Claims (11)
1. Fremgangsmåde til fotosyntetisk produktion af butanol omfattende dyrkning af en transgen designeralge i et flydende medium, hvori algen er genetisk manipuleret til at udtrykke et sæt af enzymer, der virker på et mellemprodukt af Calvin-cyklussen og konvertere mellemproduktet til butanol; hvor sættet af enzymer består af phosphoglyceratmutase, enolase, pyruvatkinase, pyruvat-ferredoxin-oxidoreductase (eller pyruvat-NADP + oxidoreductase), thiolase, 3-hydroxybutyryl-CoA-dehydrogenase, crotonase, butyryl-CoA-dehydrogenase, butyraldehyd-dehydrogenase og butanol-dehydrogenase; hvor den nævnte butyraldehyd-dehydrogenase og butanol-dehydrogenase kan bruge NADPH, som leveres af den fotodrevne elektrontransportproces; hvori de nukleotider, der koder for nævnte sæt af enzymer er genetisk manipulerede således, at enzymerne, der er udtrykt, er indsat i chloroplasteme af den fotosyntetiske værtsorganisme, direkte af et stroma-signalpeptid; og udvinding af butanol fra det nævnte flydende medium.
2. Fremgangsmåden ifølge krav 1, hvor den nævnte alge er udvalgt fra gruppen bestående af grønalger, rødalger, brunalger, blågrønne alger (oxyphotobacteria herunder cyanobakterier og oxychlorobacteria), kiselalger, marine alger, ferskvandsalger, kold-tolerante algestammer, varmetolerante algestammer, antenne-pigment-deficiente mutanter, butanol-tolerante algestammer og kombinationer deraf.
3. Fremgangsmåden ifølge krav 2, hvor algen er Chlamydomonas reinhardtii.
4. Fremgangsmåden ifølge krav 2, hvor den nævnte blå-grønne alge (oxyphotobacteria herunder cyanobakterier og oxychlorobacteria) er valgt fra gruppen bestående af Thermosynechococcus elongatus BP-1, Nostoc sp. PCC 7120, Synechococcus elongatus PCC 6301, Syncechococcus sp. stamme PCC 7942, Syncechococcus sp. stamme PCC 7002, Syncechocystis sp. stamme PCC 6803, Prochlorococcus marinus MED4, Prochlorococcus marinus MIT 9313, Prochlorococcus marinus NATL1A, Prochlorococcus SS120, Spirulina platensis (Arthrospira platensis), Spirulina pacifica, Lyngbya Majuscule, Anabaena sp., Synechocystis sp., Synechococcus elongatus, Synechococcus (MC-A), Trichodesmium sp., Richelia intracellularis, Synechococcus WH7803, Synechococcus WH8102, Nostoc punctiforme, Syncechococcus sp. stamme PCC 7943, Synechocyitis PCC 6714 hycocyanin-deficient mutant PD-1, Cyanothece stamme 51142, Cyanothece sp. CCY0110, Oscillatoria Limosa, Lyngbya majuscula, Symploca muscorum, Gloeobacter violaceus, Prochloron didemni, Prochlorothrix hollandica, Synechococcus (MC-A), Trichodesmium sp., Richelia intracellularis, Prochlorococcus marinus, Prochlorococcus SS120, Synechococcus WH8102, Lyngbya majuscula, Symploca muscorum, Synechococcus bigranulatus, cryophilic Oscillatoria sp., Phormidium sp., Nostoc sp.-1, Calothrix parietina, termofil Synechococcus bigranulatus, Synechococcus lividus, termofil Mastigocladus laminosus, Chlorogloeopsis fritschii PCC 6912, Synechococcus Vulcanus, Synechococcus sp. stamme MA4, Synechococcus sp. stamme MA19, og Thermosynechococcus elongatus.
5. Fremgangsmåden ifølge krav 1, hvor ekspressionen af det nævnte enzym reguleres af en inducerbar promotor.
6. Fremgangsmåde ifølge ethvert af kravene 1 til 4, hvor algen er genetisk manipuleret til at også indeholde et DNA-konstrukt, der koder for mindst et enzym, der fremmer NADPH/NADH omdannelsen til forøget fotobiologisk produktion af butanol.
7. Fremgangsmåde ifølge ethvert af kravene 1 til 4, hvor algecellerne er genetisk manipulerede til også at inaktivere stivelse-syntese-aktivitet.
8. Fremgangsmåde ifølge ethvert af kravene 1 til 4, hvor algen er genetisk manipuleret til at også inducerbart at udtrykke et ekstra sæt af designer enzymer, der omfatter amylase, stivelsephosphorylase, hexokinase, phosphoglucomutase, glucosephosphatisomerase, phosphofructosekinase, aldolase, triosephosphatisomerase, glyceraldehyde-3-phosphat- dehydrogenase, phosphoglyceratkinase, phosphoglyceratmutase, enolase, pyruvatkinase, og kombinationer deraf, som fremmer nedbrydning af stivelse og glycolyse i stroma-regionen af chloroplasten.
9. Transgen alge, der er anvendelig til fotosyntetisk produktion af butanol, genetisk manipuleret til inducerbart at udtrykke phosphoglyceratmutase, enolase, pyruvatkinase, pyruvat-ferredoxin-oxidoreductase (eller pyruvat-NADP + oxidoreductase), thiolase, 3-hydroxybutyryl-CoA-dehydrogenase, crotonase, butyryl-CoA-dehydrogenase , butyraldehyd-dehydrogenase og butanol-dehydrogenase i chloroplasten; hvor nævnte butyraldehyd-dehydrogenase og butanol-dehydrogenase kan bruge NADPH, som leveres af den fotodrevne elektrontransportproces; og, som også inducerbart udtrykker glyceraldehyd-3-phosphat-dehydrogenase og phosphoglycerat, aldolase og triosephosphat-isomerase, phosphofructose kinase og amylase, stivelse-phosphorylase, hexokinase, phosphoglucomutase, glucosephosphat-isomerase i chloroplasten.
10. Transgen alge ifølge krav 9, der også inducerbart udtrykker mindst en af følgende i chloroplasten: NADPH-phosphatase; NAD-kinase; en interfererende RNA-molekyle, der inhiberer ekspressionen af glucose-phosphateisomerase (G-P-isomerase) eller phosphoglucomutase; eller sættet af enzymer der helt eller delvist fremmer nedbrydning af stivelse og glycolyse bestående af amylase, stivelsephosphorylase, hexokinase, phosphoglucomutase, glucosephosphat- isomerase, phosphofructosekinase, aldolase, triosephosphat-isomerase, glyceraldehyd-3-phosphat-dehydrogenase, phosphoglyceratkinase, phosphoglycerat mutase, enolase, pyruvatkinase, og kombinationer deraf.
11. Transgen alge ifølge ethvert af kravene 9 og 10, hvor algen er en blågrøn alge (cyanobakterien eller oxychlorobacterium).
