Abstract

Purified and isolated polypeptide (I) having at least part of the primary structural conformation and a biological property (ies) of naturally occurring pluripotent granulocyte colony stimulating factor (hpG-CSF) is new when the (I) is the prod. of procaryotic or eucaryotic expression of an exogenous DNA sequence. - DNA sequence for use in securing expression of (I) in a procaryotic or eucaryotic host cell is new. The DNA sequence is either a defined sequence of its complementary strands, or a sequence that hybridises to such sequences or their fragments, or a sequence that, but for the degeneracy of the genetic code, would hydridise to any of these other sequences.

PATENTKRAV 1. Fremgangsmåde til fremstilling af et humant pluripotent granulocyt kolonistimulerende faktor{hpG-CSF)produkt med in vivo granulocytopoietiske, biologiske egenskaber som naturligt forekommende hpG-CSF, kendetegnet ved, at fremgangsmåden omfatter følgende trin: (a) under hensigtsmæssige næringsbetingelser at dyrke pattedyrsceller omfattende andet promotor DNA end hpG-CSF promotor DNA og operativt forbundet med DNA kodende for et hpg-CSF polypeptid med den modne aminosyresekvens ifølge figur Vil; og (b) isolere det af cellerne eksprimerede hpg-CSF produkt . 2. Fremgangsmåde ifølge krav 3, kende tegnet ved, at primat-cellerne er COS celler.A process for the preparation of a human pluripotent granulocyte colony-stimulating factor (hpG-CSF) product with in vivo granulocytopoietic biological properties as naturally occurring hpG-CSF, characterized in that the method comprises the following steps: (a) mammalian cells comprising promoter DNA other than hpG-CSF promoter DNA and operably linked to DNA encoding a hpg-CSF polypeptide with the mature amino acid sequence of Figure Vil; and (b) isolating the hpg-CSF product expressed by the cells. The method of claim 1, characterized in that the promoter DNA is viral promoter DNA. The method of claim 1, characterized in that the cells are primate cells. The method of claim 3, characterized in that the primate cells are COS cells.
