Den foreliggende opfindelse angår hidtil ukendte adenosin-derivater, fremgangsmåder til fremstilling af dem, farmaceutiske præparater indeholdende dem og deres brug ved fremstilling af medicin. Forbindelserne virker som lipolysein-5 hibitorer.

Der findes ikke eksempel på nogen forbindelse hvor -A-X-B er en cycloalkyl- eller cycloalkenylring som er substitueret med 20 en hydroxygruppe, og der er ikke antydning i GB patentskrift nr. 1.143.150 af at nogen af de beskrevne forbindelser har anti-lipolytisk virkning.
Fra tysk offentliggørelsesskrift nr. 2052596 kendes ligeledes 2-chlor-adenosiner, hvis 6-aminogruppe er substitueret med DK 170894 B1 2 cyclopentyl eller cyclohexyl, med antilipolytisk og anti-hyperlipidæmisk virkning.
Der er nu fundet en hidtil ukendt gruppe af N(6)-monosubstituerede adenosinderivater som afviger strukturelt fra de 5 tidligere beskrevne ved bl.a. ikke at være 2-halogenerede, og som virker signifikant mere selektivt som lipolyseinhibitorer end de kendte forbindelser.

Solvaterne kan fx være hydrater.
En foretrukken klasse af forbindelser med formel I er sådanne^ 10 hvor X er et hydrogenatom.
En anden foretrukken klasse af forbindelser med formel I er sådanne, hvor gruppen R er en cycloalkyl- eller cycloalkylen-ring indeholdende 5 eller 6 kulstofatomer og substitueret med en hydroxygruppe i enhver tilgængelig stilling. Ringen kan 15 eventuelt være substitueret med en C1_3alkylgruppe (fx methyl eller ethyl). R er især en cycloalkylring indeholdende 5 eller 6 kulstofatomer og substitueret med en hydroxygruppe i enhver tilgængelig stilling, og eventuelt tillige substitueret med en C1_3alkylgruppe (fx methyl) på det samme kul-20 stofatom som hydroxygruppen.

Forsøg med dyr har vist at forbindelser ifølge opfindelsen er lipolyseinhibitorer, dvs. at de nedsætter de plasmafrie fedtsyrekoncentrationer. Forbindelserne kan derfor bruges til 20 behandling af hyperlipidæmier. Forbindelserne kan desuden som følge af deres anti-lipolytiske virkning nedsætte forhøjede blodsukker- og ketonstofniveauer og kan derfor være værdifulde ved behandling af diabetes. Da anti-lipolytiske stoffer har hypolipidæmisk og hypofribrinogenæmisk virkning, kan 25 forbindelserne også vise anti-aterosklerotisk virkning.
Den anti-lipolytiske virkning af forbindelser ifølge opfindelsen er blevet påvist ved deres evne til at nedsætte koncentrationen af ikke-forestrede fedtsyrer (NEFA) i sultende rotter efter oral indgift.
30 En særlig vigtig gruppe af forbindelser er på grund af deres udtalte anti-lipolytiske virkning den gruppe forbindelser med formel I, hvor X er et hydrogenatom og R er en cyclopentyl- DK 170894 B1 5 ring substitueret med en hydroxygruppe og eventuelt tillige substitueret med en C^.galkylgruppe (fx methyl) .
Foruden deres anti-lipolytiske virkning, kan forbindelserne med formel I påvirke hjertefunktionen ved at nedsætte hjerte-5 frekvensen og hjerteledningen. Forbindelserne kan derfor bruges ved behandling af et antal hjerte-karsygdomme, fx hjerte-arrytmi, navnlig efter myokardie-infarkt og angina pectoris. Forbindelserne kan også inhibere frigivelse af renin og således være af nytte ved behandling af for højt blodtryk og 10 hjertesvigt. Forbindelserne kan også være nyttige til påvirkning af centralnervesystemet (CNS), fx som sovemidler, beroligende midler, smertestillende midler og/eller midler mod krampe.
Det har således vist sig, at forbindelserne ifølge opfindel-15 sen er mere selektive end kendte forbindelser såsom forbindelser kendt fra DE-A-20525296; den fra dette skrift kendte forbindelse Kal95, nemlig N-cyclopentyl-2-chloradenosin, gav følgende resultater ved sammenligning med forbindelsen fra eksempel 1 nedenfor, idet virkningerne overfor 20 adenosinreceptorundertyperne Alf A2a og A2b er udtrykt som ækvi-effektive koncentrationsforhold i forhold til standardforbindelsen N-ethyl-carboxamidoadenosin (NECA):

Det kan af disse resultater ses, at forbindelserne ifølge opfindelsen har en langt højere selektivitet end de kendte forbindelser.

