Ikke-human værtsorganisme, der omfatter et polynukleotid, der koder for et polypeptid med en aminosyresekvens, der er mindst 90 % identisk med aminosyresekvensen ifølge SEQ ID NO: 2, hvor i aminosyresekvensen glycin i position 149 er erstattet af en proteinogenisk aminosyre, hvilket polypeptid har γ-glutamylkinaseaktivitet, hvor polynukleotidet overudtrykkes ved anvendelse af en stærk promotor, således at aktiviteten eller koncentrationen af det tilsvarende protein forøges med mindst 10 % i forhold til aktiviteten eller koncentrationen af proteinet i den oprindelige mikroorganisme.
2. Værtsorganisme ifølge krav 1, hvor aminosyreudskiftningen er udvalgt fra SEQ ID NO: 2, hvor glycin i position 149 er erstattet af en proteinogenisk aminosyre.
3. Værtsorganisme ifølge krav 1 eller 2, hvor glycinen i position 149 i SEQ ID NO: 2 er erstattet af L-asparaginsyre.
4. Værtsorganisme ifølge krav 3, hvor polynukleotidet omfatter nukleotidsekvensen ifølge SEQ ID NO: 3.
5. Ikke-human værtsorganisme, der omfatter et polynukleotid, der koder for et polypeptid, der fra position 145 til 154 har aminosyresekvensen fra position 145 til 154 i SEQ ID NO: 2, hvor polynukleotidet under stringente betingelser, der omfatter et vasketrin med 0,5 SSC ved 50-68 °C, hybridiserer til komplementet til SEQ ID NO: 3, hvilket polypeptid har γ-glutamylkinaseaktivitet.
6. Værtsorganisme ifølge krav 5, hvor det kodede polypeptid fra position 145 til 154 har aminosyresekvensen fra position 145 til 154 i SEQ ID NO: 4.
7. Værtsorganisme ifølge krav 5, hvor det kodede polypeptid fra position 110 til 189 har aminosyresekvensen fra position 110 til 189 i SEQ ID NO: 2.
8. Værtsorganisme ifølge krav 6, hvor det kodede polypeptid fra position 110 til 189 har aminosyresekvensen fra position 110 til 189 i SEQ ID NO: 4.
9. Værtsorganisme ifølge krav 1 til 8, hvor værtsorganismen er udvalgt fra gruppen, der omfatter gær, E. coli, B. subtilis, koryneforme bakterier.
10. Værtsorganisme ifølge krav 9, hvor værtsorganismen er en koryneform bakterie.
11. Værtsorganisme ifølge krav 10, hvor værtsorganismen er Corynebacterium glutamicum.
12. Fremgangsmåde til fremstilling af L-prolin, der omfatter a) fermentering af værtsorganismen ifølge et eller flere af kravene 1 til 12, b) berigelse af L-aminosyren i mediet eller i bakteriecellerne og c) isolering eller opsamling af aminosyren eventuelt med bestanddele fra fermenteringssuppen og/eller biomasse.
13. Fremgansgmåde ifølge krav 12, hvor værtsorganismer fermenteres, der omfatter et eller flere overudtrykte gener, der er udvalgt fra gruppen, der omfatter: a) gdh-genet, der koder for glutamatdehydrogenase, b) proA-genet, der koder for γ-glutamylphosphatreduktase, c) proC-genet, der koder for pyrolin-5-carboxylatreduktase og d) ocd-genet, der koder for ornithincyclodeaminase.
14. Fremgansgmåde ifølge krav 12, hvor en værtsorganisme fermenteres, der omfatter et eller flere svækkede gener, der er udvalgt fra gruppen, der omfatter: a) ilvA-genet, der koder for threonindeaminase, b) putA-genet, der koder for pralindehydrogenase/pyrolin-5-carboxylatdehydrogenase, c) sucA-genet, der koder for 2-ketoglutaratdehydrogenase, d) sucB-genet, der koder for dihydrolipoamidsuccinyltransferase, og e) argD-genet, der koder for acetylornithinaminotransferase.
