Claims (5)

1. Cellekultur-beholder omfattende et hus, der definerer et kammer, der inkluderer en inverteret keglestubs-bund og et cylindrisk legeme, der strækker sig opad fra den inverterede keglestubs-bund, hvor kammeret har en vertikal akse, hvor den inverterede keglestubs-bund har en hældning, en øverste del og en modstående nederste del, hvilken nederste del har en større indre diameter end den øverste del, og hvor dimensionerne af den indre diameter af den øverste del, af den indre diameter af den nederste del, af den indre diameter af det cylindriske legeme, af højden af den inverterede keglestubs-bund og af højden af det cylindriske legeme er henholdsvis 40 mm, 250 mm, 250 mm, 90 mm og 110 mm.
2. System til dyrkning af celler, omfattende en beholder ifølge krav 1 og et orbital rystebord eller impellere.
3. Fremgangsmåde til at dyrke levende celler, omfattende trinnene: (a) at tilvejebringe en beholder ifølge krav 1; (b) at indføre et arbejdsvæske-dyrkningsmedium til beholderen; (c) at indføre til beholderen en podekultur indeholdende celler af interesse; og (d) at dyrke cellerne under en egnet betingelse, hvor forholdet mellem det 02-åndbare areal/kulturmediumvolumenet ikke er mindre end 0,14 cm2/cm3.
4. Fremgangsmåden ifølge krav 3, hvor forholdet mellem arbejdsvæske-dyrkningsmedium-volumen/podekultur-volumen ikke er større end 1/10.
5. Fremgangsmåden ifølge krav 3, hvor arbejdsvæske-dyrkningsmediet indeholder et serum-frit medium med C02-ikke-afhængigt 20 mM FIEPES eller ækvivalenter for at fastholde pH af kulturen stabil.