Forbindelserne med formel I og fysiologisk acceptable salte eller solvater kan således bruges til behandling af mennesker eller dyr der lider af en tilstand ved hvilken der er en fordel i at nedsætte den plasmafrie fedtsyrekoncentration og/el-5 ler nedsætte hjerteraten og hjerteledningen.
Ved behandling af mennesker eller dyr der lider af en tilstand ved hvilket det er fordelagtigt at nedsætte den plasmafrie fedtsyrekoncentration og/eller reducere hjertefrekvensen og hjerteledningen, indgiver man en virksom mængde af en 10 forbindelse med formel I eller et fysiologisk acceptabelt salt eller solvat deraf til patienten.
Det vil forstås, at henvisning til behandling omfatter både profylakse og lindring af etablerede symptomer.
Opfindelsen angår også farmaceutiske præparater der som virk-15 somt stof indeholder mindst én forbindelse med den almene formel I eller et fysiologisk acceptabelt salt eller solvat deraf sammen med en farmaceutisk bærer og/eller excipient til brug i human- eller veterinærmedicinen.
Præparater ifølge opfindelsen kan formuleres til oral, buc-20 cal, parenteral eller rektal indgivelse eller i en form egnet til indgift ved inhalation eller insufflation. Oral indgivelse foretrækkes.
Tabletter og kapsler til oral indgift kan indeholde almindelige excipienter som fx bindemidler såsom stivelseslim eller 25 polyvinylpyrrolidon; fyldstoffer som fx laktose, mikrokry-stallinsk cellulose eller majsstivelse; smøremidler som fx magnesiumstearat eller stearinsyre; desintegreringsmidler som fx kartoffelstivelse, natriumkroskarmellose eller natrium-glykollatstivelse; eller befugtningsmidler som fx natrium-30 laurylsulfat. Tabletterne kan være coatede ved metoder der er velkendte for de sagkyndige. Orale flydende præparater kan være i form af fx vandige eller olieagtige suspensioner, opløsninger, emulsioner, sirupper eller eliksirer, eller kan DK 170894 B1 7 foreligge som et tørt produkt til rekonstituering i vand eller andre egnede opløsningsmidler inden brug. Sådanne flydende præparater kan indeholde almindelige additiver som fx suspensionsmidler, fx sorbitolsirup, methylcellulose eller 5 carboxymethylcellulose; emulgeringsmidler som fx sorbitan-monooleat; ikke-vandige opløsningsmidler (som fx kan være spiseolier) såsom propylenglykol eller ethylenalkohol; og konserveringsmidler som fx methyl- eller propyl-p-hydroxyben-zoater eller sorbinsyre. Præparaterne kan også indeholde 10 buffersalte, smagsstoffer, farvestoffer og sødemidler (fx mannitol) efter behov.
Til buccal indgift kan præparaterne have form af tabletter eller pastiller formuleret på almindelig vis.
Forbindelserne med formel I kan oparbejdes til parenteral 15 indgift ved bolus-injektion eller kontinuerlig infusion og kan foreligge i enhedsdosisform i ampuller eller i multi-dosisbeholdere med et konserveringsmiddel tilsat. Præparaterne kan have sådanne former som fx suspensioner, opløsninger eller emulsioner i olieagtige eller vandige bærevæsker og kan 20 indeholde hjælpestoffer som fx suspenderings-, stabiliserings- og/eller dispergeringsmidler. Det virksomme stof kan også være i pulverform til sammenblanding med en egnet bære-væsker som fx sterilt pyrogenfrit vand før brug.
Forbindelserne med formel I kan også oparbejdes som stikpil-25 ler, fx indeholdende almindelige suppositoriegrundlag som fx kakaosmør eller andre glycerider.
En hensigtsmæssig dosis af forbindelserne ifølge opfindelsen, til indgift i mennesker (der vejer ca. 70 kg), er 2 mg til 2 g, fortrinsvis 10 mg til 1 g, af det virksomme stof pr.
30 enhedsdosis, som kan indgives fx l til 4 gange dagligt. Det vil forstås at det kan være nødvendigt at variere dosis rutinemæssigt i afhængighed af patientens alder og tilstand. Doseringen vil også afhænge af indgivelsesvejen.

