Claims (14)
- Indretning og fremgangsmåde til storvolumentransfektion1. Indretning (1) til påføring af et elektrisk felt på en suspension af celler, cellederivater, organeller, subcellulære partikler og/eller vesikler, hvilken indretning omfatter mindst ét kammer (6), der omfatter mindst to elektroder (4, 5), og mindst ét separeringselement (13), som kan bevæges inde i kammeret (6) mellem to terminalpunkter (14, 15), og som er et forseglingselement, der, hvis det er i en position mellem terminalpunkterne (14, 15), sikrer væsketæt og/eller gastæt separering af mindst ét første rum (26) i kammeret (6) fra mindst ét andet rum (27) i kammeret (6), hvor det første rum (26) er udformet til at rumme en alikvot af suspensionen og omfatter mindst én åbning (7, 8) til indføring eller udledning af suspensionen ind i eller ud af det første rum (26), kendetegnet ved, at separeringselementet (13) omfatter et fleksibelt og/eller elastisk materiale, og det andet rum (27) er udformet til at rumme en yderligere alikvot af suspensionen og omfatter mindst én åbning (10, 11) til indføring eller udledning af suspensionen ind i eller ud af det andet rum (27).
- 2. Indretning ifølge krav 1, hvor mindst én åbning (7, 8) er anbragt i den ene ende (9) af kammeret (6), og mindst én yderligere åbning (10, 11) er anbragt i den modsatte ende (12) af kammeret (6).
- 3. Indretning ifølge krav 1 eller 2, hvor separeringselementet (13) er koblet til mindst ét justeringselement (19), der er anbragt uden for kammeret (6).
- 4. Indretning ifølge et hvilket som helst af kravene 1 til 3, hvor separeringselementet (13) omfatter mindst to med afstand placerede dele (16, 17), hvor det indre rum (18) mellem de med afstand placerede dele (16, 17) i separeringselementet (13) omfatter et kompressibelt materiale.
- 5. Indretning ifølge et hvilket som helst af kravene 1 til 4, hvor kammeret (6) omfatter mindst to segmenter, hvor hvert segment omfatter mindst én elektrode (4, 5).
- 6. Indretning ifølge krav 5, hvor hvert segment er forsynet med mindst én første elektrode (4) og mindst én anden elektrode (5), hvor den anden elektrode (5) er en fælles elektrode for mindst to segmenter.
- 7. Indretning ifølge et hvilket som helst af kravene 1 til 6, hvor kammeret (6) omfatter tilsvarende fordybninger (3) i to komponenter, der er fastgjort til hinanden.
- 8. Indretning ifølge krav 7, hvor hver fordybning (3) er forsynet med mindst én elektrode (4, 5).
- 9. Indretning ifølge et hvilket som helst af kravene 1 til 8, hvor kammeret (6) endvidere omfatter mindst ét basiselement (2, 30), der mindst i det væsentlige er fremstillet af et isolerende materiale og indbefatter mindst én overflade, til hvilken elektroden (4, 5) er fastgjort, hvor overfladen omfatter mindst ét ledende område (32), som er udformet til at tilvejebringe en elektrisk forbindelse mellem elektroden (4, 5) og mindst ét elektrisk kontaktpunkt (33).
- 10. Indretning ifølge et hvilket som helst af kravene 3 til 9, hvor der er anbragt mindst én pakning (22) mellem justeringselementet (19) og kammeret (6).
- 11. Indretning ifølge et hvilket som helst af kravene 1 til 10, hvor kammeret (6) omfatter mindst ét forseglingsindlæg (24), der mindst delvis strækker sig langs én side af kammeret (6), hvor forseglingsindlægget (24) omfatter et kompressibelt materiale.
- 12. Fremgangsmåde til påføring af et elektrisk felt på en suspension af celler, cellederivater, organeller, subcellulære partikler og/eller vesikler, hvilken fremgangsmåde omfatter: a) indføring af en alikvot af suspensionen i mindst ét kammer i en indretning til påføring af et elektrisk felt på suspensionen, hvilket kammer omfatter mindst to elektroder, hvor et separeringselement, der er anbragt inde i kammeret, flyttes i en første retning; b) påføring af en spændingspuls på alikvoten via mindst to elektroder i kammeret; c) udledning af alikvoten fra kammeret og samtidig indføring afen yderligere alikvot af suspensionen i kammeret, hvor separeringselementet flyttes i en anden retning modsat den første retning, og hvor separeringselementet omfatter et fleksibelt og/eller elastisk materiale og er et forseglingselement, som sikrer væsketæt og/eller gastæt separering af alikvoterne fra hinanden; d) påføring af en spændingspuls på den yderligere alikvot via de mindst to elektroder i kammeret.
- 13. Fremgangsmåde ifølge krav 12, der endvidere omfatter: e) udledning af den yderligere alikvot fra kammeret og samtidig indføring af en yderligere alikvot af suspensionen i kammeret, hvor separeringselementet flyttes i den første retning modsat den anden retning, og hvor separeringselementet separerer alikvoterne fra hinanden; f) påføring af en spændingspuls på den yderligere alikvot via mindst to elektroder i kammeret; g) valgfri gentagelse af trinnene c) til f) for yderligere alikvoter af suspensionen, indtil hele suspensionen er processeret.
- 14. Fremgangsmåde ifølge krav 12 eller 13, hvor separeringselementet flyttes ved at rotere et justeringselement, der er operativt koblet med separeringselementet.
