Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af hidtil ukendte (thio)pyridoyl-imidazol-2-(thi)oner med den almene formel

10 hvor Q og T hver er et oxygenatom eller et divalent svovlatom, er lavere alkyl, og 15 Ar er usubstitueret pyridyl, idet dog Ar ikke er usubstitueret 3-pyridyl, når R^ er methyl, eller et pharmaceutisk acceptabelt syre/baseadditionssalt deraf.
Disse ifølge opfindelsen fremstillede forbindelser 20 er anvendelige som cardiotoniske midler til behandling af svigtende hjertefunktion.
Den her omhandlede analogifremgangsmåde er karakteriseret ved det i den kendetegnende del af krav 1 angivne.
Anvendt i den foreliggende beskrivelse omfatter ud-25 trykket "lavere alkyl" ligekædet eller forgrenet alkyl med 1-4 carbonatomer, dvs. f.eks. methyl, ethyl, propyl, isopropyl, n-butyl, isobutyl og tert.-butyl.
Udtrykket "halogen" omfatter i den foreliggende beskrivelse fluor, chlor og brom.
30 Anvendt her omfatter udtrykket "pyridyl" 2-, 3- og 4-pyridyl.
Fra GB nr. 2.055.364-A kendes forbindelser med en struktur svarende til formel I og med cardiotonisk virkning. Disse kendte forbindelser afviger dog fra forbindelser med 35 formel I ved substituenten Ar, hvor de kendte forbindelser DK 168627 B1 2 ikke kan indeholde pyridyl; men i stedet indeholder 2-furyl, 2-thienyl eller eventuelt substitueret phenyl.
I forhold til de kendte forbindelser har forbindelser med formel I fremstillet ved fremgangsmåden ifølge opfindel-5 sen den fordel, at de har overlegen cardiotonisk virkning efter oral indgivelse, og at deres vandopløselighed er større ved neutral pH-værdi. Dette er nærmere belyst i nedenstående eksempler 5 og 6.

hvor Ar, Q, T og % er som anført for formlen I, og M er en 20 pharmaceutisk accpetabel alkalimetalion, såsom en natriumeller kaliumion, en jordalkalimetalion, såsom en calciumeller magnesiumion, en overgangsmetalion, såsom en zinkeller jernion, eller en hovedgruppemetalion, såsom en aluminiumion. Almindeligvis er de pharmaceutisk acceptable base-25 additionssalte krystallinske materialer, der er lettere opløselige i vand og forskellige hydrophile opløsningsmidler, og som i sammenligning med deres frie syref omer almindeligvis udviser højere smeltepunkter.
Forbindelserne med fomlen I kan, da Ar er pyridyl, 30 danne pharmaceutisk aktive syreadditionssalte med både uorganiske og organiske syrer. Som eksempler på uorganiske syrer, der danner egnede salte, kan nævnes saltsyre, hydr-ogenbromidsyre, svovlsyre og phosphorsyre og syremetalsalte, såsom natriummonohydrogenorthophosphat og kaliumhydrogen-35 sulfat. Eksempler på organiske syrer, der danner egnede 3 DK 168627 B1 salte, omfatter mono-, di- og tricarboxylsyrer. Eksempler på sådanne syrer er f.eks. eddikesyre, glycolsyre, mælkesyre, pyrodruesyre, malonsyre, ravsyre, glutarsyre, fumarsyre, æblesyre, vinsyre, citronsyre, ascorbinsyre, maleinsyre, 5 hydroxymaleinsyre, benzoesyre, hydroxybenzoesyre, phenyled-dikesyre, kanelsyre, salicylsyre, 2-phenoxybenzoesyre og sulfonsyrer, såsom methansulfonsyre og 2-hydroxyethansulfonsyre. Sådanne salte kan foreligge enten i en hydratiseret eller i en i det væsentlige vandfri form. I reglen er syre- 10 additonssaltene af disse forbindelser krystallinske materialer, der er opløselige i vand og forskellige hydrophile, organiske opløsningsmidler, og som i sammenligning med de frie baseformer i reglen udviser højere smeltepunkter og forøget kemisk stabilitet.
