Claims (14)

1. Rekombinant globinmolekyle, der binder carbonmonoxid med høj affinitet, til anvendelse i en fremgangsmåde til behandling af carboxyhæmoglobinæmi hos et individ, hvilken fremgangsmåde omfatter: udvælgelse af et individ med carboxyhæmoglobinæmi; og indgivelse til individet af en terapeutisk virksom mængde af det rekombinante globinmolekyle, der binder carbonmonoxid med høj affinitet, hvor det rekombinante globinmolekyle, omfatter: et humant rekombinant neuroglobin med en H64Q-, H64L-, H64A- eller H64W-mutation, hvor nummereringen svarer til SEQ ID NO: 1; eller et humant rekombinant cytoglobin med en H81Q-, H81A-, H81L-eller H81W-mutation, hvor nummereringen svarer til SEQ ID NO: 4, hvorved carboxyhæmoglobinæmi hos individet behandles.
2. In vitro fremgangsmåde til fjernelse af carbonmonoxid fra hæmoglobin i blod eller væv, omfattende at bringe blodet eller vævet i kontakt med et rekombinant globinmolekyle, der binder carbonmonoxid med høj affinitet, hvor det rekombinante globinmolekyle omfatter: et humant rekombinant neuroglobin med en H64Q-, H64L-, H64A- eller H64W- mutation, hvor nummereringen svarer til SEQ ID NO: 1; eller et humant rekombinant cytoglobin med en H81Q-, H81A-, H81L-eller H81W- mutation, hvor nummereringen svarer til SEQ ID NO: 4, hvorved der fjernes carbonmonoxid fra hæmoglobin i blodet eller vævet.
3. Molekyle til anvendelse ifølge krav 1 eller fremgangsmåde ifølge krav 2, hvor det humane rekombinante neuroglobin yderligere omfatter en C46G-mu-tation, en C55S-mutation, en C120S-mutation eller en hvilken som helst kombination deraf.
4. Molekyle til anvendelse eller fremgangsmåde ifølge krav 3, hvor det humane rekombinante neuroglobin omfatter aminosyresekvensen med SEQ ID NO: 3.
5. Molekyle til anvendelse ifølge krav 1 eller fremgangsmåde ifølge krav 2, hvor det humane rekombinante cytoglobin yderligere omfatter en C38S-muta-tion eller en C83S-mutation, eller både en C38S- og en C83S-mutation.
6. Molekyle til anvendelse eller fremgangsmåde ifølge krav 5, hvor det humane rekombinante cytoglobin omfatter aminosyresekvensen med SEQ ID NO: 6.
7. Molekyle til anvendelse ifølge et hvilket som helst af kravene 1 og 3-6, hvor individet har mindst 3 %, mindst 5 %, mindst 10 %, mindst 15 % eller mindst 20 % carboxyhæmoglobin i blodet.
8. Molekyle til anvendelse ifølge et hvilket som helst af kravene 1 og 3-7, hvor det rekombinante globinmolekyle i fremgangsmåden indgives ved intravenøs infusion.
9. Molekyle til anvendelse ifølge et hvilket som helst af kravene 1 og 3-8, hvor den terapeutisk virksomme mængde af det rekombinante globinmolekyle er ca. 50 til ca. 800 gram.
10. Humant rekombinant neuroglobin omfattende en mutation ved rest 64 og yderligere omfattende en C46G-mutation, en C55S-mutation og en C120S-mutation, hvor nummereringen svarer til SEQ ID NO: 1.
11. Humant rekombinant neuroglobin ifølge krav 10, hvor mutationen ved rest 64 er en H64Q-, H64L-, H64A eller H64W-mutation.
12. Humant rekombinant neuroglobin ifølge krav 11, omfattende aminosyrese-kvensen med SEQ ID NO: 3.
13. Humant rekombinant cytoglobin omfattende en H81A- eller H81L-muta-tion, og yderligere omfattende en C38S-mutation og en C83S-mutation, hvor nummereringen svarer til SEQ ID NO: 4.
14. Sammensætning omfattende det humane rekombinante neuroglobin eller cytoglobin ifølge et hvilket som helst af kravene 10-13 og en farmaceutisk acceptabel bærer.