Forbindelser med formel IV og beskyttede derivater deraf er enten kendte forbindelser eller kan fremstilles ved sædvan-10 lige fremgangsmåder.
Ifølge en tredje generel fremgangsmåde (C), kan en forbindelse med formel I fremstilles ud fra andre forbindelser med formel I ved konventionelle fremgangsmåder.
Hydrogeneringen kan fx således bruges til at fremstille en 15 forbindelse med formel I, hvor R er en substitueret cycloal-kylgruppe, ud fra den tilsvarende forbindelse med formel I hvor R er en substitueret cycloalkenylgruppe.
Hydrogenering ifølge den generelle fremgangsmåde (C) kan udføres ved sædvanlige fremgangsmåder, fx ved hjælp af hydrogen 20 i nærværelse af en ædelmetalkatalysator såsom palladium, raney-nikkel, platin eller rhodium. Katalysatoren kan være båret på fx trækul eller aluminiumoxid, eller man kan bruge en homogen katalysator som fx tris(triphenylfosfin)-rhrodium-chlorid. Hydrogeneringen vil almindeligvis blive udført i et 25 opløsningsmiddel som fx en alkohol såsom metanol eller ethanol, en ether såsom tetrahydrofuran eller dioxan, en ester såsom ethylacetat eller vand, eller i en blanding af opløs- DK 170894 B1 11 ningsmidler, fx en blanding af to eller flere af de netop nævnte, ved en temperatur i området -20 til +100°C og ved et tryk på 1-10 atmosfærer.
Det vil forstås at det under de ovennævnte omdannelser kan 5 være nødvendigt eller ønskeligt at beskytte alle følsomme grupper i de pågældende forbindelser for at undgå uønskede sidereaktioner. Det kan fx være nødvendigt at beskytte hy-droxylgrupperne i en forbindelse med formel II, III eller IV eller nitrogenatomet i en forbindelse med formlen RNH2.
10 Eksempler på egnede hydroxylbeskyttende grupper er bl.a. acyl (fx kulbrintecarbonylgrupper såsom acetyl, benzoyl, pivaloyl og oktanoyl), alkyl (fx methyl, t-butyl og metoxymethyl), aralkyl (fx benzyl, diphenylmethyl, triphenylmethyl og p-me-toxyphenyldiphenylmethyl) og silyl (fx trialkylsilyl såsom t-15 butylmethylsilyl). Desuden kan to nabo-hydroxylgrupper være beskyttet med en alkylidengruppe (fx isopropyliden) eller med en disiloxanylgruppe (fx 1,1,3,3-tetraisopropyldisilox-1,3-dityl). Særligt hensigtsmæssigt beskyttede derivater af forbindelserne II, III og IV er isopropyliden-, triacetyl-, 20 tribenzoyl- eller tri-t-butyldimethylsilylderivater.
Eksempler på egnede N-beskyttende grupper til forbindelser med formlen RNH2 er bl.a. arylmethylgrupper (fx benzyl), acylgrupper (fx acetyl) og silylgrupper (fx trimethylsilyl).
Ifølge en fjerde generel fremgangsmåde (D), kan en forbindel-25 se med formel I fremstilles ved, at man fjerner alle beskyttelsesgrupper fra et beskyttet derivat af en forbindelse med formel I. Fjernelse af beskyttelsesgruppen kan udføres ved hjælp af konventionelle teknikker som de fx er beskrevet i "Protective Groups in Organic Synthesis" af T.W. Greene (John 30 Wiley and Sons, 1981).
OH-beskyttende acylgrupper kan således fx fjernes ved hjælp af metanol i nærværelse af en base som fx kaliumcarbonat, t-butylamin eller ammoniak. En isopropylidengruppe kan fjernes DK 170894 B1 12 ved syrekatalyseret hydrolyse, fx ved hjælp af trifluoreddi-kesyre eller svovlsyre. t-Butyldimethylsilylgrupper kan fjernes ved alkalisk hydrolyse, fx ved hjælp af natriumhydroxid i ethanol.
5 En N-benzylgruppe kan spaltes ved hydrogenolyse i nærværelse af en katalysator såsom palladium på trækul, fx som beskrevet under fremgangsmåde (C). En N-acylgruppe, fx en acetylgruppe eller en trimethylsilylgruppe kan fjernes under sure eller basiske forhold, fx ved hjælp af fortyndet saltsyre eller na-10 triumhydroxid. Specifikke diastereoisomerer af forbindelser med formel I kan vindes ved konventionelle metoder, fx ved syntese ud fra et passende asymmetrisk udgangsmateriale, idet der bruges en hvilken som helst af de foroven beskrevne fremgangsmåder; eller hvor det er hensigtsmæssigt ved separering 15 af en blanding af isomerer af forbindelse med formel I ved sædvanlige metoder, fx ved fraktioneret krystallisation eller kromatografering.
I de generelle fremgangsmåder beskrevet i det foregående kan forbindelser med formel I vindes i form af et salt, hensigts-20 mæssigt i form af et fysiologisk acceptabelt salt. Om ønsket, kan sådanne salte omdannes til de tilsvarende frie baser ved hjælp af sædvanlige metoder.
Fysiologisk acceptable salte af forbindelser med formel I kan fremstilles ved at man omsætter en forbindelse med formel I 25 med en tilsvarende syre eller base i nærværelse af et egnet opløsningsmiddel som fx acetonitril, acetone, chloroform, ethylacetat eller en alkohol (fx metanol, ethanol eller iso-propanol).
Fysiologisk acceptable salte kan også fremstilles ud fra 30 andre salte, heri medregnet andre fysiologisk acceptable salte af forbindelserne med formel I, idet der bruges sædvanlige metoder.