15 Det fremgår af ovennævnte formel I, at forbindelserne, der fremstilles ved fremgangsmåden ifølge opfindelsen, er 1,3-dihydro-4-picolinoyl-2H-imidazol-2-oner, 1,3-dihydro-4-nicotinoyl-2H-imidazol-2-oner og 1,3-dihydro-4-isonicotinoyl-2H-imidazol-2-oner.
20 De foretrukne forbindelser, der fremstilles ved frem gangsmåden ifølge opfindelsen, er sådanne forbindelser med formlen I, hvor Q og T hver er et oxygenatom.
De særlig foretrukne forbindelser er sådanne forbindelser med formlen I, hvori ^ er methyl eller ethyl.
25 De mest foretrukne forbindelser er forbindelser med formlen I, hvor Ar er usubstitueret 4-pyridyl.
Ved fremgangsmådevariant a) fremstilles imidazol-2-on-derivaterne med formlen I, hvor T er et oxygenatom, ved omsætning af en aminodiketon med formlen 30 .•V-« 1 NH2 35 DK 168627 B1 4 hvori og Ar har de for formlen I anførte betydninger, med et cyanat- eller thiocyanatsalt, efter ønske, fortrinsvis natrium- eller kaliumcyanat eller -thiocyanat. Denne omsætning udføres ved at blande ca. 1 molækvivalent af en 5 passende aminoketon med fra 1 til 5 molækvivalenter, fortrinsvis 1 molækvivalent, af et cyanat- eller thiocyanatsalt i et passende opløsningsmiddel. Reaktionen får lov at forløbe i fra 5 minutter til 10 timer afhængig af reaktanterne, opløsningsmidlet og temperaturen, der kan være fra 0 til 10 100°c, fortrinsvis ca. 80°C. Egnede opløsningsmidler til denne reaktion er et hvilket som helst opløsningsmiddel, der ikke er reaktionsdygtigt, såsom vand eller et med vand blandbart opløsningsmiddel, f.eks. en organisk syre, såsom eddikesyre, en alkohol, såsom methanol eller ethyanol, eller 15 en ether, såsom tetrahydrofuran eller p-dioxan. Fortrinsvis blandes eventuelt ikke-vandigt opløsningsmiddel med vand.
Det foretrukne opløsningsmiddel er vand.
Produktet fra denne reaktion kan isoleres ved en hvilken som helst kendt metode, såsom ved omdannelse til 20 det tilsvarende natrium- eller kaliumsalt og genudfældning med carbondioxid eller en mineralsyre, såsom fortyndet saltsyre.
Ved fremgangsmådevariant b) fremstilles forbindelserne med formlen I, hvor Q og T begge er et oxygenatom, ved en 25 Friedel-Crafts-acylering af en l,3-dihydro-2H-imidazol-2-on med formlen

hvor R^ har den for formlen I anførte betydning, idet der som acyleringsmiddel anvendes et usubstitueret pyridoyl-35 halogenid, fortrinsvis et pyridoylchlorid.
