hver gang 2 af de 6 Cys-grupper i positionerne 5, 13, 15, 21, 27 og 38 er forbundet via disulfid-broer, samt dets fysiologisk acceptable salte, idet polypeptidet med formlen I, hvori Η-(X) -Y er H-Val-Val og n = 0, er undtaget, fremstilles ved, at man a) fremstiller det på i og for sig kendt måde ved hjælp af fastfasesyntese, eller b) til fremstilling af et polypeptid, hvori η = 0, I. underkaster hirudin en Edman-nedbrydning to gange, II. omsætter det således tilvejebragte peptid med en aktiv ester af aminosyre eller et peptid med formlen U-(X)m-Y-0H, hvori U er en urethanbeskyttelsesgruppe, III. fraspalter phenylthio-carbamoyl-gruppen ved e-aminofunktionen af Lys ved hjælp af hydrazin, IV. og urethanbeskyttelsesgruppen U ved hjælp af en syre eller en base, og eventuelt overfører det ved a) eller b) tilvejebragte polypeptid i dets fysiologisk acceptable salt. Polypeptidet kan også udvindes fra orme af Gnathobdellida-ordenen, fortrinsvis fra orme af Hirudo--slægten ved hjælp af en kombination af ekstraktionsmetoder, fældningsmetoder, membranfiltrering og/eller chroma-tografiske fremgangsmåder, og det tilvejebragte peptid kan eventuelt overføres i dets fysiologisk acceptable salte.
Det af blodigler udvundne polypeptid har aminosyresammen-sætningen 9 Asparaginsyre, 5 Threonin, 4 Serin, 13 Gluta-minsyre, 3 Prolin, 9 Glycin, 2 Valin, 6 Cystein, 2 Iso-leucin, 4 Leucin, 2 Tyrosin, 1 Phenylalanin, 3 Lysin og 1 Histidin, hvori OH-gruppen i en af tyrosingrupperne kan være forestret med svovlsyre.
Polypeptiderne er blodkoagulationsinhibitorer og kan anvendes som diagnostika og reagenser.