Opfindelsen belyses nærmere ved de følgende mellemprodukter og eksempler. Organiske ekstrakter tørredes, hvor det angives, over vandfrit natriumsulfat. Tyndlagskromatografering (TLC) udførtes på kiselsyreanhydrid. Kolonnekromatografering 5 udførtes på kiselsyreanhydrid (Merck 7734) med mindre andet er angivet, og det kiselsyreanhydrid til hvilket reaktionsblandingen adsorberedes, var også Merck 7734 med mindre andet er angivet. Flash- eller flammekromatografering (FCC) udførtes på kiselsyreanhydrid (Merck 9385). "Florisil" var 60-100 10 mesh (BDH-vare). Følgende forkortelser anvendes:
System A: dichlormetan/ethanol/0,88 åmmoniakopløsning,
System B: ethylacetat/metanol DEA: N,N-diisopropylethylamin THF: tetrahydrofuran 15 ^-NMR-spektra opnåedes ved 250 MHz fra fortyndede opløsninger i dimethylsulfoxid.
Mellemprodukt 1 3-Aza-2-oxabicyclo[2.2.1]-heptan-3-carboxylsyre-phenylmethyl-ester 20 5,0 g azodicarbonamid omrørtes med en opløsning af 7,0 g kaliumhydroxid i 12 ml vand ved 4°C. Efter omrøring på isbad i en time fortyndedes blandingen med 30 ml is/vand og opløsningen filtreredes. Filtratet fortyndedes med kold (2°C) 100 ml ethanol og det resulterende faste stof frafiltreredes, 25 vaskedes med ethanol, metanol og ether for at tilvejebringe 6,9 g kaliumazodicarboxylat. Dette stof blev derpå blandet med 0,82 g 3-aza-2-oxabicyclo[2.2.1]-hept-5-en-3-carboxyl-syre-phenylmethylester i 60 ml tørt pyridin og der tilsattes 2,02 g eddikesyre under omrøring ved stuetemperatur. En time 30 senere tilsattes der en yderligere portion iseddikesyre (2,02 g) og reaktionsblandingen omrørtes i 15,5 timer. Blandingen inddampedes til tørhed under nedsat tryk. Der tilsattes derefter mere eddikesyre for at fjerne den resterende gule farve af diimid-forstadiet. Remanensen fordeltes mellem DK 170894 B1 14 0,5M citronsyre (75 ml) og 75 ml ethylacetat og den organiske fase fraskiltes, tørredes og koncentreredes i vakuum. Remanensen rensedes ved FCC, idet der elueredes med ethylacetat/-cyclohexan 1:2 til frembringelse af 0,69 g af den i over-5 skriften angivne forbindelse som en olie.
TLC {cyclohexan/ethylacetat 2:1), Rf = 0,25.
Mellemprodukt 2 N-(cis-3-hydroxycyclopentyl)-karbamidsyre, phenylmethylester
En opløsning af 5,0 g 3-aza-2-oxabicyclo[2.2.1]-heptan-3-10 carboxylsyre-phenylmethylester i 0,5 ml iseddikesyre sattes til en omrørt suspension af 0,35 ml zinkpulver i en blanding af eddikesyre og vand (1/1; 4 ml) og blandingen omrørtes ved 60°C i 7,5 timer. Der tilsattes mere zinkpulver (0,14 g) og 1,0 ml eddikesyre og omrøringen fortsattes i endnu 16,5 ti-15 mer. Efter afkøling til stuetemperatur filtreredes blandingen og zinkoverskuddet vaskedes med 20 ml 2M saltsyre. Filtratet og vaskevæskerne forenedes og neutraliseredes med 8%s natri-umcarbonat og ekstraheredes med 3 x 30 ml ethylacetat. De forenede organiske ekstrakter vaskedes med 30 ml saltopløs-20 ning, tørredes og koncentreredes i vakuum. Rensning ved FCC, under eluering med ethylacetat/cyclohexan 1:1 gav en olie (120 mg), som størknede ved henstillen og derefter blev omkrystalliseret fra cyclohexan til frembringelse af 70 mg af den i overskriften angivne forbindelse med smp. 62-63°C.
25 Mellemprodukt 3
Trans-N-[3-(formyloxy)-cyclopentyl]-karbamidsyre-phenylmethylester 1,78 g diethylazodicarboxylat sattes dråbevis til en omrørt opløsning af 1,19 g N-(cis-3-hydroxycyclopentyl)-karbamid-30 syre-phenylmethylester, 2,68 g triphenylfosfin og 0,47 g myresyre i 65 ml THF under nitrogen ved stuetemperatur. Den DK 170894 B1 15 resulterende opløsning omrørtes i 2 timer og koncentreredes for at give en remanens, som omrørtes i 20 ml ether ved ca. -10°C under nitrogen i 1 time. Blandingen fortyndedes med 20 ml cyclohexan og det faste stof frafiltreredes og vaskedes 5 med ether/cyclohexan 1:1 (ca. 20 ml). Filtratet og vaskevæskerne forenedes og koncentreredes og den resulterende remanens rensedes ved FCC, under eluering med ethylacetat/cyclo-hexan 2:3 for at give 1,18 g af den i overskriften angivne forbindelse med smp. 45-48°C.

