IgG-underklasserne IgGj, IgG2, IgG3 og IgG4, c) bindes mere præferentielt til det allergenspecifikke IgG end til det ikke-specifikke IgG, og 10 d) i det væsentlige ikke udviser nogen affinitet for frit humant IgG i form af en opløsning.
2. Fremgangsmåde til fremstilling af monoklonal-t antistof, HG2-25, KENDETEGNET ved, AT man sammensmelter myelomacell er og antistofpro- 15 ducerende celler fra et dyr immuniseret med humant IgG til hybridoma-frembringelse, udvælger en anti-humant IgG-antistofproducerende hybridoma, kloner hybridomaen ved aftagende fortyndinger til frembringelse af kloner, som frembringer monoklonale, antistoffer med høj affinitet for humant IgG, multiplicerer klonen og udvinder HG2-25 fra klonen, som li- 20 geledes bindes til IgGl5 IgG2, IgG3 og IgG4 og fortrinsvis til allergen-bindende humant IgG.
3. Fremgangsmåde ifølge krav 2, KENDETEGNET ved, AT klonen transplanteres i bughulen på en mus, som forud er behandlet med pristan, og 25 multipliceres til produktion af det monoklonale antistof, HG2-25, i ascites, og det producerede HG2-25 opsamles og renses.
4. Fremgangsmåde iføgle krav 2, KENDETEGNET ved, AT et råt monoklonalt antistof HG2-25 renses ved affinitetskromatografi under anvend- 30 else af en protein A bindende bærerharpiks.
