Claims (11)
- SAMMENSÆTNINGER OG FREMGANGSMÅDER ANVENDELIGE TIL DYRKNING AF DIEFERENTIERBARE CELLER1. Anvendelse af en sammensætning til dyrkning og opretholdelse af pluripotente differeniierbare celler i en stabiliseret pluripotent tilstand, hvilken sammensætning omfatter en basal saitnæringsopløsning, en ErbB3-ligand, en aktivator af insulin-lignende vækstfaktor 1 receptor (IGF1R) og indeholder mindre end 3% serum, hvor anvendelsen sker i fravær af et næringslag, hvor (a) nævnte ErbS3-!igand er udvalgt fra gruppen bestående af Neuregulin-1, Heregulin-β (HRG-β), Heregulin-a (HRG-a), Neu-differentieringsfaktor (NDF), acetylcholinreeeptor-inducerende aktivitet (ARIA), glialvækstfaktor 2 (GGF2), motor-neuron-afiedt faktor (SMDF), Neuregulin-2, Neuregu!in-2(i (NRG2-p), Kpuegnlin, Biregulin og funktionelle fragmenter deraf, især foretrukket HRG-β eller et funktionelt fragment deraf; og (b) hvor aktivatoren af IGF1R omfatter en insulin-lignende vækstfaktor eller et funktionelt fragment deraf.
- 2. Anvendelse af en sammensætning ifølge krav 1, hvor sammensætningen er fri for eksogent insulin og insulin-substitutter.
- 3. Anvendelse af en sammensætning ifølge krav 1 eller 2, hvor sammensætningen endvidere omfatter transformeringsvækstfaktor beta (TGF-β), et TGFP-familiemedlem eller et funktionelt fragment deraf, hvor TGF-P-familiemedlemmet fortrinsvis er udvalgt fra gruppen bestående af Nodal, Activin A, Activin B, knoglemorfogent protein-2 (BMP2), knoglemorfogent protein-4 (BMP4) og funktionelle fragmenter deraf, især Activin A.
- 4. Anvendelse af en sammensætning ifølge et hvilket som helst af de foregående krav, hvor sammensætningen er fri for eksogen fibroblastvækstfaktor eller endvidere omfatter mindst en fibroblasivækstfalctor (FGF) udvalgt fra gruppen bestående af FGF-2, FGF-7, FGF-10, FGF-22 og funktionelle fragmenter deraf.
- 5. Anvendelse af en sammensætning ifølge et hvilket som helst af de foregående krav, hvor sammensætningen endvidere omfatter et serumalbumin (SA), fortrinsvis bovint SA (BSA) eller human SA (I-ISA), og hvor koncentrationen af SA fortrinsvis er mere end ca. 0,2%, volumen til volumen (vol/vol) og mere fortrinsvis mindre end ca. .5 volumenprocent.
- 6. Anvendelse, af en sammensætning ifølge et hvilket som helst af de foregående krav, hvor sammensætningen endvidere omfatter mindst ét uopløseligt substrat, fortrinsvis udvalgt fra gruppen bestående af et collagen, et fibroneetin og fragmenter deraf.
- 7. Anvendelse af en sammensætning ifølge et hvilket som helst af de foregående, krav, hvor den insulin-lignende vækstfaktor er insulin-lignende vækstfaktor 1 (IGF!) eller lougR3-IGFL
- 8. Anvendelse af en sammensætning ifølge et hvilket som helst af de foregående krav, hvor sammensætningen indeholder mindre end 2% serum.
- 9. Anvendelse af en sammensætning ifølge ei hvilket som helst af de foregående krav, hvor sammensætningen indeholder mindre end 1% serum.
- 10. Anvendelse af en sammensætning ifølge et hvilket som helst af de foregående krav, hvor sammensætningen ikke indeholder serum.
- 11. Anvendelse af en sammensætning ifølge et hvilket som helst af de foregående krav, hvor sammensætningen ikke indeholder serumerstatning.