Trans-3 -aminocyclopentanol-hydrochlorid
En opløsning af 1,1 g trans-N-[3 -(formyloxy)-cyclopentyl]-karbamidsyre-phenylmethylester i 40 ml ethanol omrørtes med 0,25 g kaliumcarbonat ved stuetemperatur i 1 time. Blandingen 15 filtreredes derefter og koncentreredes i vakuum. Det resulterende halvfaste stof opløstes i 50 ml ethylacetat og filtreredes, og filtratet koncentreredes i vakuum og efterlod et fast stof, som blev hydrogeneret i 50 ml ethanol med 200 mg 5%s palladium på kulstof som katalysator ved et tryk på 1 20 atmosfære i 20 timer. Katalysatoren blev derpå udskiftet med frisk 200 mg 5%s palladium på kulstof og hydrogeneringen fortsattes i yderligere 20 timer. Reaktionsblandingen filtreredes og filtratet koncentreredes i vakuum. Remanensen rensedes ved FCC under eluering med system A (40:10:1) og gav en 25 olie (0,26 g) som fortyndedes med 40 ml ethanol, blev gjort sur med 3M saltsyre i ethanol og koncentreredes i vakuum til frembringelse af 0,34 g af den i overskriften angivne forbindelse .

Mellemprodukt 5 [la, 2a, 6/3] -2-methyl-6- [ (phenylmethyl) -amino] -cyclohexanol
En blanding af 5,0 g 2-methyl-7-oxabicyclo[4.1.0]-heptan, 5,15 g benzylamin og 0,8 ml vand opvarmedes under tilbagesva-5 ling i 7,5 timer under nitrogen. Rensning ved FCC under elue-ring med system B (40:1) gav 3,9 g af den i overskriften angivne forbindelse som et fast stof. TLC (system B, 20:1), RF = 0,33.
Mellemprodukt 6 10 [la, 2a, 6/3] -6-amino-2-methylcyclohexanol-hydrochlorid
En opløsning af 3,5 g [la,2a, 6/3]-2-methyl-6-[ (phenylmethyl) -amino]-cyclohexanol i absolut ethanol (75 ml) blev hydrogeneret over 10%s palladium på trækul (50%s vandig pasta (1,3 g) i absolut ethanol (25 ml). Blandingen filtreredes og 15 inddampedes til ca. 100 ml. Opløsningen blev gjort sur med ca. 1M ethanolisk saltsyre og opløsningsmidlet afdampedes i vakuum til dannelse af et fast stof som omkrystalliseredes to gange fra isopropanol/ethylacetat; den herved fremkomne, i overskriften angivne forbindelse, havde smp. 194-195°C.
20 Eksempel 1 N-[(IS,trans)-2-hydroxycyclopentyl]-adenosin
En blanding af 2,87 g 6-chlorpurin-/?-D-ribosid og 1,38 g (IS,trans)-2-aminocyclopentanol-hydrochlorid opvarmedes under tilbagesvaling i 100 ml isopropanol indeholdende 3,87 g DEA i 25 18 timer. En blanding af 6 g 6-chlorpurin-jS-D-ribosid, trans-3-amino-cyclopent-l-en-4-ol, 0,69 g 4-methylbenzensulfonat og 0,52 g DEA i 35 ml isopropanol opvarmedes til tilbagesvaling under omrøring natten over under nirogen. Den afkølede blanding 20 koncentreredes i vakuum og gav en olie (2,56 g), som rensedes ved kolonnekromatografering på kiselsyreanhydrid (Merck 9385, deaktiveret med 1% triethylamin) under eluering med system B 10:1; der vandtes et fast stof (0,522 g). Dette stof omkrystalliseredes fra 15 ml ethylacetat og gav 185 mg af den i 25 overskriften angivne forbindelse.