Friedel-Crafts-reaktionerne ifølge opfindelsen udføres 5 DK 168627 B1 ved i forvejen at blande ca. 1 molækvivalent af en passende imidazol-2-on med fra 1 molækvivalent til 10 molækvivalenter, fortrinsvis 3-6 molækvivalenter, af en Lewis-syre-katalysator i et passende opløsningsmiddel, f.eks. petroleumsethere, et 5 chloreret carbonhydrid, såsom carbontetrachlorid, ethylen-chlorid, 1,1,2,2-tetrachlorethan, methylenchlorid eller chloroform, en chloreret aromatisk forbindelse, såsom 1,2,4-trichlorbenzen eller o-dichlorbenzen, carbondisulfid eller nitrobenzen. Det foretrukne opløsningsmiddel er 1,1,2,2-10 tetrachlorethan (tetrachlorethan). Fra 1 molækvivalent til 10 molækvivalenter, fortrinsvis ca. 1 molækvivalent, af et pyridoylhalogenid sættes, fortrinsvis dråbevis, til blandingen af imidazol-2-on, Lewis-syre og opløsningsmiddel, og reaktionen får lov at forløbe i fra 1/2 time til 10 timer, 15 fortrinsvis fra 1 time til 5 timer, afhængigt af reaktanterne, opløsningsmidlet og temperaturen, der kan ligge på fra -78 til 15Q°C, fortrinsvis fra 0 til 100'C, særlig foretrukket på ca. 85°C. Den fremkomne pyridoylimidazol-2-on kan isoleres fra reaktionsblandingen ved en hvilken som helst 20 kendt metode, fortrinsvis ved hurtig afkøling af reaktionsblandingen med isvand eller vand efterfulgt af neutralisation med vandigt natriumbicarbonat eller en anden svag base og derefter fjernelse af produktet ved filtrering eller ekstraktion med organiske opløsningsmidler, typisk ethanol, efter-25 fulgt af fjernelse af opløsningsmiddel. Rensning sker typisk ved hjælp af chromatografi på silicagel.
Lewis-syre-katalysatorer, der er egnet til anvendelse i Friedel-Crafts-reaktionerne, der er beskrevet her, er f.eks. et metal, såsom aluminium, cerium, kobber, jern, 30 molybden, wolfram eller zink, en Brønsted-syre, såsom en phosphorsyre, svovlsyre, sulfonsyre eller en hydrohalogen-syre, såsom saltsyre eller hydrogenbromidsyre, halogen-substituerede eddikesyrer, såsom chloreddikesyre eller tri-fluoreddikesyre, eller et metalhalogenid, såsom et borhal-35 ogenid, zinkchlorid, zinkbromid, berylliumchlorid, kobber-chlorid, jern(III)bromid, jern(III)chlorid, kviksølv(II)- DK 168627 Bl 6 chlorid, kviksølv(I)chlorid, antimonbromid, antimonchlorid, titan(IV)bromid, titan(IV)chlorid, titan(III)chlorid, alumi-niumbromid eller fortrinsvis aluminiumchlorid.
Når det ønskes, at T er et divalent svovlatom, omsæt-5 tes den tilsvarende pyridoylimidazol-2-on med formlen I, hvor T er et oxygenatom, med phosphorpentasulfid, P2S5, ved alment kendte metoder. Denne omsætning udføres ved at blande ca. 1 molækvivalent af den pyridoylimidazol-2-on, hvor T er et oxygenatom, med fra 1 til 5 molækvivalenter, fortrinsvis 10 ca. 1 molækvivalent, P2S5 sammen med et passende opløsningsmiddel. Denne reaktion får lov at forløbe i fra 1 til 10 timer, fortrinsvis ca. 5 timer, afhængigt af reaktanten, opløsningsmidlet og temperaturen, der kan ligge fra 25 til ca. 125eC, fortrinsvis på ca. 80°C. Et passende opløsnings-15 middel til denne reaktion er et hvilket som helst ikke-reak-tionsdygtigt opløsningsmiddel, f.eks. tetrahydrofuran, p-dioxan, benzen, toluen eller pyridin. Det foretrukne opløsningsmiddel er toluen.
Alkalimetal-, jordalkalimetal-, overgangsmetal- eller 20 hovedgruppemetalbaseadditionssalte af imidazol-2-onerne, der fremstilles ved fremgangmsåden ifølge opfindelsen, kan fremsitlles ud fra et tilsvarende metalsalt, f.eks. et alk-oxid, såsom natriummethoxid eller kaliumethoxid, eller et hydrid, såsom calciumhydrid. Disse reaktioner kan udføres 25 med eller uden opløsningsmiddel. Egnede opløsningsmidler er f.eks. lavere alkoholer, såsom methanol, ethanol, isopropanol, n-propanol eller n-butanol, eller dimethylformamid (DMF). Imidazol-2-onen og basen får lov at reagere i fra 1 minut til 24 timer, fortrinsvis ca. 1 time, afhængigt at reak-30 tanterne og temperaturen, der kan ligge fra -78*C til 150°C, fortrinsvis fra 0 til 25°C.
