Claims (14)
1. Serumfrit, koncentreret, eukaryot enkeltdels-næringsmiddel, der omfatter små peptider, der omfatter cystein og tyrosin, idet hver af peptiderne omfatter seks eller færre aminosyrer, hvor det for hvert peptid gælder, at mindst én af aminosyreme er en cystein og/eller en tyrosin, og hvor det eukaryote koncentrerede næringsmiddel er hydrolysatfrit.
2. Serumfrit, koncentreret næringsmiddel ifølge krav 1, hvor det for hvert peptid gælder, at mindst én af de resterende aminosyrer er udvalgt fra gruppen, der består af alanin, serin, glycin, valin, prolin, asparaginsyre og glutaminsyre, fortrinsvis alanin eller glycin.
3. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1 eller 2, hvor det koncentrerede enkeltdels-næringsmiddel er ved et neutralt pH.
4. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1 til 3, som øger titeren af et rekombinant protein, der dannes i cellekultursystemet.
5. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1 til 4, der øger den levedygtige celledensitet i cellekultursystemet.
6. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1 til 5, hvor mindst ét af de små peptider er et dipeptid udvalgt fra gruppen, der består af X-tyrosin, X-cystein, tyrosin-X og cystein-X eller et salt deraf, og hvor X er udvalgt fra gruppen, der består af alanin, glycin, valin, prolin, asparaginsyre og glutaminsyre, fortrinsvis hvor X er alanin eller glycin.
7. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1 til 6, hvor den N-terminale aminosyre af de små peptider har en fri aminogruppe.
8. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1-7, hvor det koncentrerede næringsmiddel er en væskeformig opløsning eller et tørt pulver eller et granuleret tørt pulver.
9. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af ovennævnte krav, hvor de små peptider eller dipeptider er til stede ved en koncentration på 1 g/L til 16 g/L, fortrinsvis 2,5 g/L til 8,5 g/L.
10. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af ovennævnte krav, som er en væskeformig opløsning, der forbliver fri for præcipitat i over 12 måneder ved opbevaring ved 2-8 °C.
11. Serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af ovennævnte krav, hvor næringsmidlet også er proteinfrit.
12. Fremgangsmåde til fremstilling af et serumfrit, koncentreret næringsmiddel, der omfatter kombinering af små peptider, der omfatter cystein og tyrosin, ifølge et hvilket som helst af ovennævnte krav med et carbohydrat, fortrinsvis glucose, og mindst én aminosyre eller et salt deraf.
13. Kit til dyrkning af en celle in vitro, hvilket kit omfatter en eller flere beholdere, hvor en første beholder indeholder et serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1 til 11, og hvor det serumfrie, koncentrerede næringsmiddel understøtter væksten af cellen i kultur og/eller ekspressionen af rekombinant protein eller et virus og/eller en viruspartikel.
14. Anvendelse af et serumfrit, koncentreret næringsmiddel ifølge et hvilket som helst af kravene 1 til 11 eller kit ifølge krav 13 til dyrkning af celler og/eller til produktion af et virus eller et rekombinant protein, hvor cellerne er udvalgt blandt insektceller, nematodeceller, CHO-celler, COS-celler, VERO-celler, BHK-celler, AE-1-celler, SP2/0-celler, L5.1-celler, PerC6- og hybridomaceller.