Herved vandtes der 6,5 g af den i overskriften angivne forbindelse med smp. 170-180°C. Dette salts kromatografiske opførsel lignende den der konstateredes for en autentisk prøve af den frie base.
Eksempel 21 15 N-[(IS,Trans)-2-hydroxycyclopentyl]-adenosin-(IS)-(+)-10-kam-fersulfonsyresalt
En blanding af 3,51 g N-[(IS,trans)-2-hydroxycyclopentyl]-adenosin og 2,44 g (IS)-(+)-10-kamfersulfonsyre i 35 ml isopropanol opvarmedes under tilbagesvaling under nitrogen ind-20 til der opnåedes en klar opløsning. Opløsningen fortyndedes med 50 ml isopropylacetat og blandingen afkøledes til ca.
25°C under omrøring. Det resulterende krystallinske faste stof isoleredes ved filtrering, vaskedes med isopropanol/isopropylacetat 1:2 (2x15 ml) og tørredes i vakuum ved 40°C til 25 frembringelse af 5,31 g af den i overskriften angivne forbindelse med smp. 150-152°C.

20 Den virksomme bestanddel opløses i vand til injektion. Natri-umchloridet opløses deri og den resulternede opløsning titre-res til pH 7,2 med natriumhydroxidopløsning hvorpå fosfat-bufferopløsningen tilsattes. Opløsningen suppleres med vand egnet til injektion til ønsket rumfang og steriliseres ved 25 membranfiltrering. Den fyldes aseptisk på beholdere egnet til inhalation ved forstøvning.
5 Magnesiumstearat 1 mg Mannitol til en tabletkerne-vægt på 65 mg Den virksomme bestanddel sigtes og blandes med mannitol og hydroxypropylmethylcellulose. Der tilsættes passende rumfang 10 renset vand til granulering. Efter tørring sigtes kornene, blandes med magnesiumstearat og presses med passende stempler på en automatisk tabletmaskine.

Fremgangsmåde til fremstilling af en forbindelse ifølge krav 1, kendetegnet ved, at a) en forbindelse med formlen II DK 170894 Bl L _N tt OH OH hvor L er en fraspaltelig gruppe, og X har den i krav 1 angivne betydning, eller et beskyttet derivat deraf omsættes med en forbindelse med formlen RNH2, hvor R har den i krav 1 angivne betydning, eller et salt eller et beskyttet derivat 5 deraf, under basiske betingelser, om nødvendigt efterfulgt af fjernelse af enhver tilstedeværende beskyttelsesgruppe, eller b) en forbindelse med formel III \Jk_N
12. Fremgangsmåde ifølge krav 11b, kendetegnet ved, at forbindelsen med formel III 5 eller et beskyttet derivat deraf er fremstillet ved omsætning af en forbindelse med den almene formel IV j.Hj "Xf H0 OH hvor X har den i krav 1 angivne betydning, eller et beskyttet derivat deraf, med en stærk base efterfulgt af omsætning med et alkyleringsmiddel til indførelse af den ønskede gruppe R.