Syreadditionssaltene af forbindelserne med formlen I fremstilles ved gængse metoder, såsom ved at behandle en forbindelse med formlen I med en passende, uorganisk eller 35 organisk syre. F.eks. sættes 1-10 molækvivalenter syre til 1 molækvivalent af en forbindelse med formlen I ved en tempe- 7 DK 168627 B1 ratur på fra -5 til 80°C, typisk ved stuetemperatur, og reaktionen får lov at forløbe i 0,1-5 timer. Disse reaktioner kan udføres med eller uden tilsat opløsningsmiddel. Egnede opløsningsmidler er f.eks. lavere alkoholer, såsom methanol, 5 ethanol, isopropanol, n-propanol eller n-butanol, eller vand.
Aminodiketonerne med formlen III kan fremstilles ved reduktion af en passende oxim med formlen 10

15 Disse oximer reduceres ved en hvilken som helst egnet metode, der er almindelig kendt, såsom katalytisk i et surt alkoholisk medium, såsom ethanol-saltsyre, over en ædelmetalkatalysator, såsom palladium på trækul, eller med zink eller tin i en eddikesyre/eddikesyreanhydrid-opløsning.

Forbindelserne med formlen IV samt pyridoylchloriderne er almindelig kendte eller kan let fremstilles ved en teknik, der er analog med standard på området.
35 Forbindelserne med formlen I kan anvendes til behand ling af svigtende hjertefunktion, herunder sådanne lidelser DK 168627 B1 8 som kongestion, uregelmæssig hjertefunktion, svigtende funktion i ventre hjertekammer eller i højre hjertekammer eller til behandling af eventuelle andre tilstande, der kræver en styrkelse af hjertefunktionen med et cardiotonisk middel. I 5 mange henseender besidder de her omhandlede forbindelser en digitalislignende virkning.
Anvendeligheden af forbindelser med formlen I som cardiotoniske midler kan påvises ved at indgive prøveforbindelsen (0,1-10 mg/kg) intravenøst, intraperitonealt, in-10 traduodenalt eller intragastrisk i et passende bærestof til bastardhunde (begge køn). Følgende forsøgsmetoder kan anvendes (anvendt i eksempel 5):
Forsøgshundene bedøves og forberedes ved, at en egnet arterie (f.eks. femoralis eller halspulsåre) og vene (f.eks.
15 femoralis eller ydre halsvene) blotlægges, polyethylenkate-tere fyldt med 0,1% heparin-natrium indsættes, henholdsvis til overvågning af arterieblodtryk og til indgivelse af forbindelser. Brystkassen åbnes ved at gå ind ved venstre 5. interkostale mellemrum, og der dannes en pericardial 20 vugge til understøtning af hjertet. En Walton-Brodie-belast-ningsmåler sys på højre eller venstre hjertekammer til overvågning af myocardiets sammentrækningskraft. En elektromagnetisk strømningssonde kan anbringes omkring roden af opgående aorta for at måle hjerteydelsen minus cononarblod-25 strømmen. Hjertesvigt fremkaldes ved at indgive natriumpento-barbital (20-40 mg/kg) eller propranalol-hydrochlorid (3 mg/kg) i det blod, der gennemstrømmer hjertet. Efter indgivelse af et af disse midler, der nedsætter hjertefunktionen, forøges højre arteries tryk dramatisk, og hjertekapaci-30 teten reduceres stærkt. Reversering af disse virkninger ved hjælp af prøveforbindelsen påviser cardiotonisk aktivitet.
