Claims (15)
1. Sammensætning, der omfatter rekombinante adenoviruspartikler, hvor det rekombinante adenovirus omfatter et transgen og er et rekombinant humant adenovirus af serotype 5, 11a, 26, 34, 35, 49 eller 50 eller et rekombinant simian- adenovirus, der er kendetegnet ved, at genomerne i mindst 99 % af adenoviruspartiklerne i sammensætningen har nukleotidsekvensen: CTATCTAT som de 5'-terminale nukleotider.

3. Sammensætning ifølge krav 2, hvor det rekombinante adenovirus er et rekombinant humant adenovirus af serotype 26 eller 35.
4. Sammensætning ifølge et hvilket som helst af ovennævnte krav, som er en farmaceutisk sammensætning.
5. Sammensætning ifølge et hvilket som helst af ovennævnte krav, hvor det rekombinante adenovirus mangler mindst en del af El-regionen.
6. Sammensætning ifølge et hvilket som helst af ovennævnte krav, der omfatter mindst 1 x 107, fortrinsvis mindst 1 x 108, fortrinsvis mindst 1 x 109, fortrinsvis mindst 1 x 1010 rekombinante adenoviruspartikler.
7. Fremgangsmåde til fremstilling af en batch af rekombinante adenoviruspartikler, hvor mindst 99 % af adenoviruspartiklerne i batchen har identiske nukleotidsekvenser i 5'-terminalerne af deres genomer, og hvor adenovirusset omfatter et transgen, hvilken fremgangsmåde omfatter: a) udførelse af et molekylært kloningstrin til udskiftning af de naturligt forekommende 5'-terminaler, som ikke er CTATCTAT, på et adenovirusgenom med ændrede 5'-terminaler, der omfatter nukleotidsekvensen: CTATCTAT som de terminale nukleotider, b) opformering i værtsceller af det rekombinante adenovirus med de ændrede 5'-terminaler og c) høst af det rekombinante adenovirus til opnåelse af en batch af rekombinante adenoviruspartikler, hvor mindst 99 % af adenoviruspartiklerne i batchen har nukleotidsekvensen: CTATCTAT som de 5'-terminale nukleotider af deres genomer.
8. Fremgangsmåde til fremstilling af en batch af rekombinante adenoviruspartikler, hvor mindst 99 % af adenoviruspartiklerne i batchen har identiske nukleotidsekvenser i 5'-terminalerne af deres genomer, og hvor adenovirusset omfatter et transgen, hvilken fremgangsmåde omfatter: a) udførelse af en plakoprensning af et adenovirus, hvor det rekombinante adenovirus er et rekombinant humant adenovirus af serotype 5, 11a, 26, 34, 35, 49 eller 50 eller et rekombinant simian-adenovirus, til isolering af et adenovirus eller rekombinant adenovirus fra en enkelt plak, hvor adenovirusset eller det rekombinante adenovirus har nukleotidsekvensen: CTATCTAT som de 5'-terminale nukleotider af dets genom, b) opformering i værtsceller af et rekombinant adenovirus, der er opnået fra den enkelte plak i trin a) , og c) høst af det rekombinante adenovirus til opnåelse af en batch af rekombinante adenoviruspartikler, hvor mindst 99 % of adenoviruspartiklerne i batchen har nukleotidsekvensen: CTATCTAT som de 5'-terminale nukleotider af deres genomer.
9. Fremgangsmåde ifølge krav 7 eller 8, hvor batchen omfatter mindst 1 x 107 rekombinante adenoviruspartikler.
10. Fremgangsmåde ifølge et hvilket som helst af kravene 7 til 9, hvor det rekombinante adenovirus er et rekombinant humant adenovirus af serotype 5, 26, 35, 49 eller 50.
11. Fremgangsmåde ifølge et hvilket som helst af kravene 7 til 10, hvor det rekombinante adenovirus er et rekombinant humant adenovirus af serotype 26 eller 35.
12. Fremgangsmåde ifølge et hvilket som helst af kravene 7 til 11, hvor det rekombinante adenovirus mangler mindst en del af El-regionen.
13. Fremgangsmåde ifølge et hvilket som helst af kravene 7 til 12, der yderligere omfatter oprensning af det rekombinante adenovirus.
14. Fremgangsmåde ifølge krav 13, der yderligere omfatter formulering af det rekombinante adenovirus til en farmaceutisk sammensætning.