Forbindelserne kan indgives på forskellige måder for at opnå den ønskede virkning. Forbindelserne kan indgives alene eller i form af pharmaceutiske præparater til den 35 patient, der behandles, enten oralt eller parenteralt, dvs. intravenøst eller intramuskulært. Den indgivne mængde for- 9 DK 168627 B1 bindelse varierer afhængigt af patienten, hjertelidelsens sværhedsgrad og indgivelsesmåden.
Ved oral eller parenteral indgivelse ligger den car-diotonisk effektive mængde forbindelse fra 0,01 mg/kg af 5 patientens legemsvægt pr. dag til op til 500 mg/kg af patientens legemsvægt pr. dag og fortrinsvis fra 0,03 op til 200 mg/kg af patientens legemsvægt pr. dag.
Til oral indgivelse kan en doseringsenhed f.eks. indeholde fra 1,5 til 500 mg af det aktive stof, fortrinsvis 10 10-100 mg aktivt stof. Til parenteral indgivelse kan en doseringsenhed f.eks. indeholde fra 5 til 50 mg aktivt stof. Gentagen daglig indgivelse af forbindelserne kan være ønskelig og varierer alt efter patientens tilstand og indgivelsesmåden.
15 Som anvendt her skal udtrykket "patient" betyde varm blodede dyr, f.eks. fugle, såsom kyllinger og kalkuner, og pattedyr, såsom primater, mennesker, får heste, kvæg, svin, hunde, katte, rotter og mus.
Til oral indgivelse kan forbindelserne sammensættes 20 til faste eller flydende præparater, såsom kapsler, piller, tabletter, pastiller, puddere, opløsninger, suspensioner eller emulsioner. De faste enhedsdoseringsformer kan være en kapsel, der kan være af den almindelige gelatinetype indeholdende f.eks. smøremidler og inaktivt fyldstof, såsom 25 lactose, saccharose og majsstivelse. Forbindelserne kan også tabletteres med gængse tabletgrundmaterialer, såsom lactose, saccharose og majsstivelse, kombineret med bindemidler, såsom acacia, majsstivelse eller gelatine, disintegreringsmidler, såsom kartoffelstivelse eller alginsyre, 30 samt et smøremiddel, såsom stearinsyre eller magnesiumstea-rat.
Til partenteral indgivelse kan forbindelserne indgives som injicerbare doser af en opløsning eller suspension af forbindelsen i et fysiologisk acceptabelt fortyndingsmiddel 35 med et pharmaceutisk bærestof, der kan være en steril væske, såsom vand og olier, med eller uden tilsætning af et over- DK 168627 B1 10 fladeaktivt middel eller andre, pharmacutisk acceptable hjælpestoffer. Som eksempler på olier, der kan anvendes i disse præparater, kan nævnes olier af mineralsk, animalsk, vegetabilsk eller syntetisk oprindelse, f.eks. jordnøddeolie, 5 sojabønneolie og mineralolie. I almindelighed foretrækkes vand, saltvandsopløsning, vandig dextrose og beslægtede sukkerforbindelser, ethanol og glycoler, såsom propylenglycol eller polyethylenglycol, som flydende bærestoffer, især til inj icerbare opløsninger.
10 Forbindelserne kan indgives i form af en depotinjek tion eller som implantat, der kan sammensættes på en sådan måde, at der fås protraheret virkning af det aktive stof.
Det aktive stof kan komprimeres i pellets eller små cylindre og implanteres subcutant eller intramuskulært som depotin-15 jektioner eller implantater. Implantater kan være kombineret med inaktive materialer, såsom bionedbrydelige polymere eller syntetiske siliconer, f.eks. "Silastic®", siliconegummi fremstillet af Dow-Corning Corporation.
Opfindelsen vil i det følgende blive nærmere belyst 20 ved hjælp af eksempler.
Eksempel 1 1.3-Dihvdro-4-ethvl-isonicotinovl-2H-imidazol-2-on
Til 3,0 g (27 mmol) l,3-dihydro-4-ethyl-2H-imidazol-25 2-on og 4,74 g (27 mmol) isonicotinoylchlorid-hydrochlorid i 100 ml tetrachlorethan sættes 21,3 g (160 mmol) aluminium-chlorid. Blandingen omrøres ved 85’C i 4 timer og bratkøles med vand. Opløsningen neutraliseres med natriumbicarbonat, og suspensionen filtreres. Remanensen vaskes med ethanol, 30 og de forenede filtrater inddampes til tørhed. Chromatografi på silicagel giver den i overskriften nævnte forbindelse, smp. 260-263eC.

Eksempel 2 1.3- Dihvdro-4-isonicotinovl-5-methvl-2H-imidazol-2-on I 80 ml tetrachlorethan anbringes 3,87 g (39,5 mmol) 1.3- dihydro-4-methyl-2H-imidazol-2-on og 7 g (39,5 mmol) 5 isonicotinoylchlorid-hydrochlorid. 26 g aluminiumchlorid (194 mmol) tilsættes, og blandingen omrøres ved 85°C i 3 timer. Tetrachlorethanet dekanteres fra reaktionsblandingen, og remanensen bratkøles med vand og neutraliseres med na-triumbicarbonat. Suspensionen filtreres, og filtratet ind-10 dampes til tørhed. Chromatografi over silicagel giver den i overskriften nævnte forbindelse, smp. 295-296°C.

15 Eksempel 3 1.3- Dihydro-4-ethvl-5-nicotinovl-2H-imidazol-2-on
Til en godt omrørt blanding af 8,85 g (50 mmol) nico-tinoylchlorid-hydrochlorid og 5,6 g (50 mmol) 1,3-dihydro- 4-ethyl-2H-imidazol-2-on i 100 ml tetrachlorethan sættes 22 20 g (164 mmol) aluminiumchlorid. Blandingen opvarmes ved 85°C i 2 timer og behandles med vand. Opløsningen neutraliseres med natriumbicarbonat, filtreres og inddampes til tørhed. Chromatografi over silicagel giver den i overskriften nævnte forbindelse, smp. 219-221°c.

Eksempel 4
De cardiovaskulære virkninger af l,3-dihydro-4-methyl-5-30 isonicotinoyl-2H-(imidazol)-2-on (MDL 19.214), 1,3-dihydro--4-ethyl-5-isonicotinoyl-2H-imidazol-2-on (MDL 19.205) og 1.3- dihvdro-4-ethvl-5-nicotinovl-2H-imidazol-2-on fMDL 19.198^
Hunde bedøves med natriumpentobarbital (35 mg/kg 35 intravenøst) og gives kunstigt åndedræt. Der indlægges en kanyle i femoralarterien og -venen, henholdsvis til måling DK 168627 B1 12 af systemisk blodtryk og til injicering af præparater. Brystkassen åbnes, og en Walton-Brodie belastningsmålebue sys til venstre hjertekammer til måling af hjertets sammentrækningskraft. Hjertehastigheden måles ud fra et bly(II)-elek-5 trocardiogram. Alle målinger optegnes kontinuerligt på en polygraf. MDL 19.198, MDL 19.205 og MDL 19.214 indgives ved intravenøse injektioner, og de forøger alle hjertets sammentrækningskraft, hjertehastigheden og nedsætter det systemiske blodtryk. Af disse virkninger er forøgelsen af 10 hjertets sammentrækningskraft langt den mest fremherskende virkning af alle forbindelser. Den intravenøse dosis af MDL 19.198, MDL 19.205 og MDL 19.214, der forøger hjertets sammentrækningskraft 30%, er henholdsvis 0,16, 0,04 og 0,13 mg/kg. Som sammenligning er de doser af disse forbindelser, 15 der forøger hjertehastigheden 15%, henholdsvis 3,5, 1,5 og 6 mg/kg. Ved den højeste dosis, der gives, nedsætter MDL 19.198 (3 mg/kg intravenøst), MDL 19.205 (1 mg/kg intravenøst) og MDL 19.214 (1 mg/kg intravenøst) kun det systemiske blodtryk henholdsvis 11, 10 og 4%.

Eksempel 5
Cardiotonisk virkning af forbindelser med formel I sammenlig- net med kendte forbindelser_
Forsøgsmetoden er den ovenfor beskrevne med visse 25 modifikationer. Walton-Brodie-belastningsmåleren sys på venstre hjertekammer på dyr, som er bedøvet under forsøget.
På dyr, som er ved bevidsthed under forsøget, implanteres en tryk-transducer i venstre hjertekammer til optegnelse af dP/dt (hastighedsændringen i udviklet tryk under systole), 30 hvilket er en accepteret indikator for hjertets sammentrækkelighed. Der skal gå mindst 3 døgn mellem de kirurgiske indgreb og forsøget for hunde, som er ved bevidsthed under forsøget.
På forsøgsdagen indsættes der subcutant nåleelektroder 35 på både bedøvede og ikke-bedøvede hunde til optegnelse af et bly(II)-elektrocardiogram, og ud fra dette elektrocar- DK 168627 B1 13 diogram optegnes hjertefrekvensen med en tachograf.
Porsøgsforbindelserne indgives enten intravenøst eller oralt, og ændringerne i enten hjertets sammentræknings-kraft eller hastigheden af trykudvikling i venstre hjertekam-5 mer samt hjertefrekvensen og det gennemsnitlige arterieblodtryk måles. En af forsøgsforbindelsen fremkaldt forøgelse i hjertesammentrækningskraften eller i hastigheden af trykudvikling i venstre hjertekammer eller hjertefrekvensen er, med eller uden en ændring i arterieblodtrykket, et tegn på 10 cardiotonisk aktivitet.

Forbindelserne MDL 19.315, MDL 19.198, MDL 19.214 og MDL 19.205 er alle sammenlignet med den kendte MDL 19.325 5 væsentligt kraftigere til forøgelse af hjertekontraktionsstyrken ved intravenøs indgivelse (tabel I).
Forbindelserne MDL 19.316, MDL 19.315, MDL 19.214 og MDL 19.205 er alle sammenlignet med den kendte MDL 19.325 væsentligt kraftigere til forøgelse af hjertefrekvensen ved 10 intravenøs indgivelse (tabel I).
Forbindelsen MDL 19.205 er sammenlignet med den kendte MDL 17.043 en smule bedre til forøgelse af hjertesammentrækningskraften ved intravenøs indgivelse (tabel II), men væsentligt bedre ved oral indgivelse (tabel III). Oral ind-15 givelse fører også til en væsentlig forøgelse i hjertefrekvensen.
Eksempel 6
Sammenligning af opløselighedsprofiler i afhængighed af pH-20 værdien_
Den kendte forbindelse MDL 17.043 (identificeret i eksempel 5) og forbindelsen MDL 19.205 med formel I (identificeret i eksempel 4) er blevet afprøvet med henblik på en fastlæggelse af deres pH-opløselighedsprofiler. Følgende 25 forsøgsmetoder er blevet anvendt:
Prøverne fremstilles, ved at der i en flintglasampul anbringes MDL 17.043 eller MDL 19.205 i en mængde, som er i overskud i forhold til den mængde, som er nødvendig til opnåelse af en mættet opløsning. En vandig opløsning af 30 natriumhydroxid og/eller saltsyre til opnåelse af den ønskede pH-værdi tilsættes, og ampullerne flammeforsegles. Ampullerne anbringes i et vandbad, som holdes på 25°C, og omrystes i 4 døgn til opnåelse af mættethed. Prøverne fjernes fra badet, og den klare, ovenstående opløsning skilles fra fast, uopløst 35 materiale. Bestemmelse af mængden af forsøgsforbindelse i opløsning sker ved målinger af UV-absorption. På denne måde DK 168627 B1 18 opnås der opløselighedsværdier for MDL 17.043 og MDL 19.205 i passende pH-værdiområder, som dækker alle forskellige, ioniserede tilstande for begge molekyler.
De opnåede resultater er vist i nedenstående tabel IV.

Det foretrækkes at formulere pharmaceutiske præparater, som skal anvendes til intravenøs indgivelse, således at de har en pH-værdi i det normale fysiologiske pH-område 35 fra 6 til 8, medens intravenøse præparater med en pH-værdi, som ligger væsentligt uden for det normale område, anses DK 168627 B1 19 for mindre ønskelige.
På denne baggrund fremgår det af de i tabel IV viste resultater, at den kendte forbindelse MDL 17.043 er praktisk taget uopløselig i vandige medier ved pH-værdien 6,4, og at 5 denne forbindelse, hvis den skal være anvendelig til effektiv intravenøs indgivelse, vil kræve et pharmaceutisk præparat, som har en pH-værdi på mindst ca. 12.
Forbindelsen MDL 19.205 med formel I er ved en pH-værdi på 6,4 ca. 250 gange så opløselig i vand som MDL 10 17.043, og derfor vil det være let at fremstille pharmaceu- tiske præparater, som indeholder MDL 19.205, og som er egnede til effektiv intravenøs indgivelse i det normale fysiologiske pH-område på 6-8.
Opløseligheden i pH-området 6-8 indebærer desuden 15 den yderligere fordel, at forbindelsen MDL 19.205 sammenlignet med MDL 17.043 må forventes at være langt mere tilgængelig for biologisk absorption, hvis den indgives i form af et fast, pharmaceutisk præparat, som indtages oralt.

1. Analogifremgangsmåde til fremstilling af (thio)-pyridoylimidazol-2-(thi)oner med den almene formel 5 T p π I Hν' H 10 hvor Q og T hver er et oxygenatom eller et divalent svovlatom, Ri er lavere alkyl, og Ar er usubstitueret pyridyl, 15 idet dog Ar ikke er er usubstitueret 3-pyridyl, når Ri er methyl, eller et pharmaceutisk acceptabelt syre/baseaddi-tionssalt deraf, kendetegnet ved, at a) en aminoketon med formlen .-M- nh2 25 hvor Ri og Ar har de ovenfor anførte betydninger, omsættes med et cyanat- eller thiocyanatsalt; eller b) en imidazol-2-on med formlen 30 Ri7=C o 35 DK 168627 B1 21 hvor har den for formlen I anførte betydning, underkastes en Friedel-Crafts-acylering med et pyridoylhalogenid i nærvær af en Lewis-syre-katalysator til fremstilling af forbindelser med formel I, hvor Q og T begge er et oxygenatom; hvorefter 5 om ønsket (i) en forbindelse med formel I, hvor T er et oxygenatom, omsættes med phosphorpentasulfid til fremstilling af en forbindelse, hvor T er et divalent svovlatom; og/eller (ii) en forbindelse med formel I omsættes med et 10 basisk metalsalt eller en uorganisk eller organisk syre til fremstilling af et pharmaceutisk acceptabelt salt, hvorefter den dannede (thio)pyridoylimidazol-2-(thi)on isoleres .
2. Analogifremgangsmåde ifølge krav 1, kende-15 tegnet ved, at Q og T i formel I begge er et oxygenatom.
3. Analogifremgangsmåde ifølge krav 1, kendetegnet ved, at Ar i formel I er usubstitueret 2- eller 4-pyridyl, fortrinsvis usubstitueret 4-pyridyl.
4. Analogifremgangsmåde ifølge krav 1, kende tegnet ved, at i formel I er methyl eller ethyl.
5. Analogifremgangsmåde ifølge krav 1, kendetegnet ved, at den fremstillede forbindelse er 1,3-di-hydro-4-isonicotinoyl-5-methyl-2H-imidazol-2-on eller 1,3-di-25 hydro-4-ethyl-5-isonicotinoyl-2H-imidazol-2-on.
